Decrease in plasma miR-27a and miR-221 after concussion in Australian football players
Introduction: Sports-related concussion (SRC) is a common form of brain injury that lacks reliable methods to guide clinical decisions. MicroRNAs (miRNAs) can influence biological processes involved in SRC, and measurement of miRNAs in biological fluids may provide objective diagnostic and return to play/recovery biomarkers. Therefore, this prospective study investigated the temporal profile of circulating miRNA levels in concussed male and female athletes. Methods: Pre-season baseline blood samples were collected from amateur Australian rules football players (82 males, 45 females). Of these, 20 males and 8 females sustained an SRC during the subsequent season and underwent blood sampling at 2-, 6- and 13-days post-injury. A miRNA discovery Open Array was conducted on plasma to assess the expression of 754 known/validated miRNAs. miRNA target identified were further investigated with quantitative real-time PCR (qRT-PCR) in a validation study. Data pertaining to SRC symptoms, demographics, sporting history, education history and concussion history were also collected. Results: Discovery analysis identified 18 candidate miRNA. The consequent validation study found that plasma miR-221-3p levels were decreased at 6d and 13d, and that miR-27a-3p levels were decreased at 6d, when compared to baseline. Moreover, miR-27a and miR-221-3p levels were inversely correlated with SRC symptom severity. Conclusion: Circulating levels of miR-27a-3p and miR-221-3p were decreased in the sub-acute stages after SRC, and were inversely correlated with SRC symptom severity. Although further studies are required, these analyses have identified miRNA biomarker candidates of SRC severity and recovery that may one day assist in its clinical management
Delivery of Dark Material to Vesta via Carbonaceous Chondritic Impacts
NASA's Dawn spacecraft observations of asteroid (4) Vesta reveal a surface
with the highest albedo and color variation of any asteroid we have observed so
far. Terrains rich in low albedo dark material (DM) have been identified using
Dawn Framing Camera (FC) 0.75 {\mu}m filter images in several geologic
settings: associated with impact craters (in the ejecta blanket material and/or
on the crater walls and rims); as flow-like deposits or rays commonly
associated with topographic highs; and as dark spots (likely secondary impacts)
nearby impact craters. This DM could be a relic of ancient volcanic activity or
exogenic in origin. We report that the majority of the spectra of DM are
similar to carbonaceous chondrite meteorites mixed with materials indigenous to
Vesta. Using high-resolution seven color images we compared DM color properties
(albedo, band depth) with laboratory measurements of possible analog materials.
Band depth and albedo of DM are identical to those of carbonaceous chondrite
xenolith-rich howardite Mt. Pratt (PRA) 04401. Laboratory mixtures of Murchison
CM2 carbonaceous chondrite and basaltic eucrite Millbillillie also show band
depth and albedo affinity to DM. Modeling of carbonaceous chondrite abundance
in DM (1-6 vol%) is consistent with howardite meteorites. We find no evidence
for large-scale volcanism (exposed dikes/pyroclastic falls) as the source of
DM. Our modeling efforts using impact crater scaling laws and numerical models
of ejecta reaccretion suggest the delivery and emplacement of this DM on Vesta
during the formation of the ~400 km Veneneia basin by a low-velocity (<2
km/sec) carbonaceous impactor. This discovery is important because it
strengthens the long-held idea that primitive bodies are the source of carbon

Dark Matter Constraints from a Joint Analysis of Dwarf Spheroidal Galaxy Observations with VERITAS
We present constraints on the annihilation cross section of WIMP dark matter
based on the joint statistical analysis of four dwarf galaxies with VERITAS.
These results are derived from an optimized photon weighting statistical
technique that improves on standard imaging atmospheric Cherenkov telescope
(IACT) analyses by utilizing the spectral and spatial properties of individual
photon events. We report on the results of 230 hours of observations of
five dwarf galaxies and the joint statistical analysis of four of the dwarf
galaxies. We find no evidence of gamma-ray emission from any individual dwarf
nor in the joint analysis. The derived upper limit on the dark matter
annihilation cross section from the joint analysis is at 1 TeV for the bottom quark () final state,
at 1 TeV for the tau lepton

‘Like Skydiving without a Parachute’: How Class Origin Shapes Occupational Trajectories in British Acting
There is currently widespread concern that access to, and success within, the British acting profession is increasingly dominated by those from privileged class origins. This article seeks to empirically interrogate this claim using data on actors from the Great British Class Survey (N = 404) and 47 qualitative interviews. First, survey data demonstrate that actors from working-class origins are significantly underrepresented within the profession. Second, they indicate that even when those from working-class origins do enter the profession they do not have access to the same economic, cultural and social capital as those from privileged backgrounds. Third, and most significantly, qualitative interviews reveal how these capitals shape the way actors can respond to shared occupational challenges. In particular we demonstrate the profound occupational advantages afforded to actors who can draw upon familial economic resources, legitimate embodied markers of class origin (such as Received Pronunciation) and a favourable typecasting
Evidence for seasonal cycles in deep-sea fish abundances: A great migration in the deep SE Atlantic?
Animal migrations are of global ecological significance, providing mechanisms for the transport of nutrients and energy between distant locations. In much of the deep sea (>200 m water depth), the export of nutrients from the surface ocean provides a crucial but seasonally variable energy source to seafloor ecosystems. Seasonal faunal migrations have been hypothesized to occur on the deep seafloor as a result, but have not been documented.
Here, we analyse a 7.5‐year record of photographic data from the Deep‐ocean Environmental Long‐term Observatory Systems seafloor observatories to determine whether there was evidence of seasonal (intra‐annual) migratory behaviours in a deep‐sea fish assemblage on the West African margin and, if so, identify potential cues for the behaviour.
Our findings demonstrate a correlation between intra‐annual changes in demersal fish abundance at 1,400 m depth and satellite‐derived estimates of primary production off the coast of Angola. Highest fish abundances were observed in late November with a smaller peak in June, occurring approximately 4 months after corresponding peaks in primary production.
Observed changes in fish abundance occurred too rapidly to be explained by recruitment or mortality, and must therefore have a behavioural driver. Given the recurrent patterns observed, and the established importance of bottom‐up trophic structuring in deep‐sea ecosystems, we hypothesize that a large fraction of the fish assemblage may conduct seasonal migrations in this region, and propose seasonal variability in surface ocean primary production as a plausible cause. Such trophic control could lead to changes in the abundance of fishes across the seafloor by affecting secondary production of prey species and/or carrion availability for example.
In summary, we present the first evidence for seasonally recurring patterns in deep‐sea demersal fish abundances over a 7‐year period, and demonstrate a previously unobserved level of dynamism in the deep sea, potentially mirroring the great migrations so well characterized in terrestrial systems
Resistance to First-Line Anti-TB Drugs Is Associated with Reduced Nitric Oxide Susceptibility in Mycobacterium tuberculosis
Background and objective: The relative contribution of nitric oxide (NO) to the killing of Mycobacterium tuberculosis in human tuberculosis (TB) is controversial, although this has been firmly established in rodents. Studies have demonstrated that clinical strains of M. tuberculosis differ in susceptibility to NO, but how this correlates to drug resistance and clinical outcome is not known. Methods: In this study, 50 sputum smear- and culture-positive patients with pulmonary TB in Gondar, Ethiopia were included. Clinical parameters were recorded and drug susceptibility profile and spoligotyping patterns were investigated. NO susceptibility was studied by exposing the strains to the NO donor DETA/NO. Results: Clinical isolates of M. tuberculosis showed a dose- and time-dependent response when exposed to NO. The most frequent spoligotypes found were CAS1-Delhi and T3_ETH in a total of nine known spoligotypes and four orphan patterns. There was a significant association between reduced susceptibility to NO (>10% survival after exposure to 1mM DETA/NO) and resistance against first-line anti-TB drugs, in particular isoniazid (INH). Patients infected with strains of M. tuberculosis with reduced susceptibility to NO showed no difference in cure rate or other clinical parameters, but a tendency towards lower rate of weight gain after two months of treatment. Conclusion: There is a correlation between resistance to first-line anti-TB drugs and reduced NO susceptibility in clinical strains of M. tuberculosis. Further studies including the mechanisms of reduced NO susceptibility are warranted and could identify targets for new therapeutic interventions
Evolution of late-stage metastatic melanoma is dominated by aneuploidy and whole genome doubling
Although melanoma is initiated by acquisition of point mutations and limited focal copy number alterations in melanocytes-of-origin, the nature of genetic changes that characterise lethal metastatic disease is poorly understood. Here, we analyze the evolution of human melanoma progressing from early to late disease in 13 patients by sampling their tumours at multiple sites and times. Whole exome and genome sequencing data from 88 tumour samples reveals only limited gain of point mutations generally, with net mutational loss in some metastases. In contrast, melanoma evolution is dominated by whole genome doubling and large-scale aneuploidy, in which widespread loss of heterozygosity sculpts the burden of point mutations, neoantigens and structural variants even in treatment-naïve and primary cutaneous melanomas in some patients. These results imply that dysregulation of genomic integrity is a key driver of selective clonal advantage during melanoma progression
