    A Comparison of Road Structures of Six EU Countries

    The European Union has expanded significantly in recent years. Sustainable trade within the Union, leading to economic growth to the benefit of the ‘old’ and ‘new’ member states is thus extremely important. The road infrastructure is strategic and vital to such development since an uneven transport infrastructure, in terms of capacity and condition, has the potential to reinforce uneven development trends and hinder economic convergence of old and new member states. In the decades since their design and construction, loading conditions have significantly changed for many major highway infrastructure elements/networks owing primarily to increased freight volumes and vehicle sizes. This, coupled with the gradual deterioration of a significant number of highway structures due to their age, and the absence of a pan-European assessment framework, can be expected to affect the smooth functioning of the infrastructure in its as-built condition. Increased periods of reduced flow can be expected owing to planned and unplanned interventions for repair/rehabilitation. This paper reports the findings of a survey regarding the current status of the highway infrastructure elements in six countries within the European Union as reported by the owners/operators. The countries surveyed include a cross-section of ‘existing’ older countries and ‘new’ member states. The current situations for bridges, culverts, tunnels and retaining walls are reported, along with their potential replacement costs. The findings act as a departure point for further studies in support of a centralised and/or synchronised EU approach to infrastructure maintenance management. Information in the form presented in this paper is central to any future decision-making frameworks in terms of trade route choice and operations, monetary investment, optimised maintenance, management and rehabilitation of the built infrastructure and the economic integration of the newly joined member states

    Experimental Demonstration of a Mode Shape-Based Scour-Monitoring Method for Multispan Bridges with Shallow Foundations

    A vibration-based scour monitoring approach applicable to bridges with multiple simply supported spans on shallow foundations is experimentally investigated in this paper. A monitoring strategy based on the relative changes in pier mode shape amplitudes due to scour is postulated. The first global mode shape of a bridge structure with multiple spans is extracted from acceleration measurements using an output-only approach, Frequency Domain Decomposition (FDD). The relative changes of the pier mode shape amplitudes under scour are then tracked. Here, each pier mode shape value is compared with the mean values of the remaining piers in a process that creates a Mean-Normalised Mode Shape (MNMS). The approach is demonstrated on a scaled model of a bridge with four spans, supported on sprung foundations, where scour is simulated by the replacement of springs with springs of lower stiffness corresponding to a reduction in foundation stiffness. It is shown that at a given ‘scoured’ pier, significant increases in the MNMS value occur, suggesting that the location of the scour can be identified. The magnitude of the MNMS at a given pier also increases with an increase in stiffness loss due to scour. In practice, the approach would work best by carrying out a visual inspection of the bridge to establish the initial health condition at the time of sensor installation. After this initial process, the bridge can be monitored remotely for scour on an ongoing basis

    Proceedings of the CSHM-6: Structural Health Monitoring of New and Ageing Infrastructure, Queen's University Belfast, Northern Ireland, United Kingdom, 26-27 May 2016This paper showcases the importance of field testing in efforts to deal with the deteriorating infrastructure. It demonstrates a load test performed on a healthy but aging composite reinforced concrete bridges in Exeter, UK. The bridge girders were instrumented with strain transducers and static strains were recorded while a four-axle, 32 tonne lorry remained stationary in a single lane. The results obtained from the field test were used to calculate transverse load distribution factors (DFs) of the deck structure for each loading case. Additionally, a 3-D finite element model of the bridge was developed and calibrated based on field test data. Similar loading cases were simulated on the analytical model and behaviour of the structure under static loading was studied. It was concluded that the bridge support conditions had changed throughout its service life, which affected the superstructure load distribution characteristics. Finally, DFs obtained from analysis were compared with factors provided in Design Manual for Roads and Bridges Standard Specification for similar type of bridges.European Union’s Horizon 2020Marie SkƂodowska–Curie gran

    Mycobacterium ulcerans (M. ulcerans) is a necrotizing skin infection endemic to the Bellarine Peninsula, Australia. Current treatment recommendations include 8 weeks of combination antibiotics, with adjuvant surgery if necessary. However, antibiotic toxicity often results in early treatment cessation and local experience suggests that shorter antibiotic courses may be effective with concurrent surgery. We report the outcomes of patients in the Barwon Health M. ulcerans cohort who received shorter courses of antibiotic therapy than 8 weeks

    Dynamics of a ferromagnetic domain wall and the Barkhausen effect

    We derive an equation of motion for the the dynamics of a ferromagnetic domain wall driven by an external magnetic field through a disordered medium and we study the associated depinning transition. The long-range dipolar interactions set the upper critical dimension to be dc=3d_c=3, so we suggest that mean-field exponents describe the Barkhausen effect for three-dimensional soft ferromagnetic materials. We analyze the scaling of the Barkhausen jumps as a function of the field driving rate and the intensity of the demagnetizing field, and find results in quantitative agreement with experiments on crystalline and amorphous soft ferromagnetic alloys.Comment: 4 RevTex pages, 3 ps figures embedde

    Mycobacterium ulcerans disease: experience with primary oral medical therapy in an Australian cohort

    Mycobacterium ulcerans (MU) is responsible for disfiguring skin infections which are challenging to treat. The recommended treatment for MU has continued to evolve from surgery to remove all involved tissue, to the use of effective combination oral antibiotics with surgery as required. Our study describes the oral medical treatment utilised for consecutive cases of MU infection over a 15 month period at our institution, in Victoria, Australia. Managing patients primarily with oral antibiotics results in high cure rates and excellent cosmetic outcomes. The success with medical treatment reported in this study will aid those treating cases of MU infection, and will add to the growing body of knowledge about the relative roles of antibiotics and surgery for treating this infection

    Successful Outcomes with Oral Fluoroquinolones Combined with Rifampicin in the Treatment of Mycobacterium ulcerans: An Observational Cohort Study

    Buruli ulcer is a necrotizing infection of skin and subcutaneous tissue caused by Mycobacterium ulcerans and is the third most common mycobacterial disease worldwide (after tuberculosis and leprosy). In recent years its treatment has radically changed, evolving from a predominantly surgically to a predominantly medically treated disease. The World Health Organization now recommends combined streptomycin and rifampicin antibiotic treatment as first-line therapy for Mycobacterium ulcerans infections. However, alternatives are needed where recommended antibiotics are not tolerated or accepted by patients, contraindicated, or not accessible nor affordable. This study describes the use of antibiotics, including oral fluoroquinolones, in the treatment of Mycobacterium ulcerans in south-eastern Australia. It demonstrates that antibiotics combined with surgery are highly effective in the treatment of Mycobacterium ulcerans. In addition, oral fluoroquinolone-containing antibiotic combinations are shown to be as effective and well tolerated as other recommended antibiotic combinations. Fluoroquinolone antibiotics therefore offer the potential to provide an alternative oral antibiotic to be combined with rifampicin for Mycobacterium ulcerans treatment, allowing more accessible and acceptable, less toxic, and less expensive treatment regimens to be available, especially in resource-limited settings where the disease burden is greatest

    Targeting tumour re-wiring by triple blockade of mTORC1, epidermal growth factor, and oestrogen receptor signalling pathways in endocrine-resistant breast cancer

    Background Endocrine therapies are the mainstay of treatment for oestrogen receptor (ER)-positive (ER+) breast cancer (BC). However, resistance remains problematic largely due to enhanced cross-talk between ER and growth factor pathways, circumventing the need for steroid hormones. Previously, we reported the anti-proliferative effect of everolimus (RAD001-mTORC1 inhibitor) with endocrine therapy in resistance models; however, potential routes of escape from treatment via ERBB2/3 signalling were observed. We hypothesised that combined targeting of three cellular nodes (ER, ERBB, and mTORC1) may provide enhanced long-term clinical utility. Methods A panel of ER+ BC cell lines adapted to long-term oestrogen deprivation (LTED) and expressing ESR1wt or ESR1Y537S, modelling acquired resistance to an aromatase-inhibitor (AI), were treated in vitro with a combination of RAD001 and neratinib (pan-ERBB inhibitor) in the presence or absence of oestradiol (E2), tamoxifen (4-OHT), or fulvestrant (ICI182780). End points included proliferation, cell signalling, cell cycle, and effect on ER-mediated transactivation. An in-vivo model of AI resistance was treated with monotherapies and combinations to assess the efficacy in delaying tumour progression. RNA-seq analysis was performed to identify changes in global gene expression as a result of the indicated therapies. Results Here, we show RAD001 and neratinib (pan-ERBB inhibitor) caused a concentration-dependent decrease in proliferation, irrespective of the ESR1 mutation status. The combination of either agent with endocrine therapy further reduced proliferation but the maximum effect was observed with a triple combination of RAD001, neratinib, and endocrine therapy. In the absence of oestrogen, RAD001 caused a reduction in ER-mediated transcription in the majority of the cell lines, which associated with a decrease in recruitment of ER to an oestrogen-response element on the TFF1 promoter. Contrastingly, neratinib increased both ER-mediated transactivation and ER recruitment, an effect reduced by the addition of RAD001. In-vivo analysis of an LTED model showed the triple combination of RAD001, neratinib, and fulvestrant was most effective at reducing tumour volume. Gene set enrichment analysis revealed that the addition of neratinib negated the epidermal growth factor (EGF)/EGF receptor feedback loops associated with RAD001. Conclusions Our data support the combination of therapies targeting ERBB2/3 and mTORC1 signalling, together with fulvestrant, in patients who relapse on endocrine therapy and retain a functional ER
