[email protected] Abstract BACKGROUND: Gastric banding is thought to decrease appetite in addition to the mechanical effects of food restriction, although this has been difficult to demonstrate in human studies. Our aim was to investigate the changes in orexigenic signals in the obese Zucker rat after gastric banding. METHODS: Obese Zucker rats (fa/fa) were submitted to gastric banding (GBP), sham gastric banding fed ad libitum (sham), or sham operation with food restriction, pair-fed to the gastric banding group (sham-PF). Lean Zucker rats (fa/+) were used as additional controls. Body weight and food intake were daily recorded for 21 days after surgery when epididymal fat was weighed and fasting ghrelin and hypothalamic NPY mRNA expression were measured. RESULTS: Gastric banding in obese Zucker rats resulted in a significant decrease of cumulative body weight gain and food intake. Furthermore, gastric banded rats were leaner than Sham-PF, as expressed by a significantly lower epididymal fat weight. Ghrelin levels of gastric banded rats were not increased when compared to sham-operated animals fed ad libitum and were significantly lower than the levels of weight matched sham-PF rats (1116.9 +/- 103.3 g GBP vs 963.2 +/- 54.3 g sham, 3,079.5 +/- 221.6 sham-PF and 2,969.9 +/- 150.9 g lean rats, p < 0.001); hypothalamic NPY mRNA expression was not increased in GBP when compared to sham-operated rats. CONCLUSION: In obese Zucker rats, GBP prevents the increase in orexigenic signals that occur during caloric deprivation. Our data support the hypothesis that sustained weight loss observed after gastric banding does not depend solely on food restriction

    Microalgae biomass interaction in byopolymer gelled systems

    Microalgae are an enormous biological resource, representing one of the most promising sources for the development of new food products and applications. Pea protein/k-carrageenan/starch gels, interesting vegetarian alternatives to dairy desserts, served as model systems to study the addition of microalgal biomass, its effect, and subsequent rheological behaviour. Spirulina and Haematococcus gels presented a markedly different rheological behaviour compared to the control mixed biopolymer gelled system. The present goal is to clarify how these microalgae affect the gelation and interact with each biopolymer present in the complex mixed gel system. Hence, the aim of the present work is to study the effect of Spirulina and Haematococcus microalgal biomass addition on the rheological behaviour of pea protein, k- carrageenan and starch simple gels, as well as in pea protein/k-carrageenan and pea protein/starch systems. The gelation process was monitored in-situ through dynamic oscillatory measurements (temperature, time and frequency sweep tests) for a 24 h maturation period, and rheological results were supported with uorescence optical microscopy observations. The addition of Spirulina and Haematococcus to biopolymer gelled systems induced signi cant changes in the gels’ rheological behaviour and microstructure. In general, it was observed that the gelling mechanism is ruled by the biopolymers, while microalgae seem to be embedded in the gel network acting as active particle llers. The addition of Haematococcus resulted in more structured gels in comparison to the control and Spirulina systems. In the case of k-carrageenan gels, both microalgae induced a large increase in the rheological parameters, which should be related to the high ionic content of microalgal biomass. Spirulina addition on starch systems promoted a decrease in the gels’ rheological parameters. This should be related to the starch gelatinization process, probably by competing for water binding zones during the granules’ hydration proces

    Potentialities of the extraction technologies and use of bioactive compounds from winery by-products: a review from a circular bioeconomy perspective

    Agro-industrial by-products and by-products from the wine industry (pomace, peels, leaves, stems, and seeds) represent a potential economic interest because they are usually relevant natural sources of bioactive compounds, which may present significant biological activities related to human health and well-being. This article aims to review wine and winery industry by-products as potential natural sources of antioxidant, antimicrobial, anti-inflammatory, antiaging, and anticancer compounds, as well as briefly highlighting the extraction methods used to obtain these bioactive compounds and explore their potential applications in the food, cosmetic, and packaging industries. Although there are some studies of wine industry by-products with different origins, this revision will be mainly focused on the Portuguese vineyard industry since it represents an import industrial sector as proof of the diversity of the bioactive compounds identified.     Efficient Recursion Method for Inverting Overlap Matrix

    A new O(N) algorithm based on a recursion method, in which the computational effort is proportional to the number of atoms N, is presented for calculating the inverse of an overlap matrix which is needed in electronic structure calculations with the the non-orthogonal localized basis set. This efficient inverting method can be incorporated in several O(N) methods for diagonalization of a generalized secular equation. By studying convergence properties of the 1-norm of an error matrix for diamond and fcc Al, this method is compared to three other O(N) methods (the divide method, Taylor expansion method, and Hotelling's method) with regard to computational accuracy and efficiency within the density functional theory. The test calculations show that the new method is about one-hundred times faster than the divide method in computational time to achieve the same convergence for both diamond and fcc Al, while the Taylor expansion method and Hotelling's method suffer from numerical instabilities in most cases.Comment: 17 pages and 4 figure

    No âmbito de um projecto de investigação tendo como propósito o desenvolvimento de sistemas sustentáveis de produção de biomassa lenhosa para energia, foi instalado em Bragança, em 2008, um ensaio de produção intensiva de biomassa lenhosa com base em culturas de curta rotação. Este ensaio tem como objectivos específicos a avaliação dos efeitos de factores como material vegetal, densidade de plantação, rega e fertilização no crescimento e produção de biomassa lenhosa. Nesta comunicação descrevemos a instalação do ensaio relativamente ao desenho experimental seguido, estrutura interna das parcelas de estudo, material vegetal utilizado, operações de preparação do solo e de plantação e ainda gestão da cultura com respeito a fertilização, rega e protecção. Avaliamos também a sobrevivência das plantas no final da primeira estação de crescimento, bem como apresentamos os custos gerais associados à instalação. A apresentação deste trabalho permite partilhar com a comunidade científica e técnica nacional a experiência da instalação deste tipo de culturas, muito recentes no nosso país, e dessa forma contribuir para a definição de processos ajustados e eficientes de instalação e gestão de culturas lenhosas para produção de biomassa para energia em Portugal

    Apoio à decisão para a proteção integrada no combate a Tuta absoluta em tomate hortoindustrial

    A traça-do-tomateiro é atualmente uma praga-chave do tomate para indústria, na região do Ribatejo. A sua presença foi detectada em 2010, tendo causado prejuízos significativos na campanha de 2011. O objetivo deste projeto é criar ferramentas para a tomada de decisão no âmbito da proteção desta cultura contra esta praga. Durante o ano de 2013, procedeu-se à monitorização semanal de adultos de T. absoluta com recurso a armadilhas com feromona sexual, e à observação visual em 25 a 50 plantas, de acordo com o estado fenológico da cultura. O material entomológico foram analisadas, posteriormente, em laboratório para deteção de parasitismo. Além de avaliar a intensidade de ataque, identificaram-se fatores de risco prévio, para construção de mapas de risco de apoio à tomada de decisão para a limitação da praga como culturas vizinhas e respetivas datas de colheita. Os dados preliminares não apresentaram uma correlação entre o número de machos adultos capturados e o número de larvas presentes na cultura, pelo que a estimativa de risco baseada apenas na contagem de machos adultos não parece ser suficiente para a tomada de decisão. Verificaram-se dificuldades no combate à praga devido a razões de ordem estrutural (e.g. posse da terra, hospedeiros alternativos na vizinhança) e a lacunas no conhecimento da sua bioecologia.The South American tomato moth is presently an important pest on processing tomato crop in the Ribatejo region. This species was detected in 2010 and caused significant losses in 2011. The goal of this project is to develop decision-making tools to improve crop protection. During 2013, the adults were monitored weekly by sexual pheromone traps and the pest incidence was evaluated by sampling 25 to 50 plants, according to the crop age. Entomological samples were analysed in the laboratory to detect parasitism. In addition to the monitoring in the field, risk factors were also identified in order to define risk maps for decision-making support as neighbouring crops and harvest dates. The preliminary data did not reveal a correlation between the number of adult males present in the traps and the number of larvae present on the crop, so the risk assessment based only on adult monitoring is unsatisfactory for decision-making. At this stage, some difficulties were found related to structural aspects (e.g. land ownership, alternative hosts in the neighbourhood) and to a significant knowledge gap on the pest bioecology.PRODERinfo:eu-repo/semantics/publishedVersio

    Sustainable woody biomass production systems: a novel solution for energy, agriculture and the environment in Portugal

    Bioenergy is a major current issue within the agriculture and energy sectors and the society in general. The environmental and social concerns raised by first generation biofuel crops increased the interest on woody biomass. Biomass production for direct conversion to heat and/or electricity is common in northern European countries, the USA, and, more recently, southern Europe, mainly based on short rotation woody crops (SRWC). These crops are renewable sources of energy, CO2 neutral, able to preserve biodiversity and water, and to contribute to socio-economic development. In Portugal, there is an increasing demand of renewable and sustainable sources of energy. Portugal also presents conditions to expand biomass production and use for energy, namely through SRWCs. There is, however, a lack of experience in this field. With the purpose of studying short rotation woody crops dedicated to energy production, we have launched in Bragança, Portugal, a research project aiming to develop technology for the sustainable production, processing, and use of woody biomass for energy. The goal is to analyse environmental and socio-economic aspects of these crops considering carbon cycle, conservation of water, soils, and biodiversity, as well as creation of wealth locally. It also aims to evaluate the potential of northeastern Portugal for biomass production and carbon sequestration. In this presentation we address the rational behind the project, the methodologies followed, and the results obtained during the first year of the study.     Basis Functions for Linear-Scaling First-Principles Calculations

    In the framework of a recently reported linear-scaling method for density-functional-pseudopotential calculations, we investigate the use of localized basis functions for such work. We propose a basis set in which each local orbital is represented in terms of an array of `blip functions'' on the points of a grid. We analyze the relation between blip-function basis sets and the plane-wave basis used in standard pseudopotential methods, derive criteria for the approximate equivalence of the two, and describe practical tests of these criteria. Techniques are presented for using blip-function basis sets in linear-scaling calculations, and numerical tests of these techniques are reported for Si crystal using both local and non-local pseudopotentials. We find rapid convergence of the total energy to the values given by standard plane-wave calculations as the radius of the linear-scaling localized orbitals is increased.Comment: revtex file, with two encapsulated postscript figures, uses epsf.sty, submitted to Phys.
