Experimental Design Approach in Erythropoietin-alginate Microsphere Preparation with Different Concentrations of Drug and Polymer
Background: Microspheres as drug delivery system has been selected to increase stability of Erythropoietin (EPO) to achieve efficacy. Aim: Aim of this research was to determine effect of polymer and EPO concentrations on the characteristics. Materials and Method: Microspheres involved sodium alginate as polymer and CaCl as a crosslinker. The concentrations of sodium alginate used were 2% and 3%, and EPO were 5000 IU and 10000 IU. Formula of microspheres which consist of 2% and 3% of alginate and 5000 IU EPO were called F1 and F2 respectively, whereas microspheres using 2 and 3% alginate and 10000 IU EPO was named F3 and F4 respectively. Characterization including morphology, particle size, swelling index, and yield of microspheres prepared by ionotropic gelation aerosolization technique. Design of Experiment (DoE) was used to analyze the formula. Results: Results showed that particle sizes of EPO‑alginate microspheres were 3.36 ± 0.126µm, 3.42 ± 0.098µm, 3.88 ± 0.131µm and 3.95 ± 0.151µm for F1, F2, F3 and F4 respectively. The swelling index measurement based on mass and particle size of microspheres of all formulas showed an index of less than 10. Respectively, yield was 77.84 ±0.290%, 86.65 ± 0.191%, 91.89 ± 0.210%, and 94.65 ± 0.252% for F1 to F4. Using the ANOVA factorial design, it was found that increasing sodium alginate concentration significantly increased yield, while increasing EPO concentration significantly increased particle size and yield of microspheres. Both sodium alginate and EPO concentrations did not affect swelling index of microspheres. Range concentrations of sodium alginate and EPO that produced optimal characteristics of microspheres can be observed in the feasible area of design space overlaid contour plot generated from DoE study. Conclusion: EPO-alginate microspheres demonstrated the prospective as carrier and DoE is potential for further optimized formulations
Stability and Release Kinetics of an Advanced Gliclazide-Cholic Acid Formulation: The Use of Artificial-Cell Microencapsulation in Slow Release Targeted Oral Delivery of Antidiabetics
Introduction: In previous studies carried out in our laboratory, a bile acid (BA) formulation exerted a hypoglycaemic effect in a rat model of type-1 diabetes (T1D). When the antidiabetic drug gliclazide (G) was added to the bile acid, it augmented the hypoglycaemic effect. In a recent study, we designed a new formulation of gliclazide-cholic acid (G-CA), with good structural properties, excipient compatibility and exhibits pseudoplastic-thixotropic characteristics. The aim of this study is to test the slow release and pH-controlled properties of this new formulation. The aim is also to examine the effect of CA on G release kinetics at various pH values and different temperatures. Method: Microencapsulation was carried out using our Buchi-based microencapsulating system developed in our laboratory. Using sodium alginate (SA) polymer, both formulations were prepared: G-SA (control) and G-CA-SA (test) at a constant ratio (1:3:30), respectively. Microcapsules were examined for efficiency, size, release kinetics, stability and swelling studies at pH 1.5, pH 3, pH 7.4 and pH 7.8 and temperatures of 20 and 30 °C. Results: The new formulation is further optimised by the addition of CA. CA reduced microcapsule swelling of the microcapsules at pH 7.8 and pH 3 at 30 °C and pH 3 at 20 °C, and, even though microcapsule size remains similar after CA addition, percent G release was enhanced at high pH values (pH 7.4 and pH 7.8, p < 0.01). Conclusion: The new formulation exhibits colon-targeted delivery and the addition of CA prolonged G release suggesting its suitability for the sustained and targeted delivery of G and CA to the lower intestine
Incorporation of active agents into biocompatible polymers and study of their release
The present thesis explores the possibility of incorporating active agents intobiocompatible, biodegradable or not polymers that are usually found in medicalapplications. Those altered materials could be used for the synthesis of surgicalsutures or other medical implants that would release in a controlled ratepharmaceutical substances directly where they are required (like the area of a wound)but also in extremely small amounts, thus avoiding potential toxicity problems. Threecases of drug incorporation were examined: a) directly into inert, non-degradablepolymeric matrixes, b) directly into biodegradable matrixes and c) indirectly intobiocompatible nanospheres that could either be applied on the surface or into the bulkof other medical materials. In all above cases the material’s microstructure and thenature of the encapsulated drug were thoroughly studied; the drug release controlmodes and rates and therefore the dosage that reaches the patient were also explored.Polypropylene was studied as a non-degradable material. Attempts werecarried out to incorporate two non-steroidal anti-inflammatory drugs with drasticallydifferent melting points, one below and the other above the melting temperature of thepolymer; that is, Ibuprofen and Diclofenac-sodium, respectively. The mixing of thepolymer-active agent was attempted by various methods, such as solubilization,melting and combination of the two, also compared for their efficiency. Moreover, themolecular orientation achieved through uniaxial stretching of the samples wasevaluated as a parameter of tuning the drug release rate.As the biodegradable matrix, MaxonTM (a polymer found in commercialresorbable sutures) was selected and loaded with the antitumor drug Mitoxantrone-2HCl. In the case of MaxonTM, the influence of the percentage of crystallinity on therates of polymer degradation and active agent release was additionally studied.In the third drug loading case attempted, nanospheres were synthesizedout of the biopolymer chitosan via the emulsion crosslinking technique during whichthe antimicrobial drug Rifampicin was encapsulated. Moreover, the capability ofmixing the nanoparticles into a polymer matrix and having two active agents to bereleased at the same time (one directly from the matrix and one from thenanoparticles) with individual rates for each, was explored.The first part of the current thesis (chapters 1-5) constitutes an extensive reportof the technology of controlled release, the nanoparticulate systems (and moreparticularly those based on chitosan), the surgical implants/sutures and theircategories and materials used for their fabrication nowadays. Additionally, the basictheory behind the experimental techniques as well as the instrumentation used duringthis study are analyzed. Finally, the second part (chapters 6-8) records all experimentsconducted along with their results and analysis.Η παρούσα εργασία διερευνά την πιθανότητα ενσωμάτωσης φαρμακευτικώνουσιών σε βιοσυμβατά, αποικοδομήσιμα και μη πολυμερή που συναντώνται συχνά σειατρικές εφαρμογές. Τα σύνθετα πλέον αυτά υλικά θα μπορούσαν ναχρησιμοποιηθούν ως η βάση για βιοδραστικά χειρουργικά ράμματα ή άλλου είδουςιατρικά εμφυτεύματα, τα οποία θα αποδεσμεύουν με ελεγχόμενο ρυθμό δραστικέςουσίες απευθείας σε σημεία όπου αυτές απαιτούνται (όπως η περιοχή ενόςτραύματος) αλλά και στις απαραίτητες, συχνά εξαιρετικά μικρές, ποσότητεςαποφεύγοντας έτσι προβλήματα ενδεχόμενης τοξικότητας. Συνολικά εξετάστηκαντρεις περιπτώσεις ενσωμάτωσης των φαρμακευτικών ουσιών: α) απευθείας σεαδρανείς, μή αποικοδομήσιμες πολυμερικές μήτρες, β) απευθείας σεβιοαπορροφήσιμες μήτρες και γ) έμμεσα σε βιοσυμβατά νανοσφαιρίδια τα οποίαμπορούν είτε να εφαρμοστούν στην επιφάνεια είτε να ενθυλακωθούν στην κυρίωςμάζα άλλων ιατρικών υλικών. Σε όλες τις παραπάνω περιπτώσεις, μελετήθηκανδιεξοδικά η μικροδομή των υλικών, η φύση του εγκλωβισμένου φαρμάκου, ενώδιερευνήθηκαν και τρόποι ελέγχου του ρυθμού αποδέσμευσης των ενεργών ουσιώνκαι συνεπώς της δοσολογίας που κάθε στιγμή θα φτάνει στον ασθενή.Στην περίπτωση μή αποικοδομήσιμου υλικού μελετήθηκε το πολυπροπυλένιοστο οποίο πραγματοποιήθηκαν προσπάθειες ενσωμάτωσης δύο μή στεροειδώναντιφλεγμονωδών ουσιών του Ibuprofen και του Diclofenac sodium, ουσίες μεδιαφορετικά σημεία τήξεως, χαμηλότερα και υψηλότερα αντίστοιχα από αυτό τουπολυμερούς. Η ανάμειξη πολυμερούς-φαρμάκων επιχειρήθηκε μέσω διαφόρωνμεθόδων, όπως διαλυτοποίησης, τήξης και συνδυασμό των δύο, οι οποίες καισυγκρίθηκαν για την αποτελεσματικότητά τους. Επιπλέον, ο μοριακόςπροσανατολισμός που επιτεύχθηκε μέσω μονοαξονικού εφελκυσμού των δοκιμίωναξιολογήθηκε ως παράγοντας ελέγχου του ρυθμού αποδέσμευσης του φαρμάκου.Ως βιοαποικοδομήσιμη πολυμερική μήτρα επιλέχθηκε το υλικό MaxonTM(συναντάται σε εμπορικά απορροφήσιμα ράμματα) στο οποίο φορτώθηκε τοαντιογκικό φάρμακο mitoxantrone-2HCl. Στην περίπτωση του MaxonTM μελετήθηκεεπίσης η επίδραση του ποσοστού της κρυσταλλικότητας της μήτρας (μεταβαλλόμενομέσω θερμικής επεξεργασίας) στους ρυθμούς αποδέσμευσης αλλά καιαποικοδόμησης του υλικού.Στην τρίτη και τελευταία λύση φόρτωσης που επιχειρήθηκε, νανοσφαιρίδιααπό το βιοπολυμερές χιτοζάνη συντέθηκαν μέσω της τεχνικής διασύνδεσης σεγαλάκτωμα κατά την οποία εγκλωβίστηκε σε αυτά η αντιμικροβιακή ουσίαrifampicin. Σε αυτή την περίπτωση εξετάστηκε επίσης η δυνατότητα εγκλωβισμού ήεπιφανειακή εφαρμογή των ήδη φορτωμένων με μια ενεργή ουσία νανοσωματιδίωνσε μια πολυμερική μήτρα στην οποία επίσης ενσωματώνεται μια δεύτερηφαρμακευτική ουσία με ρυθμό αποδέσμευσης διαφορετικό της πρώτης. Θα έχουμελοιπόν δύο φαρμακευτικές ουσίες ταυτόχρονα, μία απευθείας στην πολυμερική μήτρακαι μία δεύτερη στα νανοσφαιρίδια οι οποίες θα έχουν τη δυνατότητα αποδέσμευσηςμε διαφορετικούς ρυθμούς προσαρμοσμένους στην εκάστοτε εφαρμογή.Στο πρώτο μέρος της παρούσας διατριβής (Κεφάλαια 1-5) γίνεται μια εκτενήςαναφορά στην τεχνολογία ελεγχόμενης αποδέσμευσης, τα συστήματανανοσωματιδίων και συγκεκριμένα αυτών που απαρτίζονται από χιτοζάνη και σταχειρουργικά εμφυτεύματα/ράμματα, τις κατηγορίες και τα υλικά αυτών πουχρησιμοποιούνται σήμερα. Επιπλέον, αναλύεται η βασική θεωρία των πειραματικώντεχνικών που χρησιμοποιήθηκαν και η οργανολογία τους. Τέλος, στο δεύτερο μέρος(Κεφάλαια 6-8) περιγράφονται οι πειραματικές προσπάθειες και παραθέτονται τααποτελέσματα αυτών και η ανάλυσή τους

In vitro release of bovine serum albumin from alginate/HPMC hydrogel beads
In recent years, the use of swelling polymeric matrices for the encapsulation and controlled release of protein drugs has received significant attention. The purpose of the present study was to investigate the release of albumin, a model protein from alginate/hydroxypropyl-methylcellulose (HPMC) gel beads. A hydrogel system comprised of two natural, hydrophilic polymers; sodium alginate and HPMC was studied as a carrier of bovine serum albumin (BSA) which was used as a model protein. The morphology, bead size and the swelling ratio were studied in different physical states; fully swollen, dried and reswollen using scanning electron microscopy and image analysis. Finally the effect of different alginate/HPMC ratios on the BSA release profile in physiological saline solution was investigated. Swelling experiments revealed that the bead diameter increases with the viscosity of the alginate solution while the addition of HPMC resulted in a significant increase of the swelling ratio. The BSA release patterns showed that the addition of HPMC increased the protein-release rate while the release mechanism fitted the Peppas model. Alginate/HPMC beads prepared using the ionic gelation exhibited high BSA loading efficiency for all formulations. The presence of HPMC increased the swelling ability of the alginate beads while the particle size remained unaffected. Incorporation of HPMC in the alginate gels also resulted in improved BSA release in physiological saline solution. All formulations presented a non-Fickian release mechanism described by the Peppas model. In addition, the implementation of non-parametric tests showed significant differences in the release patterns between the alginate/HPMC and the pure alginate beads, respectively
