    Resource availability and territorial behavior in Ruby-throated Hummingbirds (Archilochus colubris) at UMBS

    General EcologyThe goal of this study was to determine if Ruby-throated Hummingbirds (Archilochus colubris) exhibited territorial behaviors in relation to varied degrees of nectar availability. In order to examine this territorial behavior, we worked with students in the ornithology class to set-up 6 feeding sites at the edge of a field and woods at the University of Michigan Biological Station (UMBS) in Pellston, Michigan. Feeders at the sites were used to mimic low, intermediate, and high resource availability. Our group predicted that males would exhibit more territorial behavior than females at the sites with higher resource abundance. We also expected the most interactions to occur among males. We observed and recorded the territorial behavior of chasing at the feeding sites 3 times a day for hour-long periods for 4 days. Our results showed that the highest number of chasing behavior occurred at the single-feeder stations amongst females. We observed relatively low numbers of chasing between males. Furthermore, we observed that males preferred visiting sites with more feeders whereas females did not exhibit a clear preference of feeder abundance.     La qualité de vie au travail des éducatrices de la petite enfance

    Cet article traite de la qualité de vie au travail des éducatrices de la petite enfance. Il présente les résultats d’une enquête réalisée au Québec auprès de 519 éducatrices travaillant en centre de la petite enfance. Au total, cinq sources de stress (Boyle, Borg, Falzon, & Baglioni, 1995), cinq sources de bien-être psychologique (Dagenais- Desmarais, 2010) et une mesure d’autonomie (Gillet, Rosnet, & Vallerand, 2008) au travail sont considérées. Dans l’ensemble, les résultats révèlent un niveau de stress plutôt bas et des niveaux de bien-être et d’autonomie plutôt élevés. Analysés par tranche d’expérience, les résultats montrent que les cinq premières années au travail sont généralement suivies d’une augmentation du stress et d’une diminution du bien-être psychologique. Ils montrent aussi que l’autonomie est fortement associée au bien-être psychologique. Sur le plan pratique, ces résultats invitent les gestionnaires et les formateurs d’éducatrices de la petite enfance à porter une attention particulière aux premières années de vie professionnelle ainsi qu’à la préparation et à la mise en place de conditions favorables à l’exercice de l’autonomie au travail.

    The CIP2A–TOPBP1 axis safeguards chromosome stability and is a synthetic lethal target for BRCA-mutated cancer

    BRCA1/2-mutated cancer cells adapt to the genome instability caused by their deficiency in homologous recombination (HR). Identification of these adaptive mechanisms may provide therapeutic strategies to target tumors caused by the loss of these genes. In the present study, we report genome-scale CRISPR-Cas9 synthetic lethality screens in isogenic pairs of BRCA1- and BRCA2-deficient cells and identify CIP2A as an essential gene in BRCA1- and BRCA2-mutated cells. CIP2A is cytoplasmic in interphase but, in mitosis, accumulates at DNA lesions as part of a complex with TOPBP1, a multifunctional genome stability factor. Unlike PARP inhibition, CIP2A deficiency does not cause accumulation of replication-associated DNA lesions that require HR for their repair. In BRCA-deficient cells, the CIP2A-TOPBP1 complex prevents lethal mis-segregation of acentric chromosomes that arises from impaired DNA synthesis. Finally, physical disruption of the CIP2A-TOPBP1 complex is highly deleterious in BRCA-deficient tumors, indicating that CIP2A represents an attractive synthetic lethal therapeutic target for BRCA1- and BRCA2-mutated cancers

    Presence of task-1 channel in the laryngeal mucosa in the newborn lamb

    Nearly 40 potassium channels have been described in respiratory epithelial cells. Of these are found several members of the 4-transmembrane domain, 2-pore K(+) channel family (K2P family), namely Twik-1 and -2, Trek-1 and -2, Task-2, -3, and -4, Thik-1, and KCNK7. The aim of this study was to verify whether the Twik-related acid-sensitive K(+) channel, subtype 1 (Task-1) (also known as KCNK3), is present in the laryngeal mucosa in the newborn lamb. Through the use of immunohistochemistry and nested polymerase chain reaction (PCR) amplification, results indicate that Task-1 protein and mRNA are present in the laryngeal mucosa, in both the ciliated, pseudostratified columnar (respiratory) epithelium and the nonkeratinized, stratified squamous epithelium. The complete ovine Task-1 protein sequence showed high homology levels with previously reported mouse, bovine, and human Task-1 sequences. This includes a complete homology for the C-terminal amino acid sequence, which is mandatory for protein trafficking to the cell membrane. These results represent the first demonstration that Task-1, a pH-sensitive channel responsible for setting membrane potential, is present in the laryngeal mucosa of a newborn mammal

    Vascular and blood-brain barrier-related changes underlie stress responses and resilience in female mice and depression in human tissue

    Prevalence, symptoms, and treatment of depression suggest that major depressive disorders (MDD) present sex differences. Social stress-induced neurovascular pathology is associated with depressive symptoms in male mice; however, this association is unclear in females. Here, we report that chronic social and subchronic variable stress promotes blood-brain barrier (BBB) alterations in mood-related brain regions of female mice. Targeted disruption of the BBB in the female prefrontal cortex (PFC) induces anxiety- and depression-like behaviours. By comparing the endothelium cell-specific transcriptomic profiling of the mouse male and female PFC, we identify several pathways and genes involved in maladaptive stress responses and resilience to stress. Furthermore, we confirm that the BBB in the PFC of stressed female mice is leaky. Then, we identify circulating vascular biomarkers of chronic stress, such as soluble E-selectin. Similar changes in circulating soluble E-selectin, BBB gene expression and morphology can be found in blood serum and postmortem brain samples from women diagnosed with MDD. Altogether, we propose that BBB dysfunction plays an important role in modulating stress responses in female mice and possibly MDD
