    Perencanaan struktur dalam dunia teknik sipil harus selalu memperhatikan tiga aspek penting dalam penerapannya, yaitu fungsional, ekonomis, serta aman. Ketiga aspek ini menjadi sangat penting karena masing – masing memiliki keterkaitan antara satu dengan yang lain. Suatu struktur yang fungsional dan aman namun tidak mengindahkan aspek ekonomis maka akan sangat sulit diwujudkan, begitu pula apabila struktur tersebut fungsional dan ekonomis tanpa mengindahkan aspek keamanan maka struktur tersebut akan meningkatkan resiko dalam mewujudkannya. Dengan dilandasi hal tersebut maka perencanaan struktur atas gedung kampus universitas pelita harapan Surabaya ini mencoba menerapkan ketiga aspek tersebut dengan menggunakan Tata Cara Perhitungan Struktur Beton Untuk Gedung (SNI 03 – 2847 – 2002) dan Tata Cara Perencanaan Ketahanan Gempa Untuk Bangunan Gedung (SNI 03 – 1726 – 2002) yang diakui di negara Indonesia sebagai acuan dalam perencanaan struktur. Struktur yang direncanakan adalah struktur gedung sebagai fasilitas perkuliahan 5 lantai. Struktur berada di Surabaya yang terletak pada wilayah gempa 2. Gedung fasilitas perkuliahan ini akan direncanakan dengan menggunakan sistem struktur rangka pemikul momen berupa Sistem Rangka Pemikul Momen Menengah (SRPMM) dengan daktilitas parsial. Analisis struktur dilakukan dengan memodelkan struktur dalam bentuk tiga dimensi memanfaatkan program bantu ETABS. Hasil output program ini akan digunakan untuk memperoleh gaya gaya dalam yang bekerja pada struktur yang kemudian akan digunakan dalam perencanaan lentur, geser maupun torsi. Perencanaan struktur ini akan mencakup struktur atas gedung yang berupa perencanaan pelat atap maupun pelat lantai, balok, kolom, serta tangga. Beban yang dianalisis berupa kombinasi akibat beban mati, beban hidup, beban gempa dan beban hujan. Perencanaan memperoleh hasil dimensi balok yang digunakan berukuran 400 × 700 mm2 untuk balok induk dan 300 × 500 mm2 untuk balok anak, kolom berukuran 850 × 850 mm2, pelat dengan ketebalan 100 mm untuk pelat atap dan ketebalan 120 mm untuk pelat lantai. Hasil perencanaan menunjukkan bahwa struktur memenuhi persyaratan Tata Cara Perhitungan Struktur Beton Untuk Gedung (SNI 03 – 2847 – 2002) untuk tinjauan terhadap lentur, geser maupun torsi. Kinerja struktur atas gedung juga memenuhi persyaratan kinerja batas layan dan kinerja batas ultimit seperti yang disyaratkan Tata Cara Perencanaan Ketahanan Gempa Untuk Bangunan Gedung SNI (03-1726-2002) sehingga gedung aman untuk digunakan

    Dengan semakin banyaknya USAha makanan berdiri dan hampir semuanya dipenuhi pengunjung, menyebabkan semakin banyaknya pengusaha yang mulai menekuni bisnis makanan mulai dari tempat makan sederhana hingga mewah. Tujuan dari penelitian ini adalah untuk mengetahui pengaruh bauran pemasaran (produk, harga, tempat dan proses) terhadap minat beli ulang konsumen pada Warung Bu Darmi Siwalankerto Surabaya. Penelitian ini menggunakan metode kuantitatif eksplanatif. Berdasarkan hasil penelitian, diperoleh pengaruh yang signifikan pada bauran pemasaran produk, harga, tempat dan proses terhadap minat beli ulang konsumen. Bauran pemasaran produk berpengaruh dominan terhadap minat beli ulang konsumen pada Warung Bu Darmi Siwalankerto Surabaya

    Redox changes during the cell cycle in the embryonic root meristem of Arabidopsis thaliana

    Aims: The aim of this study was to characterize redox changes in the nuclei and cytosol occurring during the mitotic cell cycle in the embryonic roots of germinating Arabidopsis seedlings, and to determine how redox cycling was modified in mutants with a decreased capacity for ascorbate synthesis. Results: Using an in vivo reduction-oxidation (redox) reporter (roGFP2), we show that transient oxidation of the cytosol and the nuclei occurred at G1 in the synchronized dividing cells of the Arabidopsis root apical meristem, with reduction at G2 and mitosis. This redox cycle was absent from low ascorbate mutants in which nuclei were significantly more oxidized than controls. The cell cycle-dependent increase in nuclear size was impaired in the ascorbate-deficient mutants, which had fewer cells per unit area in the root proliferation zone. The transcript profile of the dry seeds and size of the imbibed seeds was strongly influenced by low ascorbate but germination, dormancy release and seed aging characteristics were unaffected. Innovation: These data demonstrate the presence of a redox cycle within the plant cell cycle and that the redox state of the nuclei is an important factor in cell cycle progression. Conclusions: Controlled oxidation is a key feature of the early stages of the plant cell cycle. However, sustained mild oxidation restricts nuclear functions and impairs progression through the cell cycle leading to fewer cells in the root apical meristem

    Dark matter distribution in Milky Way-analog galaxies

    Our current understanding of how dark matter (DM) is distributed within the Milky Way (MW) halo, particularly in the solar neighborhood, is based on either careful studies of the local stellar orbits or model assumptions on the global shape of the MW halo. In this work, we undertake a study of external galaxies, with the intent of providing insight to the DM distribution in MW-analog galaxies. For this, we carefully select a sample of galaxies similar to the MW, based on maximum atomic hydrogen (HI) rotational velocity (v=200-280 km s^{-1}) and morphological type (Sab-Sbc) criteria. With a need for deep, highly-resolved HI, our resulting sample is composed of 5 galaxies from the VIVA and THINGS surveys. To perform our baryonic analysis, we use deep Spitzer mid-IR images at 3.6 and 4.5 {\mu}m from the S4G survey. Based on the dynamical three-dimensional modeling software 3D-Barolo, we construct RCs and derive the gas and stellar contributions from the galaxy\'s gaseous- and stellar-disks mass surface density profiles. Through a careful decomposition of their rotation curves into their baryonic (stars, gas) and DM components, we isolate the DM contribution by using an MCMC-based approach. Based on the Sun\'s location and the MW\'s R_{25}, we define the corresponding location of the solar neighborhood in these systems. We put forward a window for the DM density (\rho=0.21-0.46 GeV cm^{-3}) at these galactocentric distances in our MW analog sample, consistent with the values found for the MW\'s local DM density, based on more traditional approaches found in the literature.Comment: 16 pages, 6 figures, 3 tables, submitted to Ap

    Didactic Sequence for Teaching Exponential Function

    This paper presents a methodological proposal for the teaching of exponential function, resulting from the application of a didactic sequence involving exponential function, where evidence of learning and the consolidation and application of mathematical concepts in problem solving were identified and analyzed. The Didactic Engineering of Michèle Artigue (1988) was used as a research methodology. As theoretical contributions that guided and enabled the development of the research, we chose the use of Mathematical Investigation in the classroom; Didactic Sequence in the conception of Zabala (1999); the Articulated Units of Conceptual Reconstruction proposed by Cabral (2017) and assumptions of Vygotsky\u27s theory. A didactic sequence composed of five UARC\u27s was elaborated to work the exponential function, with a view to minimizing the difficulties naturally imposed by the content to be explained. Microgenetic analysis of verbal interactions between teacher and students was used to analyze the results of the application. The results show that the students participating in the experiment showed evidence of learning, recorded during the process, and began to have a good understanding of the concepts and properties related to the topic, in addition to a good performance in carrying out the activities, facts that corroborate the potential of the didactic sequence proposed herein

    Inhibition of PbGP43 expression may suggest that gp43 is a virulence factor in Paracoccidioides brasiliensis

    ABSTARCT: Glycoprotein gp43 is an immunodominant diagnostic antigen for paracoccidioidomycosis caused by Paracoccidioides brasiliensis. It is abundantly secreted in isolates such as Pb339. It is structurally related to beta-1,3-exoglucanases, however inactive. Its function in fungal biology is unknown, but it elicits humoral, innate and protective cellular immune responses; it binds to extracellular matrix-associated proteins. In this study we applied an antisense RNA (aRNA) technology and Agrobacterium tumefaciens-mediated transformation to generate mitotically stable PbGP43 mutants (PbGP43 aRNA) derived from wild type Pb339 to study its role in P. brasiliensis biology and during infection. Control PbEV was transformed with empty vector. Growth curve, cell vitality and morphology of PbGP43 aRNA mutants were indistinguishable from those of controls. PbGP43 expression was reduced 80-85% in mutants 1 and 2, as determined by real time PCR, correlating with a massive decrease in gp43 expression. This was shown by immunoblotting of culture supernatants revealed with anti-gp43 mouse monoclonal and rabbit polyclonal antibodies, and also by affinity-ligand assays of extracellular molecules with laminin and fibronectin. In vitro, there was significantly increased TNF-α production and reduced yeast recovery when PbGP43 aRNA1 was exposed to IFN-γ-stimulated macrophages, suggesting reduced binding/uptake and/or increased killing. In vivo, fungal burden in lungs of BALB/c mice infected with silenced mutant was negligible and associated with decreased lung ΙΛ-10 and IL-6. Therefore, our results correlated low gp43 expression with lower pathogenicity in mice, but that will be definitely proven when PbGP43 knockouts become available.
