Agile at scale : a summary of the 8th International Workshop on Large-Scale Agile Development
The Large-Scale Agile Development workshop explored the
main research challenges in large-scale software development. We considered multi-site organisations with large-scale projects that include a large
number of teams adopting agile methods. Such topics include inter-team
coordination, knowledge sharing, large project organisation, agile transformation, agile teamwork quality, project models that facilitate several
self-organising teams, and practices for scaling agile methods. We accepted five full research papers, which are included in this volume. The
accepted papers report empirical research studies using surveys, observations and case studies. Also, an interactive online discussion session was
conducted to compare the two approaches, SAFe and Spotify. The workshop participants, which were around a hundred people, joined this discussion to compare the two approaches and suggest some future research
questions about the hybridisation of SAFe and Spotify. This workshop
summary contributes as a current snapshot of research along with some
results from an interactive discussion about SAFe and Spotify
LNK (SH2B3): paradoxical effects in ovarian cancer.
LNK (SH2B3) is an adaptor protein studied extensively in normal and malignant hematopoietic cells. In these cells, it downregulates activated tyrosine kinases at the cell surface resulting in an antiproliferative effect. To date, no studies have examined activities of LNK in solid tumors. In this study, we found by in silico analysis and staining tissue arrays that the levels of LNK expression were elevated in high-grade ovarian cancer. To test the functional importance of this observation, LNK was either overexpressed or silenced in several ovarian cancer cell lines. Remarkably, overexpression of LNK rendered the cells resistant to death induced by either serum starvation or nutrient deprivation, and generated larger tumors using a murine xenograft model. In contrast, silencing of LNK decreased ovarian cancer cell growth in vitro and in vivo. Western blot studies indicated that overexpression of LNK upregulated and extended the transduction of the mitogenic signal, whereas silencing of LNK produced the opposite effects. Furthermore, forced expression of LNK reduced cell size, inhibited cell migration and markedly enhanced cell adhesion. Liquid chromatography-mass spectroscopy identified 14-3-3 as one of the LNK-binding partners. Our results suggest that in contrast to the findings in hematologic malignancies, the adaptor protein LNK acts as a positive signal transduction modulator in ovarian cancers
Nuclear Receptor Rev-erb Alpha (Nr1d1) Functions in Concert with Nr2e3 to Regulate Transcriptional Networks in the Retina
The majority of diseases in the retina are caused by genetic mutations affecting the development and function of photoreceptor cells. The transcriptional networks directing these processes are regulated by genes such as nuclear hormone receptors. The nuclear hormone receptor gene Rev-erb alpha/Nr1d1 has been widely studied for its role in the circadian cycle and cell metabolism, however its role in the retina is unknown. In order to understand the role of Rev-erb alpha/Nr1d1 in the retina, we evaluated the effects of loss of Nr1d1 to the developing retina and its co-regulation with the photoreceptor-specific nuclear receptor gene Nr2e3 in the developing and mature retina. Knock-down of Nr1d1 expression in the developing retina results in pan-retinal spotting and reduced retinal function by electroretinogram. Our studies show that NR1D1 protein is co-expressed with NR2E3 in the outer neuroblastic layer of the developing mouse retina. In the adult retina, NR1D1 is expressed in the ganglion cell layer and is co-expressed with NR2E3 in the outer nuclear layer, within rods and cones. Several genes co-targeted by NR2E3 and NR1D1 were identified that include: Nr2c1, Recoverin, Rgr, Rarres2, Pde8a, and Nupr1. We examined the cyclic expression of Nr1d1 and Nr2e3 over a twenty-four hour period and observed that both nuclear receptors cycle in a similar manner. Taken together, these studies reveal a novel role for Nr1d1, in conjunction with its cofactor Nr2e3, in regulating transcriptional networks critical for photoreceptor development and function
Spectral weight transfer in a disorder-broadened Landau level
In the absence of disorder, the degeneracy of a Landau level (LL) is
, where is the magnetic field, is the area of the sample
and is the magnetic flux quantum. With disorder, localized states
appear at the top and bottom of the broadened LL, while states in the center of
the LL (the critical region) remain delocalized. This well-known phenomenology
is sufficient to explain most aspects of the Integer Quantum Hall Effect (IQHE)
[1]. One unnoticed issue is where the new states appear as the magnetic field
is increased. Here we demonstrate that they appear predominantly inside the
critical region. This leads to a certain ``spectral ordering'' of the localized
states that explains the stripes observed in measurements of the local inverse
compressibility [2-3], of two-terminal conductance [4], and of Hall and
longitudinal resistances [5] without invoking interactions as done in previous

Exoplanet phase curves: observations and theory
Phase curves are the best technique to probe the three dimensional structure
of exoplanets' atmospheres. In this chapter we first review current exoplanets
phase curve observations and the particular challenges they face. We then
describe the different physical mechanisms shaping the atmospheric phase curves
of highly irradiated tidally locked exoplanets. Finally, we discuss the
potential for future missions to further advance our understanding of these new

MAESTRI Toolkit for Industrial Symbiosis: overview, lessons learnt and implications
This paper presents a structured approach to support the development of self-organized industrial symbiosis, the Toolkit for Industrial Symbiosis. Developed within MAESTRI project, it provides a set of tools and methods to help companies gain value from wasted resources and contributes to MAESTRI goal of advancing the sustainability of European manufacturing and process industry. A participatory approach was taken for its development. The ultimate objective of this work is to encourage companies to change their attitude and consider waste as a resource and potential source for value creation
Expression quantitative trait loci are highly sensitive to cellular differentiation state
Blood cell development from multipotent hematopoietic stem cells to specialized blood cells is accompanied by drastic changes in gene expression for which the triggers remain mostly unknown. Genetical genomics is an approach linking natural genetic variation to gene expression variation, thereby allowing the identification of genomic loci containing gene expression modulators (eQTLs). In this paper, we used a genetical genomics approach to analyze gene expression across four developmentally close blood cell types collected from a large number of genetically different but related mouse strains. We found that, while a significant number of eQTLs (365) had a consistent “static” regulatory effect on gene expression, an even larger number were found to be very sensitive to cell stage. As many as 1,283 eQTLs exhibited a “dynamic” behavior across cell types. By looking more closely at these dynamic eQTLs, we show that the sensitivity of eQTLs to cell stage is largely associated with gene expression changes in target genes. These results stress the importance of studying gene expression variation in well-defined cell populations. Synthesis, Biological Evaluation and Mechanism Studies of Deoxytylophorinine and Its Derivatives as Potential Anticancer Agents
Previous studies indicated that (+)-13a-(S)-Deoxytylophorinine (1) showed profound anti-cancer activities both in vitro and in vivo and could penetrate the blood brain barrier to distribute well in brain tissues. CNS toxicity, one of the main factors to hinder the development of phenanthroindolizidines, was not obviously found in 1. Based on its fascinating activities, thirty-four derivatives were designed, synthesized; their cytotoxic activities in vitro were tested to discover more excellent anticancer agents. Considering the distinctive mechanism of 1 and interesting SAR of deoxytylophorinine and its derivatives, the specific impacts of these compounds on cellular progress as cell signaling transduction pathways and cell cycle were proceeded with seven representative compounds. 1 as well as three most potent compounds, 9, 32, 33, and three less active compounds, 12, 16, 35, were selected to proform this study to have a relatively deep view of cancer cell growth-inhibitory characteristics. It was found that the expressions of phospho-Akt, Akt, phospho-ERK, and ERK in A549 cells were greater down-regulated by the potent compounds than by the less active compounds in the Western blot analysis. To the best of our knowledge, this is the first report describing phenanthroindolizidines alkaloids display influence on the crucial cell signaling proteins, ERK. Moreover, the expressions of cyclin A, cyclin D1 and CDK2 proteins depressed more dramatically when the cells were treated with 1, 9, 32, and 33. Then, these four excellent compounds were subjected to flow cytometric analysis, and an increase in S-phase was observed in A549 cells. Since the molecular level assay results of Western blot for phospho-Akt, Akt, phospho-ERK, ERK, and cyclins were relevant to the potency of compounds in cellular level, we speculated that this series of compounds exhibit anticancer activities through blocking PI3K and MAPK signaling transduction pathways and interfering with the cell cycle progression
