    Variabilidad de la Respuesta de las Células Dendríticas Estimuladas in vitro con Porphyromonas gingivalis y Aggregatibacter actinomycetemcomitans

    ResumenLas células dendríticas son células presentadoras de antígeno capaces de inducir la activación y maduración de linfocitos T CD4+ vírgenes hacia un fenotipo efector especifico Th1 o Th2, dependiendo del tipo de antigeno presentado, las senales co-estimuladoras que expresan y el ambiente de citoquinas en el cual se produce la interaccion celula-celula. El objetivo del presente trabajo es analizar la respuesta de las celulas dendriticas estimuladas in vitro con distintas concentraciones de las bacterias periodontopatógenas Pg y Aa. En celulas dendriticas derivadas de monocitos de sangre periferica estimuladas con 101 a 109 bacterias/mL de Pg y Aa se evaluo la expresion del marcador de maduracion CD80 mediante citometria de flujo y de las citoquinas IL1β, IL2, IL5, IL6, IL10, IL12, IL13, IFNγ, TNFα y TNFβ mediante RT-PCR cuantitativa. Aa y Pg indujeron maduracion de las celulas dendriticas, detectandose significativamente mayor expresion de CD80 con la estimulacion de Aa, e indujeron predominantemente la expresion de citoquinas propias de una respuesta Th1. Dependiendo de la carga bacteriana, fueron detectados distintos umbrales de induccion de expresion de citoquinas. Aa indujo la sintesis de IL1β, IL12, IFNγ, TNFα y TNFβ a menor carga bacteriana que Pg. Tomados en conjunto, estos datos nos permiten especular un mayor potencial antigenico y proyectar una mayor capacidad patogenica durante la infeccion periodontal de Aa en comparación a Pg.AbstractDendritic cells are potent antigen-presenting cells able to prime naive T cells and polarize them towards a Th1 or Th2 response, depending on the type of the antigen presented to the TCR, the type of costimulatory signals, and the cytokine pattern in the environment. The aim of this work was to analyze the response of dendritic cells to in vitro stimulation with Pg and Aa. In monocyte-derived dendritic cells stimulated with 101 to 109 bacteria/mL of Pg or Aa were evaluated both the expression of the maturation marker CD80 by flow cytometry and the expression of the cytokines IL1β, IL2, IL5, IL6, IL10, IL12, IL13, IFNγ, TNFα and TNFβ by quantitative RT-PCR. Both Pg and Aa led to dendritic cell maturation, detecting higher CD80 expression upon Aa-stimulation, and induced a Th1 pattern of cytokine expression. Aa-stimulated dendritic cells expressed IL1β, IL12, IFNγ, TNFα and TNFβ mRNAs with lower bacterial charge than with Pg. Furthermore, our data indicated the existence of distinct thresholds for the induction of the different cytokines analyzed. Taken together, these data allow us to speculate a higher antigenic potential and higher pathogenic capacity of Aa than Pg during periodontal infections

    Nuclear data requirements for the ADS conceptual design EFIT: Uncertainty and sensitivity study

    In this paper, we assess the impact of activation cross-section uncertainties on relevant fuel cycle parameters for a conceptual design of a modular European Facility for Industrial Transmutation (EFIT) with a “double strata” fuel cycle. Next, the nuclear data requirements are evaluated so that the parameters can meet the assigned design target accuracies. Different discharge burn-up levels are considered: a low burn-up, corresponding to the equilibrium cycle, and a high burn-up level, simulating the effects on the fuel of the multi-recycling scenario. In order to perform this study, we propose a methodology in two steps. Firstly, we compute the uncertainties on the system parameters by using a Monte Carlo simulation, as it is considered the most reliable approach to address this problem. Secondly, the analysis of the results is performed by a sensitivity technique, in order to identify the relevant reaction channels and prioritize the data improvement needs. Cross-section uncertainties are taken from the EAF-2007/UN library since it includes data for all the actinides potentially present in the irradiated fuel. Relevant uncertainties in some of the fuel cycle parameters have been obtained, and we conclude with recommendations for future nuclear data measurement programs, beyond the specific results obtained with the present nuclear data files and the limited available covariance information. A comparison with the uncertainty and accuracy analysis recently published by the WPEC-Subgroup26 of the OECD using BOLNA covariance matrices is performed. Despite the differences in the transmuter reactor used for the analysis, some conclusions obtained by Subgroup26 are qualitatively corroborated, and improvements for additional cross sections are suggested

    Multiyear Life Cycle Assessment of switchgrass (Panicum virgatum L.) production in the Mediterranean region of Spain: A comparative case study

    [EN] An LCA of the cultivation of switchgrass in the Mediterranean region of Spain is carried out, based on 2010-2013 inventory data from experimental plots of two sites, Moncofar and Orihuela. Thus, a 4-year cycle is evaluated, considering different sources of variability. The functional unit is 1 t of switchgrass (dry basis) for electricity generation. Besides typical impact categories, blue and green water consumption impacts are also addressed by using watershed-specific characterization factors. In 2010, the production in Orihuela is more input-intensive than it is in Moncofar, while the biomass yield is lower. This causes greater climate change as CO2-eq. (709.1 vs. 65.0 kg t(-1)) and greater metal depletion as Fe-eq. (8.1 vs. 1.5 kg t(-1)). In the subsequent years, the yields are higher in Orihuela, and Moncofar performs worse for some specific impact categories, mainly the toxicity-related ones, and also metal depletion as Fe-eq., but only in 2011 (2.0 vs. 1.2 kg t(-1)). Due to larger irrigation doses, the blue water impact as ecosystem-eq. water is always higher in Orihuela (e.g. in 2010, 2020 vs. 390 m(3) t(-1)). On the contrary, the green water impact, also as ecosystem-eq. water, is greater in Moncofar, except for the first year (86.8 vs. 15.2 m(3) t(-1)). Switchgrass from the two locations could be eligible for bioelectricity production in the European Union in accordance with the sustainability requirements for greenhouse gas savings.     An insight into transfer hydrogenation reactions catalysed by iridium(iii) bis-n-heterocyclic carbenes

    A variety of [M(L)2(L')2{kC,C'-bis(NHC)}]BF4 complexes (M = Rh or Ir; L = CH3CN or wingtip group; L' = I– or CF3COO–; NHC=N-heterocyclic carbene) have been tested as pre-catalysts for the transfer hydrogenation of ketones and imines. The conversions and TOF's obtained are closely related to the nature of the ligand system and metal centre, more strongly coordinating wingtip groups yielding more active and recyclable catalysts. Theoretical calculations at the DFT level support a classic stepwise metal-hydride pathway against the concerted Meerwein–Ponndorf–Verley (MPV) mechanism. The calculated catalytic cycle involves a series of ligand rearrangements due to the high trans effect of the carbene and hydrido ligands, which are more stable when situated in mutual cis positions. The reaction profiles obtained for the complexes featuring an iodide or a trifluoroacetate in one of the apical positions agree well with the relative activity observed for both catalysts

    Strength tests on asphalt mixes attacked by motor fuels. Recommendations on bath immersion times

    [Abstract:] One of the limitations of asphalt mixes as pavement materials is their poor resistance to attack from crude oil-based motor fuels. Several procedures exist for determining the resistance of asphalt materials to motor fuel action. Different test standards can be found for fillers and surface dressings in both the US and European series. However, where the assessment of asphalt mixes themselves are concerned, there is a considerable lack of standardisation. This paper reports on laboratory studies on certain essential aspects of the procedures for assessing the strength of asphalt mixes in respect of motor fuel attack, chiefly the time the mix is exposed to the fuel. Based on the research results, the paper makes some good-practice recommendations for weight-loss procedures after immersion in motor fuel, with or without subsequent brushing, and in respect of the Marshall stability preserved after immersion. In relation to the immersion period and in view of the laboratory findings and their subsequent statistical processing, one of the factors with the greatest influence is the recommendation that the immersion period should be 24 hours, except in the weight-loss test subseuent to immersion without any brushing where the results obtained prove to be more significant if the immersion period is seven days

    Confined dynamics in poly(ethylene terephthalate): A coherent and incoherent neutron scattering study

    We show that the combination of dielectric relaxation with neutron spin echo and incoherent neutron backscattering measurements performed in deuterated and protonated poly(ethylene terephthalate) suggests that the intrinsic dynamics of semicrystalline polymers occurs in an homogeneous scenario, similar to that valid to describe the dynamics of totally amorphous polymers. The quasielastic neutron scattering data are satisfactorily described by a theoretical model that considers that the proton mobility follows a random jump-diffusion in a restricted environment

    HST PanCET program: A Cloudy Atmosphere for the promising JWST target WASP-101b

    We present results from the first observations of the Hubble Space Telescope (HST) Panchromatic Comparative Exoplanet Treasury (PanCET) program for WASP-101b, a highly inflated hot Jupiter and one of the community targets proposed for the James Webb Space Telescope (JWST) Early Release Science (ERS) program. From a single HST Wide Field Camera 3 (WFC3) observation, we find that the near-infrared transmission spectrum of WASP-101b contains no significant H2_2O absorption features and we rule out a clear atmosphere at 13{\sigma}. Therefore, WASP-101b is not an optimum target for a JWST ERS program aimed at observing strong molecular transmission features. We compare WASP-101b to the well studied and nearly identical hot Jupiter WASP-31b. These twin planets show similar temperature-pressure profiles and atmospheric features in the near-infrared. We suggest exoplanets in the same parameter space as WASP-101b and WASP-31b will also exhibit cloudy transmission spectral features. For future HST exoplanet studies, our analysis also suggests that a lower count limit needs to be exceeded per pixel on the detector in order to avoid unwanted instrumental systematics.Comment: 7 pages, 4 figures, 1 table, Accepted to ApJ

    The IFMIF-EVEDA accelerator beam dump design

    The IFMIF-EVEDA accelerator will be a 9 MeV, 125 mA cw deuteron accelerator prototype for verifying the validity of the 40 MeV accelerator design for IFMIF. A beam dump designed for maximum power of 1.12 MW will be used to stop the beam at the accelerator exit. The conceptual design for the IFMIF-EVEDA accelerator beam dump is based on a conical beam stop made of OFE copper. The cooling system uses an axial high velocity flow of water pressurized up to 3.4 × 105 Pa to avoid boiling. The design has been shown to be compliant with ASME mechanical design rules under nominal full power conditions. A sensitivity analysis has been performed to take into account the possible margin on the beam properties at the beam dump entrance. This analysis together with the study of the maintenance issues and the mounting and dismounting operations has led to the complete design definition
