    Anthocyanins inhibit tumor necrosis alpha-induced loss of Caco-2 cell barrier integrity

    An increased permeability of the intestinal barrier is proposed as a major event in the pathophysiology of conditions characterized by chronic gut inflammation. This study investigated the capacity of pure anthocyanins (AC), and berry and rice extracts containing different types and amounts of AC, to inhibit tumor necrosis alpha (TNFα)-induced permeabilization of Caco-2 cell monolayers. Caco-2 cells differentiated into intestinal epithelial cell monolayers were incubated in the absence/presence of TNFα, with or without the addition of AC or AC-rich plant extracts (ACRE). AC and ACRE inhibited TNFα-induced loss of monolayer permeability as assessed by changes in transepithelial electrical resistance (TEER) and paracellular transport of FITC-dextran. In the range of concentrations tested (0.25–1 μM), O-glucosides of cyanidin, and delphinidin, but not those of malvidin, peonidin and petunidin protected the monolayer from TNFα-induced decrease of TEER and increase of FITC-dextran permeability. Cyanidin and delphinidin acted by mitigating TNFα-triggered activation of transcription factor NF-κB, and downstream phosphorylation of myosin light chain (MLC). The protective actions of the ACRE on TNFα-induced TEER increase was positively correlated with the sum of cyanidins and delphinidins (r2 = 0.83) content in the ACRE. However, no correlation was observed between TEER and ACRE total AC, malvidin, or peonidin content. Results support a particular capacity of cyanidins and delphinidins in the protection of the intestinal barrier against inflammation-induced permeabilization, in part through the inhibition of the NF-κB pathway.Fil: Cremonini, Eleonora.     A GIS-based hydrographic resurvey strategy of the Belgian Continental Shelf

    Using a GIS-based approach, bathymetric surveys and maritime traffic records were combined to determine resurvey priorities within the Belgian Continental Shelf (BCS). Four reference layers were produced: (1) water depth; (2) maximum absolute change in water depth; (3) ship traffic intensity; (4) maximum ship draught. The reference layers were reclassified and merged into a weighted overlay analysis. Two combinations of layers and weight factors were used and resulted in priority maps differing greatly from each other. The reliability of the analysis depends on the way weight factors are assigned, and on the availability and accuracy of the data. These are limited for bathymetric surveys. This empirical GIS-based methodology can be applied as a whole to a zone showing various morphodynamic patterns. It can also be automated: additional datasets can be included in the analysis, and different scenarios and assumptions can be easily tested

    Exploring the drivers of malnutrition in West Africa from health and social science perspectives : a comparative methodological review

    West Africa has a high burden of malnutrition and the drivers are often complex, highly context-specific, and cut across individual, social, political and environmental domains. Public health research most often considers immediate individual health and diet drivers, at the expense of wider considerations that may fall outside of a health agenda. The objective of this systematic mapping review is to map the broad drivers of malnutrition in West Africa, from public health and social science perspectives, and to evaluate the additional value of an interdisciplinary approach. Evidence was gathered from one public health (MEDLINE) and one social science (International Bibliography of Social Science) database using a detailed search syntax tailored to each disciplinary configuration. Literature was screened against pre-determined eligibility criteria and extracted from abstracts. Studies published in English or French between January 2010 and April 2018 were considered for inclusion. Driver categories (immediate, underlying and basic drivers) were coded against the UNICEF conceptual framework of malnutrition. A total of 358 studies were included; 237 were retrieved from the public health database and 124 from the social science database, 3 studies were included in both. The public health and social science literature document different drivers, with MEDLINE most often reporting immediate drivers of malnutrition and the International Bibliography of Social Science database reporting underlying and basic drivers. The combined literature offers more balanced representation across categories. An interdisciplinary approach proved successful in achieving complementarity in search results while upholding rigorous methods. We recommend that interdisciplinary approaches are utilised to bridge recognised gaps between defined disciplines

    Impact of pollen on throughfall biochemistry in European temperate and boreal forests

    Pollen is known to affect forest throughfall biochemistry, but underlying mechanisms are not fully understood. We used generalized additive mixed modelling to study the relationship between long-term series of measured throughfall fluxes in spring (April–June) at forest plots and corresponding airborne pollen concentrations (Seasonal Pollen Integral, SPIn) from nearby aerobiological monitoring stations. The forest plots were part of the intensive long term monitoring (Level II) network of the UNECE International Co-operative Programme on Assessment and Monitoring of Air Pollution Effects on Forests (ICP Forests) with dominant tree genera Fagus, Quercus, Pinus and Picea, and were distributed all across Europe. We also conducted a 7-day laboratory dissolution experiment with bud scales and flower stalks of European beech (Fagus sylvatica L.), pollen of beech, common oak (Quercus robur L.), silver birch (Betula pendula L.), Scots pine (Pinus sylvestris L.), Corsican pine (Pinus nigra Arnold ssp. laricio (Poiret) Maire), Norway spruce (Picea abies (L.) Karst.) and sterilized pollen of silver birch in a nitrate (NO3--N) solution (11.3 mg N L-1). Throughfall fluxes of potassium (K+), ammonium (NH4+-N), dissolved organic carbon (DOC) and dissolved organic nitrogen (DON) showed a positive relationship with SPIn whereas NO3--N fluxes showed a negative relationship with SPIn. In years with massive seed production of beech and oak SPIn and throughfall fluxes of K+ and DOC were higher, but fluxes of NO3--N were lower. The experiment broadly confirmed the findings based on field data. Within two hours, pollen released large quantities of K+, phosphate, DOC and DON, and lesser amounts of sulphate, sodium and calcium. After 24-48 hours, NO3--N started to disappear, predominantly in the treatments with broadleaved pollen, while concentrations of nitrite and NH4+-N increased. At the end of the experiment, the inorganic nitrogen (DIN) was reduced, presumably because it was lost as gaseous nitric oxide (NO). There was no difference for sterilized pollen, indicating that the involvement of microbial activity was limited in above N transformations.     Improved SIFTER v2 algorithm for long-term GOME-2A satellite retrievals of fluorescence with a correction for instrument degradation

    Solar-induced fluorescence (SIF) data from satellites are increasingly used as a proxy for photosynthetic activity by vegetation and as a constraint on gross primary production. Here we report on improvements in the algorithm to retrieve mid-morning (09:30 LT) SIF estimates on the global scale from the GOME-2 sensor on the MetOp-A satellite (GOME-2A) for the period 2007-2019. Our new SIFTER (Sun-Induced Fluorescence of Terrestrial Ecosystems Retrieval) v2 algorithm improves over a previous version by using a narrower spectral window that avoids strong oxygen absorption and being less sensitive to water vapour absorption, by constructing stable reference spectra from a 6-year period (2007-2012) of atmospheric spectra over the Sahara and by applying a latitude-dependent zero-level adjustment that accounts for biases in the data product. We generated stable, good-quality SIF retrievals between January 2007 and June 2013, when GOME-2A degradation in the near infrared was still limited. After the narrowing of the GOME-2A swath in July 2013, we characterised the throughput degradation of the level-1 data in order to derive reflectance corrections and apply these for the SIF retrievals between July 2013 and December 2018. SIFTER v2 data compare well with the independent NASA v2.8 data product. Especially in the evergreen tropics, SIFTER v2 no longer shows the underestimates against other satellite products that were seen in SIFTER v1. The new data product includes uncertainty estimates for individual observations and is best used for mostly clear-sky scenes and when spectral residuals remain below a certain spectral autocorrelation threshold. Our results support the use of SIFTER v2 data being used as an independent constraint on photosynthetic activity on regional to global scales.</p

    Follow-up of newborns treated with extracorporeal membrane oxygenation: a nationwide evaluation at 5 years of age

    INTRODUCTION: Extracorporeal membrane oxygenation (ECMO) is a supportive cardiopulmonary bypass technique for babies with acute reversible cardiorespiratory failure. We assessed morbidity in ECMO survivors at the age of five years, when they start primary school and major decisions for their school careers must be made. METHODS: Five-year-old neonatal venoarterial-ECMO survivors from the two designated ECMO centres in The Netherlands (Erasmus MC – Sophia Children's Hospital in Rotterdam, and University Medical Center Nijmegen) were assessed within the framework of an extensive follow-up programme. The protocol included medical assessment, neuromotor assessment, and psychological assessment by means of parent and teacher questionnaires. RESULTS: Seventeen of the 98 children included in the analysis (17%) were found to have neurological deficits. Six of those 17 (6% of the total) showed major disability. Two of those six children had a chromosomal abnormality. Three were mentally retarded and profoundly impaired. The sixth child had a right-sided hemiplegia. These six children did not undergo neuromotor assessment. Twenty-four of the remaining 92 children (26%) showed motor difficulties: 15% actually had a motor problem and 11% were at risk for this. Cognitive delay was identified in 11 children (14%). The mean IQ score was within the normal range (IQ = 100.5). CONCLUSION: Neonatal ECMO in The Netherlands was found to be associated with considerable morbidity at five years of age. It appeared feasible to have as many as 87% of survivors participate in follow-up assessment, due to cooperation between two centres and small travelling distances. Objective evaluation of the long-term morbidity associated with the application of this highly invasive technology in the immediate neonatal period requires an interdisciplinary follow-up programme with nationwide consensus on timing and actual testing protocol

    Recognizing recurrent neural networks (rRNN): Bayesian inference for recurrent neural networks

    Recurrent neural networks (RNNs) are widely used in computational neuroscience and machine learning applications. In an RNN, each neuron computes its output as a nonlinear function of its integrated input. While the importance of RNNs, especially as models of brain processing, is undisputed, it is also widely acknowledged that the computations in standard RNN models may be an over-simplification of what real neuronal networks compute. Here, we suggest that the RNN approach may be made both neurobiologically more plausible and computationally more powerful by its fusion with Bayesian inference techniques for nonlinear dynamical systems. In this scheme, we use an RNN as a generative model of dynamic input caused by the environment, e.g. of speech or kinematics. Given this generative RNN model, we derive Bayesian update equations that can decode its output. Critically, these updates define a 'recognizing RNN' (rRNN), in which neurons compute and exchange prediction and prediction error messages. The rRNN has several desirable features that a conventional RNN does not have, for example, fast decoding of dynamic stimuli and robustness to initial conditions and noise. Furthermore, it implements a predictive coding scheme for dynamic inputs. We suggest that the Bayesian inversion of recurrent neural networks may be useful both as a model of brain function and as a machine learning tool.
