    Conditioned place preference and locomotor activity in response to methylphenidate, amphetamine and cocaine in mice lacking dopamine D4 receptors

    Methylphenidate (MP) and amphetamine (AMPH) are the most frequently prescribed medications for the treatment of attention-deficit/hyperactivity disorder (ADHD). Both drugs are believed to derive their therapeutic benefit by virtue of their dopamine (DA)-enhancing effects, yet an explanation for the observation that some patients with ADHD respond well to one medication but not to the other remains elusive. The dopaminergic effects of MP and AMPH are also thought to underlie their reinforcing properties and ultimately their abuse. Polymorphisms in the human gene that codes for the DA D4 receptor (D4R) have been repeatedly associated with ADHD and may correlate with the therapeutic as well as the reinforcing effects of responses to these psychostimulant medications. Conditioned place preference (CPP) for MP, AMPH and cocaine were evaluated in wild-type (WT) mice and their genetically engineered littermates, congenic on the C57Bl/6J background, that completely lack D4Rs (knockout or KO). In addition, the locomotor activity in these mice during the conditioning phase of CPP was tested in the CPP chambers. D4 receptor KO and WT mice showed CPP and increased locomotor activity in response to each of the three psychostimulants tested. D4R differentially modulates the CPP responses to MP, AMPH and cocaine. While the D4R genotype affected CPP responses to MP (high dose only) and AMPH (low dose only) it had no effects on cocaine. Inasmuch as CPP is considered an indicator of sensitivity to reinforcing responses to drugs these data suggest a significant but limited role of D4Rs in modulating conditioning responses to MP and AMPH. In the locomotor test, D4 receptor KO mice displayed attenuated increases in AMPH-induced locomotor activity whereas responses to cocaine and MP did not differ. These results suggest distinct mechanisms for D4 receptor modulation of the reinforcing (perhaps via attenuating dopaminergic signalling) and locomotor properties of these stimulant drugs. Thus, individuals with D4 receptor polymorphisms might show enhanced reinforcing responses to MP and AMPH and attenuated locomotor response to AMPH.Fil: Thanos, P. K..     Educational Strategies to Promote Adherence to Treatment in Patients with Cardiovascular Disease

    Introduction: Educational material is a key strategy for primary health care promotion. Purpose: To design and validate educational material adapted to the population and aimed to increase knowledge about adherence to the treatment of arterial hypertension and diabetes mellitus. Methodology: Methodological study for the design of educational material for people with diabetes mellitus and high blood pressure. For the design, content validity tests were carried out, with the participation of six experts in health education and six patients with chronic diseases. Validation was performed pursuant to the attraction, understanding, engagement, and acceptance criteria. Results: The validation confirmed that all items and criteria were above the minimal expected range. Conclusion: The design and validation of educational material provide elements that improve the education of patients about their pathologies and their adherence to treatment

    From absent to present pasts: civil society, democracy and the shifting place of memory in Brazil

    This paper takes Alexis de Tocqueville’s concern with the emotional life of citizens as a cue for exploring the role of collective memory within ‘the self-organizing sphere’ and asking how the invocation of memory affects progress towards democracy. The paper hones in on the Brazilian experience, re-assessing Brazil’s amnesiac past as well as its much lauded ‘turn to memory’. Against common assertions that Brazil’s ‘turn to memory’ will enhance the country’s democratic credentials, this paper argues that the move from an ‘absent’ to a ‘present’ past in Brazil in fact bodes rather mixed prospects for the country’s democratic deepening

    Could the 2017 ILAE and the four-dimensional epilepsy classifications be merged to a new "Integrated Epilepsy Classification"?

    Over the last few decades the ILAE classifications for seizures and epilepsies (ILAE-EC) have been updated repeatedly to reflect the substantial progress that has been made in diagnosis and understanding of the etiology of epilepsies and seizures and to correct some of the shortcomings of the terminology used by the original taxonomy from the 1980s. However, these proposals have not been universally accepted or used in routine clinical practice. During the same period, a separate classification known as the "Four-dimensional epilepsy classification" (4D-EC) was developed which includes a seizure classification based exclusively on ictal symptomatology, which has been tested and adapted over the years. The extensive arguments for and against these two classification systems made in the past have mainly focused on the shortcomings of each system, presuming that they are incompatible. As a further more detailed discussion of the differences seemed relatively unproductive, we here review and assess the concordance between these two approaches that has evolved over time, to consider whether a classification incorporating the best aspects of the two approaches is feasible. To facilitate further discussion in this direction we outline a concrete proposal showing how such a compromise could be accomplished, the "Integrated Epilepsy Classification". This consists of five categories derived to different degrees from both of the classification systems: 1) a "Headline" summarizing localization and etiology for the less specialized users, 2) "Seizure type(s)", 3) "Epilepsy type" (focal, generalized or unknown allowing to add the epilepsy syndrome if available), 4) "Etiology", and 5) "Comorbidities & patient preferences"

    From North Africa to Latin America and back: comparative findings and theoretical reflections

    Taking the different case studies of the book together, one general observation stands out: Key agents of socioeconomic contention, including movements by organized labor and the unemployed that were important in the run-up to the uprisings and that saw their political opportunities open up in the immediate aftermath of the revolutions, have since been effectively marginalized as political actors. The concluding chapter reflects on the causes of this weakness of socioeconomic contention by identifying comparative insights that emerge from the contributions to this volume and by situating them in the context of broader comparative and theoretical debates on the relationship between social movements and political change. More specifically, the chapter first discusses Egypt’s and Tunisia’s post-revolutionary trajectories from a comparative perspective. Second, it discusses these comparative findings in the light of experiences in Latin America. Third, drawing again on comparative scholarship on Latin America, the chapter offers a theoretical interpretation of some of the main dynamics observed in Egypt and Tunisia based on the notion of a popular-sector incorporation crisis. Fourth and finally, the chapter concludes with general implications and an outlook

    Weak-lensing mass calibration of redMaPPer galaxy clusters in Dark Energy Survey Science Verification data

    We use weak-lensing shear measurements to determine the mean mass of optically selected galaxy clusters in Dark Energy Survey Science Verification data. In a blinded analysis, we split the sample of more than 8000 redMaPPer clusters into 15 subsets, spanning ranges in the richness parameter 5 ≤ λ ≤ 180 and redshift 0.2 ≤ z ≤ 0.8, and fit the averaged mass density contrast profiles with a model that accounts for seven distinct sources of systematic uncertainty: shear measurement and photometric redshift errors; clustermember contamination; miscentring; deviations from the NFW halo profile; halo triaxiality and line-of-sight projections. We combine the inferred cluster masses to estimate the joint scaling relation between mass, richness and redshift, M(λ, z) ∝ M0λF (1 + z) G. We find M0 ≡ (M200m | λ = 30, z = 0.5) = [2.35 ± 0.22 (stat) ± 0.12 (sys)] × 1014 M., with F = 1.12 ± 0.20 (stat) ± 0.06 (sys) and G = 0.18 ± 0.75 (stat) ± 0.24 (sys). The amplitude of the mass–richness relation is in excellent agreement with the weak-lensing calibration of redMaPPer clusters in SDSS by Simet et al. and with the Saro et al. calibration based on abundance matching of SPT-detected clusters. Our results extend the redshift range over which the mass–richness relation of redMaPPer clusters has been calibrated with weak lensing from z ≤ 0.3 to z ≤ 0.8. Calibration uncertainties of shear measurements and photometric redshift estimates dominate our systematic error budget and require substantial improvements for forthcoming studies
