    Evaluation of mannan-oligosaccharides (MOS) in broiler chicken during hot humid summer using zoo technical, molecular and physio-biochemical tools

    Climate resilient poultry production is a major challenge especially for hot regions like India. Accordingly, the efficacy of mannan-oligosaccharides as anti-heat stressor was studied. Broiler chicks were reared on a standard diet up to 14th day of age. Thereafter, the chicks were randomly distributed into three dietary treatment groups viz. T1 (Control group: Standard diet, T2 (Standard diet with MOS @ 0.3%) and T3 (Standard diet with MOS @ 0.5%) each with 40 birds divided in 5 replicates of 8 birds each upto 42 days of age. Experiment was carried out during hot-humid (August-September, 26.0±0.12° to 34.25±0.37°C, Rh%: 76.95±0.90 to 86.15±0.61) summer. Growth performance, immunity (4th week), physiological (4th and 6th week), biochemical (4th and 6th wk) and molecular parameters (4th and 6th wk) were recorded. Feed intake, live weight gain and FCR improved significantly (P<0.001) on MOS supplementation. The relative yield of immune organs at 4th and 6th week of age, humoral (P<0.001) as well as cellular (P<0.01) immunity also improved significantly. The percentage of haemoglobin, protein, aspartate transaminase and alanine transaminase increased significantly (P<0.001) due to MOS supplementation at 4th as well as 6th week of age. While H:L ratio, serum corticosterone and serum cholesterol decreased significantly (P<0.001) in MOS supplemented groups. Supplementation of MOS at both the levels (0.3 and 0.5%) caused significant down regulation of relative expression of HSP70 in jejunum tissues during 28th or 42nd day of age. From the results of the present study, it can be concluded that MOS supplementation @ 0.3% or 0.5% in diet of heat stressed broilers improved performance as well as welfare

    Liposomal nanotheranostics for multimode targeted in vivo bioimaging and near‐infrared light mediated cancer therapy

    Developing a nanotheranostic agent with better image resolution and high accumulation into solid tumor microenvironment is a challenging task. Herein, we established a light mediated phototriggered strategy for enhanced tumor accumulation of nanohybrids. A multifunctional liposome based nanotheranostics loaded with gold nanoparticles (AuNPs) and emissive graphene quantum dots (GQDs) were engineered named as NFGL. Further, doxorubicin hydrochloride was encapsulated in NFGL to exhibit phototriggered chemotherapy and functionalized with folic acid targeting ligands. Encapsulated agents showed imaging bimodality for in vivo tumor diagnosis due to their high contrast and emissive nature. Targeted NFGL nanohybrids demonstrated near infrared light (NIR, 750 nm) mediated tumor reduction because of generated heat and Reactive Oxygen Species (ROS). Moreover, NFGL nanohybrids exhibited remarkable ROS scavenging ability as compared to GQDs loaded liposomes validated by antitumor study.     EFFECT OF PENETRATION ENHANCER ON THE IN VITRO EX VIVO PERMEATION OF DICLOFENAC GEL

      Objective: The objective of this study was to compare and evaluate in vitro ex vivo transdermal potential of the diclofenac sodium (DS) gel by different permeation enhancers using Franz-type diffusion cells.Methods: DS gel was prepared with carbapol, dimethyl sulfoxide, oleic acid (OA), and menthol with 1% w/w concentration each as the penetration enhancer. The in vitro, ex vivo permeability were determined using cellophane membrane and abdominal rat skin. Steady-state flux, permeability coefficient (kp), diffusion coefficient, and lag time were calculated.Results: Menthol, at the concentration of 1% w/w, shows maximum kp, diffusion coefficient permeation enhancement effect with an enhancement ratio 40%, as compared to dimethyl sulfoxide and OA.Conclusion: This finding predicts that DS gel 1% w/w containing menthol 1% w/w can possibly deliver therapeutically relevant dose of DS.Keywords: Diclofenac sodium gel, Menthol, Penetration enhancer, Permeation coefficient. Â

    Quality by design approach for anti-hyperthyroid immediate release tablet of carbimazole

    The current study to formulate, evaluate and optimize carbimazole immediate release tablet by using quality by design approach. The look at of optimization formula and experimental design had been done with the aid of using design parameter primarily based totally on Box-Behnken design. Immediate release dosage form for rapid efficacy and inhibit the peroxidase enzymes and act as thyroid inhibitor. Method: Immediate release carbimazole tablets were manufactured by dry granulation using Quality by Design. The optimization of the formulation of tablets is done by examining the profile of the objective quality product. Microcrystalline cellulose, anhydrous lactose and magnesium stearate were all included in experiments designed using the Box-Behnken design. Their concentrations are influenced by the drug disintegration and release factor f2. Result: Optimized batch formulation shows immediate drug release with short disintegration time. In anova models are significant. Conclusion: The utilization of bound together quality by design approach for successful advancement of an IR carbimazole tablet.
