    Extreme climatic events down-regulate the grassland biomass response to elevated carbon dioxide

    Terrestrial ecosystems are considered as carbon sinks that may mitigate the impacts of increased atmospheric CO2 concentration ([CO2]). However, it is not clear what their carbon sink capacity will be under extreme climatic conditions. In this study, we used long-term (1998–2013) data from a C3 grassland Free Air CO2 Enrichment (FACE) experiment in Germany to study the combined effects of elevated [CO2] and extreme climatic events (ECEs) on aboveground biomass production. CO2 fertilization effect (CFE), which represents the promoted plant photosynthesis and water use efficiency under higher [CO2], was quantiffied by calculating the relative differences in biomass between the plots with [CO2] enrichment and the plots with ambient [CO2]. Down-regulated CFEs were found when ECEs occurred during the growing season, and the CFE decreases were statistically significant with p well below 0.05 (t-test). Of all the observed ECEs, the strongest CFE decreases were associated with intensive and prolonged heat waves. These findings suggest that more frequent ECEs in the future are likely to restrict the mitigatory effects of C3 grassland ecosystems, leading to an accelerated warming trend.     The VLTI/MIDI survey of massive young stellar objects - Sounding the inner regions around intermediate- and high-mass young stars using mid-infrared interferometry

    We aim to characterize the distribution and composition of circumstellar material around young massive stars, and to investigate exactly which physical structures in these objects are probed by long-baseline mid-infrared interferometric observations. We used the two-telescope interferometric instrument MIDI of the Very Large Telescope Interferometer of the European Southern Observatory to observe a sample of 24 intermediate- and high-mass young stellar objects in the N band (8-13 micron). We had successful fringe detections for 20 objects, and present spectrally-resolved correlated fluxes and visibility levels for projected baselines of up to 128 m. We fit the visibilities with geometric models to derive the sizes of the emitting regions, as well as the orientation and elongation of the circumstellar material. Fourteen objects in the sample show the 10 micron silicate feature in absorption in the total and correlated flux spectra. For 13 of these objects, we were able to fit the correlated flux spectra with a simple absorption model, allowing us to constrain the composition and absorptive properties of the circumstellar material. Nearly all of the massive young stellar objects observed show significant deviations from spherical symmetry at mid-infrared wavelengths. In general, the mid-infrared emission can trace both disks and outflows, and in many cases it may be difficult to disentangle these components on the basis of interferometric data alone, because of the sparse spatial frequency coverage normally provided by current long-baseline interferometers. For the majority of the objects in this sample, the absorption occurs on spatial scales larger than those probed by MIDI. Finally, the physical extent of the mid-infrared emission around these sources is correlated with the total luminosity, albeit with significant scatter.Comment: 36 pages, 22 figures.     Modern X-ray spectroscopy:XAS and XES in the laboratory

    X-ray spectroscopy is an important tool for scientific analysis. While the earliest demonstration experiments were realised in the laboratory, with the advent of synchrotron light sources most of the experiments shifted to large scale synchrotron facilities. In the recent past there is an increased interest to perform X-ray experiments also with in-house laboratory sources, to simplify access to X-ray absorption and X-ray emission spectroscopy, in particular for routine measurements. Here we summarise the recent developments and comment on the most representative example experiments in the field of in-house laboratory X-ray spectroscopy. We first give an introduction and some historic background on X-ray spectroscopy. This is followed by an overview of the detection techniques used for X-ray absorption and X-ray emission measurements. A short paragraph also puts related high energy resolution and resonant techniques into context, though they are not yet feasible in the laboratory. At the end of this section the opportunities using wavelength dispersive X-ray spectroscopy in the laboratory are discussed. Then we summarise the relevant details of the recent experimental laboratory setups split into two separate sections, one for the recent von Hamos setups, and one for the recent Johann/Johansson type setups. Following that, focussing on chemistry and catalysis, we then summarise some of the notable X-ray absorption and X-ray emission experiments and the results accomplished with in-house setups. In a third part we then discuss some applications of laboratory X-ray spectroscopy with a particular focus on chemistry and catalysis.</p

    Taurolidine reduces the tumor stimulating cytokine interleukin-1beta in patients with resectable gastrointestinal cancer: a multicentre prospective randomized trial

    Background The effect of additional treatment strategies with antineoplastic agents on intraperitoneal tumor stimulating interleukin levels are unclear. Taurolidine and Povidone-iodine have been mainly used for abdominal lavage in Germany and Europe. Methods In the settings of a multicentre (three University Hospitals) prospective randomized controlled trial 120 patients were randomly allocated to receive either 0.5% taurolidine/2,500 IU heparin (TRD) or 0.25% povidone-iodine (control) intraperitoneally for resectable colorectal, gastric or pancreatic cancers. Due to the fact that IL-1beta (produced by macrophages) is preoperatively indifferent in various gastrointestinal cancer types our major outcome criterion was the perioperative (overall) level of IL-1beta in peritoneal fluid. Results Cytokine values were significantly lower after TRD lavage for IL-1beta, IL-6, and IL-10. Perioperative complications did not differ. The median follow-up was 50.0 months. The overall mortality rate (28 vs. 25, p = 0.36), the cancer-related death rate (17 vs. 19, p = .2), the local recurrence rate (7 vs. 12, p = .16), the distant metastasis rate (13 vs. 18, p = 0.2) as well as the time to relapse were not statistically significant different. Conclusion Reduced cytokine levels might explain a short term antitumorigenic intraperitoneal effect of TRD. But, this study analyzed different types of cancer. Therefore, we set up a multicentre randomized trial in patients undergoing curative colorectal cancer resection. Trial registration : ISRCTN6647853

    The collateral network concept: Remodeling of the arterial collateral network after experimental segmental artery sacrifice

    ObjectiveA comprehensive strategy to prevent paraplegia after open surgical or endovascular repair of thoracoabdominal aortic aneurysms requires a thorough understanding of the response of the collateral network to extensive segmental artery sacrifice.MethodsTen Yorkshire pigs underwent perfusion with a low-viscosity acrylic resin. With the use of cardiopulmonary bypass, 2 animals each were perfused in the native state and immediately, 6 hours, 24 hours, and 5 days after sacrifice of all segmental arteries (T4–L5). After digestion of surrounding tissue, the vascular cast of the collateral network underwent analysis of arterial and arteriolar diameters and the density and spatial orientation of the vasculature using light and scanning electron microscopy.ResultsWithin 24 hours, the diameter of the anterior spinal artery had increased significantly, and within 5 days the anterior spinal artery and the epidural arterial network had enlarged in diameter by 80% to 100% (P < .0001). By 5 days, the density of the intramuscular paraspinous vessels had increased (P < .0001), a shift of size distribution from small to larger arterioles was seen (P = .0002), and a significant realignment of arterioles parallel to the spinal cord had occurred (P = .0005).ConclusionsWithin 5 days after segmental artery occlusion, profound anatomic alterations in the intraspinal and paraspinous arteries and arterioles occurred, providing the anatomic substrate for preservation of spinal cord blood flow via collateral pathways

    Incidence and Outcome of Progressive Multifocal Leukoencephalopathy over 20 Years of the Swiss HIV Cohort Study

    Background. We investigated the incidence and outcome of progressive multifocal leukoencephalopathy (PML) in human immunodeficiency virus (HIV)-infected individuals before and after the introduction of combination antiretroviral therapy (cART) in 1996. Methods. From 1988 through 2007, 226 cases of PML were reported to the Swiss HIV Cohort Study. By chart review, we confirmed 186 cases and recorded all-cause and PML-attributable mortality. For the survival analysis, 25 patients with postmortem diagnosis and 2 without CD4+ T cell counts were excluded, leaving a total of 159 patients (89 before 1996 and 70 during 1996-2007). Results. The incidence rate of PML decreased from 0.24 cases per 100 patient-years (PY; 95% confidence interval [CI], 0.20-0.29 cases per 100 PY) before 1996 to 0.06 cases per 100 PY (95% CI, 0.04-0.10 cases per 100 PY) from 1996 onward. Patients who received a diagnosis before 1996 had a higher frequency of prior acquired immunodeficiency syndrome-defining conditions (P=.007) but similar CD4+ T cell counts (60 vs. 71 cells/µL; P=.25), compared with patients who received a diagnosis during 1996 or thereafter. The median time to PML-attributable death was 71 days (interquartile range, 44-140 days), compared with 90 days (interquartile range, 54-313 days) for all-cause mortality. The PML-attributable 1-year mortality rate decreased from 82.3 cases per 100 PY (95% CI, 58.8-115.1 cases per 100 PY) during the pre-cART era to 37.6 cases per 100 PY (95% CI, 23.4.-60.5 cases per 100 PY) during the cART era. In multivariate models, cART was the only factor associated with lower PML-attributable mortality (hazard ratio, 0.18; 95% CI, 0.07-0.50; P<.001), whereas all-cause mortality was associated with baseline CD4+ T cell count (hazard ratio per increase of 100 cells/µL, 0.52; 95% CI, 0.32-0.85; P=.010) and cART use (hazard ratio, 0.37; 95% CI, 0.19-0.75; P=.006). Conclusions. cART reduced the incidence and PML-attributable 1-year mortality, regardless of baseline CD4+ T cell count, whereas overall mortality was dependant on cART use and baseline CD4+ T cell coun

    Reference state, structure, regime shifts, and regulatory drivers in a coastal sea over the last century : The Central Baltic Sea case

    The occurrence of regime shifts in marine ecosystems has important implications for environmental legislation that requires setting reference levels and targets of quantitative restoration outcomes. The Baltic Sea ecosystem has undergone large changes in the 20(th) century related to anthropogenic pressures and climate variability, which have caused ecosystem reorganization. Here, we compiled historical information and identified relationships in our dataset using multivariate statistics and modeling across 31 biotic and abiotic variables from 1925 to 2005 in the Central Baltic Sea. We identified a series of ecosystem regime shifts in the 1930s, 1970s, and at the end of the 1980s/beginning of the 1990s. In the long term, the Central Baltic Sea showed a regime shift from a benthic to pelagic-dominated state. Historically, benthic components played a significant role in trophic transfer, while in the more recent productive system pelagic-benthic coupling was weak and pelagic components dominated. Our analysis shows that for the entire time period, productivity, climate, and hydrography mainly affected the functioning of the food web, whereas fishing became important more recently. Eutrophication had far-reaching direct and indirect impacts from a long-term perspective and changed not only the trophic state of the system but also affected higher trophic levels. Our study also suggests a switch in regulatory drivers from salinity to oxygen. The "reference ecosystem" identified in our analysis may guide the establishment of an ecosystem state baseline and threshold values for ecosystem state indicators of the Central Baltic Sea.Peer reviewe

    Activation of the tryptophan/serotonin pathway is associated with severity and predicts outcomes in pneumonia: results of a long-term cohort study

    As part of the immune defense during infection, an increase in enzyme activity of indoleamine 2,3-dioxygenase (IDO) leads to a breakdown of tryptophan to kynurenine. In previous animal studies, therapeutic antagonism of IDO resulted in reduced sepsis mortality. We investigated the prognostic ability of tryptophan, serotonin, kynurenine and IDO (represented by the ratio of kynurenine/tryptophan) to predict adverse clinical outcomes in patients with community-acquired pneumonia (CAP).; We measured tryptophan, serotonin and kynurenine on admission plasma samples from CAP patients included in a previous multicenter trial by liquid chromatography coupled with tandem mass spectrometry (LC-MS/MS). We studied their association with inflammation (C-reactive protein), infection (procalcitonin) and clinical outcome.; Mortality in the 268 included patients was 45% within 6 years of follow-up. IDO and kynurenine showed a strong positive correlation with markers of infection (procalcitonin) and inflammation (C-reactive protein) as well as sepsis and CAP severity scores. Tryptophan showed similar, but negative correlations. In a multivariate regression analysis adjusted for age and comorbidities, higher IDO activity and lower tryptophan levels were strongly associated with short-term adverse outcome defined as death and/or ICU admission within 30 days with adjusted odds ratios of 9.1 [95% confidence interval (CI) 1.4-59.5, p=0.021] and 0.11 (95% CI 0.02-0.70, p=0.021). Multivariate analysis did not reveal significant associations for kynurenine and serotonin.; In hospitalized CAP patients, higher IDO activity and lower tryptophan levels independently predicted disease severity and short-term adverse outcome. Whether therapeutic modulation of IDO has positive effects on outcome needs further investigation

    A new MR-SAD algorithm for the automatic building of protein models from low-resolution X-ray data and a poor starting model

    Determining macromolecular structures from X-ray data with resolution worse than 3 Å remains a challenge. Even if a related starting model is available, its incompleteness or its bias together with a low observation-to-parameter ratio can render the process unsuccessful or very time-consuming. Yet, many biologically important macromolecules, especially large macromolecular assemblies, membrane proteins and receptors, tend to provide crystals that diffract to low resolution. A new algorithm to tackle this problem is presented that uses a multivariate function to simultaneously exploit information from both an initial partial model and low-resolution single-wavelength anomalous diffraction data. The new approach has been used for six challenging structure determinations, including the crystal structures of membrane proteins and macromolecular complexes that have evaded experts using other methods, and large structures from a 3.0 Å resolution F1-ATPase data set and a 4.5 Å resolution SecYEG–SecA complex data set. All of the models were automatically built by the method to Rfree values of between 28.9 and 39.9% and were free from the initial model bias
