    Cryptosporidium Infection in Cats; Epidemiology and Cross Transmission Studies

    Cryptosporidium species are important gastrointestinal and respiratory coccidian parasites which affect a wide range of host species including man. The aim of the studies described in this thesis was to determine the prevalence of Cryptosporidium infection in cats in the Glasgow area. Staining and concentration techniques for detection of Cryptosporidium oocysts in cat faecal specimens were first evaluated and used in the subsequent prevalence study. Modified Ziehl-Neelsen (MZN) and auramine phenol (A-P) techniques were found to be suitable for the screening of cat faeces for Cryptosporidium oocysts, and a fluorescein isothiocyanate (FITC) labelled monoclonal antibody (MAb) was used for confirmation of the diagnosis. Formol-ether (F-E) sedimentation and sucrose flotation techniques were effective for the concentration of Cryptosporidium oocysts in cat faecal specimens. In the prevalence study, 26 out of 294 (8.8%) cats from Glasgow and the surrounding area, were positive for Cryptosporidium infection. Infection was more common in kittens than in adult cats, and in farm and feral than in domestic cats. However, 74% of the serum samples from cats in the same area were positive for specific anti-Cryptosporidium antibody. The prevalence of IgM and IgA isotypes was higher in sick than in healthy groups of cats. Experimental transmission studies using Cryptosporidium spp. isolated from a farm cat into lambs and mice suggested that farm cats may be infected with the same Cryptosporidium isolates infecting other animals and that infection may be transmitted between various animal species on farms leading to a wide spread of the parasite. This thesis concludes that Cryptosporidium infection is common in cats in the Glasgow area and that these animals should be regarded as a potential reservoir of infection for other animals and humans, in particular children and immunodeficient individuals

    South African societies have been characterized by the prolific incidence of illicit drug use in recent times.  The paper attempts to examine the legislations implemented on the use of illicit drugs in South Africa. The study adopts a review of the literature to identify and describe the most common illicit drugs used around South Africa communities, placing emphasis on the policies developed by the South African government in combating these situations. The research follows a critical   discussion on the issues associated with drug use, its causes, as well as its effects on humans and the environment. The study recommends relevant initiatives to combat all intricacies associated with drug use within the country. This approach will be appropriate in facilitating a clear-cut   understanding of the possible remedies to quench the burning flame of illicit drug use across a broad range of South African communities

    Assessment of the Quality of coffee parchments in Southern Highlands of Tanzania: A case of Mbozi District in Songwe Region

    It’s unfortunate that even a single defective of a coffee bean can contribute to affect the receptive-lot of coffee. Coffee parchments or beans with high quality fetches high price in the world market. The study aimed at assessing the quality of coffee parchments along the post-harvest stages. Main focus was on improvement of post-harvest practices for coffee quality of coffee from farm level and to the processing under supervision of primary societies. The study involved coffee farmers in 6 Agricultural Marketing and Cooperatives Societies (AMCOS). Questionnaire, check list tools and physical observation were used for data collection. The result indicates that coffee farmers in Tanzania failed to fetch a good price from their produce due to low grades and classes of coffee parchments. Low grades and classes were observed to be attributed by application of poor practices along the coffee value chain. Our study suggests that, coffee stakeholders and the government should support the development of coffee sector along the value chain by up-scaling of good practices.     Genetic structure among the local chicken ecotypes of Tanzania based on microsatellite DNA typing

    A study was conducted to evaluate the genetic structure of local chicken ecotypes of Tanzania using 20 polymorphic microsatellite DNA markers. A standard PCR was followed by manual genotyping (6% native polyacrylamide gel visualized by silver staining). Phylogenetic analysis of 13 individuals from each of the nine ecotypes named Ching'wekwe, Kuchi, Mbeya, Morogoro-medium, N'zenzegere, Pemba, Singamagazi, Unguja and a White Leghorn breed was performed. The ten populations assorted into nine clusters with chickens from the same ecotype often clustering together. It is concluded that there is high genetic relatedness within indigenous chicken ecotype than between ecotypes. This offers a basic step towards rational decision-making on the modalities of selective breeding without compromising the existence of each unique genetic resource.Key words: Local-chicken, ecotypes, microsatellites, DNA, PCR.African Journal of Biotechnology Vol. 4 (8), pp. 768-77

    Implementing poultry vaccination and biosecurity at the village level in Tanzania: a social strategy to promote health in free-range poultry populations

    A social strategy was tested for implementing Newcastle disease (ND) vaccination and biosecurity improvements among free-ranging chicken at village level in Tanzania. In addition to training the local poultry vaccinators, data recorders and poultry-keepers, the strategy involved training and empowering leaders at the district, ward and village level. The trainings covered poultry health, management, and marketing of village chickens, with an emphasis on ND vaccination and improving biosecurity against avian influenza (AI), The study sites included villages in one ward in each of three each three districts (Iringa, Mtwara-Mikindani, and Mvomero) of mainland Tanzania. Ninety-six local leaders at district level and 101 leaders at ward levels were trained. In addition, 196 farmers (households) were trained, as well as 86 vaccinators and 26 data recorders. Data recorders were also trained as poultry first aid workers. ND vaccination was conducted by the vaccinators, supervised by their local leaders with technical assistance from veterinarians. A total of 158,343 village chickens were vaccinated in three rounds of vaccination three months apart. The training and empowerment of local leaders and local implementers was the key element for success as it fostered the feeling of local ownership of the program and prevented conflicts with other development activities within the villages. We conclude that most animal health programs will increase their odds of success by involving local leaders and by addressing the current challenges facing the farmers. Further assessment on the usefulness of this approach is needed

    Unveiling the Inhibitory Potentials of Peptidomimetic Azanitriles and Pyridyl Esters towards SARS-CoV-2Main Protease: A MolecularModelling Investigation

    The severe acute respiratory syndrome coronavirus-2 (SARS-CoV-2) is responsible for COVID-19, which was declared a global pandemic in March 2020 by the World Health Organization (WHO). Since SARS-CoV-2 main protease plays an essential role in the virus’s life cycle, the design of small drug molecules with lower molecular weight has been a promising development targeting its inhibition. Herein, we evaluated the novel peptidomimetic azatripeptide and azatetrapeptide nitriles against SARS-CoV-2 main protease. We employed molecular dynamics (MD) simulations to elucidate the selected compounds’ binding free energy profiles against SARS-CoV-2 and further unveil the residues responsible for the drug-binding properties. Compound 8 exhibited the highest binding free energy of 49.37 0.15 kcal/mol, followed by compound 7 (39.83 0.19 kcal/mol), while compound 17 showed the lowest binding free energy (23.54 0.19 kcal/mol). In addition, the absorption, distribution, metabolism, and excretion (ADME) assessment was performed and revealed that only compound 17 met the drug-likeness parameters and exhibited high pharmacokinetics to inhibit CYP1A2, CYP2C19, and CYP2C9 with better absorption potential and blood-brain barrier permeability (BBB) index. The additional intermolecular evaluations suggested compound 8 as a promising drug candidate for inhibiting SARS-CoV-2 Mpro. The substitution of isopropane in compound 7 with an aromatic benzene ring in compound 8 significantly enhanced the drug’s ability to bind better at the active site of the SARS-CoV-2 Mpro

    Exploring the risk factors of child malnutrition in Sub-Sahara Africa: a scoping review

    Background: Malnutrition prevents children from reaching their full physical and mental potential. Health and physical consequences of prolonged states of malnourishment among children are: delay in their physical growth and motor development ; lower intellectual quotient (IQ), greater behavioural problems and deficient social skills; susceptibility to contracting diseases. According to the 2015 Millennium development goal (MDG) report, sub-Saharan Africa (SSA) accounts for one third of all undernourished children globally, highlighting that malnutrition still remains a major health concern for children under 5 years in the sub-region, thus buttressing the need for urgent intervention. Aim: The aim of this study is to explore the risk factors of child malnutrition in sub-Saharan Africa through a scoping review. Methods: The scoping review was conducted using the following specific subject databases: EBSCOhost, google scholar, Pub med, demographic research and research gate. Attention was paid to keywords during navigation to ensure consistency of searches in each database. Two limiters were applied in all five databases. These included the use of the English language and articles published on child malnutrition in sub Saharan Africa. Results: The researchers identified eight themes for inclusion in the findings. The themes fell into four major categories being maternal related, family related, child related as well as context related factors. These themes reflect factors associated with child malnutrition. Conclusion: This scop-ing review revealed that there are quite a number of risk factors that lead to child malnutrition. Therefore, there is an urgency for strategic interventions aimed at improving child nutrition through female education if the 2030 end malnutrition SDG 2.2 are to be achieved

    Mitochondrial phylogeography and population structure of the cattle tick Rhipicephalus appendiculatus in the African Great Lakes region

    Abstract Background The ixodid tick Rhipicephalus appendiculatus is the main vector of Theileria parva, wich causes the highly fatal cattle disease East Coast fever (ECF) in sub-Saharan Africa. Rhipicephalus appendiculatus populations differ in their ecology, diapause behaviour and vector competence. Thus, their expansion in new areas may change the genetic structure and consequently affect the vector-pathogen system and disease outcomes. In this study we investigated the genetic distribution of R. appendiculatus across agro-ecological zones (AEZs) in the African Great Lakes region to better understand the epidemiology of ECF and elucidate R. appendiculatus evolutionary history and biogeographical colonization in Africa. Methods Sequencing was performed on two mitochondrial genes (cox1 and 12S rRNA) of 218 ticks collected from cattle across six AEZs along an altitudinal gradient in the Democratic Republic of Congo, Rwanda, Burundi and Tanzania. Phylogenetic relationships between tick populations were determined and evolutionary population dynamics models were assessed by mismach distribution. Results Population genetic analysis yielded 22 cox1 and 9 12S haplotypes in a total of 209 and 126 nucleotide sequences, respectively. Phylogenetic algorithms grouped these haplotypes for both genes into two major clades (lineages A and B). We observed significant genetic variation segregating the two lineages and low structure among populations with high degree of migration. The observed high gene flow indicates population admixture between AEZs. However, reduced number of migrants was observed between lowlands and highlands. Mismatch analysis detected a signature of rapid demographic and range expansion of lineage A. The star-like pattern of isolated and published haplotypes indicates that the two lineages evolve independently and have been subjected to expansion across Africa. Conclusions Two sympatric R. appendiculatus lineages occur in the Great Lakes region. Lineage A, the most diverse and ubiquitous, has experienced rapid population growth and range expansion in all AEZs probably through cattle movement, whereas lineage B, the less abundant, has probably established a founder population from recent colonization events and its occurrence decreases with altitude. These two lineages are sympatric in central and eastern Africa and allopatric in southern Africa. The observed colonization pattern may strongly affect the transmission system and may explain ECF endemic instability in the tick distribution fringes

    To beat or not to beat a tick: comparison of DNA extraction methods for ticks ( Ixodes scapularis

    Background. Blacklegged ticks (Ixodes scapularis) are important disease vectors in the United States, known to transmit a variety of pathogens to humans, including bacteria, protozoa, and viruses. Their importance as a disease vector necessitates reliable and comparable methods for extracting microbial DNA from ticks. Furthermore, to explore the population genetics or genomics of this tick, appropriate DNA extraction techniques are needed for both the vector and its microbes. Although a few studies have investigated different methods of DNA isolation from ticks, they are limited in the number and types of DNA extraction and lack species-specific quantification of DNA yield.Methods. Here we determined the most efficient and consistent method of DNA extraction from two different developmental stages of I. scapularis—nymph and adult—that are the most important for disease transmission. We used various methods of physical disruption of the hard, chitinous exoskeleton, as well as commercial and non-commercial DNA isolation kits. To gauge the effectiveness of these methods, we quantified the DNA yield and confirmed the DNA quality via PCR of both tick and microbial genetic material.Results. DNA extraction using the Thermo GeneJET Genomic DNA Purification Kit resulted in the highest DNA yields and the most consistent PCR amplification when combined with either cutting or bead beating with select matrices across life stages. DNA isolation methods using ammonium hydroxide as well as the MoBio PowerSoil kit also produced strong and successful PCR amplification, but only for females.Discussion. We contrasted a variety of readily available methods of DNA extraction from single individual blacklegged ticks and presented the results through a quantitative and qualitative assessment
