    Quantification of phosphorus loss from soil to water.

    End of Project ReportThe methods, results and discussion of the project are in five separate sections, 4.1) Phosphorus (P) export from agricultural grassland with overland flow and drainage water (Johnstown Castle); 4.2) Phosphorus export from farm in Dripsey catchment, Co. Cork (NMP); 4.3) Hydrometeorological aspects of farm in Dripsey Catchment (NMP); 4.4) Phosphorus desorption from Irish soils; 4.5) National phosphorus model. Most of the field and laboratory studies were carried out at Johnstown Castle, at UCC and the field site in the Dripsey catchment. The main aim of the project was to quantify the loss of P from soil to water where point source contributions from farmyards were not high. This involved the construction of hydrologically isolated field sites where the quantity of overland flow and the P concentrations for different runoff events from the fields could be measured. In addition, 90 soil samples representative of Irish soils were collected and analysed for the different factors influencing soil adsorption and desorption of P. These results, in addition to catchment data, were used as a first attempt at developing a model that could be used to help predict P loss from soil to water at a catchment scale. The study in the Dripsey was on a farm where water flow and P levels at two points in a stream were measured. The hydrometeorology at this site was also studied. At Johnstown Castle, three overland flow sites, of the order of one hectare each, and one subsurface flow site were studied for P loss to water.Environmental Protection Agency

    Trans and gender diverse young people's attitudes towards game-based digital mental health interventions: A qualitative investigation

    Background Trans and gender diverse (TGD) young people are at high risk for mental health difficulties. Previous research has shown that three in four TGD young people have been diagnosed with an anxiety disorder and/or depression and almost one in two have attempted suicide. TGD young people experience barriers to traditional mental health services, commonly faced with inexperienced providers and discrimination. Video and computer games, as well as online spaces, are sources of resilience for TGD young people. Digital mental health interventions are a feasible, but understudied, approach to consider for this population. Methods N = 14 TGD young people aged 11–18 years were recruited to take part in focus groups as part of a multistage project. The focus groups were transcribed verbatim and analysed using a general inductive thematic analysis approach. This paper reports on their attitudes towards digital games and game-based digital mental health interventions. Discussion Aspects of game-based digital mental health interventions should be inclusive of diverse genders and sexuality, moderated appropriately and include content such as storylines and characters who are of diverse sexualities and/or gender. Participants were of the opinion that games should be preventative rather than treatment-focused; however, considering the high rate of mental health difficulties in this population, interventions should ideally address symptoms along the continuum from sub-clinical to clinical

    Targeting HOX-PBX interactions causes death in oral potentially malignant and squamous carcinoma cells but not normal oral keratinocytes

    YesBackground: High HOX gene expression has been described in many cancers, including oral squamous cell carcinoma and the functional roles of these genes are gradually being understood. The pattern of overexpression suggests that inhibition may be useful therapeutically. Inhibition of HOX protein binding to PBX cofactors by the use of synthetic peptides, such as HXR9, results in apoptosis in multiple cancers. Methods: Activity of the HOX-PBX inhibiting peptide HXR9 was tested in immortalised normal oral (NOK), potentially-malignant (PMOL) and squamous cell carcinoma (OSCC) cells, compared to the inactive peptide CXR9. Cytotoxicity was assessed by LDH assay. Expression of PBX1/2 and c-Fos was assessed by qPCR and western blotting. Apoptosis was assessed by Annexin-V assay. Results: PMOL and OSCC cells expressed PBX1/2. HOX-PBX inhibition by HXR9 caused death of PMOL and OSCC cells, but not NOKs. HXR9 treatment resulted in apoptosis and increased expression of c-Fos in some cells, whereas CXR9 did not. A correlation was observed between HOX expression and resistance to HXR9.     Modal scattering at an impedance transition in a lined flow duct

    An explicit Wiener-Hopf solution is derived to describe the scattering of duct modes at a hard-soft wall impedance transition in a circular duct with uniform mean flow. Specifically, we have a circular duct r = 1,-8 <x <8 with mean flow Mach number M > 0 and a hard wall along x <0 and a wall of impedance Z along x > 0. A minimum edge condition at x = 0 requires a continuous wall streamline r = 1 + h(x, t ), no more singular than h = O(x1/2) for x ¿ 0. A mode, incident from x <0, scatters at x = 0 into a series of reflected modes and a series of transmitted modes. Of particular interest is the role of a possible instability along the lined wall in combination with the edge singularity. If one of the "upstream" running modes is to be interpreted as a downstream-running instability, we have an extra degree of freedom in the Wiener-Hopf analysis that can be resolved by application of some form of Kutta condition at x = 0, for example a more stringent edge condition where h = O(x3/2) at the downstream side. The question of the instability requires an investigation of the modes in the complex frequency plane and therefore depends on the chosen impedance model, since Z = Z(¿) is essentially frequency dependent. The usual causality condition by Briggs and Bers appears to be not applicable here because it requires a temporal growth rate bounded for all real axial wave numbers. The alternative Crighton-Leppington criterion, however, is applicable and confirms that the suspected mode is usually unstable. In general, the effect of this Kutta condition is significant, but it is particularly large for the plane wave at low frequencies and should therefore be easily measurable. For ¿ ¿ 0, the modulus tends to |R001| ¿ (1 + M)/(1 - M) without and to 1 with Kutta condition, while the end correction tends to8without and to a finite value with Kutta condition. This is exactly the same behaviour as found for reflection at a pipe exit with flow, irrespective if this is uniform or jet flow

    Forest processes from stands to landscapes: exploring model forecast uncertainties using cross-scale model comparison

    Forest management practices conducted primarily at the stand scale result in simplified forests with regeneration problems and low structural and biological diversity. Landscape models have been used to help design management strategies to address these problems. However, there remains a great deal of uncertainty that the actual management practices result in the desired sustainable landscape structure. To investigate our ability to meet sustainable forest management goals across scales, we assessed how two models of forest dynamics, a scaled-up individual-tree model and a landscape model, simulate forest dynamics under three types of harvesting regimes: clearcut, gap, and uniform thinning. Althougth 50– 100 year forecasts predicted average successional patterns that differed by less than 20% between models, understory dynamics of the landscape model were simplified relative to the scaled-up tree model, whereas successional patterns of the scaled-up tree model deviated from empirical studies on the driest and wettest landtypes. The scale dependencies of both models revealed important weaknesses when the models were used alone; however, when used together, they could provide a heuristic method that could improve our ability to design sustainable forest management practices

    In vivo x-ray imaging reveals improved airway surface hydration after a therapy designed for cystic fibrosis

    Sufficient airway surface liquid (ASL) depth is critical to ensure that the mucociliary transport system can effectively clear inhaled pathogens from the lungs. In those with cystic fibrosis (CF), the CF transmembrane conductance regulator gene defect decreases airway surface hydration (1, 2), compromising lung defense mechanisms, resulting in progressive lung infection and early death (3). Aneffective CF respiratory therapy should increase the ASL depth, restoring clearance mechanisms, but it is difficult to measure this depth in vivo. Current clinical health measures used for CF drug development studies include lung function testing and structural lung imaging (i.e., computed tomography), both indirect and delayed measures of airway hydration. Although the ASL depth has been measured by confocal microscopy in epithelial cultures(1, 4) and histologically in fragments of trachea (5), these technologies are not feasible in vivo

    Quantification of muco-obstructive lung disease variability in mice via laboratory X-ray velocimetry

    To effectively diagnose, monitor and treat respiratory disease clinicians should be able to accurately assess the spatial distribution of airflow across the fine structure of lung. This capability would enable any decline or improvement in health to be located and measured, allowing improved treatment options to be designed. Current lung function assessment methods have many limitations, including the inability to accurately localise the origin of global changes within the lung. However, X-ray velocimetry (XV) has recently been demonstrated to be a sophisticated and non-invasive lung function measurement tool that is able to display the full dynamics of airflow throughout the lung over the natural breathing cycle. In this study we present two developments in XV analysis. Firstly, we show the ability of laboratory-based XV to detect the patchy nature of cystic fibrosis (CF)-like disease in β-ENaC mice. Secondly, we present a technique for numerical quantification of CF-like disease in mice that can delineate between two major modes of disease symptoms. We propose this analytical model as a simple, easy-to-interpret approach, and one capable of being readily applied to large quantities of data generated in XV imaging. Together these advances show the power of XV for assessing local airflow changes.     Aquatic food security:insights into challenges and solutions from an analysis of interactions between fisheries, aquaculture, food safety, human health, fish and human welfare, economy and environment

    Fisheries and aquaculture production, imports, exports and equitability of distribution determine the supply of aquatic food to people. Aquatic food security is achieved when a food supply is sufficient, safe, sustainable, shockproof and sound: sufficient, to meet needs and preferences of people; safe, to provide nutritional benefit while posing minimal health risks; sustainable, to provide food now and for future generations; shock-proof, to provide resilience to shocks in production systems and supply chains; and sound, to meet legal and ethical standards for welfare of animals, people and environment. Here, we present an integrated assessment of these elements of the aquatic food system in the United Kingdom, a system linked to dynamic global networks of producers, processors and markets. Our assessment addresses sufficiency of supply from aquaculture, fisheries and trade; safety of supply given biological, chemical and radiation hazards; social, economic and environmental sustainability of production systems and supply chains; system resilience to social, economic and environmental shocks; welfare of fish, people and environment; and the authenticity of food. Conventionally, these aspects of the food system are not assessed collectively, so information supporting our assessment is widely dispersed. Our assessment reveals trade-offs and challenges in the food system that are easily overlooked in sectoral analyses of fisheries, aquaculture, health, medicine, human and fish welfare, safety and environment. We highlight potential benefits of an integrated, systematic and ongoing process to assess security of the aquatic food system and to predict impacts of social, economic and environmental change on food supply and demand

    Molybdenum blue nano-rings: an effective catalyst for the partial oxidation of cyclohexane

    Molybdenum blue (MB), a multivalent molybdenum oxide with a nano-ring morphology is well-known in analytical chemistry but, to date it has been largely ignored in other applications. In the present work, MB has been characterized by STEM-HAADF imaging for the first time, showing the nano-ring morphology of this complex molybdenum oxide and the ordered super-molecular framework crystals that can result from the self-assembly of these MB nano-ring units. The potential of MB as an oxidation catalyst has also been investigated, where it is shown to have excellent catalytic activity and stability in the selective oxidation of cyclohexane to cyclohexanol and cyclohexanone which are important intermediates in the production of nylon
