    Radio-echo sounding and waveform modeling reveal abundant marine ice in former rifts and basal crevasses within Crary Ice Rise, Antarctica

    Crary Ice Rise formed after the Ross Ice Shelf re-grounded ~1 kyr BP. We present new ice-penetrating radar data from two systems operating at center frequencies of 7 and 750 MHz that confirm the ice rise is composed of a former ice shelf buried by subsequent accumulation. Stacks of englacial diffraction hyperbolas are present almost everywhere across the central ice rise and extend up to ~350 m above the bed. In many cases, bed reflections beneath the diffraction hyperbolas are obscured for distances up to 1 km. Waveform modeling indicates that the diffraction hyperbolas are likely caused by marine ice deposits in former basal crevasses and rifts. The in-filling of rifts and basal crevasses may have strengthened the connection between the ice rise and the surrounding ice shelf, which could have influenced local and regional ice dynamics. Three internal reflection horizons mark the upper limit of disturbed ice and diffraction hyperbolas in different sections of the ice rise, indicating at least three stages of flow stabilization across the ice rise. A surface lineation visible in MODIS imagery corresponds spatially to deepening and strong deformation of these layers, consistent with the characteristics of former grounding lines observed elsewhere in Antarctica

    Solution structure of a bacterial microcompartment targeting peptide and its application in the construction of an ethanol bioreactor

    Targeting of proteins to bacterial microcompartments (BMCs) is mediated by an 18-amino-acid peptide sequence. Herein, we report the solution structure of the N-terminal targeting peptide (P18) of PduP, the aldehyde dehydrogenase associated with the 1,2-propanediol utilization metabolosome from Citrobacter freundii. The solution structure reveals the peptide to have a well-defined helical conformation along its whole length. Saturation transfer difference and transferred NOE NMR has highlighted the observed interaction surface on the peptide with its main interacting shell protein, PduK. By tagging both a pyruvate decarboxylase and an alcohol dehydrogenase with targeting peptides, it has been possible to direct these enzymes to empty BMCs in vivo and to generate an ethanol bioreactor. Not only are the purified, redesigned BMCs able to transform pyruvate into ethanol efficiently, but the strains containing the modified BMCs produce elevated levels of alcohol

    Beyond clinical trials: extending the role of the clinical research nurse into social care and homeless research

    Aim: Clinical research nurses work at the fulcrum of clinical trials with clearly defined roles and responsibilities. In England, the National Institute for Health Research (the main funder of health research) has broadened its scope to encompass social care research. The expectation is that clinical research nurses will expand their skill set to support these new studies, many of which will employ qualitative and mixed methods. This discussion paper explores the challenges of facilitating this clinical academic workforce development through a case study of a homeless health and social care research project. This was one of the first studies to engage clinical research nurses in this new and expanded role. Background: Much of what is known about the research nurse workforce has been generated through studies of clinical trials in oncology. The ‘caring-recruiting’ dichotomy has been used as a heuristic device for identifying workforce issues that can impact on study delivery such as how intense pressure to recruit study participants leads to low job satisfaction. Design: This case study reflects on the authors' experiences of employing a clinical research nurse in a social care research project concerned with the discharge of homeless people from hospital. The ‘caring-recruiting’ dichotomy is used to generate new information about the relationship between workforce development and the successful delivery of social care research. Conclusion: The case study illuminates how social care research can generate different pressures and ethical challenges for research nurses. The time and skill it took to recruit study participants identified as ‘hard to reach’ was suggestive of the need to move beyond performance measures that prioritise recruitment metrics. The need for different types of staff supervision and training was also warranted as supporting study participants who were homeless was often distressing, leading to professional boundary issues. Relevance to workforce development: This study highlights that performance management, training and supervisory arrangements must be tailored to the characteristics of each new study coming onto the portfolio to ensure research nurses are fully supported in this new and expanded role

    Broadcast-enhanced key predistribution schemes

    We present a formalisation of a category of schemes that we refer to as broadcast-enhanced key predistribution schemes (BEKPSs). These schemes are suitable for networks with access to a trusted base station and an authenticated broadcast channel. We demonstrate that the access to these extra resources allows for the creation of BEKPSs with advantages over key predistribution schemes such as flexibility and more efficient revocation. There are many possible ways to implement BEKPSs, and we propose a framework for describing and analysing them. In their paper “From Key Predistribution to Key Redistribution,” Cichoń et al. [2010] propose a scheme for “redistributing” keys to a wireless sensor network using a broadcast channel after an initial key predistribution. We classify this as a BEKPS and analyse it in that context. We provide simpler proofs of some results from their paper, give a precise analysis of the resilience of their scheme, and discuss possible modifications. We then study two scenarios where BEKPSs may be particularly desirable and propose a suitable family of BEKPSs for each case. We demonstrate that they are practical and efficient to implement, and our analysis shows their effectiveness in achieving suitable trade-offs between the conflicting priorities in resource-constrained networks

    Arrival of Paleo-Indians to the Southern Cone of South America: New Clues from Mitogenomes

    With analyses of entire mitogenomes, studies of Native American mitochondrial DNA (mtDNA) variation have entered the final phase of phylogenetic refinement: the dissection of the founding haplogroups into clades that arose in America during and after human arrival and spread. Ages and geographic distributions of these clades could provide novel clues on the colonization processes of the different regions of the double continent. As for the Southern Cone of South America, this approach has recently allowed the identification of two local clades (D1g and D1j) whose age estimates agree with the dating of the earliest archaeological sites in South America, indicating that Paleo-Indians might have reached that region from Beringia in less than 2000 years. In this study, we sequenced 46 mitogenomes belonging to two additional clades, termed B2i2 (former B2l) and C1b13, which were recently identified on the basis of mtDNA control-region data and whose geographical distributions appear to be restricted to Chile and Argentina. We confirm that their mutational motifs most likely arose in the Southern Cone region. However, the age estimate for B2i2 and C1b13 (11–13,000 years) appears to be younger than those of other local clades. The difference could reflect the different evolutionary origins of the distinct South American-specific sub-haplogroups, with some being already present, at different times and locations, at the very front of the expansion wave in South America, and others originating later in situ, when the tribalization process had already begun. A delayed origin of a few thousand years in one of the locally derived populations, possibly in the central part of Chile, would have limited the geographical and ethnic diffusion of B2i2 and explain the present-day occurrence that appears to be mainly confined to the Tehuelche and Araucanian-speaking grou

    Investigation of the Young’s modulus and thermal expansion of amorphous titania-doped tantala films

    The current generation of advanced gravitational wave detectors utilize titania-doped tantala/silica multilayer stacks for their mirror coatings. The properties of the low-refractive-index silica are well known; however, in the absence of detailed direct measurements, the material parameters of Young’s modulus and coefficient of thermal expansion (CTE) of the high refractive index material, titania-doped tantala, have been assumed to be equal to values measured for pure tantala coatings. In order to ascertain the true values necessary for thermal noise calculations, we have undertaken measurements of Young’s modulus and CTE through the use of nanoindentation and thermal-bending measurements. The measurements were designed to assess the effects of titania-doping concentration and post-deposition heat-treatment on the measured values in order to evaluate the possibility of optimizing material parameters to further improve thermal noise in the detector. Young’s modulus measurements on pure tantala and 25% and 55% titania-doped tantala show a wide range of values, from 132 to 177 GPa, which are dependent on both titania concentration and heat-treatment. Measurements of CTE give values of (3.9±0.1)×10^(−6)  K^(−1) and (4.9±0.3)×10^(−6)  K^(−1) for 25% and 55% titania-doped tantala, respectively, without dependence on post-deposition heat-treatment

    Physical exercise has been identified as a major non-pharmacological agent, assuming a protective role in the treatment of chronic non-communicable diseases, especially in hypertension and diabetes mellitus. The levels of physical activity as well as its intensity are of great impact for the full benefits to come of this practice. We know that the level of physical activity is a relevant factor for maintaining good health, however, little attention has been paid to what is established in relation to high-intensity daily exercise adjusted to cardiac damage or adaptation. The objective was to reflect on the prescription of physical activity in the treatment of hypertensive and diabetic patients and how eccentric exercise is related to the cardiomyocyte tissue in the damage process. We observed that the highest level of physical activity was associated with better quality of life, and that active patients have an 80% chance of survival when compared to sedentary ones. Thus, despite the potential benefits from the metabolic point of view, public health efforts should also focus on maintaining and protocols of physical activity levels since our study showed that intense eccentric physical activity had harmful effects at the cardiac level
