    Measuring the ringdown scalar polarization of gravitational waves in Einstein scalar Gauss-Bonnet gravity

    We model the scalar waves produced during the ringdown stage of binary black hole coalescence in Einstein scalar Gauss-Bonnet (EsGB) gravity, using numerical relativity simulations of the theory in the decoupling limit. Through a conformal coupling of the scalar field to the metric in the matter-field action, we show that the gravitational waves in this theory can have a scalar polarization. We model the scalar quasi-normal modes of the ringdown signal in EsGB gravity, and quantify the extent to which current and future gravitational wave detectors could observe the spectrum of scalar radiation emitted during the ringdown phase of binary black hole coalescence. We find that within the limits of the theory's coupling parameters set by current theoretical and observational constraints, the scalar ringdown signal from black hole remnants in the 101−103 M⊙10^1 - 10^3 \, M_{\odot} mass range is expected to be well below the detectability threshold with the current network of gravitational-wave detectors (LIGO-Virgo-KAGRA), but is potentially measurable with next-generation detectors such as the Einstein Telescope.Comment: 27 pages, 19 figures, to match published version in Phys.     The basic chemistry of exercise-induced DNA oxidation:oxidative damage, redox signalling and their interplay

    Acute exercise increases reactive oxygen and nitrogen species generation. This phenomenon is associated with two major outcomes: (1) redox signalling and (2) macromolecule damage. Mechanistic knowledge of how exercise-induced redox signalling and macromolecule damage are interlinked is limited. This review focuses on the interplay between exercise-induced redox signalling and DNA damage, using hydroxyl radical (·OH) and hydrogen peroxide (H2O2) as exemplars. It is postulated that the biological fate of H2O2 links the two processes and thus represents a bifurcation point between redox signalling and damage. Indeed, H2O2 can participate in two electron signalling reactions but its diffusion and chemical properties permit DNA oxidation following reaction with transition metals and ·OH generation. It is also considered that the sensing of DNA oxidation by repair proteins constitutes a non-canonical redox signalling mechanism. Further layers of interaction are provided by the redox regulation of DNA repair proteins and their capacity to modulate intracellular H2O2 levels. Overall, exercise-induced redox signalling and DNA damage may be interlinked to a greater extent than was previously thought but this requires further investigation

    Influence of vitamin C and vitamin E on redox signalling:implications for exercise adaptations

    The exogenous antioxidants vitamin C (ascorbate) and vitamin E (α-tocopherol) often blunt favourable cell signalling responses to exercise, suggesting that redox signalling contributes to exercise adaptations. Current theories posit that this antioxidant paradigm interferes with redox signalling by attenuating exercise-induced reactive oxygen species (ROS) and reactive nitrogen species (RNS) generation. The well-documented in vitro antioxidant actions of ascorbate and α-tocopherol and characterisation of the type and source of the ROS/RNS produced during exercise theoretically enables identification of the redox-dependent mechanism responsible for the blunting of favourable cell signalling responses to exercise. This review aimed to apply this reasoning to determine how the aforementioned antioxidants might attenuate exercise-induced ROS/RNS production. The principal outcomes of this analysis are (1) neither antioxidant is likely to attenuate nitric oxide signalling either directly (reaction with nitric oxide) or indirectly (reaction with derivatives, e.g. peroxynitrite) (2) neither antioxidant reacts appreciably with hydrogen peroxide, a key effector of redox signalling (3) ascorbate but not α-tocopherol has the capacity to attenuate exercise-induced superoxide generation and (4) alternate mechanisms, namely pro-oxidant side reactions and/or reduction of bioactive oxidised macromolecule adducts, are unlikely to interfere with exercise-induced redox signalling. Out of all the possibilities considered, ascorbate mediated suppression of superoxide generation with attendant implications for hydrogen peroxide signalling is arguably the most cogent explanation for blunting of favourable cell signalling responses to exercise. However, this mechanism is dependent on ascorbate accumulating at sites rich in NADPH oxidases, principal contributors to contraction mediated superoxide generation, and outcompeting nitric oxide and superoxide dismutase isoforms. The major conclusions of this review are: (1) direct evidence for interference of ascorbate and α-tocopherol with exercise-induced ROS/RNS production is lacking (2) theoretical analysis reveals that both antioxidants are unlikely to have a major impact on exercise-induced redox signalling and (3) it is worth considering alternate redox-independent mechanisms

    Trial of canakinumab, an IL-1β receptor antagonist, in patients with inclusion body myositis.

    Objective: To assess whether canakinumab, a monoclonal antibody against IL-1β approved for autoinflammatory diseases, is effective as target-specific therapy in patients with sporadic inclusion body myositis (sIBM). Methods: Because in sIBM IL-1β colocalizes with amyloid precursor protein and upregulates amyloid aggregates enhancing degeneration, targeting IL-1β with canakinumab may arrest disease progression. On this basis, 5 ambulatory patients with sIBM participated in an institutional review board--approved open-labeled study with 150 mg canakinumab [4 bimonthly, then monthly subcutaneous injections] for a mean period of 15.8 months. Patients were assessed bimonthly with a manual dynamometer in 12 proximal and distal muscles and with grip force (GF) in both hands. Total muscle strength (TMS) was expressed in kilograms. Efficacy was defined as \u3e15% increased strength after 12 months. Results: Patient 1 stopped at month 5 because of 23% loss in TMS and 32.35% in GF; patient 2 showed 37.1% increase in TMS and 13% in GF by month 9; patient 3 exhibited 26.7% reduction in TMS and 10% in GF at month 33; patient 4 showed 6.5% reduction in TMS and 1.6% in GF after 15 months, denoting relative stability; and patient 5 showed 30.4% loss in TMS and 20.8% in GF after 18 months. In patients 2 and 4, in whom 3-year longitudinal data were available, no effect on disease progression was noted. Conclusions: In this long-term, open-label study, canakinumab showed small, but not clinically appreciable, stabilizing benefits in 2 of 5 patients with sIBM over 1 year, was ineffective in 2 others, and might have worsened one.     Community Networks and Sustainability: a Survey of Perceptions, Practices, and Proposed Solutions

    Community network (CN) initiatives have been around for roughly two decades, evangelizing a distinctly different paradigm for building, maintaining, and sharing network infrastructure but also defending the basic human right to Internet access. Over this time they have evolved into a mosaic of systems that vary widely with respect to their network technologies, their offered services, their organizational structure, and the way they position themselves in the overall telecommunications’ ecosystem. Common to all these highly differentiated initiatives is the sustainability challenge. We approach sustainability as a broad term with an economical, political, and cultural context. We first review the different perceptions of the term. These vary both across and within the different types of stakeholders involved in CNs and are reflected in their motivation to join such initiatives. Then, we study the diverse approaches of CN operators towards the sustainability goal. Given the rich context of the term, these range all the way from mechanisms to fund their activities, to organizational structures and social activities serving as incentives for the engagement of their members. We iterate on incentive mechanisms that have been proposed and theoretically analyzed in the literature for CNs as well as tools and processes that have been actually implemented in them. Finally, we enumerate lessons that have been learned out of these two decades of CNs’ operation and discuss additional technological and regulatory issues that are key to their longer-term sustainability

    Towards a Digital Twin of Coronary Stenting: A Suitable and Validated Image-Based Approach for Mimicking Patient-Specific Coronary Arteries

    Considering the field of application involving stent deployment simulations, the exploitation of a digital twin of coronary stenting that can reliably mimic the patient-specific clinical reality could lead to improvements in individual treatments. A starting step to pursue this goal is the development of simple, but at the same time, robust and effective computational methods to obtain a good compromise between the accuracy of the description of physical phenomena and computational costs. Specifically, this work proposes an approach for the development of a patient-specific artery model to be used in stenting simulations. The finite element model was generated through a 3D reconstruction based on the clinical imaging (coronary Optical Coherence Tomography (OCT) and angiography) acquired on the pre-treatment patient. From a mechanical point of view, the coronary wall was described with a suitable phenomenological model, which is consistent with more complex constitutive approaches and accounts for the in vivo pressurization and axial pre-stretch. The effectiveness of this artery modeling method was tested by reproducing in silico the stenting procedures of two clinical cases and comparing the computational results with the in vivo lumen area of the stented vessel

    Beyond retail: New ways of classifying UK shopping and consumption spaces

    Early attempts to classify shopping activity often took a relatively simple approach, largely driven by the lack of reliable data beyond fascia name and retail outlet counts by centre. There seems to be a consensus amongst contemporary scholars, commercial research consultancies and retailers that more comprehensive classifications would generate better-informed debate on changes in the urban economic landscape, as well as providing the basis for a more effective comparison of retail centres across time and space, particularly given the availability of new data sources and techniques and in the context of the transformational changes presently affecting the retail sector. This paper seeks to demonstrate the interrelationship between supply and demand for retailing services by integrating newly available data sources within a rigorously specified classification methodology. This in turn provides new insight into the multidimensional and dynamic taxonomy of consumption spaces within Great Britain. First, such a contribution is significant in that it moves debate within the literature past simple linear scaling of retail centre function to a more nuanced understanding of multiple functional forms; and second, in that it provides a nationally comparative and dynamic framework through which the evolution of retail structures can be evaluated. Using non-hierarchical clustering techniques, the results are presented in the form of a two-tier classification with 5 distinctive ‘coarse’ clusters and 15 more detailed and nested sub-clusters. The paper concludes that more nuanced and dynamic classifications of this kind can help deliver more effective insights into changing role of retailing and consumer services in urban areas across space and through time and will have implications for a variety of stakeholders

    Historical variation and trends in storminess along the Portuguese South Coast

    This work investigates historical variation and trends in storm climate for the South Portugal region, using data from wave buoy measurements and from modelling, for the period 1952 to 2009. Several storm parameters (annual number of storms; annual number of days with storms; annual maximum and mean individual storm duration and annual 99.8th percentile of significant wave height) were used to analyse: (1) historical storminess trends; (2) storm parameter variability and relationships; and (3) historical storminess and its relationship to the North Atlantic Oscillation (NAO). No statistically significant linear increase or decrease was found in any of the storm parameters over the period of interest. The main pattern of storm characteristics and extreme wave heights is an oscillatory variability with intensity peaks every 7–8 yr, and the magnitude of recent variations is comparable with that of variations observed in the earlier parts of the record. In addition, the results reveal that the NAO index is able to explain only a small percentage of the variation in storm wave height, suggesting that more local factors may be of importance in controlling storminess in this region.info:eu-repo/semantics/publishedVersio
