The Production of Epistemic Culture and Agency during a First-Grade Engineering Design Unit in an Urban Emergent School
Primary school practices are often bound by traditions that perpetuate compliance and skills-based, decontextualized, rote memorization activities. These histories of practice, prevalent in schools serving mostly Black and Brown children, make it inordinately difficult for students to author themselves as knowledge builders (i.e., with epistemic agency), which is a form of injustice. Engineering is a potentially fertile context to support the creation of epistemic culture, whereby young students’ assets are recognized, named, and leveraged as they create and shape the group’s disciplinary knowledge. The authors investigated this potential. The primary research question was: How do first-grade students in an urban emergent school author themselves as epistemic agents during an engineering design unit? Using a social practice theory lens and ethnographic methods, the authors studied 29 days of a materials engineering unit focusing on the teacher’s epistemic commitments, implicit meanings of knowledge in classroom discourse, and practices that opened space for students’ epistemic agency. Data collection included fieldnotes and video of class activities and teacher and student interviews. Class discussions about the properties and uses of materials yielded a rich context for studying epistemic culture. The teacher’s epistemic commitments included an eschewing of disciplinary silos, recognizing the nonlinear nature of knowledge-building about engineering, and acknowledging children’s thinking as an asset for engineering knowledge production. Examples of students’ discursive moves demonstrating epistemic agency included: reminding others about the relevance of previous lessons to the current topic, mirroring the teacher’s instructional moves, claiming voice, space, time, and material resources for knowledge-building, translating one another’s ideas, and making unsolicited connections to their lives. Young children’s intellectual assets can too easily be overlooked in traditional learning contexts. This study demonstrates the affordances of responsive engineering instruction in recognizing and building on youths’ intellectual curiosity and enthusiasm for substantively contributing to the classroom’s knowledge-generating practices
Channeling your inner science warrior: the nature of teachers’ professional agency in high-needs schools
High-needs schools in the United States contend with a disparity in resource allocation, a higher rate of teacher and administrator turnover, an always narrowing curriculum due to increased accountability measures, and a heightened culture of surveillance. These realities shape what has happened and what seems possible for teachers, especially impacting their ability to integrate and enact science and engineering in elementary schools. Increased accountability pressures heighten the culture of surveillance, narrow the curriculum, and reduce teacher autonomy and professionalism. These realities impact science and engineering instruction by dictating how and what teachers teach. Science often gets pushed to the margins in favor of tested subjects and institutional demands constraining teachers’ practice. There are few, if any, accounts of teachers who exercise their professional agency to navigate these tensions productively. The purpose of this study is to raise teachers’ voices, their ideas, the dilemmas they face, and how they navigate those dilemmas with their professional agency. This study employs a narrative inquiry design to explore elementary teachers’ professional visions for science and engineering in high-needs schools, the dilemmas they encounter when working to enact science and engineering, and the nature of teachers’ STEM-linked professional agency as they creatively wrestle with and work through these dilemmas. Data include over 65 interviews and 18 STEM journey maps. Data analysis included coding teachers’ narratives using in vivo codes, construction of themes based on emergent patterns, and cross-narrative analysis. This analysis led to the description of elementary teachers’ professional visions for science and engineering in high-needs schools as ideal images of engagement, instructional and curricular connection, instruction that broaden student pathways, and teaching for altruistic reasons. As teachers worked to enact their professional visions, various sociocultural conditions of their schools and districts facilitate and constrained that work. When teachers experienced conflict between their professional visions and institutional ideals and demands, they encountered dilemmas focused on the roles of teachers and ideal curriculum and pedagogy. As teachers wrestled with dilemmas, they enacted STEM-linked professional agency. Moments of STEM-linked professional agency were visible in moments when teachers described science and engineering as both highly thinkable and doable. This led to the understanding of STEM-linked professional agency as a phenomenon within certain contexts of schooling when teachers created change within their workplace to align practice with vision in ways that were innovative and responsive
Psychosocial factors and health as determinants of quality of life in community-dwelling older adults.
PURPOSE: It is important to understand the determinants of differences in quality of life in old age and to include a wide range of possible predictors. The present study investigated the determinants of quality of life in two groups of older adults for whom there was an unusually informative set of possible predictor variables. METHOD: Participants were members of the Lothian Birth Cohorts of 1921 (n = 550) or 1936 (n = 1,091). Four facets of quality of life (QoL) and general QoL were measured using the WHOQOL-BREF. Possible determinants included personality traits, measured with the International Personality Item Pool (IPIP) scales; childhood and old age general cognitive ability, measured with the Moray House Test; minor psychological symptoms, measured with the Hospital Anxiety and Depression Scale (HADS); physical health, assessed by grip strength and cardiovascular disease history; and sociodemographic factors, assessed by interview. RESULTS: Linear regression analyses revealed that HADS depression had the greatest influence on quality of life. Personality traits, most notably Emotional Stability, also predicted quality of life to varying degrees, along with factors reflecting current life circumstances. There were differences between the two cohorts in the variables which predicted quality of life. There were different, conceptually relevant, contributions to the different QoL facets. CONCLUSIONS: Personality traits and minor depressive symptoms have an important influence on self-reported quality of life in old age. Quality of life may be influenced more by current than past circumstances, and this relationship may change with age
Histone H3.3 beyond cancer: Germline mutations in Histone 3 Family 3A and 3B cause a previously unidentified neurodegenerative disorder in 46 patients
Although somatic mutations in Histone 3.3 (H3.3) are well-studied drivers of oncogenesis, the role of germline mutations remains unreported. We analyze 46 patients bearing de novo germline mutations in histone 3 family 3A (H3F3A) or H3F3B with progressive neurologic dysfunction and congenital anomalies without malignancies. Molecular modeling of all 37 variants demonstrated clear disruptions in interactions with DNA, other histones, and histone chaperone proteins. Patient histone posttranslational modifications (PTMs) analysis revealed notably aberrant local PTM patterns distinct from the somatic lysine mutations that cause global PTM dysregulation. RNA sequencing on patient cells demonstrated up-regulated gene expression related to mitosis and cell division, and cellular assays confirmed an increased proliferative capacity. A zebrafish model showed craniofacial anomalies and a defect in Foxd3-derived glia. These data suggest that the mechanism of germline mutations are distinct from cancer-associated somatic histone mutations but may converge on control of cell proliferation
Adding 6 months of androgen deprivation therapy to postoperative radiotherapy for prostate cancer: a comparison of short-course versus no androgen deprivation therapy in the RADICALS-HD randomised controlled trial
Background
Previous evidence indicates that adjuvant, short-course androgen deprivation therapy (ADT) improves metastasis-free survival when given with primary radiotherapy for intermediate-risk and high-risk localised prostate cancer. However, the value of ADT with postoperative radiotherapy after radical prostatectomy is unclear.
Methods
RADICALS-HD was an international randomised controlled trial to test the efficacy of ADT used in combination with postoperative radiotherapy for prostate cancer. Key eligibility criteria were indication for radiotherapy after radical prostatectomy for prostate cancer, prostate-specific antigen less than 5 ng/mL, absence of metastatic disease, and written consent. Participants were randomly assigned (1:1) to radiotherapy alone (no ADT) or radiotherapy with 6 months of ADT (short-course ADT), using monthly subcutaneous gonadotropin-releasing hormone analogue injections, daily oral bicalutamide monotherapy 150 mg, or monthly subcutaneous degarelix. Randomisation was done centrally through minimisation with a random element, stratified by Gleason score, positive margins, radiotherapy timing, planned radiotherapy schedule, and planned type of ADT, in a computerised system. The allocated treatment was not masked. The primary outcome measure was metastasis-free survival, defined as distant metastasis arising from prostate cancer or death from any cause. Standard survival analysis methods were used, accounting for randomisation stratification factors. The trial had 80% power with two-sided α of 5% to detect an absolute increase in 10-year metastasis-free survival from 80% to 86% (hazard ratio [HR] 0·67). Analyses followed the intention-to-treat principle. The trial is registered with the ISRCTN registry, ISRCTN40814031, and ClinicalTrials.gov, NCT00541047.
Findings
Between Nov 22, 2007, and June 29, 2015, 1480 patients (median age 66 years [IQR 61–69]) were randomly assigned to receive no ADT (n=737) or short-course ADT (n=743) in addition to postoperative radiotherapy at 121 centres in Canada, Denmark, Ireland, and the UK. With a median follow-up of 9·0 years (IQR 7·1–10·1), metastasis-free survival events were reported for 268 participants (142 in the no ADT group and 126 in the short-course ADT group; HR 0·886 [95% CI 0·688–1·140], p=0·35). 10-year metastasis-free survival was 79·2% (95% CI 75·4–82·5) in the no ADT group and 80·4% (76·6–83·6) in the short-course ADT group. Toxicity of grade 3 or higher was reported for 121 (17%) of 737 participants in the no ADT group and 100 (14%) of 743 in the short-course ADT group (p=0·15), with no treatment-related deaths.
Interpretation
Metastatic disease is uncommon following postoperative bed radiotherapy after radical prostatectomy. Adding 6 months of ADT to this radiotherapy did not improve metastasis-free survival compared with no ADT. These findings do not support the use of short-course ADT with postoperative radiotherapy in this patient population
Duration of androgen deprivation therapy with postoperative radiotherapy for prostate cancer: a comparison of long-course versus short-course androgen deprivation therapy in the RADICALS-HD randomised trial
Background
Previous evidence supports androgen deprivation therapy (ADT) with primary radiotherapy as initial treatment for intermediate-risk and high-risk localised prostate cancer. However, the use and optimal duration of ADT with postoperative radiotherapy after radical prostatectomy remains uncertain.
Methods

Participants were randomly assigned (1:1) to add 6 months of ADT (short-course ADT) or 24 months of ADT (long-course ADT) to radiotherapy, using subcutaneous gonadotrophin-releasing hormone analogue (monthly in the short-course ADT group and 3-monthly in the long-course ADT group), daily oral bicalutamide monotherapy 150 mg, or monthly subcutaneous degarelix. Randomisation was done centrally through minimisation with a random element, stratified by Gleason score, positive margins, radiotherapy timing, planned radiotherapy schedule, and planned type of ADT, in a computerised system. Findings
Between Jan 30, 2008, and July 7, 2015, 1523 patients (median age 65 years, IQR 60–69) were randomly assigned to receive short-course ADT (n=761) or long-course ADT (n=762) in addition to postoperative radiotherapy at 138 centres in Canada, Denmark, Ireland, and the UK. With a median follow-up of 8·9 years (7·0–10·0), 313 metastasis-free survival events were reported overall (174 in the short-course ADT group and 139 in the long-course ADT group; HR 0·773 [95% CI 0·612–0·975]; p=0·029). 10-year metastasis-free survival was 71·9% (95% CI 67·6–75·7) in the short-course ADT group and 78·1% (74·2–81·5) in the long-course ADT group. Toxicity of grade 3 or higher was reported for 105 (14%) of 753 participants in the short-course ADT group and 142 (19%) of 757 participants in the long-course ADT group (p=0·025), with no treatment-related deaths.
Interpretation
Compared with adding 6 months of ADT, adding 24 months of ADT improved metastasis-free survival in people receiving postoperative radiotherapy. For individuals who can accept the additional duration of adverse effects, long-course ADT should be offered with postoperative radiotherapy.
