    Synthesis of heterocyclic compounds by radical electrochemical approach in environmentally friendly media

    Radical cyclisation is rapidly becoming an important method for the formation of cyclic systems. Hence, some electrochemical results obtained in the study of electroreductive intramolecular cyclisation of ethyl 2-bromo-3-(3,4-methylenedioxophenyl)-3-(propargyloxy)propanoate (1a), [1-bromo-2-methoxy-2-(prop-2’-ynyloxy) ethyl] benzene (1b) and 1-[2-bromo-2-phenyl-1-(prop-2’-ynyloxy)ethyl]-4-methoxybenzene (1c) promoted by (1,4,8,11-tetramethyl-1,4,8,11-tetraazacyclotetra-decane)nickel(I), [Ni(tmc)]+, electrogenerated at glassy carbon cathodes in ethanol, ethanol / water and microemulsions made with cationic and anionic surfactants are presented. The results obtained indicate that the reaction proceeds via cleavage of the carbon–bromine bond to form a radical intermediate that undergoes cyclization on the unsaturated C-C bond to afford the substituted tetrahydrofurans.Fundação para a Ciência e a Tecnologia (FCT

    Conference Report XVI International Symposium on Marine Natural Products | XI European Conference on Marine Natural Products, 1-5 September 2019, Peniche, Portugal.Seaweed have promising applications within food, cosmetic and health industries, which led to an increased interest in studying these organisms [1]. In several coastal areas, thousands of tons of macroalgae are cast on beaches and shorelines and it would be very interesting if this biomass could be managed, allowing the extraction of added-value compounds. In this context, polar extracts (methanol and ethanol) of a macroalgal beach cast sample mainly composed of “Lobophora variegate” were prepared and the anti-aging and antioxidant activities were evaluated. The preliminary results showed interesting results, and thus these crude extracts were then fractionated sequentially by their solubility in dichloromethane, acetone and ethyl acetate, resulting in 4 semi-pure fractions each, which were also tested. Fractions A1.1.1 and A1.2.3 were very good tyrosinase inhibitors (IC₅₀ = 37.87 and 24.01 μg/mL, respectively) and fractions A1.1.2 and A1.2.2 presented very good inhibition of elastase (IC₅₀ = 44.76 and 20.86 μg/mL, respectively). However, none of the fractions was active against collagenase.     Radical cyclisation continues to be a central methodology for the preparation of natural products containing heterocyclic rings. Hence, some electrochemical results obtained by cyclic voltammetry and controlled-potential electrolysis in the study of electroreductive intramolecular cyclisation of ethyl (2S, 3R)-2- bromo-3-propargyloxy-3-(2’,3’,4’,6’-tetra-O-acetyl-β-D-glucopyranosiloxy) propanoate (1) promoted by (1,4,8,11-tetramethyl-1,4,8,11-tetraazacyclotetra-decane)nickel(I), [Ni(tmc)]+, electrogenerated at glassy carbon cathodes in ethanol and ethanol/water mixtures containing tetraalkylammonium salts, are presented.     The electrochemical reductive cyclisation of unsaturated organic halides in the presence of Ni(II) complexes as catalysts was examined in aprotic solvents such as DMF and in protic solvents such as ethanol, butanol or ethanolewater mixtures.     Nursing work of nursing at a Day Hospital in João Pessoa in the perspective of the psychiatric reform

    This is the report of an investigation that was carried out in order to apprehend themes that explain nursing work at a day hospital in João Pessoa, State of Paraíba, in the perspective of the psychiatric reform. The authors used the referential of dialectical and historical materialism and work as an analytical category. The empirical material was analyzed through the technique of discourse analysis. The study revealed a coincident theme - to change the model of traditional psychiatric assistance - and, in this sense, the work of nursing incorporates, in addition to its formal, technically specialized quality, a political quality, thus indicating a new meaning to the nursing work process.Relatamos uma investigação para apreender temas que explicam o trabalho de enfermagem, no Hospital-Dia, na perspectiva da reforma psiquiátrica em João Pessoa-PB. Utilizamos o referencial do materialismo histórico e dialético e o trabalho como categoria analítica. O material empírico foi analisado pela técnica de análise do discurso. O estudo revelou um tema coincidente, mudar o modelo de assistência psiquiátrica tradicional, e, nesse sentido, o trabalho de enfermagem incorpora além da qualidade formal, técnica-especializada, uma qualidade política indicando um novo significado ao processo de trabalho de enfermagem.Relatamos una investigación para capturar temas que explican el trabajo de enfermería, en un Hospital de Día, en la perspectiva de la reforma psiquiátrica en João Pessoa-PB.     "A evidência mostra que os artrópodes constituem uma fracção importante da biodiversidade estrutural e funcional dos habitats terrestres (Kim, 1993). Por outro lado, só é possível conservar os processos ecológicos associados aos artrópodes promovendo a gestão correcta dos seus habitats. Para tal, é necessária uma correcta identificação e caracterização das suas comunidades de forma a promover a sua boa gestão.     2,3-Diphosphoglycerate and the Protective Effect of Pyruvate Kinase Deficiency against Malaria Infection—Exploring the Role of the Red Blood Cell Membrane

    Malaria remains a major world public health problem, contributing to poverty and inequality. It is urgent to find new efficacious tools with few adverse effects. Malaria has selected red blood cell (RBC) alterations linked to resistance against infection, and understanding the protective mechanisms involved may be useful for developing host-directed tools to control Plasmodium infection. Pyruvate kinase deficiency has been associated with resistance to malaria. Pyruvate kinase-deficient RBCs display an increased concentration of 2,3-diphosphoglycerate (2,3-DPG).We recently showed that 2,3-DPG impacts in vitro intraerythrocytic parasite growth, induces a shift of the metabolic profile of infected cells (iRBCs), making it closer to that of noninfected ones (niRBCs), and decreases the number of parasite progenies that invade new RBCs. As an increase of 2,3-DPG content may also have an adverse effect on RBC membrane and, consequently, on the parasite invasion, in this study, we explored modifications of the RBC morphology, biomechanical properties, and RBC membrane on Plasmodium falciparum in vitro cultures treated with 2,3-DPG, using atomic force microscopy (AFM)-based force spectroscopy and other experimental approaches. The presence of infection by P. falciparum significantly increased the rigidity of parasitized cells and influenced the morphology of RBCs, as parasitized cells showed a decrease of the area-to-volume ratio. The extracellular addition of 2,3-DPG also slightly affected the stiffness of niRBCs, making it more similar to that of infected cells. It also changed the niRBC height, making the cells appear more elongated. Moreover, 2,3-DPG treatment influenced the cell surface charge, becoming more negative in treated RBCs than in untreated ones. The results indicate that treatment with 2,3-DPG has only a mild effect on RBCs in comparison with the effect of the presence of the parasite on the host cell. 2,3-DPG is an endogenous host metabolite, which may, in the future, originate a new antimalarial tool with few adverse effects on noninfected cells.publishersversionpublishe

    Cyclic voltammetry and constant-current electrolysis in a one-compartment cell with a sacrificial anode has been used to study the indirect electroreduction of N-allyl- -haloamides by electrogenerated Ni(I) complexes conducted inN,N-dimethylformamide (DMF) and acetonitrile (ACN) and the resultswere compared with those obtained in protic solvents such as EtOH and EtOH–H2O mixtures. It was observed that the indirect electrochemical reduction of N-allyl- -haloamides led to the corresponding lactams and the yields and selectivities obtained in EtOH and EtOH–H2O were much better than those obtained in DMF or ACN.Fundação para a Ciência e a Tecnologia (FCT

    Extracellular electrophysiological measurements of cooperative signals in astrocytes populations

    Astrocytes are neuroglial cells that exhibit functional electrical properties sensitive to neuronal activity and capable of modulating neurotransmission. Thus, electrophysiological recordings of astroglial activity are very attractive to study the dynamics of glial signaling. This contribution reports on the use of ultra-sensitive planar electrodes combined with low noise and low frequency amplifiers that enable the detection of extracellular signals produced by primary cultures of astrocytes isolated from mouse cerebral cortex. Recorded activity is characterized by spontaneous bursts comprised of discrete signals with pronounced changes on the signal rate and amplitude. Weak and sporadic signals become synchronized and evolve with time to higher amplitude signals with a quasi-periodic behavior, revealing a cooperative signaling process.
