Food intake is related to the nutritional state of the organism, which is
characterized by the availability of energy, partly in form of glucose. It is
known for decades that a fall in blood glucose level is a potent stimulus to
initiate food intake, but little is known so far about feeding related changes
in brain glucose concentrations. There are neurons in the hypothalamus and in
the brain stem, which change their action potential frequency in response to
changes in ambient extracellular glucose concentrations. Interestingly, these
neurons are linked to other hypothalamic structures, which are involved in the
control of food intake. The ability of neurons to response to changing local
glucose concentrations has been discussed recently with regard to the control
of ingestive behavior. The aim of the present in vivo microdialysis study was
to describe the relation between feeding and changes in glucose concentrations
in the VMH of rats. First, the method of ZNF microdialysis was used to
evaluate interstitial glucose concentrations in VMH under baseline conditions,
both in food deprived and non-deprived rats. The results demonstrate that in
fed animals absolute glucose levels in the VMH are higher (1,43 mM) compared
to rats after a 24-h food deprivation (0,94 mM). Additionally, a control group
of non-deprived rats was used to measure blood glucose in order to compare VMH
and blood glucose concentrations. As expected blood glucose concentration in
control rats was significantly higher compared to interstitial glucose
concentration observed in the VMH of non-deprived rats. To examine whether
feeding influences hypothalamic glucose, changes of glucose concentration over
time were determined relative to a baseline. Experiments were conducted in
relation to both, nutritional state (food deprived rats / non-deprived rats)
and feeding conditions throughout the experiment (free feeding rats / rats
without access to food). The experimental design took into account that rats
are mainly nocturnal animals and start to feed shortly after beginning of the
dark phase. Therefore, experiments enclosed light and dark phase. Sampling of
microdialysates started 90 Minutes before and ended 120 Minutes after
beginning of the dark phase of the experiment. Whereas no significant
alterations in VMH glucose concentration before meal initiation were observed
in the light phase of the experiment, the results of this microdialysis study
show clearly that glucose level in the VMH of rats increase significantly in
relation to food intake. The data demonstrate that food intake following a
24-h food deprivation further augments this increase as compared to non-
deprived conditions. However, food related increase in VMH glucose does not
correlate with the individual amount of food intake. In addition, feeding
related changes in VMH glucose and blood glucose over time have been compared.
Whereas hypothalamic glucose increased significantly during feeding, blood
glucose concentrations of free feeding control animals did not change
significantly over the same experimental period. Since lactate is closely
linked to glucose metabolism, alterations in interstitial lactate
concentrations in the VMH were examined. Independent of both, nutritional
state and feeding conditions, hypothalamic lactate level in the VMH of rats
increased significantly compared to baseline over the whole time of the
experiment. As with glucose, the increase in interstitial lactate in the VMH
of free feeding rats did not correlate with meal size. In contrast, lactate
/pyruvate-ratio did not change significantly compared to baseline over the
time of the experiment. In conclusion, the present study shows for the first
time that ventromedial hypothalamic glucose concentrations increase with food
intake. After meal initiation a significant increase in glucose was observed,
which could be intensified by a preceding food deprivation. Although numerous
in vitro studies have demonstrated that changes in ambient extracellular
glucose concentrations influence the action potential frequency of
hypothalamic neurons, the present in vivo microdialysis study provides
evidence that such changes actually occur under physiological conditions.
Results further indicate that the feeding related increase in VMH glucose
depends on the nutritional state of the organism. Adaptive adjustments of
brain glucose transporters to a changing nutritional status or/and
compensatory responses of the brain, e.g. mobilization of brain glycogen
reserves to sustain cerebral function, may possibly be involved. The data
further indicate that feeding related alterations in cerebral glucose
concentration over time do not parallel changes in blood glucose level. This
is evidence to suggest that mechanisms regulating glucose supply in the brain
are dissociated from that in the periphery, possibly to ensure predominantly
glucose supply of the brain when energy levels are diminished. The nature of
the relation between food intake, hypothalamic glucose concentrations and
involved regulatory mechanisms as well as their physiological importance
appears to be complex. The present study demonstrates that changes in
hypothalamic glucose concentrations occur in relation to food intake. One of
the mechanisms by which brain glucose acts on feeding behavior could be by
inducing gene expression of both, orexigenic and anorexigenic peptides, as
suggested by in vitro studies. It was important, therefore, to demonstrate
with the present study that hypothalamic glucose changes in relation to
feeding under in vivo conditions
Sensory Difference of Bio-Dynamically, Organically and Conventionally Produced Wheat from the DOK Long-Term Field Trial
There is a need of scientific evidence on the differentiation of organic from conventional produce concerning health, nutrition and sensory related qualities (Leifert et al., 2007). Analysis of wheat from the DOK long-term system comparison trial near Basel, Switzerland (Mäder et al., 2002) showed that organic wheat differed in contents of 16 “diagnostic” proteins from conventional wheat (Zörb et al., 2009a), had higher concentrations of K+ and Mg2+ cations and lower concentrations of six amino acids, and a different seed ripening metabolism (Zörb et al., 2009b). In a previous sensory test with cooked porridge of wheat (cv. Tamaro) from the DOK trial (harvest 1999), the biodynamic samples had been preferred (Arncken et al., 2007). In the present work we aimed to corroborate these results with dry samples of three harvest years

Scenario on the evolution of the arthropod brain. Animation, swf file. Anterior is up. The compound brain of arthropods (sensu Richter et al. [5]) most likely evolved by a fusion of initially separate proto-, deuto- and tritocerebral ganglia. Note that the tripartite brain of arthropods alternatively might have evolved several times independently, as some groups, including branchiopod crustaceans, show a bipartite brain (e.g. [29])
M-DC8+leukocytes - A novel human dendritic cell population
Dendritic cells (DC) constitute a heterogeneous leukocyte population having in common a unique capacity to induce primary T cell responses and are therefore most attractive candidates for immunomodulatory strategies. Two populations of blood DC (CD11c+ CD123(dim) and CD11c- CD123(high)) have been defined so far. However, their direct isolation for experimental purposes is hampered by their low frequency and by the lack of selective markers allowing large scale purification from blood. Here we describe the monoclonal antibody (mAb) M-DC8, which was generated by immunizing mice with highly enriched blood DC. This mAb specifically reacts with 0.2-1% of blood leukocytes and enables their direct isolation by a one-step immunomagnetic procedure from fresh mononuclear cells. These cells can be differentiated from T cells, B cells, NK cells and monocytes using lineage-specific antibodies. M-DC8+ cells express HLA class It molecules, CD33 and low levers of the costimulatory molecules CD86 and CD40. Upon in vitro culture M-DC8+ cells spontaneously mature into cells with the phenotype of highly stimulatory cells as documented by the upregulation of HLA-DR, CD86 and CD40; in parallel CD80 expression is induced. M-DC8+ cells display an outstanding capacity to present antigen. In particular, they proved to be excellent stimulators of autologous mixed leukocyte reaction and to activate T cells against primary antigens such as keyhole limpet hemocyanin. Furthermore, they induce differentiation of purified allogeneic cytotoxic T cells into alloantigen-specific cytotoxic effector cells. While the phenotypical analysis reveals similarities with the two known blood DC populations, the characteristic expression of Fc gamma RIII (CD16) and the M-DC8 antigen clearly defines them as a novel population of blood DC. The mAb M-DC8 might thus be a valuable tool to determine circulating DC for diagnostic purposes and to isolate these cells for studies of antigen-specific T cell priming. Copyright (C) 2000 S. Conservation Helps Limit Rate Increases for a Colorado Utility
There is a commonly held belief in the water industry that declining per capita usage due to water conservation is forcing rate increases to compensate for fewer units of volume billed. But the rate increases necessitated by conservation are actually much smaller than the rate increases that would be necessary to account for population growth in the absence of conservation. The City of Westminster, CO examined the effects of their conservation program on customer rates and found startling results
Nitrogen Budgets and Soil Nitrogen Stocks of Organic and Conventional Cropping Systems: Trade-Off between Efficiency and Sustainability of Nitrogen Use
Organic and conventional cropping systems differ in the nature and amounts of nitrogen (N) inputs, which may affect efficiency and sustainability of N use. In the DOK (bio-Dynamic, bio-Organic, Konventionell) field experiment, organic and conventional cropping systems have been compared since 1978 at two fertilization levels. Nitrogen inputs via manure and/or mineral fertilizers, and N exports from plots with harvested products have throughout been recorded. For all treatments, N outputs with harvests have exceeded the inputs with fertilizers. Over the past years, symbiotic N2 fixation by soybean and clover grown in the trial has additionally been assessed, indicating average annual inputs of about 100 kg ha-1 yr-1 of N fixed from the atmosphere. Soil surface budgets opposing N inputs via fertilization, symbiotic fixation, seeds and deposition to N outputs via harvested products have been computed at the plot level for the duration from 1985 to 2012. The resulting balances range from negative values of about -20 kg N ha-1 yr-1 (where outputs exceed the sum of said N inputs) to surpluses of about +50 kg N ha-1 yr-1. The budget based N use efficiency (NUE; N output via harvested products divided by sum of N inputs) in the case of negative balances suggests irrationally high NUE (>100%), while positive balances are related to lower NUE for treatments with inputs exceeding outputs. Negative balances, however, indicate soil N mining, while surpluses point to a risk of N losses, and/or N accumulation in the soil. Estimation of soil N stock changes based on yearly total N concentration measurements in the topsoil layer is currently ongoing. Preliminary results suggest that soil N stocks in the topsoil decreased under all treatments more than expected from the N balance, and that positive N balances are needed to maintain topsoil N stocks. An increase in soil N concentration was observed in none of the treatments. In conclusion, the results indicate an efficiency-sustainability trade-off. Treatments with a higher NUE lose more soil stock N than those with a lower NUE. Treatments with lower NUE indicate higher N losses from the studied crop-topsoil system. Sustainable soil N management in addition to organic fertilizer inputs might at this site require reduced soil tillage. The significance of N contained in deeper soil layers, and deep rooting crops in recovering leached N should as well be investigated
Despite women’s presence in the education and instruction of girls in the early modern period, teaching as a sector for women’s professional activity only developed in the first half of the nineteenth century with the growth of schools for middle-class girls. The development of possibilities was handicapped, however, by the total absence of professional training for women in the various German states. This article uses a case study to explore the first attempts to train women through the creation of a seminar for schoolteachers. It shows the opposition to Doris Lutkens’ project to provide more systematic training for women teacher, and uses her example to provide a history of women teacher’s training in Hamburg at mid century. This local study also shows that although the major criticism directed against girl’s education was the absence of serious training of their teachers, critics nonetheless refused to address this specific problem.
Why is there music?
How and why music has evolved in human cultures remains an unsolved ques-tion even today. Recently a broad discussion has emerged within the related sciences. First, the anatomical prerequisites are considered that make vocaliza-tions possible. Other important facts came from archaeological findings. Possi-bly the oldest music instrument is a bone flute, found in Slovenia and dated to 50.000 years. (Kunej & Turk, 2000)
The next step will be exploring the different hypotheses about the origins of mu-sic. (1) Is it possible that music is an adaptation, or is it rather a byproduct (Pinker, 1997)? (2) If music is an adaptation, are the origins of music caused by sexual, group or kin selection? (3) Is music an analogous trait or a homologous one? For example, did music evolve independently in different species or do we share a common ancestor with primates who were already singing?
I will show that most of the reviewed scientists argue that music is an adaptation caused by group selection and not a byproduct. Fewer scientists argue for mu-sic caused by sexual selection – most of them are even against sexual selec-tion. The same is the case for kin selection. Additionally they argue for analogy and against homology. But they are using a lot of different kind of evidence. Most of them used evidence from animals for the origins of music in humans.
These points are still open and unanswered or subject to dispute. Scientists are using different evidence for their argumentation without reconsidering their ar-guments in other contexts. There is only one universal known in human music: lullabies
