Biological and immunological characterization of recombinant Yellow Fever 17D Viruses expressing a Trypanosoma cruzi Amastigote Surface Protein-2 CD8+ T cell epitope at two distinct regions of the genome
<p>Abstract</p> <p>Background</p> <p>The attenuated Yellow fever (YF) 17D vaccine virus is one of the safest and most effective viral vaccines administered to humans, in which it elicits a polyvalent immune response. Herein, we used the YF 17D backbone to express a <it>Trypanosoma cruzi </it>CD8<sup>+ </sup>T cell epitope from the Amastigote Surface Protein 2 (ASP-2) to provide further evidence for the potential of this virus to express foreign epitopes. The TEWETGQI CD8<sup>+ </sup>T cell epitope was cloned and expressed based on two different genomic insertion sites: in the <it>fg </it>loop of the viral Envelope protein and the protease cleavage site between the NS2B and NS3. We investigated whether the site of expression had any influence on immunogenicity of this model epitope.</p> <p>Results</p> <p>Recombinant viruses replicated similarly to vaccine virus YF 17D in cell culture and remained genetically stable after several serial passages in Vero cells. Immunogenicity studies revealed that both recombinant viruses elicited neutralizing antibodies to the YF virus as well as generated an antigen-specific gamma interferon mediated T-cell response in immunized mice. The recombinant viruses displayed a more attenuated phenotype than the YF 17DD vaccine counterpart in mice. Vaccination of a mouse lineage highly susceptible to infection by <it>T. cruzi </it>with a homologous prime-boost regimen of recombinant YF viruses elicited TEWETGQI specific CD8<sup>+ </sup>T cells which might be correlated with a delay in mouse mortality after a challenge with a lethal dose of <it>T. cruzi</it>.</p> <p>Conclusions</p> <p>We conclude that the YF 17D platform is useful to express <it>T. cruzi </it>(Protozoan) antigens at different functional regions of its genome with minimal reduction of vector fitness. In addition, the model <it>T. cruzi </it>epitope expressed at different regions of the YF 17D genome elicited a similar T cell-based immune response, suggesting that both expression sites are useful. However, the epitope as such is not protective and it remains to be seen whether expression of larger domains of ASP-2, which include the TEWETGQI epitope, will elicit better T-CD8+ responses to the latter. It is likely that additional antigens and recombinant virus formulations will be necessary to generate a protective response.</p
Performance of Beef Heifers of Various Genetic Groups, Supplemented or Not, in Coastcross Pastures
The objective of this study was to determine whether the performance of beef heifers of different genetic groups was affected by breed x nutritional environment interactions. Sixty four weaned heifers, 16 per genetic group: ½ Angus + ½ Nellore (AN), ½ Canchim + ½ Nellore (CN), ½ Simmental + ½ Nellore (SN) and pure Nellore (NE), were used with or without 3.0 kg of concentrate.animal-1 .day-1 in a fertilized coastcross pasture under rotational grazing system. There were effects of genetic group and supplementation (P\u3c 0.05) on the weight and age at first estrus, but there was no interaction between them. In a rotational grazing system with 4000 kg of available dry matter per hectare with 13% of crude protein, the crossbred AN, supplemented or not, was more precocious (111 days) than Nellore heifers, showing the first estrus at 356 days of age and 324 kg of live weight
The Mediating Role of Physical Inactivity on the Relationship between Inflammation and Artery Thickness in Prepubertal Adolescents
Objective: To analyze the relationship between inflammatory markers and the lipid profile, blood flow, and artery structure in prepubertal adolescents stratified according to sports practice.Study design The sample was composed of 120 adolescents (57 boys and 63 girls) with a mean age of 11.7 +/- 0.7 years (ranging from 11 to 13 years). Intima-media thickness (IMT) and blood flow were measured with ultrasonography. The lipid profile and high-sensitivity C-reactive protein were measured after the subjects had fasted for 12 hours overnight. Trunk fatness was measured by dual-energy x-ray absorptiometry. Organized sports participation was analyzed as a categorical variable. Biological maturation was determined via the age at peak height velocity. Results: In the adjusted model, high-sensitivity C-reactive protein was significantly related to high-density lipoprotein-cholesterol (beta = -5.797 [-11.500 to -0.093]), femoral IMT (beta = 0.062 [0.008-0.116]), and the sum of femoral and carotid IMT (beta = 1.107 [0.223-1.919]), but only in the group without sports participation. Slopes of the crude linear regression were greater in the group without sports participation for femoral IMT (t = 2.621; P = .009) and the sum of femoral and carotid IMT (t = 2.876; P = .004) when compared with the group with sports participation. Microstructure and morphoquantitative study of pineal gland in santa ines sheep
O presente estudo avaliou os aspectos morfoquantitativos e qualitativos da glândula pineal em fêmeas de ovinos da raça Santa Inês em atividade reprodutiva e anestro fisiológico. Foram utilizados sete ovinos, fêmeas e adultas, coletados em frigoríficos da região de Brasília-DF. Estes não apresentavam sinais clínicos relacionados a afecções do sistema nervoso. Os encéfalos foram retirados e suas dimensões mensuradas. Posteriormente foram seccionados para expor a glândula pineal e mensurar suas dimensões. Os fragmentos do diencéfalo, contendo a glândula pineal, foram submersos em solução aquosa a 20% de formaldeído e posteriormente submetidos a técnicas histológicas convencionais. Para a avaliação morfoquantitativa desta glândula, foram empregadas as colorações Hematoxilia-Eosina, Azul de Toluidina e Tricrômio de Gomori. O encéfalo teve um comprimento médio de 67,25 ± 1,75 mm e a largura média de 58,97 ± 4,0 mm. Já a glândula pineal apresentou o comprimento médio de 6,98 ± 0,79 mm e a largura média de 6,40 ± 1,35 mm. Diante da análise microscópica, foi obtida uma média de 86,27 ± 30,44 pinealócitos por campo. Ao ser aplicado o teste de correlação de Pearson, o número de pinealócitos apresentou fraca correlação linear negativa (r = -0,11) em relação ao comprimento da glândula pineal e uma fraca correlação linear positiva (r = 0,39) em relação à largura da mesma. Portanto o número de pinealócitos apresenta uma maior correlação com a largura da glândula do que com o seu comprimento. Mastócitos estiveram presentes em apenas um animal (14,28%) e as concreções calcareas foram observadas em dois animais (28,57%). A glândula se mostrou envolta por uma cápsula de tecido conjuntivo com ausência de projeções para o parênquima. _________________________________________________________________________________ ABSTRACTThis study examined morphologic aspects of the pineal gland in Santa Ines sheep in anoestrus physiological and
reproductive activity. Seven female adult sheep were collected at slaughterhouses in the region of Brasilia-DF, with no
clinical signs related to diseases of the nervous system were used. The brains were removed and their dimensions were
measured. The brains were sectioned to expose and measure the pineal gland. The fragments of the diencephalon
containing the pineal gland were immersed in 20% formaldehyde solution. Subsequently, the fragments were submitted
to conventional histological techniques. The Hematoxylin-eosin, Toluidine blue and Gomori trichrome staining were
used for a morphoquantitative analysis. The brain had an average length of 67.25 ± 1.75 mm and average width of
58.97 ± 4.0 mm. The pineal gland had an average length of 6.98 ± 0.79 mm and average width of 6.40 ± 1.35 mm. The
quantitative analysis showed an average 86.27 ± 30.44 of pinealocytes per field. According to the Pearson correlation
test, the number of pinealocytes showed a weak negative linear correlation (r = -0.11) with the length of the pineal
gland and a weak positive linear correlation (r = 0.39) with the width. Therefore, the number of pinealocytes has a
stronger correlation with the width of the gland compared to the length. Mastocytes were present in only one animal
(14.28%). Calcareous concretions were observed in two animals (28.57%). The connective tissue formed the capsule
surrounding the gland with no projections into the parenchyma
VARIAÇÃO ENTRE E DENTRO DE POPULAÇÕES EM TAMANHO E DORMÊNCIA DE SEMENTES DE Schizolobium parahyba (Vell.) Blake NA FLORESTA ATLÂNTICA
http://dx.doi.org/10.5902/1980509820592Seed size and dormancy level were studied in 20 trees from two populations of Schizolobium parahybatoevaluate how these characteristics occurbetween and within populations, and whether seed germinationwas affected by its morphometry. These two populations are located in the coastal (Paraty) or mountain(Miguel Pereira) regions in the state of Rio de Janeiro, Brazil. The seed morphometrictraits (length,width, thickness and weight) were measured. Germination with or without seedcoatdormancy treatment(mechanical scarification) wasassessedbya randomized emergence test in nursey. All morphometric traitsdiffered significantly among individuals and between populations. The trees from the mountain regionshowed a larger seed size and a lower dormancy level than that of trees from the coastal region. Seed sizehad no effect on seed germination or seedling development. As populações são procedentes da região litorânea (Paraty) e montanhosa (Miguel Pereira) no Estado do Rio de Janeiro. As características morfométricas (comprimento, largura, espessura e massa) foram medidas e a germinação com e sem tratamento de quebra de dormência (escarificação mecânica) foi avaliada em um teste de emergência com delineamento inteiramente casualizado. Todas as variáveis morfométricas diferiram significativamente entre indivíduos e entre populações. As matrizes da região montanhosa apresentaram maior tamanho da semente e um menor nível de dormência do que aquelas localizadas na região litorânea. O tamanho da semente não influenciou a germinação das sementes e o desenvolvimento das plântulas. A influência do clima na seleção do comportamento germinativo da espécie foi discutido com base nos resultados obtidos
System of Web-Based Electronic Medical Record
Nowadays, the information systems are considered a tool to make-decision support in several areas. One of the applications of this system could be in the development of a web-based Electronic Medical Record. The attention to standards, naming, accurate measuring and the system security in the sense of information privacy are fundamental elements in the development of a web-based electronic medical record. Therefore, based on the solidarity and maturity of web applications, this work presents a solution that could supply the construction of electronic medical records by the internet. Recently, in the Brazilian market there have been few successful initiatives. Taking this into account, this work proposes the use of proven software development methodologies. How a study case was used the tengiology and vascular surgery. Currently the medical consultation processes of the angiology and vascular surgery specialties are operated manually. The final product provides automatization of these procedures
utjecaj sezonskih promjena tropske klime na reprodukcijske pokazatelje i hormonski status jaraca sanske pasmine
This study evaluated the effects different seasons (rainy, rainy-dry transition, dry, dry-rainy transition) throughout the year on the reproduction and endocrine profile of Saanen bucks reared in a tropical climate. During these seasons environmental data were recorded (ambient temperature and relative air humidity) to calculate the temperature and humidity index. Reproductive parameters were also evaluated in the same periods to check seasonal variations, wherey semen was collected twice a month from nine Saanen bucks using an artificial vagina. Furthermore, scrotal circumference, scrotal surface temperature and serum levels of testosterone, triiodothyronine (T3), thyroxine (T4) and cortisol were measured. The climate parameters influenced the reproductive parameters, mainly during the dry period, when higher values of ambient temperature reduced sperm quality, however, scrotal circumference and surface temperature, as well as serum levels of testosterone increased. In conclusion, Saanen bucks reared in a tropical climate are subject to heat stress, evinced by transient seasonal variations in reproductive parameters, where the first season is the most favorable to reproduction.U ovom je radu istražen utjecaj različitih sezona (kišna, prijelaz iz kišne u suhu, suha, prijelaz iz suhe u kišnu) tijekom godine na reprodukcijski i endokrini status jaraca sanske pasmine uzgojenih u tropskoj klimi. Zabilježeni su podaci iz okoliša (temperatura i relativna vlažnost zraka) za vrijeme različitih sezona kako bi se izračunao indeks topline. Za ista su razdoblja procijenjeni reprodukcijski pokazatelji kako bi se istražile njihove sezonske varijacije u koju je svrhu od 9 jaraca sanske pasmine primjenom umjetne vagine, prikupljano sjeme dva puta mjesečno. Osim toga mjereni su obujam skrotuma, temperatura površine skrotuma te razina testosterona, trijodotironina (T3), tiroksina (T4) i kortizola u serumu. Klimatski uvjeti su utjecali na reprodukcijske pokazatelje, najviše za vrijeme suhog razdoblja, kad veće vrijednosti temperature okoliša smanjuju kvalitetu sjemena jaraca. Obujam i temperatura površine skrotuma, kao i serumska razina testosterona, u navedenoj sezoni su povećani. Zaključeno je da su sanski jarci uzgojeni u tropskoj klimi pod utjecajem toplinskog stresa, što se vidi iz sezonskih varijacija njihovih reprodukcijskih pokazatelja, pri čemu je kišna sezona najpovoljnija za reprodukciju
End TB strategy: the need to reduce risk inequalities.
BACKGROUND: Diseases occur in populations whose individuals differ in essential characteristics, such as exposure to the causative agent, susceptibility given exposure, and infectiousness upon infection in the case of infectious diseases. DISCUSSION: Concepts developed in demography more than 30 years ago assert that variability between individuals affects substantially the estimation of overall population risk from disease incidence data. Methods that ignore individual heterogeneity tend to underestimate overall risk and lead to overoptimistic expectations for control. Concerned that this phenomenon is frequently overlooked in epidemiology, here we feature its significance for interpreting global data on human tuberculosis and predicting the impact of control measures. We show that population-wide interventions have the greatest impact in populations where all individuals face an equal risk. Lowering variability in risk has great potential to increase the impact of interventions. Reducing inequality, therefore, empowers health interventions, which in turn improves health, further reducing inequality, in a virtuous circle
