    Spatial distribution of demersal fishery resources, environmental factors and fishing activities in GSA 15 (Malta Island)

    The FAO–MedSudMed Regional Project has promoted research on the improvement of knowledge on fishery ecosystems, with a view to the sustainable management of living marine resources in the central Mediterranean (Straits of Sicily). A pilot study focused on the waters around the Maltese Islands (GSA 15), with the aim of providing a comprehensive overview of the spatial distribution of the different life stages of exploitable demersal fishes in relation to the type and distribution of fishing, as well as to the oceanographic factors characteristic of the area of study. Critical zones for their role in the ecology of the main demersal fishery target species were investigated using species abundance data disaggregated by life stage. Spatial analysis and the application of GIS techniques allowed the identification of preferred habitats (e.g. nursery, feeding and spawning areas) for Merluccius merluccius, Mullus barbatus, Parapenaeus longirostris, Raja clavata and Raja miraletus. The impact of fishing on these species was assessed, particularly in terms of fish assemblages. The transport paths of early life stages were also hypothesized on the basis of oceanographic factors typical of the area. The results revealed that the spatial distribution of the main fishery resources overlaps the limits of the current GSAs. The analysis of oceanographic factors showed that some fishery resources are sustained by young individuals transported from adjacent GSAs. The results also demonstrated that the spatial distribution of the main demersal fishery resources in the Mediterranean GSA 15 straddle other GSAs, in particular as concerns nursery and spawning areas, indicating that some fishery resources are shared. This implies that harmonized fishery management should be applied over a larger area than is implied by the GSA concept.     The challenge of sustainability in healthcare systems: Frequency and cost of inappropriate patterns of breast cancer care (the E.Pic.A study).

    Abstract Objectives In a context of decreasing economic health resources and a rise in health needs, it is urgent to face this sustainability crisis through the analysis of healthcare expenditures. Wastages, deriving from inappropriate interventions, erode resources which could be reallocated to high-value activities. To identify these areas of wastages, we developed a method for combining and analyzing data from multiple sources. Here we report the preliminary results of a retrospective cohort study evaluating the performance of breast cancer (BC) care at IRST, an Italian cancer institute. Materials and methods Four data sources gathered in a real-world setting (a clinical database, two administrative databases and a cancer registry) were linked. Essential Key Performance Indexes (KPIs) in the pattern of BC diagnosis (KPI 1 and 2) and treatment (KPI 3 and 4) based on current guidelines were developed by a board of professionals. The costs of inappropriate examinations were associated with the diagnostic KPIs. Results We found that 2798 patients treated at IRST from January 2010 to June 2016 received a total of 2516 inappropriate examinations accounting for € 573,510.80. Linkage from multiple routine healthcare data sources is feasible: it allows the measurement of important KPIs specifically designed for BC care, and the identification of areas of low-value use of the resources. Conclusion If systematically applied, this method could help provide a complete picture of inappropriateness and waste, redirect these resources to higher-value interventions for patients, and fill the gap between proper use of the resources and the best clinical results

    Myocardial-specific R-spondin3 drives proliferation of the coronary stems primarily through the Leucine Rich Repeat G Protein coupled receptor LGR4

    Coronary artery anomalies are common congenital disorders with serious consequences in adult life. Coronary circulation begins when the coronary stems form connections between the aorta and the developing vascular plexus. We recently identified the WNT signaling modulator R-spondin 3 (Rspo3), as a crucial regulator of coronary stem proliferation. Using expression analysis and tissue-specific deletion we now demonstrate that Rspo3 is primarily produced by cardiomyocytes. Moreover, we have employed CRISPR/Cas9 technology to generate novel Lgr4-null alleles that showed a significant decrease in coronary stem proliferation and thus phenocopied the coronary artery defects seen in Rspo3 mutants. Interestingly, Lgr4 mutants displayed slightly hypomorphic right ventricles, an observation also made after myocardial specific deletion of Rspo3. These results shed new light on the role of Rspo3 in heart development and demonstrate that LGR4 is the principal Rspondin 3 receptor in the heart

    Endothelial colony forming cells (ECFCs) represent a population of truly endothelial precursors that promote the angiogenic switch in solid tumors, such as breast cancer (BC). The intracellular Ca2+ toolkit, which drives the pro-angiogenic response to VEGF, is remodelled in tumor-associated ECFCs such that they are seemingly insensitive to this growth factor. This feature could underlie the relative failure of anti-VEGF therapies in cancer patients. Herein, we investigated whether and how VEGF uses Ca2+ signalling to control angiogenesis in BC-derived ECFCs (BCECFCs). Although VEGFR-2 was normally expressed, VEGF failed to induce proliferation and in vitro tubulogenesis in BC-ECFCs. Likewise, VEGF did not trigger robust Ca2+ oscillations in these cells. Similar to normal cells, VEGF-induced intracellular Ca2+ oscillations were triggered by inositol-1,4,5-trisphosphate-dependent Ca2+ release from the endoplasmic reticulum (ER) and maintained by store-operated Ca2+ entry (SOCE). However, InsP3-dependent Ca2+ release was significantly lower in BC-ECFCs due to the down-regulation of ER Ca2+ levels, while there was no remarkable difference in the amplitude, pharmacological profile and molecular composition of SOCE. Thus, the attenuation of the pro-angiogenic Ca2+ response to VEGF was seemingly due to the reduction in ER Ca2+ concentration, which prevents VEGF from triggering robust intracellular Ca2+ oscillations. However, the pharmacological inhibition of SOCE prevented BC-ECFC proliferation and in vitro tubulogenesis. These findings demonstrate for the first time that BC-ECFCs are insensitive to VEGF, which might explain at cellular and molecular levels the failure of anti-VEGF therapies in BC patients, and hint at SOCE as a novel molecular target for this disease

We present a new simple tool based on a scatter plot of the modulation curve in which the counts in each angular bin are reported after a shifting by 1/4 of the period. The sinusoidal pattern is thus transformed in a circular plot whose radius is equal to the amplitude of the modulation, while for a not polarized radiation the scatter plot is reduced to a random point distribution centred at the mean frequency value. The advantage of this tool is that one can easily evaluate the statistical significance of the polarimetric detection and can obtain useful information on the quality of the measurement

    EXTRAVERTED CHILDREN SWIM FASTER COMPARED TO INTROVERTED COUNTERPARTS REGARDLESS OF LIGHT AND SOUND NOISE LEVELS

    Individual differences of personality are thought to influence motor performance. In terms of cortical arousal levels, because extraverts are infra-activated and introverts are hyper-activated, environment stimuli might enhance the impact of the extraversion trait on task performance. This study investigated the effect of light and sound noise on the swimming performance of extraverted and introverted children. 19 extraverts (12 boys, 7 girls) and 22 introverts (12 boys, 10 girls), ages 8.2 ± 0.9 years, adapted to water and swimming at intermediate levels. Participants performed two trials of the task (swimming 15 meters as fast as possible in crawl style) under two environment conditions: bright light/loud noise (A) and dim light/slight noise (B). Movements were filmed to allow calculation of time to complete the task and the stroke cycle. There was a significant effect for the group factor, with extraverts swimming faster than introverts. No effect was detected for the environment factor or the interaction group/environment. Regarding stroke cycle, no differences were found for group, environment or interaction. Although extraversion has not affected mechanical aspects of crawl style, compared to introverts, extraverts swan faster, showing a more effective process of reacting and executing movements in time-constraints tasks.Diferenças individuais de personalidade podem influenciar o desempenho motor. O presente estudo investigou o efeito da luminosidade e do ruído sonoro no desempenho natatório de crianças extrovertidas (19; 12 meninos, 7 meninas) e introvertidos (22; 12 meninos, 10 meninas), com idade de 8.2 ± 0.9 anos, adaptadas à água e com nível intermediário de natação. As crianças executaram duas tentativas da tarefa (nadar 15 metros o mais depressa possível em estilo crawl) sob duas condições ambientais: luz forte/ruído alto (A) e luz fraca/ruído baixo (B). Os movimentos foram filmados para cálculo de tempo para completar a tarefa e de ciclo de braçada. Houve efeito significativo para o fator grupo, com extrovertidos nadando mais rapidamente que introvertidos. Não houve efeito para o fator ambiente ou interação grupo/ambiente. Quanto ao ciclo de braçada, não houve diferenças para qualquer fator ou interação. Embora a extroversão não tenha afetado aspectos mecânicos do nado crawl, comparados aos introvertidos, os extrovertidos nadaram mais rapidamente, o que demonstra um processo mais efetivo para reagir e executar movimentos com restrições de tempo

    ScienzAperta is an outreach science venue that the Istituto Nazionale di Geofisica e Vulcanologia started in 2011 as the spring of science: the doors of the headquarters of science were finally opened to public. A number of events, conferences, seminars, guided tours through the Institute and its laboratories are every year offered to general public. The venue is held in most of the cities where the Institute is located, priority to high seismic and/or volcanic risk regions. On May 2014 we held ScienzAperta for the first time in Milano and open up the doors to schools specifically dealing mostly with seismic hazard in a region where general public not necessarily think it might We offered students conferences, seminars and educational activities to highlight the fun of science and jet raise awareness on proper behaviours in case of earthquake shaking. We asked students and teachers, from elementary to high schools, to fill in a questionnaire that we use to evaluate the appreciation the venue had.     Can Urine Metabolomics Be Helpful in Differentiating Neuropathic and Nociceptive Pain? A Proof-of-Concept Study

    The diagnosis of pain nature is a troublesome task and a wrong attribution often leads to an increase of costs and to avoidable pharmaceutical adverse reactions. An objective and specific approach to achieve this diagnosis is highly desirable. The aim of this work was to investigate urine samples collected from patients suffering from pain of different nature by a metabolomics approach based on 1 H NMR spectroscopy and multivariate statistical analysis. We performed a prospective study on 74 subjects: 37 suffering from pain (12 with nociceptive and 25 with neuropathic pain), and 37 controls not suffering from any kind of chronic pain. The application of discriminant analysis on the urine spectral profiles allowed us to classify these two types of pain with high sensibility and specificity. Although the classification relies on the global urine metabolic profile, the individual contribution in discriminating neuropathic pain patients of metabolites such as choline and phosphocholine, taurine and alanine, suggests potential lesions to the nervous system. To the best of our knowledge, this is the first time that a urine metabolomics profile is used to classify these two kinds of pain. This methodology, although based on a limited sample, may constitute the basis for a new helpful tool in the clinical diagnosis
