Dying to be thin: The social construction of the female beauty ideal and eating disorders.
Although there has been a substantial amount of research conducted on females and eating disorders, little emphasis has been placed on social factors. Exploratory research was performed in an attempt to answer the question, Can eating disorders be scientifically explained by psychological and/or biochemical factors at the expense of undermining certain social factors and their explanatory power in eating disorders. In short, should the explanation of eating disorders be viewed as a combination of psychological and/or biochemical factors as well as social factors, whose explanatory significance should not be overlooked? Following the theory of social construction, and an Interpretive Interactionist approach, 10 women in therapy for an eating disorder were interviewed, utilizing open ended interview questions. The interviewees were asked 38 questions in accordance to five social variables used for the study. The findings of this thesis were discussed, and conclusions were made. Quantification of MUCIN 1, cell surface associated and MUCIN16, cell surface associated proteins in tears and conjunctival epithelial cells collected from postmenopausal women
Srinivasan, S., Heynen, M. L., Martell, E., Ritter, R., Jones, L., & Senchyna, M. (2013). Quantification of MUCIN 1, cell surface associated and MUCIN16, cell surface associated proteins in tears and conjunctival epithelial cells collected from postmenopausal women. Molecular Vision, 19, 970–979.Purpose: To quantify the expression of mucin 1, cell surface associated (MUC1) and mucin 16, cell surface associated (MUC16) proteins and messenger ribonucleic acid (mRNA) in a cohort of postmenopausal women (PMW), to explore the relationship between mucin expression, dry eye symptomology, and tear stability. Methods: Thirty-nine healthy PMW (>50 years of age) were enrolled in this study. No specific inclusion criteria were used to define dry eye; instead, a range of subjects were recruited based on responses to the Allergan Ocular Surface Disease Index (OSDI) questionnaire and tear stability measurements as assessed by non-invasive tear breakup time (NITBUT). Tears were collected from the inferior tear meniscus using a disposable glass capillary tube, and total RNA and total protein were isolated from conjunctival epithelial cells collected via impression cytology. Expression of membrane-bound and soluble MUC1 and MUC16 were quantified with western blotting, and expression of MUC1 and MUC16 mRNA was assessed with real-time PCR.Results: OSDI responses ranged from 0 to 60, and NITBUT ranged from 18.5 to 2.9 s. Only two statistically significant correlations were found: soluble MUC16 protein concentration and MUC16 mRNA expression with OSDI vision related (−0.47; p=0.01) and ocular symptom (0.39; p=0.02) subscores, respectively. Post hoc exploratory analysis on absolute expression values was performed on two subsets of subjects defined as asymptomatic (OSDI ≤6, n=12) and moderate to severe symptomatic (OSDI ≥20, n=12). The only significant difference between the two subgroups was a significant reduction in MUC16 mRNA expression found in the symptomatic dry eye group (1.52±1.19 versus 0.57±0.44; p=0.03).Conclusions: A broad exploration of mucin expression compared to either a sign (NITBUT) or symptoms of dry eye failed to reveal compelling evidence supporting a significant relationship, other than a potential association between MUC16 with specific symptoms. Furthermore, comparison of mucin protein and expression levels between the asymptomatic and moderate to severe symptomatic subgroups revealed only one significant difference, a reduction in MUC16 mRNA expression in the symptomatic subgroup.Funding for this study was provided by Alcon Research Ltd and the Natural Sciences and Engineering Research Council of Canada (NSERC)

Stoichiometry of mercury-thiol complexes on bacterial cell envelopes
We have examined the speciation of Hg(II) complexed with intact cell suspensions (1013 cells L− 1) of Bacillus subtilis, a common gram-positive soil bacterium, Shewanella oneidensis MR-1, a facultative gram-negative aquatic organism, and Geobacter sulfurreducens, a gram-negative anaerobic bacterium capable of Hg-methylation at Hg(II) loadings spanning four orders of magnitude (120 nM to 350 μM) at pH 5.5 (± 0.2). The coordination environments of Hg on bacterial cells were analyzed using synchrotron based X-ray Absorption Near Edge Structure (XANES) and Extended X-ray Absorption Fine Structure (EXAFS) spectroscopy at the Hg LIII edge. The abundance of thiols on intact cells was determined by a fluorescence-spectroscopy based method using a soluble bromobimane, monobromo(trimethylammonio)bimane (qBBr) to block thiol sites, and potentiometric titrations of biomass with and without qBBr treatment. The chemical forms of S on intact bacterial cells were determined using S k-edge XANES spectroscopy. Hg(II) was found to complex entirely with cell bound thiols at low Hg:biomass ratios. For Bacillus subtilis and Shewanella oneidensis MR-1 cells, the Hg—S stoichiometry changed from Hg—S₃ to Hg—S₂ and Hg—S (where ‘S’ represents a thiol site such as is present on cysteine) progressively as the Hg(II) loading increased on the cells. However, Geobacter sulfurreducens did not form Hg—S₃ complexes. Because the abundance of thiol was highest for Geobacter sulfurreducens (75 μM/g wet weight) followed by Shewanella oneidensis MR-1 (50 μM/g wet weight) and Bacillus subtilis (25 μM/g wet weight), the inability of Hg(II) to form Hg—S₃ complexes on Geobacter sulfurreducens suggests that the density and reactivity of S-amino acid containing cell membrane proteins on Geobacter sulfurreducens are different from those of Bacillus subtilis and Shewanella oneidensis MR-1. Upon saturation of the high affinity thiol sites at higher Hg:biomass ratios, Hg(II) was found to form a chelate with α-hydroxy carboxylate anion. The stoichiometry of cell envelope bound Hg-thiol complexes and the associated abundance of thiols on the cell envelopes provide important insights for understanding the differences in the rate and extent of uptake and redox transformations of Hg in the environment
Active Tuberculosis among Homeless Persons, Toronto, Ontario, Canada, 1998–2007
While tuberculosis (TB) in Canadian cities is increasingly affecting foreign-born persons, homeless persons remain at high risk. To assess trends in TB, we studied all homeless persons in Toronto who had a diagnosis of active TB during 1998–2007. We compared Canada-born and foreign-born homeless persons and assessed changes over time. We identified 91 homeless persons with active TB; they typically had highly contagious, advanced disease, and 19% died within 12 months of diagnosis. The proportion of homeless persons who were foreign-born increased from 24% in 1998–2002 to 39% in 2003–2007. Among foreign-born homeless persons with TB, 56% of infections were caused by strains not known to circulate among homeless persons in Toronto. Only 2% of infections were resistant to first-line TB medications. The rise in foreign-born homeless persons with TB strains likely acquired overseas suggests that the risk for drug-resistant strains entering the homeless shelter system may be escalating
