    Response to fungal exudates of the rhizosphere isolate Pseudomonas sp. UMAF110 involves a GGDEF/EAL domain-containing protein

    Pseudomonas sp. UMAF110, isolated from rhizosphere soil in Spain, display in vitro antagonism towards the pythopathogenic fungus Rosellinia necatrix and is able grow in fungal exudates (BM-RE medium). A transposon mutant library of this strain was constructed and several mutants were selected by their reduced competitiveness in BM-RE medium. Pseudomonas sp. UMAF110-G3, which contains the transposon into a gene encoding a putative REC/PAS/GGDEF/EAL protein, was selected for further characterization. Blastn searches using the sequence of the gene interrupted by the transposon in UMAF110-G3, here called cmpA (c-di-GMP Metabolizing Protein), yielded a single positive hit (98% cover, 78% identity) with a gene from a terpene-degrading Pseudomonas sp. strain isolated from soil. Context analysis of the cmpA gene in Pseudomonas sp. UMAF110 showed that this gene is located downstream from several genes involved in flagellar motility/chemotaxis. RT-PCR experiments further confirmed that cmpA form a transcriptional unit with the che gene cluster. Expression analysis of cmpA by qRT-PCR clearly showed upregulation of this gene after transfer of Pseudomonas sp. UMAF110 cells to BM-RE medium, suggesting a role for this operon in response to fungal exudates. Deletion of cmpA in Pseudomonas sp. UMAF110 did not affect the ability of the strain to form biofilms under the conditions tested. However, overexpression of wild type CmpA in Pseudomonas putida KT2440 negatively regulated biofilm formation in this strain. Together, these results suggest that CmpA could be involved in signal transduction pathways regulating flagellar motility/chemotaxis in response to fungal exudates.Universidad de Málaga.     On the extraction of cellulose nanowhiskers from food by-products and their comparative reinforcing effect on a polyhydroxybutyrate-co-valerate polymer

    The present work reports on the characterization of cellulose nanowhiskers (CNW) extracted from three different food by-products, i.e., wheat straw (WSCNW), Brewers spent grains (BGCNW) and olive pomace (OPCNW), by using an optimized hydrolysis method similar to that developed to extract bacterial cellulose nanowhiskers (BCNW). WSCNW and BGCNW were seen to present optimal properties, with aspect ratio, crystallinity and thermal stability values comparable to those of BCNW. Additionally, the optimized hydrolysis treatment led to extraction yields higher than those previously reported for food by-products. The CNW were subsequently incorporated into a commercial polyhydroxybutyrate-co-valerate polymer (PHBV) by solution casting, and the produced nanocomposites were characterized. Although the addition of BGCNW and WSCNW was advantageous in terms of mechanical performance in comparison with OPCNW, no significant enhancement of the pure PHBV mechanical properties was reported because of the low nanofiller loadings used and the inherent difficulty of achieving a high degree of dispersion by the casting method. Interestingly, BGCNW and WSCNW presented reduced moisture sensitivity as compared with BCNW, leading to greater barrier performance and resulting in oxygen permeability reductions up to 26 % with WSCNW and 44 % with BGCNW.Noelle Peutat, on leave from the University of Grenoble in France, is acknowledged for her great dedication and support in the experimental work.     Describimos una experiencia de colaboración entre el Parque de las Ciencias de Granada y el centro educativo Ciencias sobre Ruedas de Puerto Rico para llevar a siete centros educativos de Andalucía la tecnología de experimentación asistida por ordenador, EXAO, conectándola con temas de investigación real en el área de química. El proceso comenzó con la formación del profesorado en el uso de esta tecnología dentro de un enfoque de enseñanza orientado hacia la investigación en el aula. La actividad ha estado a cargo de profesores universitarios de Puerto Rico que desarrollan actualmente sus investigaciones de doctorado. Se ofrece la experiencia como un modelo de actualización y formación del profesorado no universitario de ciencias

    Phase profile analysis of transparent objects through the use of a two windows interferometer based on a one beam splitter configuration

    AbstractIn this research we implemented a two windows interferometer based on polarization phase shifting and grating interferometry techniques in order to retrieve the phase data profile of the object in a single capture. The optical configuration has two optical beams with circular polarization in opposite directions, and it is coupled with a 4-f system. An amplitude grid is used as a filter which is placed at the Fourier plane to obtain replicas of each beam which can properly interfere, depending on the separation between beams. The interferometer presents the capability of changing the beam separation in order to make different orders interfere properly. The interference patterns produced can be separately modulated through the operation of linear polarizer's placed on each interference replica.     Towards an unsupervised speaking style voice building framework: multi-style speaker diarization

    Current text-to-speech systems are developed using studio-recorded speech in a neutral style or based on acted emotions. However, the proliferation of media sharing sites would allow developing a new generation of speech-based systems which could cope with spontaneous and styled speech. This paper proposes an architecture to deal with realistic recordings and carries out some experiments on unsupervised speaker diarization. In order to maximize the speaker purity of the clusters while keeping a high speaker coverage, the paper evaluates the F-measure of a diarization module, achieving high scores (>85%) especially when the clusters are longer than 30 seconds, even for the more spontaneous and expressive styles (such as talk shows or sports)
