    Dietary calcium: strategies to optimize intake

    Calcium is an essential nutrient required for numerous biological functions. Studies have demonstrated an association between low calcium intake and chronic diseases, such as osteoporosis, colon cancer, hypertension, and obesity. However, most Brazilians do not meet the adequate intake for calcium. This review focuses on the endogenous (age, hormonal state) and exogenous (phytate, oxalate, sodium, bioactive compounds and vitamin D) factors that can influence calcium absorption. The main methods used for evaluating calcium absorption and bioavailability. The potential factors for the low calcium intake: 1) Food habits - substitution of milk for soft drinks, eating away from home and skipping meals specially breakfast; 2) High cost of dairy food. Besides, this article discuss strategies to optimize calcium intake: 1) Increase knowledge of the relationship between calcium and health and the main food sources; 2) Increase availability of calcium-fortified foods; 3) Supplement use for target groups - when and how administrate calcium salts.O cálcio é um nutriente essencial necessário em diversas funções biológicas. Estudos têm demonstrado a associação entre o baixo consumo de cálcio e doenças crônicas, entre elas osteoporose, câncer de colón, hipertensão arterial e obesidade. Entretanto, grande parte da população brasileira apresenta consumo de cálcio abaixo do recomendado. Este artigo objetiva revisar os fatores endógenos (idade e estado hormonal) e exógenos (fitatos, oxalatos, sódio, compostos bioativos e vitamina D) que influenciam a absorção do cálcio, bem como as principais metodologias utilizadas para avaliar a absorção e biodisponibilidade desse nutriente. Discorre-se sobre os possíveis fatores para o baixo consumo de cálcio: 1) Hábito alimentar - substituição de leite por bebidas com baixo teor de cálcio como o refrigerante, refeições realizadas fora de casa e a não realização de refeições como o café da manhã; 2) Alto custo dos alimentos fontes de cálcio.     Deep reefs are not refugium for shallow-water fish communities in the southwestern Atlantic

    Unidad de excelencia María de Maeztu CEX2019-000940-M1. The deep reef refugia hypothesis (DRRH) predicts that deep reef ecosystems may act as refugium for the biota of disturbed shallow waters. Because deep reefs are among the most understudied habitats on Earth, formal tests of the DRRH remain scarce. If the DRRH is valid at the community level, the diversity of species, functions, and lineages of fish communities of shallow reefs should be encapsulated in deep reefs. 2. We tested the DRRH by assessing the taxonomic, functional, and phylogenetic diversity of 22 Brazilian fish communities between 2 and 62 m depth. We partitioned the gamma diversity of shallow (30 m) into independent alpha and beta components, accounted for species' abundance, and assessed whether beta patterns were mostly driven by spatial turnover or nestedness. 3. We recorded 3,821 fishes belonging to 85 species and 36 families. Contrary to DRRH expectations, only 48% of the species occurred in both shallow and deep reefs. Alpha diversity of rare species was higher in deep reefs as expected, but alpha diversity of typical and dominant species did not vary with depth. Alpha functional diversity was higher in deep reefs only for rare and typical species, but not for dominant species. Alpha phylogenetic diversity was consistently higher in deep reefs, supporting DRRH expectations. 4. Profiles of taxonomic, functional, and phylogenetic beta diversity indicated that deep reefs were not more heterogeneous than shallow reefs, contradicting expectations of biotic homogenization near sea surface. Furthermore, pairwise beta-diversity analyses revealed that the patterns were mostly driven by spatial turnover rather than nestedness at any depth. 5. Conclusions. Although some results support the DRRH, most indicate that the shallow-water reef fish diversity is not fully encapsulated in deep reefs. Every reef contributes significantly to the regional diversity and must be managed and protected accordingly

    Mechanistic and biological characterisation of novel N5-substituted paullones targeting the biosynthesis of trypanothione in Leishmania

    Trypanothione synthetase (TryS) produces N1,N8-bis(glutathionyl)spermidine (or trypanothione) at the expense of ATP. Trypanothione is a metabolite unique and essential for survival and drug-resistance of trypanosomatid parasites. In this study, we report the mechanistic and biological characterisation of optimised N5-substituted paullone analogues with anti-TryS activity. Several of the new derivatives retained submicromolar IC50 against leishmanial TryS. The binding mode to TryS of the most potent paullones has been revealed by means of kinetic, biophysical and molecular modelling approaches. A subset of analogues showed an improved potency (EC50 0.5-10 µM) and selectivity (20-35) against the clinically relevant stage of Leishmania braziliensis (mucocutaneous leishmaniasis) and L. infantum (visceral leishmaniasis). For a selected derivative, the mode of action involved intracellular depletion of trypanothione. Our findings shed light on the molecular interaction of TryS with rationally designed inhibitors and disclose a new set of compounds with on-target activity against different Leishmania species

    Crescimento e desenvolvimento da cultura do melão sob diferentes lâminas de irrigação e salinidade da água

    Avaliar o crescimento, o desenvolvimento foliar e o acúmulo de matéria seca da cultura de melão Gold mine , submetido a diferentes lâminas de irrigação, utilizando-se água com dois níveis de salinidade, foi o objetivo deste trabalho. O experimento foi conduzido em condições de campo, na Fazenda São João, município de Mossoró, RN, cujo delineamento experimental adotado foi o de blocos casualizados, em esquema fatorial 6 x 2. Os tratamentos consistiram na introdução de seis lâminas de irrigação (0,55; 0,70; 0,85; 1,00; 1,15 e 1,30 da evapotranspiração máxima da cultura) e dois níveis de salinidade da água de irrigação 0,55 e 2,65 dS m-1. A falta de água no período crítico afetou significativamente o restante do ciclo da cultura, causando decréscimo, no peso da fitomassa seca, para lâminas abaixo do tratamento da lâmina padrão (266 mm). O acúmulo de fitomassa foi afetado pela água de maior salinidade (2,65 dS m-1) ao longo de todo ciclo.The aim of this study was to evaluate the growth, vegetative development and accumulation of dry matter of the "Gold mine" melon submitted to different depths of irrigation using two levels of salinity. The experiment was conducted under field conditions at Fazenda São João, Mossoró, RN, in random blocks and 6 x 2 factorial experimental design. The treatments consisted of 6 depths of irrigation (0.55; 0.70; 0.85; 1.00; 1.15; 1.3 of the maximum crop evapotranspiration) and two levels of salinity of the irrigation water (0.55 and 2.65 dS m-1). The absence of water in the critical period significantly affected the rest of the cycle, causing decrease in the dry weight in the treatment of irrigation depth considered as of the most appropriate depth (266 mm). The water of higher salinity (2.65 dS m-1) affected the accumulation of dry matter in the cycle
