    Dissolved organic matter as a confounding factor in the determination of pollution-induced community tolerance (PICT) of bacterial communities to heavy metals using the leucine incorporation method

    PICT methodology using the leucine incorporation method (Leu-PICT) is useful for assessing heavy metal contamination in soils. First, bacterial community is exposed to metal in the soil (selection phase), developing tolerance if metal exerts toxicity. Secondly, in detection phase, bacterial suspensions are obtained, and tolerance is quantified by a second exposition of bacterial community to the metal using Leu-PICT methodology. However, during detection phase when Leu-PICT is performed, some characteristics of bacterial suspensions may change metal bioavailability. We assess the influence of dissolved organic matter (DOM) in bacterial suspensions, as humic acids (HA), on Leu-PICT determination to Cr, Cu, Ni, Pb and Zn. Results showed that the presence of HA in bacterial suspensions causes underestimations of bacterial community tolerance to Cr (increasing Cr toxicity), and overestimations of bacterial community tolerance to Cu, Ni, Pb, and Zn (reducing metal toxicity).     Adolescents, online gambling, problematic internet use and substance consumption

    Recent research has warned of the growing participation of minors in online gambling, an illegal behaviour with an enormous addictive potential. The present study was proposed with a double objective: (1) having updated data about online gambling among adolescents and, (2) analysing its relationship with substance use and Proble-matic Internet Use [PIU]. For this purpose, a sample of 3188 Spanish adolescents between 12 and 17 years of age (Mean=14.44; SD=1.67) was gathered, to whom was applied an ad hoc questionnaire with items regarding their internet use and substance consumption, as well as specific screening instruments (Alcohol Use Disorders Identification Test -AUDIT-, Cannabis Abuse Screening Test -CAST-, the Substance Use and Abuse subscale of the Problem Oriented Screening Instrument for Teenagers -POSITuas-, and the Problematic Internet Use Scale for Ado-lescents-PIUSa-). The results showed that 8.4% of the sample had participated in online gambling during the prior year. It was also found that those who gambled on the Internet had higher rates of PIU and different online risky behaviours, as well as higher rates of substance consumption. Therefore, these are not isolated problems, making it necessary to employ an integral preventive approach to address them.Recientes investigaciones han alertado de la creciente participación de los menores en el juego online, una conducta ilegal con un enorme potencial adictivo. Para ello, se recogió una muestra de 3188 adolescentes españoles de entre 12 y 17 años (Media=14.44; DT=1.67), a los que se les aplicó un cuestionario ad hoc con ítems relativos al uso de Internet y consumo de sustancias, así como instrumentos de cribado específicos (Alcohol Use Disorders Identification Test -AUDIT-, Cannabis Abuse Screening Test -CAST-, Substance Use and Abuse subscale of the Problem Oriented Scree-ning Instrument for Teenagers -POSITuas- y la Escala de Uso Problemático de Internet para Adolescentes -EUPIa-).     Analysis of the relationship between school bullying, cyberbullying, and substance use

    The use of alcohol and other substances among underage adolescents continues to cause social concern nowadays, but it is not the only challenge that professionals face while working with this population. School bullying and cyberbullying remain another main issue affecting the well-being and development of the students. The main objective of the present study was to analyse the possible relationship between both school bullying and cyberbullying with substance use. A selective methodology was used, administering a survey among students from state funded Secondary schools in the Autonomous Community of Galicia (Spain), and obtaining a sample of 3,173 adolescents aged 12 to 17 years. The questionnaire included items referring to the consumption of several substances, specific screening scales for the evaluation of risky consumption (Alcohol Use Disorders Identification Test, Cannabis Abuse Screening Test and CRAFFT Abuse Screening Test) and validated multi-item scales to assess school bullying and cyberbullying involvement (European Bullying Intervention Project Questionnaire and European Cyberbullying Intervention Project Questionnaire). The results showed that the most consumed substances were alcohol, tobacco and cannabis, finding also high risky consumption rates. Adolescents involved in either school bullying or cyberbullying in any role (victims, perpetrators and bully-victims) presented significantly higher rates in the consumption habits and risky consumptions analysed. These results highlight the need for a comprehensive prevention approach that addresses both school bullying, cyberbullying, and addictions at the same time.S

    Norovirus GII.3[P12] Outbreak Associated with the Drinking Water Supply in a Rural Area in Galicia, Spain, 2021

    On 30 September 2021, the city council of Muxia, Spain (population of 4,564 inhabitants), reported an unusual increase of patients with acute gastroenteritis (AGE). Because geographically widespread villages belonging to the same water supply were affected, a waterborne outbreak was suspected. Overall, 115 probable cases were ascertained during epidemiological investigations carried out by the local health authority (attack rate, 5.7%); the age range was 0 to 92 years, and 54% were female. The main symptoms were vomiting (78.1%) and diarrhea (67.5%). Primary cases peaked on 29 September and subsided on 1 October, compatible with a point-source outbreak followed by possible secondary cases until 7 October. We conducted an unmatched case-control study using phone surveys. The case-control study included 62 cases and 46 controls. Univariate analysis showed that cases had a higher exposure to tap water through direct consumption (odds ratio [OR] = 86; 95% confidence interval [CI], 18 to 409) or vegetable washing (OR = 27; 95% CI, 7 to 98). Norovirus GII was detected in two terminal points of the water supply system, and 14 cases were laboratory confirmed after detection of GII in stool samples. A unique genotype (GII.3[P12]) was identified in stool samples. On 1 October, a tap water ban was put in place and the water was purged and chlorinated. The rapid increase in the number of cases and its decline after implementing control measures suggested a waterborne point-source outbreak among the residents of Muxia sharing the same water distribution system. IMPORTANCE: Noroviruses are likely to be underrecognized in most suspected waterborne outbreaks. Therefore, effective norovirus detection and the early recognition of water as a possible source of infection are important to reduce morbidity as appropriate steps are taken to control the source. In our study, we combined epidemiological, environmental, and microbiological investigations to demonstrate that it was a waterborne outbreak caused by norovirus. Metagenomic sequencing in one norovirus-positive stool sample confirmed norovirus etiology and the absence of other potential pathogens. Detection of fecal indicator bacteria and the fact that the drinking water was not chlorinated suggest a breakdown in chlorination as the cause of the outbreak. This outbreak investigation also demonstrated the importance of timely communication to the public about the risk linked to tap water consumption.This study was partially funded through the project PI20CIII/00005.S

    Vertical zonation of bacterial assemblages attributed to physical stratification during the summer relaxation of the coastal upwelling off Galicia (NW Spain)

    Research articleWe combined flow cytometry, CARD-FISH, and 16S rRNA gene tag pyrosequencing to investigate bacterioplankton dynamics along a transect in shelf waters off A Coruña (Galicia, NW Spain). Over five days (16-20th July 2012) we sampled during the relaxation of a summer upwelling pulse, providing an opportunity to examine the impact of pulses of cold nutrient-rich water into coastal microbial communities. The hydrographic conditions, characterized by intense density stratification of surface waters and the presence of a deep chlorophyll maximum (DCM) at 20–30 m, were relatively maintained over the sampling period. Indeed, bacterial abundance and composition displayed low day to day variation. Alpha diversity analysis suggested that species richness and diversity increased from coastal to shelf stations and from the surface down to the coastal DCM, which could be caused by the mixing of upwelled bacteria with the coastal surface waters. SAR11, SAR86, and Roseobacter were the most abundant bacteria detected in the samples by using CARD-FISH. The assemblages observed by pyrosequencing displayed a strong vertical zonation along the transect. Rhodobacteraceae (under class Alphaproteobacteria) and Bacteriodetes dominated the surface waters and decreased during the upwelling pulse, while SAR 86 (under class Gammaproteobacteria), Actinobacteria and SAR11 clade increased their relative abundance at the coastal DCM with upwelling relaxation, particularly at the shelf stations. Bacterial assemblages from surface waters were associated with higher temperature and light conditions, while coastal DCM assemblages were rather associated to salinity, inorganic nutrients and a diatom-bloom leading to high chlorophyll-a.     Diversity and abundance of planktonic communities in the deep waters off the galician coast (NW Spain)

    Comunicación oralPlanktonic communities play pivotal roles within marine ecosystems, affecting their structure, functioning and services. Although they have been extensively studied in the epipelagic ocean, the knowledge about these communities in the dark ocean is rather short. In this study, we explored patterns of abundance and biomass of a wide variety of taxonomic groups from the prokaryotes to mesozooplankton in the epi-, meso- and bathypelagic waters off the Galician coast. As expected, ciliate and zooplankton abundances are depleted in the bathypelagic waters relative to abundances of prokaryotes and nanoflagellates. The rate of decrease of zooplankton biomass with depth is twice as that of prokaryotes and nanoflagellates, indicating that relative contribution of mesozooplancton to the total plankton biomass decreases with depth. Overall, the diversity of prokaryotes in the dark ocean is almost as high as in the epipelagic layer, although the phylotypes are different. The major fraction of epipelagic ciliates belongs to alloricate genera, whereas tintinnids dominate the deep ciliate populations. Small copepods were dominant in the epi- and meso-pelagic zone. By contrast, foraminiferans, big copepods and myctophic fishes were more abundant in the deep ocean

    Pentraxin 3 (PTX3): A Molecular Marker of Endothelial Dysfunction in Chronic Migraine

    Even though endothelial dysfunction is known to play a role in migraine pathophysiology, studies regarding levels of endothelial biomarkers in migraine have controversial results. Our aim was to evaluate the role of pentraxin 3 (PTX3) and soluble tumour necrosis factor-like weak inducer of apoptosis (sTWEAK) as potential biomarkers of endothelial dysfunction in chronic migraine (CM). We performed a case-control study including 102 CM patients and 28 control subjects and measured serum levels of markers of endothelial dysfunction (PTX3 and sTWEAK) and inflammation [high-sensitivity C-reactive protein (hs-CRP)] as well as brachial artery flow-mediated dilation (FMD) during interictal periods. Interictal serum levels of PTX3 and sTWEAK were higher in CM patients than in controls (1350.6 ± 54.8 versus 476.1 ± 49.4 pg/mL, p < 0.001 and 255.7 ± 21.1 versus 26.4 ± 2.6 pg/mL, p < 0.0001; respectively). FMD was diminished in CM patients compared to controls (9.6 ± 0.6 versus 15.2 ± 0.9%, p < 0.001). Both PTX3 and sTWEAK were negatively correlated with FMD (r = −0.508, p < 0.001 and r = −0.188, p = 0.033; respectively). After adjustment of confounders, PTX3 remained significantly correlated to FMD (r = −0.250, p = 0.013). Diagnosis of CM was 68.4 times more likely in an individual with levels of PTX3 ≥ 832.5 pg/mL, suggesting that PTX3 could be a novel biomarker of endothelial dysfunction in CMThis research was funded by Spanish Ministry of Economy and Competitiveness—Institute of Health Carlos III, Grant/Award Numbers: PI15/01578S

    Extra-Virgin Olive Oil Modifies the Changes Induced in Non-Nervous Organs and Tissues by Experimental Autoimmune Encephalomyelitis Models

    This study reveals the existence of oxidative stress (reactive oxygen species (ROS)) in non-nervous organs and tissues in multiple sclerosis (MS) by means of a model of experimental autoimmune encephalomyelitis (EAE) in rats. This model reproduces a similar situation to MS, as well as its relationship with intestinal microbiota starting from the changes in bacterial lipopolysaccharide levels (LPS) in the outer wall of the gram-negative bacteria. Finally, the administration of extra-virgin olive oil (EVOO), hydroxytirosol (HT), and oleic acid (OA) exert beneficial effects. Twenty-five Dark Agouti two-month-old male rats, weighing around 190 g, were distributed into the following groups: Control, EAE (experimental autoimmune encephalomyelitis group), EAE + EVOO, EAE + HT, and EAE + OA. The glutathione redox system with the EAE was measured in heart, kidney, liver, and small and large intestines. The LPS and the correlation with oxidative stress in the small and large intestines were also investigated. The results showed that (1) the oxidative damage in the EAE model affects non-nervous organs and tissues; (2) The LPS is related to inflammatory phenomena and oxidative stress in the intestinal tissue and in other organs; (3) The administration of EVOO, HT, and OA reduces the LPS levels at the same time as minimizing the oxidative damage; (4) EVOO, HT, and OA improve the disease’s clinical score; and (5) on balance, EVOO offers a better neuroprotective effect
