    The biological properties of OGI surfaces positively act on osteogenic and angiogenic commitment of mesenchymal stem cells

    Osteogenesis process displays a fundamental role during dental implant osteointegration. In the present work, we studied the influence of Osteon Growth Induction (OGI) surface properties on the angiogenic and osteogenic behaviors of Mesenchymal Stem cells (MSC). MSC derived from dental pulp and HUVEC (Human Umbilical Vein Endothelial Cells) were grown in on OGI titanium surfaces, and cell proliferation and DNA synthesis were evaluated by MTT [3-(4,5-dimethylthiazol-2yl)-2,5-diphenyltetrazolium bromide] test and DNA quantification. Gene expression has been performed in order to evaluate the presence of mRNA related to endothelial and osteogenesis markers. Moreover, morphological and biochemical analyses of osteogenesis commitments has been performed. On OGI surfaces, MSC and HUVEC are able to proliferate. Gene expression profiler confirms that MSC on OGI surfaces are able to express endothelial and osteogenic markers, and that these expression are higher compared the expression on control surfaces. In conclusion On OGI surfaces proliferation, expression and morphological analyses of angiogenesis-associated markers in MSC are promoted.     Long-term follow-up of Philadelphia chromosome-positive (Ph+) chronic myeloid leukaemia (CML) in children and adolescents managed at a single institution over a 20-year period

    Chronic myeloid leukaemia (CML) is rare in childhood. In our Institution we managed 30 consecutive Ph+CML patients aged <18 years, according to our adults’ guidelines. Patients with HLA-identical related donor (RD) underwent stem cell transplant (SCT). Since 1989, patients without RD were systematically treated with -interferon (IFN) (median dosage: 6 MU/day). Of 18/19 evaluable patients, 17 (94.5%) achieved haematologic response (HR), 11/17 (65%) cytogenetic response (CyR), complete (CCyR) in 4 (23.5%). Three patients remain in CCyR, 2 achieved BCR-ABL transcript disappearance. Of 13 patients without CCyR, 5 underwent SCT, 4 switched to STI571, 4 progressed. All patients receiving STI571 in chronic phase (CP) obtained sustained CCyR and 3 a persistent molecular response. 8-year survival among IFN-treated patients, censored or not for subsequent therapies, is 62% and 63%. Overall, 13/30 patients underwent SCT: 5 HLA-identical-RD, 5 matched unrelated donor, 2 mismatched-RD, 1 unrelated mismatched umbilical cord blood. Eight allotransplanted patients (6/6 in 1st CP) are in cytogenetic and molecular remission with 8-year survival of 61% from SCT and 69% from diagnosis. In our 20-year experience, the use of IFN in children without matched RD led to prolonged cytogenetic and molecular responses and long-term survival, without impairing the outcome of subsequent SCT

Programa de Pós-Graduação em Psicologia.A Orientação Profissional é uma prática social comprometida com o desenvolvimento humano, ao auxiliar os jovens na ampliação de possibilidades profissionais e na construção de projetos de vida. Mediante uma abordagem qualitativa, esta pesquisa procurou compreender a juventude como processo social, e não como fenômeno natural. Seu propósito é investigar quais são os projetos de vida de participantes do Programa Jovem Aprendiz do Centro Cultural Escrava Anastácia sob o ponto de vista da Orientação Profissional. Para atingi-lo, realizaram-se oito entrevistas individuais e quatro grupos focais, com 33 jovens entre 14 e 21 anos. A análise de conteúdo foi organizada em quatro categorias: projeto de vida; identificação familiar; importância do Programa Jovem Aprendiz; e escolha profissional. A categoria projeto de vida relaciona-se com a dinâmica sócio-histórica e com o movimento de construção da identidade, entendida como metamorfose. Ao abordarem seus projetos, os jovens expuseram aspectos das trajetórias educacional e profissional de seus familiares, a importância do trabalho e dos estudos para o desenvolvimento profissional e as exigências do mercado de trabalho. O projeto profissional é o elemento principal de seus projetos de vida e relaciona-se com a continuação dos estudos, considerada fundamental para a obtenção de melhores condições de vida. O principal meio de efetivação dos projetos de vida dos jovens é o trabalho, que auxilia na transformação em realidade de um projeto futuro. O Programa Jovem Aprendiz foi considerado uma oportunidade profissional e motivador para os estudos e para o crescimento pessoal. Quanto à escolha profissional, os jovens apontam aspectos relacionados aos significantes #ser bem-sucedido#, #fazer o que gosta#, #ser alguém superior#, retratando o quanto o trabalho é um aspecto central para a construção da identidade deles, além de possibilitar a efetivação de seus projetos de vida e alcançar a satisfação pessoal.Career Counseling is a social practice committed to human development, helping young people to expand professional possibilities and build life projects. Using a qualitative approach, this research sought to understand youth as a social process, and not as a natural phenomenon. Its purpose is to explore which are the life projects of the Young Apprentice Program participants of the Centro Cultural Escrava Anastácia from the Career Counseling point of view. To reach it, there were eight individual interviews and four focal groups, with 33 young people between the age of 14 and 21. Content analysis was organized into four categories: life project; family identification; Young Apprentice Program importance; and career choice. The category life project relates to the social-historical dynamic and to the identity formation movement, understood as metamorphosis. By addressing their projects, young people presented the aspects of educational and professional trajectories of their relatives, the importance of work and study to the professional development and the demands of the world of work. The professional project is the main element of their life projects and relates to the continuation of studies, considered essential for the achievement of better living conditions. The primary means of realization of young people life projects is work, which helps the transformation of a future project into reality. The Young Apprentice Program was considered a professional opportunity and motivation for study and for personal growth. As for career choice, young people point out aspects related to the significant "be successful", "do what you like", "be somebody superior", depicting how work is a central aspect to their identity formation, besides allowing the realization of their life projects and achieving personal satisfaction

    The Role of Design in the CE Transition of the Furniture Industry—The Case of the Italian Company Cassina

    Abstract: The literature on circular economy has highlighted the need for more studies focused on investigating the journey of individual companies in the transition toward sustainable processes. This paper addresses this need by focusing on the furniture design industry, showing how the transition requires the re-organization of knowledge regarding materials, processes, technologies, and product quality. This assumption is demonstrated through the design research activity conducted in 2019–2020 as the first part of broader research by Cassina LAB, a collaboration between Cassina Research and Development Centre and POLI.design of Politecnico di Milano. Based on the analysis of the Italian furniture industry between constraints and opportunities, the aim of the research is to identify critical issues and propose sustainable and circular solutions, tailor-made for Cassina. Through this example, the paper contributes to the literature in two ways. First, it adds to the understanding of how companies are adopting the circular economy paradigm. Secondly, it contributes to defining tools to implement new forms of knowledge of materials and re-design processes to deliver products that are compatible with a circular economy model

    Early changes in brain structure correlate with language outcomes in children with neonatal encephalopathy.

    Global patterns of brain injury correlate with motor, cognitive, and language outcomes in survivors of neonatal encephalopathy (NE). However, it is still unclear whether local changes in brain structure predict specific deficits. We therefore examined whether differences in brain structure at 6 months of age are associated with neurodevelopmental outcomes in this population. We enrolled 32 children with NE, performed structural brain MR imaging at 6 months, and assessed neurodevelopmental outcomes at 30 months. All subjects underwent T1-weighted imaging at 3 T using a 3D IR-SPGR sequence. Images were normalized in intensity and nonlinearly registered to a template constructed specifically for this population, creating a deformation field map. We then used deformation based morphometry (DBM) to correlate variation in the local volume of gray and white matter with composite scores on the Bayley Scales of Infant and Toddler Development (Bayley-III) at 30 months. Our general linear model included gestational age, sex, birth weight, and treatment with hypothermia as covariates. Regional brain volume was significantly associated with language scores, particularly in perisylvian cortical regions including the left supramarginal gyrus, posterior superior and middle temporal gyri, and right insula, as well as inferior frontoparietal subcortical white matter. We did not find significant correlations between regional brain volume and motor or cognitive scale scores. We conclude that, in children with a history of NE, local changes in the volume of perisylvian gray and white matter at 6 months are correlated with language outcome at 30 months. Quantitative measures of brain volume on early MRI may help identify infants at risk for poor language outcomes

    Standardization of a protocol to extract and analyze chlorophyll a and carotenoids in Gracilaria tenuistipitata Var. Liui. Zhang and Xia (Rhodophyta)

    Chlorophyll a and carotenoids are important pigments in photosynthesis. Several studies have been published describing extraction and analysis protocols of these pigments, mainly in vascular plant species. This study standardizes an extraction and analysis protocol of these substances in Gracilaria tenuistipitata var. liui, a red seaweed. Apical portions grown in vitro were triturated in liquid nitrogen. Extracts were prepared in 1.5 mL solvent and centrifuged. Quantitative and qualitative analyses of pigments were performed by UV/visible light spectrophotometry and high performance liquid chromatography (HPLC) and HPLC coupled to mass spectrometry (HPLC-MS). The parameters assessed were: minimum biomass, best extraction solvent, and number of extraction steps. Methanol was the most efficient solvent, and 50 mg fresh biomass was the amount of sample indicated, submitted to one single extraction step. No significant differences were observed in levels of these pigments by UV-visible light spectrophotometry and HPLC. However, HPLC or HPLC-MS are required to identify the different carotenoids present in this seaweed species.Clorofila a e carotenoides são importantes pigmentos da fotossíntese. Na literatura são encontrados vários protocolos de extração e análise desses pigmentos utilizando, principalmente, plantas vasculares. O objetivo deste estudo foi padronizar uma metodologia de extração e análise dessas substâncias em uma macroalga vermelha, Gracilaria tenuistipitata var. liui. Amostras de talos gametofíticos cultivados in vitro foram trituradas em nitrogênio líquido, extraídas em 1,5 mL de solvente, centrifugadas e os pigmentos analisados quantitativamente e qualitativamente através de espectrofotometria de UV/visível, cromatografia liquida de alta eficiência (CLAE) e CLAE-acoplada a espectrometria de massas (CLAE-EM). Foram testados os parâmetros massa mínima, solvente para extração e número de extrações. Dentre os solventes testados, o metanol foi o mais eficiente, sendo 50mg de material fresco a massa mínima indicada para ser submetida a somente uma extração. Não foram encontradas diferenças significativas na quantificação desses pigmentos comparando-se os dados obtidos em espectrofotometria de UV/visível com os de CLAE.     Multi-object Data Integration in the Study of Primary Progressive Aphasia

    This article focuses on a multi-modal imaging data application where structural/anatomical information from grey matter (GM) and brain connectivity information in the form of a brain connectome network from functional magnetic resonance imaging (fMRI) are available for a number of subjects with different degrees of primary progressive aphasia (PPA), a neurodegenerative disorder (ND) measured through a speech rate measure on motor speech loss. The clinical/scientific goal in this study becomes the identification of brain regions of interest significantly related to the speech rate measure to gain insight into ND pathways. Viewing the brain connectome network and GM images as objects, we develop a flexible joint object response regression framework of network and GM images on the speech rate measure. A novel joint prior formulation is proposed on network and structural image coefficients in order to exploit network information of the brain connectome, while leveraging the topological linkages among connectome network and anatomical information from GM to draw inference on brain regions significantly related to the speech rate measure. The principled Bayesian framework allows precise characterization of the uncertainty in ascertaining a region being actively related to the speech rate measure.     Neurocognitive Basis of Repetition Deficits in Primary Progressive Aphasia

    Previous studies indicate that repetition is affected in primary progressive aphasia (PPA), particularly in the logopenic variant, due to limited auditory-verbal short-term memory (avSTM). We tested repetition of phrases varied by length (short, long) and meaning (meaningful, non-meaningful) in 58 participants (22 logopenic, 19 nonfluent, and 17 semantic variants) and 21 healthy controls using a modified Bayles repetition test. We evaluated the relation between cortical thickness and repetition performance and whether sub-scores could discriminate PPA variants. Logopenic participants showed impaired repetition across all phrases, specifically in repeating long phrases and any phrases that were non-meaningful. Nonfluent, semantic, and healthy control participants only had difficulty repeating long, non-meaningful phrases. Poor repetition of long phrases was associated with cortical thinning in left temporo-parietal areas across all variants, highlighting the importance of these areas in avSTM. Finally, Bayles repetition phrases can assist classification in PPA, discriminating logopenic from nonfluent/semantic participants with 89% accuracy
