    CyanoEXpress: A web database for exploration and visualisation of the integrated transcriptome of cyanobacterium Synechocystis sp. PCC6803.

    UNLABELLED: Synechocystis sp. PCC6803 is one of the best studied cyanobacteria and an important model organism for our understanding of photosynthesis. The early availability of its complete genome sequence initiated numerous transcriptome studies, which have generated a wealth of expression data. Analysis of the accumulated data can be a powerful tool to study transcription in a comprehensive manner and to reveal underlying regulatory mechanisms, as well as to annotate genes whose functions are yet unknown. However, use of divergent microarray platforms, as well as distributed data storage make meta-analyses of Synechocystis expression data highly challenging, especially for researchers with limited bioinformatic expertise and resources. To facilitate utilisation of the accumulated expression data for a wider research community, we have developed CyanoEXpress, a web database for interactive exploration and visualisation of transcriptional response patterns in Synechocystis. CyanoEXpress currently comprises expression data for 3073 genes and 178 environmental and genetic perturbations obtained in 31 independent studies. At present, CyanoEXpress constitutes the most comprehensive collection of expression data available for Synechocystis and can be freely accessed. The first establishes the emergence of the concept of online learning and the development of Information Technology Literacy, and how they interact in the context of Online Knowledge Management in Work and Organizational Psychology. It also determines and analyzes the learning and production process within this urea. The second part describes a web-based training experience with a sample of 160 students, which lasted a semester. It outlines the objectives,practical exercises, methods, results and main findings in terms ofskills enhanced through the performance patterns followed by the students in the learning process.Este artículo se ha dividido en dos secciones. La primera establece la emergencia conceptual del espacio en línea de aprendizaje y el desarrollo de la alfabetización en las tecnologías de la información, y cómo ambos se dan la mano en el contexto de la gestión del conocimiento en línea y la psicología del trabajo. También fija y analiza los procesos deaprendizaje y produccidn en este espacio. La segunda describe una experiencia de formación basada en Internet en la que se implicó a una muestra de 160 alumnos durante un semestre. Se reseñan los objetivos, los ejercicios prácticos, los resultados y las constataciones más importantes en términos de destrezas a través de los patrones de desempeño seguidos por el alumnado en el proceso de aprendizaje

    The ability of electrochemical oxidation with a BDD anode to inactivate Gram-negative and Gram-positive bacteria in low conductivity sulfate medium

    The disinfection of 100 mL of synthetic water containing 7 mM Na2SO4 with 106 CFU mL-1 of either Gram-negative or Gram-positive bacteria has been studied by electrochemical oxidation. The electrolytic cell was a stirred tank reactor equipped with a boron-doped diamond (BDD) anode and a stainless steel cathode and the trials were performed at acidic and neutral pH, at 33 mA cm-2 and 25 ºC. Reactive oxygen species, pre-eminently hydroxyl radicals, were efficiently produced in both media from water oxidation at the BDD anode and the bacteria concentration was reduced by ≥ 5 log units after 60 min of electrolysis, thus constituting a good chlorine-free disinfection treatment. All the inactivation kinetics were described by a logistic model, with no significant statistical differences between acidic and neutral suspensions. The electrochemical disinfection with BDD was very effective for Gram-negative bacilli like E. coli and P. aeruginosa and Gram-positive ones like B. atrophaeus, whereas the Gram-positive cocci S. aureus and E. hirae were more resistant. Thus, the latter organisms are a better choice than E. coli as process indicators. Scanning electron microscopy highlighted a transition from initial cells with standard morphology supported on clean filters to inactivated cells with a highly altered morphology lying on dirty filters with plenty of cellular debris. Larger damage was observed for Gram-negative cells compared to Gram-positive ones. The inactivation effect could then be related to the chemical composition of the outer layers of the cell structure along with the modification of the transmembrane potentials upon current passage

    Relação entre status social subjetivo e saúde percebida entre mulheres imigrantes latino-americanas

    OBJECTIVE: to explore the relationship between socioeconomic status and subjective social status and explain how subjective social status predicts health in immigrant women. METHODS: cross-sectional study based on data from 371 Latin American women (16-65 years old) from a total of 7,056 registered immigrants accesse through community parthers between 2009-2010. Socioeconomic status was measured through education, income and occupation; subjective social status was measured using the MacArthur Scale, and perceived health, using a Likert scale. RESULTS: a weak correlation between socioeconomic and subjective social status was found. In the bivariate analysis, a significantly higher prevalence of negative perceived health in women with no education, low income, undocumented employment was observed. In the multivariate analysis, higher odds of prevalence of negative perceptions of health in the lower levels of the MacArthur scale were observed. No significant differences with the rest of the variables were found. CONCLUSIONS: the study suggests that subjective social status was a better predictor of health status than the socioeconomic status measurements. Therefore, the use of this measurement may be relevant to the study of health inequalities, particularly in socially disadvantaged groups such as immigrants.OBJETIVO: explorar la relación entre el estatus socioeconómico y el estatus social subjetivo y explicar en qué medida el estatus social subjetivo predice la salud en mujeres inmigrantes. MÉTODOS: estudio transversal. Observaciones basadas en 371 latinoamericanas (16-65 años) de un total de 7.056 empadronadas, captadas a través de asociaciones entre 2009-2010. El estatus socioeconómico se midió a través de educación, ingresos y ocupación; el estatus social subjetivo usando la Escala MacArthur; y la salud percibida mediante una escala de likert. RESULTADOS: se encontró una correlación débil entre el estatus socioeconómico y el social subjetivo. En el análisis bivariante se observó significativamente una prevalencia mayor de salud percibida negativa en las mujeres sin estudios, con ingresos bajos, desempleadas e indocumentadas. En el análisis multivariante, se observaron Odds de prevalencia de salud percibida negativa más elevadas en los niveles de la escala MacArthur más bajos. No se observaron diferencias significativas con el resto de las variables. CONCLUSIONES: el estudio sugiere que el estatus social subjetivo es un predictor mejor del estado de salud que las medidas del estatus socioeconómico. Por tanto, el uso de esta medida puede ser relevante para el estudio de las desigualdades en salud, particularmente en los grupos en desventaja social como los inmigrantes.OBJETIVO: explorar a relação entre nível socieconômico e status social subjetivo e explicar como o status social subjetivo prediz a saúde em mulheres imigrantes. RESULTADOS: encontrou-se fraca correlação entre o nível socioeconômico e o status social subjetivo. Na análise bivariada, observou-se prevalência significativamente mais alta de saúde percebida negativamente em mulheres sem escolaridade, baixa renda, desempregadas e com emprego informal. Na análise multivariada, observaram-se maiores chances de prevalência de saúde percebida negativamente, nos níveis mais baixos da escala MacArthur. Não foram encontradas diferenças significativas nas demais variáveis. CONCLUSÕES: o estudo sugere que o status social subjetivo foi um melhor preditor de status de saúde do que as mensurações de status econômico. Portanto, o uso dessa medida pode ser relevante para o estudo das desigualdades em saúde, particularmente nos grupos em desvantagem social, como os imigrantes

    Iron Deprivation in Synechocystis : Inference of Pathways, Non-coding RNAs, and Regulatory Elements from Comprehensive Expression Profiling

    Iron is an essential cofactor in many metabolic reactions. Mechanisms controlling iron homeostasis need to respond rapidly to changes in extracellular conditions, but they must also keep the concentration of intracellular iron under strict control to avoid the generation of damaging reactive oxygen species. Due to its role as a redox carrier in photosynthesis, the iron quota in cyanobacteria is about 10 times higher than in model enterobacteria. The molecular details of how such a high quota is regulated are obscure. Here we present experiments that shed light on the iron regulatory system in cyanobacteria. We measured time-resolved changes in gene expression after iron depletion in the cyanobacterium Synechocystis sp. PCC 6803 using a comprehensive microarray platform, monitoring both protein-coding and non-coding transcripts. In total, less than a fifth of all protein-coding genes were differentially expressed during the first 72 hr. Many of these proteins are associated with iron transport, photosynthesis, or ATP synthesis. Comparing our data with three previous studies, we identified a core set of 28 genes involved in iron stress response. Among them were genes important for assimilation of inorganic carbon, suggesting a link between the carbon and iron regulatory networks. Nine of the 28 genes have unknown functions and constitute key targets for further functional analysis. Statistical and clustering analyses identified 10 small RNAs, 62 antisense RNAs, four 59UTRs, and seven intragenic elements as potential novel components of the iron regulatory network in Synechocystis.     Identification of the direct regulon of NtcA during early acclimation to nitrogen starvation in the cyanobacterium Synechocystis sp. PCC 6803

    In cyanobacteria, nitrogen homeostasis is maintained by an intricate regulatory network around transcription factor NtcA. Although mechanisms controlling NtcA activity appear to be well understood, its regulon remains poorly defined. To determine the NtcA regulon during the early stages of nitrogen starvation for the model cyanobacterium Synechocystis sp. PCC 6803, we performed chromatin immunoprecipitation, followed by sequencing (ChIP-seq), in parallel with transcriptome analysis (RNA-seq). Through combining these methods, we determined 51 genes activated and 28 repressed directly by NtcA. In addition to genes associated with nitrogen and carbon metabolism, a considerable number of genes without current functional annotation were among direct targets providing a rich reservoir for further studies. The NtcA regulon also included eight non-coding RNAs, of which Ncr1071, Syr6 and NsiR7 were experimentally validated, and their putative targets were computationally predicted. Surprisingly, we found substantial NtcA binding associated with delayed expression changes indicating that NtcA can reside in a poised state controlled by other factors. Indeed, a role of PipX as modulating factor in nitrogen regulation was confirmed for selected NtcA-targets.     HDTQ: Managing RDF Datasets in Compressed Space

    HDT (Header-Dictionary-Triples) is a compressed representation of RDF data that supports retrieval features without prior decompression. Yet, RDF datasets often contain additional graph information, such as the origin, version or validity time of a triple. Traditional HDT is not capable of handling this additional parameter(s). This work introduces HDTQ (HDT Quads), an extension of HDT that is able to represent quadruples (or quads) while still being highly compact and queryable. Two HDTQ-based approaches are introduced: Annotated Triples and Annotated Graphs, and their performance is compared to the leading open-source RDF stores on the market. Results show that HDTQ achieves the best compression rates and is a competitive alternative to well-established systems

    Influence of PPh3 moiety in the anticancer activity of new organometallic ruthenium complexes

    The effect of the PPh3 group in the antitumor activity of some new organometallic Ruthenium (II) complexes has been investigated. Several complexes of the type [Ru(II)(Cl)(PPh3)(Lig-N)], [Ru(II)(Cl)2(Lig-N)] (where Lig-N=pyridine derivate) and [Ru(II)(Cl)(PPh3)2], have been synthesized and characterized, and an important increment of the antitumor activity and cytotoxicity of the complexes due to the presence of PPh3 moiety has been demonstrated, affording IC50 values of 5.2 μM in HL-60 tumour cell lines. Atomic Force Microscopy, Circular Dichroism and Electrophoresis experiments have proved that these complexes can bind DNA resulting in a distortion of both secondary and tertiary structures. Ethidium bromide displacement Fluorescence Spectroscopy studies and Viscosity measurements support that the presence of PPh3 group induces intercalation interactions with DNA. Indeed, crystallographic analysis, suggest that intra-molecular π-π interactions could be involved in the intercalation within DNA base pairs. Furthermore, HPLC-MS studies have confirmed a strong interaction between Ruthenium complexes and proteins (Ubiquitin and Potato Carboxypeptidase Inhibitor -PCI-) including slower kinetic due to the presence of PPh3 moiety, which could have an important role in detoxification mechanism and others. Finally, Ion Mobility Mass Spectrometry (IMMS) experiments have proved that there is no change in the structural conformation of the proteins owing to their bonding to Ruthenium complexes. This seems particularly important in the case of PCI, that may be a suitable candidate for vehiculizing these complexes in a selective manner into tumour cells. In agreement with these results, further investigations should be carried out to clarify either there is a favoured binding to DNA or to specific proteins, thus to elucidate their main biological target
