    The potential of portable luminescence readers in geomorphological investigations : a review

    The development of functional portable optically stimulated luminescence (OSL) readers over the last decade has provided practitioners with the capability to acquire luminescence signals from geological materials relatively rapidly, which allows for expedient preliminary chronostratigraphic insight when working with complex depositional systems of late Quaternary age. Typically, when using the portable OSL reader, infrared (IR) or blue post-IR OSL signals are acquired from bulk unprocessed materials, in contrast to regular luminescence dating which is usually based on measurements on pure quartz or feldspar mineral separates, or on select silt-sized polymineralic portions. To demonstrate the utility of portable OSL measurements, this paper outlines the basic features of portable OSL readers and their constraints. Afterwards, case studies in which the instrument has been used to elucidate cryptostratigraphic variations in sedimentary sequences for geomorphological applications are reviewed. The studies can generally be grouped into three main categories. The first includes studies where the variation of portable OSL reader luminescence signal intensities with depth are plotted to generate profiles that contextualise sediment stratigraphy. In the second group, portable OSL reader luminescence signal intensities are used to interpret sediment processes that shed light on depositional histories. In the last category, luminescence signals from the portable OSL reader are calibrated to approximate numerical burial ages of depositional units.     A guide to northern sea cucumbers: The biology and management of Cucumaria frondosa

    This document synthesising knowledge on the northern sea cucumber Cucumaria frondosa was prepared for all stakeholders, including industry participants, government scientists, policymakers, and academic researchers. Its aim is to highlight the uniqueness of this marine resource to guide the industry forward and to emphasize areas that deserve further investigation. Available data from eastern and northern Canada, eastern United States of America, Greenland, northern Europe and the Russian Federation are presented. Topics covered include the taxonomy, distribution, biology, and ecology of the species, the natural threats it faces, the current harvesting, processing and marketing practices, and the prospects for aquaculture development. Relying on a knowledge base gathered over more than 40 years, this contribution compares C. frondosa with other common commercial species of sea cucumbers to tease out the major aspects that set it apart. A final section provides a number of key recommendations for its management and conservation

    Small-Molecule Activators of Insulin-Degrading Enzyme Discovered through High-Throughput Compound Screening

    Background: Hypocatabolism of the amyloid β-protein (Aβ) by insulin-degrading enzyme (IDE) is implicated in the pathogenesis of Alzheimer disease (AD), making pharmacological activation of IDE an attractive therapeutic strategy. However, it has not been established whether the proteolytic activity of IDE can be enhanced by drug-like compounds. Methodology/Principal Findings: Based on the finding that ATP and other nucleotide polyphosphates modulate IDE activity at physiological concentrations, we conducted parallel high-throughput screening campaigns in the absence or presence of ATP and identified two compounds—designated Ia1 and Ia2—that significantly stimulate IDE proteolytic activity. Both compounds were found to interfere with the crosslinking of a photoaffinity ATP analogue to IDE, suggesting that they interact with a bona fide ATP-binding domain within IDE. Unexpectedly, we observed highly synergistic activation effects when the activity of Ia1 or Ia2 was tested in the presence of ATP, a finding that has implications for the mechanisms underlying ATP-mediated activation of IDE. Notably, Ia1 and Ia2 activated the degradation of Aβ by ∼700% and ∼400%, respectively, albeit only when Aβ was presented in a mixture also containing shorter substrates. Conclusions/Significance: This study describes the first examples of synthetic small-molecule activators of IDE, showing that pharmacological activation of this important protease with drug-like compounds is achievable. These novel activators help to establish the putative ATP-binding domain as a key modulator of IDE proteolytic activity and offer new insights into the modulatory action of ATP. Several larger lessons abstracted from this screen will help inform the design of future screening campaigns and facilitate the eventual development of IDE activators with therapeutic utility

    Chemicals released by male sea cucumber mediate aggregation and spawning behaviours

    The importance of chemical communication in reproduction has been demonstrated in many marine broadcast spawners. However, little is known about the use of chemical communication by echinoderms, the nature of the compounds involved and their mechanism(s) of action. Here, the hypothesis that the sea cucumber Holothuria arguinensis uses chemical communication for aggregation and spawning was tested. Water conditioned by males, but not females, attracted both males and females; gonad homogenates and coelomic fluid had no effect on attraction. Male spawning water, but not female spawning water, stimulated males and females to release their gametes; the spermatozoa alone did not induce spawning. H. arguinensis male spawning water also induced spawning in the phylogenetically related H. mammata. This indicates that males release pheromones together with their gametes that induce spawning in conspecifics and possibly sympatric species. Finally, the male pheromone seems to be a mixture with at least one labile compound (biological activity is lost after four hours at ambient temperature) possibly including phosphatidylcholines.     What we talk about when we talk about "global mindset": managerial cognition in multinational corporations

    Recent developments in the global economy and in multinational corporations have placed significant emphasis on the cognitive orientations of managers, giving rise to a number of concepts such as “global mindset” that are presumed to be associated with the effective management of multinational corporations (MNCs). This paper reviews the literature on global mindset and clarifies some of the conceptual confusion surrounding the construct. We identify common themes across writers, suggesting that the majority of studies fall into one of three research perspectives: cultural, strategic, and multidimensional. We also identify two constructs from the social sciences that underlie the perspectives found in the literature: cosmopolitanism and cognitive complexity and use these two constructs to develop an integrative theoretical framework of global mindset. We then provide a critical assessment of the field of global mindset and suggest directions for future theoretical and empirical research

    Assessing control of postural stability in community-living older adults using performance-based limits of stability

    <p>Abstract</p> <p>Background</p> <p>Balance disability measurements routinely used to identify fall risks in frail populations have limited value in the early detection of postural stability deficits in community-living older adults. The objectives of the study were to 1) measure performance-based limits of stability (LOS) in community-living older adults and compare them to theoretical LOS computed from data proposed by the Balance Master<sup>® </sup>system, 2) explore the feasibility of a new measurement approach based on the assessment of postural stability during weight-shifting tasks at performance-based LOS, 3) quantify intra-session performance variability during multiple trials using the performance-based LOS paradigm.</p> <p>Methods</p> <p>Twenty-four healthy community-living older adults (10 men, 14 women) aged between 62 to 85 (mean age ± sd, 71.5 ± 6 yrs) participated in the study. Subjects' performance-based LOS were established by asking them to transfer their body weight as far as possible in three directions (forward, right and left) without changing their base of support. LOS were computed as the maximal excursion of the COP in each direction among three trials. Participants then performed two experimental tasks that consisted in controlling, with the assistance of visual feedback, their centre of pressure (COP) within two predefined targets set at 100% of their performance-based LOS. For each tasks 8 trials were performed. Ground reaction forces and torques during performance-based LOS evaluation and experimental tasks were recorded with a force plate. Sway area and medio-lateral mean COP displacement speed variables were extracted from force plate recordings.</p> <p>Results</p> <p>Significant differences between theoretical LOS computed from maximum leaning angles derived from anthropometric characteristics and performance-based LOS were observed. Results showed that a motor learning effect was present as the participants optimized their weight-shifting strategy through the first three trials of each task using the visual biofeedback provided on their COP. Reliable measures of control of postural stability at performance-based LOS can be obtained after two additional trials after the learning phase (0.69 > ICC > 1.0).</p> <p>Conclusion</p> <p>Establishing performance-based LOS instead of relying on estimations of theoretical LOS offers a more individualized and realistic insight on the true LOS of an individual. Performance-based LOS can be used as targets during weight-shifting postural tasks with real time visual feedback of the COP displacement to assess postural stability of community-living older adults. In order to obtain reliable results, a learning phase allowing subjects to learn how to control their COP displacement is needed.</p
