Toll-like receptor 9 modifies intestinal serotonergic system.

Introduction: Toll-like receptor 9 (TLR9) is expressed in intestinal epithelial cells, which recognize microbiota developing different responses 1. Several studies have shown that TLR9 seems to be involved in Inflammatory Bowel Diseases (IBD) due to an inappropriate defensive response against microorganisms 2. Moreover, intestinal serotonergic system is also altered in IBD, where extracellular serotonin (5–HT) levels are increased 3. 5-HT bioavailability is mainly regulated by the serotonin transporter (SERT), expressed in enterocytes 4. Aims & Methods: The aim of the present study was to analyse whether TLR9 activation affects SERT expression and activity, and expression of other elements from the serotonergic system (TPH1, TPH2 and 5-HT receptors). Human enterocyte-like Caco-2 cells, and ileum and colon from TLR9-/- mice and Dextran Sulphate Sodium (DSS) mouse colitis model were used as experimental models. mRNA expression was determined by RT-qPCR, and protein expression by western blot..

Toll-like receptors 2 and 4 cooperate in the control of the emerging pathogen Brucella microti

Toll-like receptors (TLRs) recognize pathogen-derived molecules and play a critical role during the host innate and adaptive immune response. Brucella spp. are intracellular gram-negative bacteria including several virulent species, which cause a chronic zoonotic infection in a wide range of mammalian hosts known as brucellosis. A new Brucella species, Brucella microti, was recently isolated from wild rodents and found to be highly pathogenic in mice. Using this species-specific model, it was previously found that CD8+ T cells are required to control this infection. In order to find out the role of TLR-mediated responses in the control of this pathogen, the course of infection of B. microti was analyzed over 3 weeks in wild-type (WT) and TLR knock out (KO) mice including TLR2-/-, TLR4-/-, TLR9-/-, TLR2×4-/- and TLR2×4×9-/-. WT and single TLR2, TLR4 and TLR9 KO mice similarly control infection in liver and spleen. In contrast, bacterial clearance was delayed in TLR2×4-/- and TLR2×4×9-/- mice at 7 and 14 days post-infection. This defect correlated with impaired maturation and pro-inflammatory cytokine production in B. microti-infected dendritic cells from TLR2×4-/- and TLR2×4×9-/- mice. Finally, it was found that Tc cells from TLR2×4-/- and TLR2×4×9-/- mice showed reduced ability to inhibit growth of B. microti in macrophages, suggesting the involvement of TLR2 and 4 in the generation of specific Tc cells. Our findings indicate that TLR2 and TLR4 are required to control B. microti infection in mice and that this effect could be related to its participation in the maturation of dendritic cells and the generation of specific CD8+ Tc cells

Reaction rate for carbon burning in massive stars

    Get PDF
Carbon burning is a critical phase for nucleosynthesis in massive stars. The conditions for igniting this burning stage, and the subsequent isotope composition of the resulting ashes, depend strongly on the reaction rate for C12+C12 fusion at very low energies. Results for the cross sections for this reaction are influenced by various backgrounds encountered in measurements at such energies. In this paper, we report on a new measurement of C12+C12 fusion cross sections where these backgrounds have been minimized. It is found that the astrophysical S factor exhibits a maximum around Ecm=3.5-4.0 MeV, which leads to a reduction of the previously predicted astrophysical reaction rate

In this paper we present evidence of anthropophagic behaviour amongst hunter-gatherer groups of the Mediterranean Mesolithic. Thirty human remains have been found in the Mesolithic levels of the Santa Maira Caves. As well as describing the anthropogenic marks identified, this paper contextualizes them within the archaeological context and subsistence patterns described for Mesolithic groups in the region, and hypothesizes about their intentionality

Fusion measurements of 12C+12C at energies of astrophysical interest

The cross section of the 12C+12C fusion reaction at low energies is of paramount importance for models of stellar nucleosynthesis in different astrophysical scenarios, such as Type Ia supernovae and Xray superbursts, where this reaction is a primary route for the production of heavier elements. In a series of experiments performed at Argonne National Laboratory, using Gammasphere and an array of Silicon detectors, measurements of the fusion cross section of 12C+12C were successfully carried out with the γ and charged-particle coincidence technique in the center-of-mass energy range of 3-5 MeV. These were the first background-free fusion cross section measurements for 12C+12C at energies of astrophysical interest. Our results are consistent with previous measurements in the high-energy region; however, our lowest energy measurement indicates a fusion cross section slightly lower than those obtained with other techniques

The technological advance and consequent incorporation of IT in the teaching field has entailed the appearance of new tools to facilitate teachers the process of monitoring and evaluating students. First direct observation of enhanced octupole collectivity in 146Ba

The octupole strength present in the neutron-rich, radiocative nucleus 146Ba has been experimentally determined for the first time using Coulomb excitation. To achieve this, A=146 fission fragments from CARIBU were post-accelerated by the Argonne Tandem Linac Accelerator System (ATLAS) and impinged on a thin 208Pb target. Using the GRETINA γ-ray spectrometer and the CHICO2 heavy-ion counter, the reduced transition probability B(E3; 3-→0+) was determined as 48(+21-29) W.u. The new result provides further experimental evidence for the presence of a region of octupole deformation surrounding the neutron-rich barium isotopes

Calculated phase diagrams, iron tolerance limits, and corrosion of Mg-Al alloys

The factors determining corrosion are reviewed in this paper, with an emphasis on iron tolerance limit and the production of high-purity castings. To understand the iron impurity tolerance limit, magnesium phase diagrams were calculated using the Pandat software package. Calculated phase diagrams can explain the iron tolerance limit and the production of high-purity castings by means of control of melt conditions; this is significant for the production of quality castings from recycled magnesium. Based on the new insight, the influence of the microstructure on corrosion of magnesium alloys is reviewed
