Adolescent Sexuality and Culture: South African Mothers` Perspective
This study explores South African mothers’ perspective on adolescent sexuality within their cultural prism. Forty-four (44) mothers were purposefully selected for the study. Data was collected through six focus group discussions. The results indicated that culturally embedded reasons inhibited mothers from effectively imparting knowledge about sexuality to their daughters.Insufficient information, lack of role models and fear of sensitising their daughters to sexuality issues were other inhibitory factors.Keywords: Adolescent, sexuality, mothers, HIV and AIDS, sexual behaviour, culture
Edge-functionalized and substitutional doped graphene nanoribbons: electronic and spin properties
Graphene nanoribbons are the counterpart of carbon nanotubes in
graphene-based nanoelectronics. We investigate the electronic properties of
chemically modified ribbons by means of density functional theory. We observe
that chemical modifications of zigzag ribbons can break the spin degeneracy.
This promotes the onset of a semiconducting-metal transition, or of an
half-semiconducting state, with the two spin channels having a different
bandgap, or of a spin-polarized half-semiconducting state -where the spins in
the valence and conduction bands are oppositely polarized. Edge
functionalization of armchair ribbons gives electronic states a few eV away
from the Fermi level, and does not significantly affect their bandgap. N and B
produce different effects, depending on the position of the substitutional
site. In particular, edge substitutions at low density do not significantly
alter the bandgap, while bulk substitution promotes the onset of
semiconducting-metal transitions. Pyridine-like defects induce a

UV-B Physiological Changes Under Conditions of Distress and Eustress in Sweet Basil
UV-B radiation has been previously reported to induce protective or deleterious effects on plants depending on the UV-B irradiation doses. To elucidate how these contrasting events are physiologically coordinated, we exposed sweet basil plants to two UV-B doses: low (8.5 kJ m-2 day-1, 30 min exposure) and high (68 kJ m-2 day-1, 4 h exposure), with the plants given both doses once continuously in a single day. Physiological tests during and after both UV-B exposures were performed by comparing the stress-induced damage and adverse effects on photosynthetic activity, the concentration and composition of photosynthetic and non-photosynthetic pigments, and stress-related hormones biosynthesis in basil plants. Our results showed that upon receiving a high UV-B dose, a severe inactivation of oxygen evolving complex (OEC) activity at the PSII donor side and irreversible PSII photodamage caused primarily by limitation of the acceptor side occurred, which overloaded protective mechanisms and finally led to the death of the plants. In contrast, low UV-B levels did not induce any signs of UV-B stress injuries. The OEC partial limitation and the inactivation of the electron transport chain allowed the activation of photoprotective mechanisms, avoiding irreversible damage to PSII. Overall results indicate the importance of a specific response mechanisms regulating photoprotection vs irreversible photoinhibition in basil that were modulated depending on the UV-B doses
A temperature dependent virus binding assay reveals the presence of neutralising antibodies in human cytomegalovirus gB vaccine recipients’ sera
Human cytomegalovirus (HCMV) remains an important cause of mortality in immune-compromised transplant patients and following congenital infection. Such is the burden, an effective vaccine strategy is considered to be of the highest priority. The most successful vaccines to date have focused on generating immune responses against glycoprotein B (gB) – a protein essential for HCMV fusion and entry. We have previously reported that an important component of the humoral immune response elicited by gB/MF59 vaccination of patients awaiting transplant is the induction of non-neutralizing antibodies that target cell-associated virus with little evidence of concomitant classical neutralizing antibodies. Here we report that a modified neutralization assay that promotes prolonged binding of HCMV to the cell surface reveals the presence of neutralizing antibodies in sera taken from gB-vaccinated patients that cannot be detected using standard assays. We go on to show that this is not a general feature of gB-neutralizing antibodies, suggesting that specific antibody responses induced by vaccination could be important. Although we can find no evidence that these neutralizing antibody responses are a correlate of protection in vivo in transplant recipients their identification demonstrates the utility of the approach in identifying these responses. We hypothesize that further characterization has the potential to aid the identification of functions within gB that are important during the entry process and could potentially improve future vaccine strategies directed against gB if they prove to be effective against HCMV at higher concentrations
The residues of fruit and vegetable pro-cessing: From “waste” to “resource” of natural phytochemical compounds
The project of Sant’Anna School, in line with the Italian legislation on limiting waste and promoting the redistribution of surpluses and unused goods, aimed to study the potential healthy value of residues obtained from the trans-formation of fruit and vegetable products that represent a cost, as they must be handled, stored and disposed according to stringent actual regulations. Two “model” species (potato and apple) were considered to test the possibility of using industrial processing waste for food applications. The extracts, obtained with “green” methods from potato and apple peels, were evaluated as natural antioxidants in the preparation of minimally processed freshcut apple. Results suggest the possibility to use these novel byproduct extracts as valuable alter-native treatments to traditional chemical additives employed for minimally processed apples
Case report of an atypical early onset X-linked retinoschisis in monozygotic twins
Background: X-linked Retinoschisis (XLRS) is one of the most common macular degenerations in young males, with a worldwide prevalence ranging from 1:5000 to 1:20000. Clinical diagnosis of XLRS can be challenging due to the highly variable phenotypic presentation and limited correlation has been identified between mutation type and disease severity or progression.
Case presentation: We report the atypical early onset of XLRS in 3-month-old monozygotic twins. Fundus examination was characterized by severe bullous retinal schisis with pre-retinal and intraretinal haemorrhages. Molecular genetic analysis of the RS1 was performed and the c.288G > A (p. Trp96Ter) mutation was detected in both patients.
Conclusions: Early onset XLRS is associated with a more progressive form of the disease, characterized by large bullous peripheral schisis involving the posterior pole, vascular abnormalities and haemorrhages. The availability of specific technology permitted detailed imaging of the clinical picture of unusual cases of XLRS. The possible relevance of modifying genes should be taken into consideration for the future development of XLRS gene therapy
Seronegative patients vaccinated with cytomegalovirus gB-MF59 vaccine have evidence of neutralising antibody responses against gB early post-transplantation
Background
Human cytomegalovirus (HCMV) causes a ubiquitous infection which can pose a significant threat for immunocompromised individuals, such as those undergoing solid organ transplant (SOT). Arguably, the most successful vaccine studied to date is the recombinant glycoprotein-B (gB) with MF59 adjuvant which, in 3 Phase II trials, demonstrated 43–50% efficacy in preventing HCMV acquisition in seronegative healthy women or adolescents and reduction in virological parameters after SOT. However, the mechanism of vaccine protection in seronegative recipients remains undefined.
Methods
We evaluated samples from the cohort of seronegative SOT patients enroled in the Phase II glycoprotein-B/MF59 vaccine trial who received organs from seropositive donors. Samples after SOT (0–90 days) were tested by real-time quantitative PCR for HCMV DNA. Anti-gB antibody levels were measured by ELISA. Neutralization was measured as a decrease in infectivity for fibroblast cell cultures revealed by expression of immediate-early antigens.
Findings
Serological analyses revealed a more rapid increase in the humoral response against gB post transplant in vaccine recipients than in those randomised to receive placebo. Importantly, a number of patient sera displayed HCMV neutralising responses – neutralisation which was abrogated by pre-absorbing the sera with recombinant gB.
Interpretation
We hypothesise that the vaccine primed the immune system of seronegative recipients which, when further challenged with virus at time of transplant, allowed the host to mount rapid immunological humoral responses even under conditions of T cell immune suppression during transplantation
Nutrient sensor O-GlcNAc transferase controls cancer lipid metabolism via SREBP-1 regulation
Elevated O-GlcNAcylation is associated with disease states such as diabetes and cancer. O-GlcNAc transferase (OGT) is elevated in multiple cancers and inhibition of this enzyme genetically or pharmacologically inhibits oncogenesis. Here we show that O-GlcNAcylation modulates lipid metabolism in cancer cells. OGT regulates expression of the master lipid regulator the transcription factor sterol regulatory element binding protein 1 (SREBP-1) and its transcriptional targets both in cancer and lipogenic tissue. OGT regulates SREBP-1 protein expression via AMP-activated protein kinase (AMPK). SREBP-1 is critical for OGT-mediated regulation of cell survival and of lipid synthesis, as overexpression of SREBP-1 rescues lipogenic defects associated with OGT suppression, and tumor growth in vitro and in vivo. These results unravel a previously unidentified link between O-GlcNAcylation, lipid metabolism and the regulation of SREBP-1 in cancer and suggests a crucial role for O-GlcNAc signaling in transducing nutritional state to regulate lipid metabolism
