    Temporal development and collapse of an Arctic plant-pollinator network

    <p>Abstract</p> <p>Background</p> <p>The temporal dynamics and formation of plant-pollinator networks are difficult to study as it requires detailed observations of how the networks change over time. Understanding the temporal dynamics might provide insight into sustainability and robustness of the networks and how they react to environmental changes, such as global warming. Here we study an Arctic plant-pollinator network in two consecutive years using a simple mathematical model and describe the temporal dynamics (daily assembly and disassembly of links) by random mechanisms.</p> <p>Results</p> <p>We develop a mathematical model with parameters governed by the probabilities for entering, leaving and making connections in the network and demonstrate that A. The dynamics is described by very similar parameters in both years despite a strong turnover in the composition of the pollinator community and different climate conditions, B. There is a drastic change in the temporal behaviour a few days before the end of the season in both years. This change leads to the collapse of the network and does not correlate with weather parameters, C. We estimate that the number of available pollinator species is about 80 species of which 75-80% are observed in each year, D. The network does not reach an equilibrium state (as defined by our model) before the collapse set in and the season is over.</p> <p>Conclusion</p> <p>We have shown that the temporal dynamics of an Arctic plant-pollinator network can be described by a simple mathematical model and that the model allows us to draw biologically interesting conclusions. Our model makes it possible to investigate how the network topology changes with changes in parameter values and might provide means to study the effect of climate on plant-pollinator networks.</p

    Berry size variability in Vitis vinifera L.

    In order to study the mechanisms of berry growth in Vitis vinifera L. we analysed cell division and pericarp enlargement of 6 genotypes showing high variability in berry size (range: 49-90 %. Both, cell number and cell volume were involved in the small-sized berries of the flb mutant producing wild-type-like berries. The differences of berry size observed in other small-sized clones were only due to cell expansion. The data suggest that the variability of berry size in V. vinifera cultivars predominantly results from modification of cell enlargement. Conversely, in wild-type-like berries, the variability of berry size could result from both, cell division and cell enlargement.

    Report of an early case of Shy-Drager syndrome in a 67 year-old woman patient. Autonomic failure was diagnosed by functional evaluation as well as laboratory tests. MR imaging disclosed a prominent putamina hypodensity in T2-weighted images at high field strength due to iron increased depositing in this basal ganglia. MR imaging evidences confirm Shy-Drager syndrome diagnosis, and contributes for differential diagnosis of idiopathic hypotension (pure autonomic failure) in special in SDS early cases.É relatado o caso de unm paciente de 67 anos de idade com quadro inicial da síndrome de Shy-Drager. O diagnóstico foi possível por provas funcionais autonômicas e exames laboratoriais. A ressonância magnética cerebral (contraste baseado na densidade de prótons e em T2) objetivou proeminente hipodensidade putaminal em T2, secundária ao aumento do depósito do ferro nesta região. Esse achado da RM confirma o diagnóstico da síndrome de Shy-Drager e permite diferenciá-la da hipotensão ortostática idiopática, particularmente na fase de início da SSD quando os sinais de comprometimento do SNC são discretos ou estão ausentes.34234

    Cellular localisation of VvRops and VvRabA5e, small GTPases developmentally regulated in grape berries

    VvRops, in particular VvRop9, and VvRabA5e are small GTPases which are developmentally regulated in grape berries. In an attempt to help elucidate the role of these proteins during fruit development and ripening, we investigated their localisation in the fruit by immunocytofluorescence. These proteins were observed at a perinuclear location, at cell periphery and around vesicles. In particular VvRops were found to be located in the nucleus and likely on the plasma membrane. VvRop9 and VvRabA5e cDNAs were introduced separately into S. cerevisiae mutants with RHO1 and YPT31/YPT32 defective genes respectively. Neither cDNAs could complement these temperature-sensitive mutants, suggesting that the functions of the VvRop9 and VvRabA5e genes in grapevine likely differ from the functions of RHO1 and YPT31/YPT32 genes in yeast.

    MuSCA: A multi-scale source-sink carbon allocation model to explore carbon allocation in plants. An application to static apple tree structures

    Background and aims: Carbon allocation in plants is usually represented at a topological scale, specific to each model. This makes the results obtained with different models, and the impact of their scales of representation, difficult to compare. In this study, we developed a multi-scale carbon allocation model (MuSCA) that allows the use of different, user-defined, topological scales of a plant, and assessment of the impact of each spatial scale on simulated results and computation time. Methods: Model multi-scale consistency and behaviour were tested on three realistic apple tree structures. Carbon allocation was computed at five scales, spanning from the metamer (the finest scale, used as a reference) up to first-order branches, and for different values of a sap friction coefficient. Fruit dry mass increments were compared across spatial scales and with field data. Key Results: The model was able to represent effects of competition for carbon assimilates on fruit growth. Intermediate friction parameter values provided results that best fitted field data. Fruit growth simulated at the metamer scale differed of ~1 % in respect to results obtained at growth unit scale and up to 60 % in respect to first order branch and fruiting unit scales. Generally, the coarser the spatial scale the more predicted fruit growth diverged from the reference. Coherence in fruit growth across scales was also differentially impacted, depending on the tree structure considered. Decreasing the topological resolution reduced computation time by up to four orders of magnitude. Conclusions: MuSCA revealed that the topological scale has a major influence on the simulation of carbon allocation. This suggests that the scale should be a factor that is carefully evaluated when using a carbon allocation model, or when comparing results produced by different models. Finally, with MuSCA, trade-off between computation time and prediction accuracy can be evaluated by changing topological scales

    Exploring the isopycnal mixing and helium–heat paradoxes in a suite of Earth system models

    This paper uses a suite of Earth system models which simulate the distribution of He isotopes and radiocarbon to examine two paradoxes in Earth science, each of which results from an inconsistency between theoretically motivated global energy balances and direct observations. The helium–heat paradox refers to the fact that helium emissions to the deep ocean are far lower than would be expected given the rate of geothermal heating, since both are thought to be the result of radioactive decay in Earth's interior. The isopycnal mixing paradox comes from the fact that many theoretical parameterizations of the isopycnal mixing coefficient ARedi that link it to baroclinic instability project it to be small (of order a few hundred m² s⁻¹) in the ocean interior away from boundary currents. However, direct observations using tracers and floats (largely in the upper ocean) suggest that values of this coefficient are an order of magnitude higher. Helium isotopes equilibrate rapidly with the atmosphere and thus exhibit large gradients along isopycnals while radiocarbon equilibrates slowly and thus exhibits smaller gradients along isopycnals. Thus it might be thought that resolving the isopycnal mixing paradox in favor of the higher observational estimates of ARedi might also solve the helium paradox, by increasing the transport of mantle helium to the surface more than it would radiocarbon. In this paper we show that this is not the case. In a suite of models with different spatially constant and spatially varying values of ARedi the distribution of radiocarbon and helium isotopes is sensitive to the value of ARedi. However, away from strong helium sources in the southeastern Pacific, the relationship between the two is not sensitive, indicating that large-scale advection is the limiting process for removing helium and radiocarbon from the deep ocean. The helium isotopes, in turn, suggest a higher value of ARedi below the thermocline than is seen in theoretical parameterizations based on baroclinic growth rates. We argue that a key part of resolving the isopycnal mixing paradox is to abandon the idea that ARedi has a direct relationship to local baroclinic instability and to the so-called "thickness" mixing coefficient AGM

    The GCC repeat length in the 5'UTR of MRP1 gene is polymorphic: a functional characterization of its relevance for cystic fibrosis

    BACKGROUND: Among the members of the ATP binding cassette transporter superfamily, MRPs share the closest homology with the CFTR protein, which is defective in CF disease. MRP1 has been proposed as a potential modifier gene and/or as novel target for pharmacotherapy of CF to explain the clinical benefits observed in some CF patients treated with the macrolide AZM. The 5'UTR of the MRP1 gene contains a GCC triplet repeat that could represent a polymorphic site and affect the activity of the promoter. METHODS: The MRP1 5' flanking region was amplified by PCR from 36 CF patients and 100 non-CF subjects and the number of GCC triplets of each allele was determined by sequence and electrophoretic analysis. We performed gene reporter studies in CF airway epithelial cells 16HBE14o-AS3, in basal conditions and in the presence of AZM. RESULTS: We found that the GCC repeat is polymorphic, ranging from 7 to 14 triplets either in CF or in non-CF subjects. Our data are preliminary and have to be confirmed on a larger population of CF subjects. The transcriptional activity of the proximal MRP1 5' regulatory region revealed no statistically significant correlations between the number of repeats and treatment with AZM. CONCLUSION: We identified a novel polymorphism in the 5'UTR of MRP1 gene that provides multiple alleles in a gene relevant for multidrug resistance as well as for CF, determining that this region is transcriptionally active and that this activity does not appear to be influenced by AZM treatment

    Tribology and its growing use towards the study of food oral processing and sensory perception

    Here we provide a comprehensive review of the knowledge base of soft tribology, the study of friction, lubrication and wear on deformable surfaces, with consideration for its application towards oral tribology and food lubrication. Studies on 'soft-tribology' have emerged to provide knowledge and tools to predict oral behaviour and assess the performance of foods and beverages. We have shown that there is a now a comprehensive set of fundamental literature, mainly based on soft contacts in the Mini-traction machine with rolling ball on disk configuration, which provides a baseline for interpreting tribological data from complex food systems. Tribology-sensory relationships do currently exist. However, they are restricted to the specific formulations and tribological configuration utilised, and cannot usually be applied more broadly. With a careful and rigorous formulation/experimental design, we envisage tribological tools to provide insights into the sensory perception of foods in combination with other in vitro techniques such as rheology, particle sizing or characterisation of surface interactions. This can only occur with the use of well characterised tribopairs and equipment; a careful characterisation of simpler model foods before considering complex food products; the incorporation of saliva in tribological studies; the removal of confounding factors from the sensory study and a global approach that considers all regimes of lubrication
