    Effect of chemical and thermochemical treatments on the surface properties of commercially pure titanium

    Chemical and physical methods have been generally used for modifying the surface of titanium implants with the aim of achieving better osseointegration. In this work, an evaluation of the effect of different chemicals and thermochemical treatments on the surface modification of titanium was carried out. Plates of commercially pure titanium were polished and treated with four different methods. Scanning electron microscopy, atomic force microscopy, energy-dispersive X-ray spectroscopy, infrared spectroscopy, X-ray diffraction and drop shape analysis were used to characterize the modified titanium surface. Our results showed that the AEPTTT surface exhibited the highest Ra values followed by AEAT, AEPT, polished Ti and AE samples. In addition, the AEAT and AEPTTT treatments developed the largest surface area, while the AE treatment produced the lowest surface area. Furthermore, the AEAT treatment created the most hydrophilic surface, which is known to positively affect the osseointegration rate of dental implants.Keywords: Titanium, surface modification, hydrogen peroxide treatment, alkali treatment, thermochemical treatment.

    Delta-9-tetrahydrocannabinol (THC), the main psychoactive compound of marijuana, induces numerous undesirable effects, including memory impairments, anxiety, and dependence. Conversely, THC also has potentially therapeutic effects, including analgesia, muscle relaxation, and neuroprotection. However, the mechanisms that dissociate these responses are still not known. Using mice lacking the serotonin receptor 5-HT2A, we revealed that the analgesic and amnesic effects of THC are independent of each other: while amnesia induced by THC disappears in the mutant mice, THC can still promote analgesia in these animals. In subsequent molecular studies, we showed that in specific brain regions involved in memory formation, the receptors for THC and the 5-HT2A receptors work together by physically interacting with each other. Experimentally interfering with this interaction prevented the memory deficits induced by THC, but not its analgesic properties. Our results highlight a novel mechanism by which the beneficial analgesic properties of THC can be dissociated from its cognitive side effects

    Analysis of shared common genetic risk between amyotrophic lateral sclerosis and epilepsy

    Because hyper-excitability has been shown to be a shared pathophysiological mechanism, we used the latest and largest genome-wide studies in amyotrophic lateral sclerosis (n = 36,052) and epilepsy (n = 38,349) to determine genetic overlap between these conditions. First, we showed no significant genetic correlation, also when binned on minor allele frequency. Second, we confirmed the absence of polygenic overlap using genomic risk score analysis. Finally, we did not identify pleiotropic variants in meta-analyses of the 2 diseases. Our findings indicate that amyotrophic lateral sclerosis and epilepsy do not share common genetic risk, showing that hyper-excitability in both disorders has distinct origins

    The Helicobacter pylori Genome Project : insights into H. pylori population structure from analysis of a worldwide collection of complete genomes

    Helicobacter pylori, a dominant member of the gastric microbiota, shares co-evolutionary history with humans. This has led to the development of genetically distinct H. pylori subpopulations associated with the geographic origin of the host and with differential gastric disease risk. Here, we provide insights into H. pylori population structure as a part of the Helicobacter pylori Genome Project (HpGP), a multi-disciplinary initiative aimed at elucidating H. pylori pathogenesis and identifying new therapeutic targets. We collected 1011 well-characterized clinical strains from 50 countries and generated high-quality genome sequences. We analysed core genome diversity and population structure of the HpGP dataset and 255 worldwide reference genomes to outline the ancestral contribution to Eurasian, African, and American populations. We found evidence of substantial contribution of population hpNorthAsia and subpopulation hspUral in Northern European H. pylori. The genomes of H. pylori isolated from northern and southern Indigenous Americans differed in that bacteria isolated in northern Indigenous communities were more similar to North Asian H. pylori while the southern had higher relatedness to hpEastAsia. Notably, we also found a highly clonal yet geographically dispersed North American subpopulation, which is negative for the cag pathogenicity island, and present in 7% of sequenced US genomes. We expect the HpGP dataset and the corresponding strains to become a major asset for H. pylori genomics

    The biomimetic method is used for the deposition of calcium phosphate coatings (Ca - P) on the surface of different biomaterials. However, the application of this method requires long exposure times in order to obtain a suitable layer thickness for its use in medical devices. In this paper, we present a fast approach to obtain apatite coatings on titanium, using a combination of supersaturated calcification solution (SCS) with chemical modification of the titanium surface. Also, it was evaluated the effect of four different surface treatments on the apatite deposition rate. Commercially pure titanium plates were activated by chemical or thermochemical treatments. Then, the activated samples were immersed in a solution with high content of calcium and phosphate ions at 37 ÂșC for 24 h, mimicking the physiological conditions. The coatings were studied by Fourier transform infrared spectroscopy (FTIR), X-ray diffraction (XRD), scanning electron microscopy (SEM) and energy dispersive X-ray spectroscopy (EDX). The use of SCS solutions allowed the formation of crystalline hydroxyapatite coatings within a period of 24 h with a thickness between 1 and 5.3 ”m. Besides, precipitates of hydroxyapatite nanoparticles with a globular configuration, forming aggregates with submicrometer size, were found in SCS solutions

    Grapevine (Vitis vinifera L.) was genetically modified with a construct containing a cDNA insert encoding the stilbene synthase gene (Vst1) from grapevine, under the control of the cauliflower mosaic virus 35S promoter in order to test the potential of over-production of resveratrol to protect plants from fungal attack. Southern blot hybridization and quantitative real-time PCR analysis demonstrated the presence and integration of one copy of exogenous DNA sequences in two grapevine-modified lines. Relative expression of the Vst1 gene in different modified lines was confirmed by using gene-specific quantitative real-time PCR. Compared to the control, the concentration of trans-resveratrol quantified by HPLC was up to 7.5 fold higher in the modified plants. The necrotic lesion size of leaves of intact modified plants inoculated by Botrytis cinerea B05.10 strain was consistently smaller and significantly different (p B 0.05) than in control plants, showing that modified grapevine plants were more resistant to the pathogen than the control plants
