    A representation theorem for integral rigs and its applications to residuated lattices

    We prove that every integral rig in Sets is (functorially) the rig of global sections of a sheaf of really local integral rigs. We also show that this representation result may be lifted to residuated integral rigs and then restricted to varieties of these. In particular, as a corollary, we obtain a representation theorem for pre-linear residuated join-semilattices in terms of totally ordered fibers. The restriction of this result to the level of MV-algebras coincides with the Dubuc-Poveda representation theorem.Comment: Manuscript submitted for publicatio

    De la coexistencia a la exclusión competitiva: ¿Puede la sobrepesca cambiar el resultado de la competencia en rayas (Chondrichthyes, Rajidae)?

    Competition for food could be a major force driving changes in the community structure of skates (Rajidae) subjected to fishing exploitation. Under this hypothesis, small skates are released from competition with larger skates after fishing has depleted the larger species. Here, we compare the abundance patterns of two sympatric skates with similar niches but different life histories, Bathyraja albomaculata (larger and slow-reproducing) and Bathyraja macloviana (smaller and faster-reproducing), before (1971, 1978) and after (1998-2004) a 108% increase in industrial bottom trawling on the southeastern South American shelf in order to test the prediction that B. macloviana should competitively exclude B. albomaculata after the increase in fishing mortality. In 1971 and 1978, there was no relationship between the abundance of both species, indicating that they coexisted over large scales. In 1998-2004, the relationship between the abundances of these skates was bell-shaped, indicating that both species increased in abundance at low densities until peaking, after which B. albomaculata decreased when B. macloviana became more abundant, consistent with resource competition. We tested whether food may be a potential limiting resource by comparing the diet of both species. The two species consumed mostly polychaetes, differing only in the consumption of polychaetes from the family Nephthyidae, which was much higher for B. macloviana. Bathyraja macloviana could replace B. albomaculata at high densities when food resources may become scarce. These results support the hypothesis that competition release is an important factor explaining the changes in skate communities in overexploited areas.La competencia por el alimento podría ser una fuerza importante detrás de los cambios en la estructura de las comunidades de rayas (Rajidae) bajo explotación pesquera. Según esta hipótesis, las rayas pequeñas son liberadas de la competencia por las rayas de mayor tamaño, al disminuir la abundancia de éstas últimas por la pesca. En este trabajo, se comparan los patrones de abundancia de dos rayas simpátricas con nichos similares pero con diferentes historias de vida, Bathyraja albomaculata (mayor y de reproducción lenta) y Bathyraja macloviana (más pequeña y de reproducción más rápida), antes (1971, 1978) y después (1998-2004) de un incremento del 108% en el arrastre de fondo industrial en la plataforma sudeste de América del Sur, para evaluar la predicción que B. macloviana excluiría competitivamente a B. albomaculata después del aumento en la mortalidad por pesca. En 1971 y 1978, no hubo relación entre las abundancias de ambas especies, indicando que, a escalas grandes, coexistían. En 1998-2004, la relación entre las abundancias de ambas especies tuvo forma de campana, indicando que ambas incrementaron su abundancia a densidades bajas hasta alcanzar un máximo, a partir del cual la abundancia de B. albomaculata disminuyó a medida que aumentaba la de B. macloviana, patrón consistente con la competencia por uso de recursos. Se evaluó si el alimento puede ser un recurso limitante mediante la comparación de la dieta de ambas especies. Ambas especies consumieron predominantemente poliquetos y difirieron sólo en el consumo de poliquetos Nephthyidae, mucho mayor en B. macloviana. B. macloviana remplazaría a B. albomaculata a altas densidades, cuando el alimento podría ser escaso.     Metabolites of milk intake: a metabolomic approach in UK twins with findings replicated in two European cohorts

    Purpose: Milk provides a significant source of calcium, protein, vitamins and other minerals to Western populations throughout life. Due to its widespread use, the metabolic and health impact of milk consumption warrants further investigation and biomarkers would aid epidemiological studies.  Methods: Milk intake assessed by a validated food frequency questionnaire was analyzed against fasting blood metabolomic profiles from two metabolomic platforms in females from the TwinsUK cohort (n = 3559). The top metabolites were then replicated in two independent populations (EGCUT, n = 1109 and KORA, n = 1593), and the results from all cohorts were meta-analyzed.  Results: Four metabolites were significantly associated with milk intake in the TwinsUK cohort after adjustment for multiple testing (P < 8.08 × 10−5) and covariates (BMI, age, batch effects, family relatedness and dietary covariates) and replicated in the independent cohorts. Among the metabolites identified, the carnitine metabolite trimethyl-N-aminovalerate (β = 0.012, SE = 0.002, P = 2.98 × 10−12) and the nucleotide uridine (β = 0.004, SE = 0.001, P = 9.86 × 10−6) were the strongest novel predictive biomarkers from the non-targeted platform. Notably, the association between trimethyl-N-aminovalerate and milk intake was significant in a group of MZ twins discordant for milk intake (β = 0.050, SE = 0.015, P = 7.53 × 10−4) and validated in the urine of 236 UK twins (β = 0.091, SE = 0.032, P = 0.004). Two metabolites from the targeted platform, hydroxysphingomyelin C14:1 (β = 0.034, SE = 0.005, P = 9.75 × 10−14) and diacylphosphatidylcholine C28:1 (β = 0.034, SE = 0.004, P = 4.53 × 10−16), were also replicated.  Conclusions: We identified and replicated in independent populations four novel biomarkers of milk intake: trimethyl-N-aminovalerate, uridine, hydroxysphingomyelin C14:1 and diacylphosphatidylcholine C28:1. Together, these metabolites have potential to objectively examine and refine milk-disease associations

    Omega-3 fatty acids correlate with gut microbiome diversity and production of N-carbamylglutamate in middle aged and elderly women

    Omega-3 fatty acids may influence human physiological parameters in part by affecting the gut microbiome. The aim of this study was to investigate the links between omega-3 fatty acids, gut microbiome diversity and composition and faecal metabolomic profiles in middle aged and elderly women. We analysed data from 876 twins with 16S microbiome data and DHA, total omega-3, and other circulating fatty acids. Estimated food intake of omega-3 fatty acids were obtained from food frequency questionnaires. Both total omega-3and DHA serum levels were significantly correlated with microbiome alpha diversity (Shannon index) after adjusting for confounders (DHA Beta(SE) = 0.13(0.04), P = 0.0006 total omega-3: 0.13(0.04), P = 0.001). These associations remained significant after adjusting for dietary fibre intake. We found even stronger associations between DHA and 38 operational taxonomic units (OTUs), the strongest ones being with OTUs from the Lachnospiraceae family (Beta(SE) = 0.13(0.03), P = 8 × 10-7). Some of the associations with gut bacterial OTUs appear to be mediated by the abundance of the faecal metabolite N-carbamylglutamate. Our data indicate a link between omega-3 circulating levels/intake and microbiome composition independent of dietary fibre intake, particularly with bacteria of the Lachnospiraceae family. These data suggest the potential use of omega-3 supplementation to improve the microbiome composition

    Metabolomic profiling to dissect the role of visceral fat in cardiometabolic health

    OBJECTIVE: Abdominal obesity is associated with increased risk of type 2 diabetes (T2D) and cardiovascular disease. The aim of this study was to assess whether metabolomic markers of T2D and blood pressure (BP) act on these traits via visceral fat (VF) mass. METHODS: Metabolomic profiling of 280 fasting plasma metabolites was conducted on 2,401 women from TwinsUK. The overlap was assessed between published metabolites associated with T2D, insulin resistance, or BP and those that were identified to be associated with VF (after adjustment for covariates) measured by dual‐energy X‐ray absorptiometry. RESULTS: In addition to glucose, six metabolites were strongly associated with both VF mass and T2D: lactate and branched‐chain amino acids, all of them related to metabolism and the tricarboxylic acid cycle; on average, 38.5% of their association with insulin resistance was mediated by their association with VF mass. Five metabolites were associated with BP and VF mass including the inflammation‐associated peptide HWESASXX, the steroid hormone androstenedione, lactate, and palmitate. On average, 29% of their effect on BP was mediated by their association with VF mass. CONCLUSIONS: Little overlap was found between the metabolites associated with BP and those associated with insulin resistance via VF mass

    Guanine a-carboxy nucleoside phosphonate (G-a-CNP) shows a different inhibitory kinetic profile against the DNA polymerases of human immunodeficiency virus (HIV) and herpes viruses

    α-Carboxy nucleoside phosphonates (α-CNPs) are modified nucleotides that represent a novel class of nucleotide-competing reverse transcriptase (RT) inhibitors (NcRTIs). They were designed to act directly against HIV-1 RT without the need for prior activation (phosphorylation). In this respect, they differ from the nucleoside or nucleotide RTIs [N(t)RTIs] that require conversion to their triphosphate forms before being inhibitory to HIV-1 RT. The guanine derivative (G-α-CNP) has now been synthesized and investigated for the first time. The (L)-(+)-enantiomer of G-α-CNP directly and competitively inhibits HIV-1 RT by interacting with the substrate active site of the enzyme. The (D)-(−)-enantiomer proved inactive against HIV-1 RT. In contrast, the (+)- and (−)-enantiomers of G-α-CNP inhibited herpes (i.e. HSV-1, HCMV) DNA polymerases in a non- or uncompetitive manner, strongly indicating interaction of the (L)-(+)- and the (D)-(−)-G-α-CNPs at a location different from the polymerase substrate active site of the herpes enzymes. Such entirely different inhibition profile of viral polymerases is unprecedented for a single antiviral drug molecule. Moreover, within the class of α-CNPs, subtle differences in their sensitivity to mutant HIV-1 RT enzymes were observed depending on the nature of the nucleobase in the α-CNP molecules. The unique properties of the α-CNPs make this class of compounds, including G-α-CNP, direct acting inhibitors of multiple viral DNA polymerases
