    Hyperpolarized 13 C magnetic resonance spectroscopy detects toxin-induced neuroinflammation in mice.

    Lipopolysaccharide (LPS) is a commonly used agent for induction of neuroinflammation in preclinical studies. Upon injection, LPS causes activation of microglia and astrocytes, whose metabolism alters to favor glycolysis. Assessing in vivo neuroinflammation and its modulation following therapy remains challenging, and new noninvasive methods allowing for longitudinal monitoring would be highly valuable. Hyperpolarized (HP) 13 C magnetic resonance spectroscopy (MRS) is a promising technique for assessing in vivo metabolism. In addition to applications in oncology, the most commonly used probe of [1-13 C] pyruvate has shown potential in assessing neuroinflammation-linked metabolism in mouse models of multiple sclerosis and traumatic brain injury. Here, we aimed to investigate LPS-induced neuroinflammatory changes using HP [1-13 C] pyruvate and HP 13 C urea. 2D chemical shift imaging following simultaneous intravenous injection of HP [1-13 C] pyruvate and HP 13 C urea was performed at baseline (day 0) and at days 3 and 7 post-intracranial injection of LPS (n = 6) or saline (n = 5). Immunofluorescence (IF) analyses were performed for Iba1 (resting and activated microglia/macrophages), GFAP (resting and reactive astrocytes) and CD68 (activated microglia/macrophages). A significant increase in HP [1-13 C] lactate production was observed at days 3 and 7 following injection, in the injected (ipsilateral) side of the LPS-treated mouse brain, but not in either the contralateral side or saline-injected animals. HP 13 C lactate/pyruvate ratio, without and with normalization to urea, was also significantly increased in the ipsilateral LPS-injected brain at 7 days compared with baseline. IF analyses showed a significant increase in CD68 and GFAP staining at 3 days, followed by increased numbers of Iba1 and GFAP positive cells at 7 days post-LPS injection. In conclusion, we can detect LPS-induced changes in the mouse brain using HP 13 C MRS, in alignment with increased numbers of microglia/macrophages and astrocytes. This study demonstrates that HP 13 C spectroscopy has substantial potential for providing noninvasive information on neuroinflammation

    Upper body motor function and swallowing impairments and its association in survivors of head and neck cancer: A cross-sectional study

    Background Upper body motor function and swallowing may be affected after curative treatment for head and neck cancer. The aims of this study are to compare maximum mouth opening (MMO), temporomandibular dysfunction (TMD), cervical and shoulder active range of motion (AROM) and strength, and swallowing difficulty between survivors of head and neck cancer (sHNC) and healthy matched controls (HMC) and to examine the correlations between these outcomes in sHNC. Methods Thirty-two sHNC and 32 HMC participated on the study. MMO, TMD, cervical and shoulder AROM, cervical and shoulder strength, the SPADI shoulder pain and disability indices, the Eating Assessment Tool (EAT-10) score, swallowing difficulty as determined using a visual analogue scale (VAS), and the location of disturbances in swallowing, were recorded. Results MMO and cervical and shoulder AROM and strength were significantly lower in sHNC, whereas FAI, SPADI score, EAT-10 and VAS were higher. The MMO, TMD, cervical and shoulder AROM, and cervical shoulder strength values showed significant correlations (some direct, others inverse) with one another. Swallowing difficulty was inversely associated with the MMO, cervical AROM and shoulder strength. Conclusion Compared with controls, sHNC present smaller MMO, lower cervical and shoulder AROM, lower cervical and shoulder strength and higher perception of TMD, shoulder pain and disability and swallowing difficulty. sHNC suffer impaired swallowing related to lower MMO, presence of TMD, cervical AROM and shoulder strength values.     Dominantly inherited distal nemaline/cap myopathy caused by a large deletion in the nebulin gene

    We report the first family with a dominantly inherited mutation of the nebulin gene (NEB). This 100kb in-frame deletion encompasses NEB exons 14-89, causing distal nemaline/cap myopathy in a three-generation family. It is the largest deletion characterized in NEB hitherto. The mutated allele was shown to be expressed at the mRNA level and furthermore, for the first time, a deletion was shown to cause the production of a smaller mutant nebulin protein. Thus, we suggest that this novel mutant nebulin protein has a dominant-negative effect, explaining the first documented dominant inheritance of nebulin-caused myopathy. The index patient, a young man, was more severely affected than his mother and grandmother. His first symptom was foot drop at the age of three, followed by distal muscle atrophy, slight hypomimia, high-arched palate, and weakness of the neck and elbow flexors, hands, tibialis anterior and toe extensors. Muscle biopsies showed myopathic features with type 1 fibre predominance in the index patient and nemaline bodies and cap-like structures in biopsies from his mother and grandmother. The muscle biopsy findings constitute a further example of nemaline bodies and cap-like structures being part of the same spectrum of pathological changes. (C) 2019 Elsevier B.V.     Tropical peatlands and their conservation are important in the context of COVID-19 and potential future (zoonotic) disease pandemics.

    The COVID-19 pandemic has caused global disruption, with the emergence of this and other pandemics having been linked to habitat encroachment and/or wildlife exploitation. High impacts of COVID-19 are apparent in some countries with large tropical peatland areas, some of which are relatively poorly resourced to tackle disease pandemics. Despite this, no previous investigation has considered tropical peatlands in the context of emerging infectious diseases (EIDs). Here, we review: (i) the potential for future EIDs arising from tropical peatlands; (ii) potential threats to tropical peatland conservation and local communities from COVID-19; and (iii) potential steps to help mitigate these risks. We find that high biodiversity in tropical peat-swamp forests, including presence of many potential vertebrate and invertebrate vectors, combined, in places, with high levels of habitat disruption and wildlife harvesting represent suitable conditions for potential zoonotic EID (re-)emergence. Although impossible to predict precisely, we identify numerous potential threats to tropical peatland conservation and local communities from the COVID-19 pandemic. This includes impacts on public health, with the potential for haze pollution from peatland fires to increase COVID-19 susceptibility a noted concern; and on local economies, livelihoods and food security, where impacts will likely be greater in remote communities with limited/no medical facilities that depend heavily on external trade. Research, training, education, conservation and restoration activities are also being affected, particularly those involving physical groupings and international travel, some of which may result in increased habitat encroachment, wildlife harvesting or fire, and may therefore precipitate longer-term negative impacts, including those relating to disease pandemics. We conclude that sustainable management of tropical peatlands and their wildlife is important for mitigating impacts of the COVID-19 pandemic, and reducing the potential for future zoonotic EID emergence and severity, thus strengthening arguments for their conservation and restoration. To support this, we list seven specific recommendations relating to sustainable management of tropical peatlands in the context of COVID-19/disease pandemics, plus mitigating the current impacts of COVID-19 and reducing potential future zoonotic EID risk in these localities. Our discussion and many of the issues raised should also be relevant for non-tropical peatland areas and in relation to other (pandemic-related) sudden socio-economic shocks that may occur in future

    Since the hiring of its first Africanist linguist Carleton Hodge in 1964, Indiana University’s Department of Linguistics has had a strong and continuing presence in the study of African languages and linguistics through the work of its faculty and of its graduates on the faculties of many other universities. Research on African linguistics at IU has covered some of the major language groups spoken on the African continent. Carleton Hodge’s work on Ancient Egyptian and Hausa, Paul Newman’s work on Hausa and Chadic languages, and Roxanna Ma Newman’s work on Hausa language structure and pedagogy have been some of the most important studies on Afro-Asiatic linguistics. With respect to Niger-Congo languages, the work of Charles Bird on Bambara and the Mande languages, Robert Botne’s work on Bantu structure (especially tense and aspect), Samuel Obeng and Colin Painter’s work on Ghanaian Languages (phonetics, phonology, and pragmatics), Robert Port’s studies on Swahili, and Erhard Voeltz's studies on Bantu linguistics are considered some of the most influential studies in the sub-field. On Nilo Saharan languages, the work of Tim Shopen on Songhay stands out. IU Linguistics has also forwarded theoretical work on African languages, such as John Goldsmith’s seminal research on tone in African languages. The African linguistics faculty at IU have either founded or edited important journals in African Studies, African languages, and African linguistics, including Africa Today, Studies in African Linguistics, and Journal of African Languages and Linguistics. In 1972, the Indiana University Department of Linguistics hosted the Third Annual Conference of African Linguistics. Proceedings of that conference were published by Indiana University Publications (African Series, vol. 7). In 1986, IU hosted the Seventeenth Annual Conference of African Linguistics with Paul Newman and Robert Botne editing the proceedings in a volume entitled Current Approaches to African Linguistics, vol. 5. In 2016, Indiana University hosted the 48th Annual Conference on African Linguistics with the theme African Linguistics Across the Disciplines. Proceedings of that meeting are published in this volume. The papers presented in this volume reflect the diversity of opportunities for language study in Africa. This collection of descriptive and theoretical work is the fruit of data gathering both in-country and abroad by researchers of languages spoken across the continent, from Sereer-sin in the west to Somali in the northeast to Ikalanga in the south. The range of topics in this volume is also broad, representative of the varied field work in country and abroad that inspires research in African linguistics. This collection of papers spans the disciplines of phonology (both segmental and suprasegmental), morphology (both morphophonological and morphosyntactic), syntax, semantics, and language policy. The data and analyses presented in this volume offer a cross-disciplinary view of linguistic topics from the many under-resourced languages of Africa
