    The stratigraphic basis of the Anthropocene Event

    This paper outlines the stratigraphic basis of a proposed Anthropocene Event. It considers a diachronous event framework to be more appropriate for understanding the Anthropocene than treating it as a new geological series/epoch. Four general categories of material evidence are identified as of particular relevance: ‘artificial’ strata with natural constituents; humanly modified ground; legacy sediments; and ‘natural’ geo-deposits containing artefactual material. All these arise from the interaction and mixing of human, natural, and hybrid human-natural forces. Taken together, such stratigraphic evidence supports the case for recognising the Anthropocene as an unfolding event

    Evaluation of cystatin C for the detection of chronic kidney disease in cats

    BackgroundSerum cystatin C (sCysC) and urinary cystatin C (uCysC) are potential biomarkers for early detection of chronic kidney disease (CKD) in cats. An in-depth clinical validation is required. ObjectivesTo evaluate CysC as a marker for CKD in cats and to compare assay performance of the turbidimetric assay (PETIA) with the previously validated nephelometric assay (PENIA). AnimalsNinety cats were included: 49 CKD and 41 healthy cats. MethodsSerum CysC and uCysC concentrations were prospectively evaluated in cats with CKD and healthy cats. Based on plasma exo-iohexol clearance test (PexICT), sCysC was evaluated to distinguish normal, borderline, and low GFR. Sensitivity and specificity to detect PexICT<1.7mL/min/kg were calculated. Serum CysC results of PENIA and PETIA were correlated with GFR. Statistical analysis was performed using general linear modeling. ResultsCats with CKD had significantly higher meanSD sCysC (1.4 +/- 0.5mg/L) (P<.001) and uCysC/urinary creatinine (uCr) (291 +/- 411mg/mol) (P<.001) compared to healthy cats (sCysC 1.0 +/- 0.3 and uCysC/uCr 0.32 +/- 0.97). UCysC was detected in 35/49 CKD cats. R-2 values between GFR and sCysC or sCr were 0.39 and 0.71, respectively (sCysC or sCr=+GFR+epsilon). Sensitivity and specificity were 22 and 100% for sCysC and 83 and 93% for sCr. Serum CysC could not distinguish healthy from CKD cats, nor normal from borderline or low GFR, in contrast with sCr. ConclusionSerum CysC is not a reliable marker of reduced GFR in cats and uCysC could not be detected in all CKD cats

    Ultrastructural elastic deformation of cortical bone tissue probed by NIR Raman spectroscopy

    Raman spectroscopy is used as a probe of ultrastructural (molecular) changes in both the mineral and matrix (protein and glycoprotein, predominantly type I collagen) components of murine cortical bone as it responds to loading in the elastic regime. At the ultrastructural level, crystal structure and protein secondary structure distort as the tissue is loaded. These structural changes are followed as perturbations to tissue spectra. We load tissue in a custom-made dynamic mechanical tester that fits on the stage of a Raman microprobe and can accept hydrated tissue specimens. As the specimen is loaded in tension and/or compression, the shifts in mineral P-O4v 1 and relative band heights in the Amide III band envelope are followed with the microprobe. Average load is measured using a load cell while the tissue is loaded under displacement control. Changes occur in both the mineral and matrix components of bone as a response to elastic deformation. We propose that the mineral apatitic crystal lattice is deformed by movement of calcium and other ions. The matrix is proposed to respond by deformation of the collagen backbone. Raman microspectroscopy shows that bone mineral is not a passive contributor to tissue strength. The mineral active response to loading may function as a local energy storage and dissipation mechanism, thus helping to protect tissue from catastrophic damage

    Using cloud ice flux to parametrise large-scale lightning

    Lightning is an important natural source of nitrogen oxide especially in the middle and upper troposphere. Hence, it is essential to represent lightning in chemistry transport and coupled chemistry climate models. Using ERA-Interim meteorological reanalysis data we compare the lightning flash density distributions produced using several existing lightning parametrisations, as well as a new parametrisation developed on the basis of upward cloud ice flux at 440 hPa. The use of ice flux forms a link to the non-inductive charging mechanism of thunderstorms. Spatial and temporal distributions of lightning flash density are compared to tropical and subtropical observations for 2007-2011 from the Lightning Imaging Sensor (LIS) on the Tropical Rainfall Measuring Mission (TRMM) satellite. The well-used lightning flash parametrisation based on cloud-top height has large biases but the derived annual total flash density has a better spatial correlation with the LIS observations than other existing parametrisations. A comparison of flash density simulated by the different schemes shows that the cloud-top height parametrisation has many more instances of moderate flash densities and fewer low and high extremes compared to the other parametrisations. Other studies in the literature have shown that this feature of the cloud-top height parametrisation is in contrast to lightning observations over certain regions. Our new ice flux parametrisation shows a clear improvement over all the existing parametrisations with lower root mean square errors (RMSEs) and better spatial correlations with the observations for distributions of annual total, and seasonal and interannual variations. The greatest improvement with the new parametrisation is a more realistic representation of the zonal distribution with a better balance between tropical and subtropical lightning flash estimates. The new parametrisation is appropriate for testing in chemistry transport and chemistry climate models that use a lightning parametrisation

    The impact of lightning on tropospheric ozone chemistry using a new global lightning parametrisation

    A lightning parametrisation based on upward cloud ice flux is implemented in a chemistry–climate model (CCM) for the first time. The UK Chemistry and Aerosols model is used to study the impact of these lightning nitric oxide (NO) emissions on ozone. Comparisons are then made between the new ice flux parametrisation and the commonly used, cloud-top height parametrisation. The ice flux approach improves the simulation of lightning and the temporal correlations with ozone sonde measurements in the middle and upper troposphere. Peak values of ozone in these regions are attributed to high lightning NO emissions. The ice flux approach reduces the overestimation of tropical lightning apparent in this CCM when using the cloud-top approach. This results in less NO emission in the tropical upper troposphere and more in the extratropics when using the ice flux scheme. In the tropical upper troposphere the reduction in ozone concentration is around 5–10 %. Surprisingly, there is only a small reduction in tropospheric ozone burden when using the ice flux approach. The greatest absolute change in ozone burden is found in the lower stratosphere, suggesting that much of the ozone produced in the upper troposphere is transported to higher altitudes. Major differences in the frequency distribution of flash rates for the two approaches are found. The cloud-top height scheme has lower maximum flash rates and more mid-range flash rates than the ice flux scheme. The initial Ox (odd oxygen species) production associated with the frequency distribution of continental lightning is analysed to show that higher flash rates are less efficient at producing Ox; low flash rates initially produce around 10 times more Ox per flash than high-end flash rates. We find that the newly implemented lightning scheme performs favourably compared to the cloud-top scheme with respect to simulation of lightning and tropospheric ozone. This alternative lightning scheme shows spatial and temporal differences in ozone chemistry which may have implications for comparison between models and observations, as well as for simulation of future changes in tropospheric ozone

    3D Computer simulations and experiments are employed to study random packings of compressible spherical grains under external confining stress. Of particular interest is the rigid ball limit, which we describe as a continuous transition in which the applied stress vanishes as (\phi-\phi_c)^\beta, where \phi is the (solid phase) volume density. This transition coincides with the onset of shear rigidity. The value of \phi_c depends, for example, on whether the grains interact via only normal forces (giving rise to random close packings) or by a combination of normal and friction generated transverse forces (producing random loose packings). In both cases, near the transition, the system's response is controlled by localized force chains. As the stress increases, we characterize the system's evolution in terms of (1) the participation number, (2) the average force distribution, and (3) visualization techniques.Comment: 4 pages, 7 figures, to appear in Phys.     A morphological method has been developed allowing measurement of the effect on intestinal epithelia of vincristine. In routinely prepared tissue sections the proportion of mitotic events progressing beyond metaphase is counted by microscopy. When estimated over a range of doses of vincristine this post-metaphase index (PMI) can be used to compare the sensitivity of differing intact tissues. Intestinal tumours were induced in rats by chemical carcinogenesis. Administration of vincristine in the presence or absence of verapamil was performed in these tumour-bearing animals. Sections were prepared from colonic and small-bowel tumours and from normal mucosa. The results show that verapamil increases the sensitivity of the tissues studied to vincristine. A dose dependent effect of verapamil on vincristine sensitisation was demonstrated in colonic tissues. These findings indicate a shared pharmacological property between the resistance of primary tumour tissue and the multidrug-resistance phenotype
