    The Role of Selective Attention in the Positivity Offset: Evidence from Event Related Potentials

    Some research suggests that positive and negative valence stimuli may be processed differently. For example, negative material may capture and hold attention more readily than equally arousing positive material. This is called the negativity bias, and it has been observed as both behavioural and electroencephalographic (EEG) effects. Consequently, it has been attributed to both automatic and elaborative processes. However, at the lowest levels of arousal, faster reaction times and stronger EEG responses to positive material have been observed. This is called the positivity offset, and the underlying cognitive mechanism is less understood. To study the role of selective attention in the positivity offset, participants completed a negative affective priming (NAP) task modified to dissociate priming for positive and negative words. The task required participants to indicate the valence of a target word, while simultaneously ignoring a distractor. In experiment 1, a behavioural facilitation effect (faster response time) was observed for positive words, in stark contrast to the original NAP task. These results were congruent with a previously reported general categorization advantage for positive material. In experiment 2, participants performed the task while EEG was recorded. In additional to replicating the behavioural results from experiment 1, positive words elicited a larger Late Positive Potential (LPP) component on ignored repetition relative to control trials. Surprisingly, negative words elicited a larger LPP than positive words on control trials. These results suggest that the positivity offset may reflect a greater sensitivity to priming effects due to a more flexible attentional set

    Se pretende evaluar, en este estudio, la temperatura alcanzada en los esterilizadores a bolas de vidrio, su fluctuación y su uniformidad en los diferentes puntos del recipiente que contiene las bolas de vidrio. Una vez determinada la temperatura mínima alcanzada por cada uno de ellos, se valora la seguridad en la esterilización del instrumental estandarizado de endodoncia, según las referencias de otros estudios publicados. Se llega a la conclusión de que la fiabilidad de los mismos, en muchos casos, no es la precisa que se desearía y se apuntan normativas que pensamos deberían disponer tales esterilizadores

    A consensus research agenda for optimising nasal drug delivery

    Nasal drug delivery has specific challenges which are distinct from oral inhalation, alongside which it is often considered. The next generation of nasal products will be required to deliver new classes of molecule, e.g. vaccines, biologics and drugs with action in the brain or sinuses, to local and systemic therapeutic targets. Innovations and new tools/knowledge are required to design products to deliver these therapeutic agents to the right target at the right time in the right patients. We report the outcomes of an expert meeting convened to consider gaps in knowledge and unmet research needs in terms of (i) formulation and devices, (ii) meaningful product characterization and modeling, (iii) opportunities to modify absorption and clearance. Important research questions were identified in the areas of device and formulation innovation, critical quality attributes for different nasal products, development of nasal casts for drug deposition studies, improved experimental models, the use of simulations and nasal delivery in special populations. We offer these questions as a stimulus to research and suggest that they might be addressed most effectively by collaborative research endeavors

    An siRNA Screen in Pancreatic Beta Cells Reveals a Role for Gpr27 in Insulin Production

    The prevalence of type 2 diabetes in the United States is projected to double or triple by 2050. We reasoned that the genes that modulate insulin production might be new targets for diabetes therapeutics. Therefore, we developed an siRNA screening system to identify genes important for the activity of the insulin promoter in beta cells. We created a subclone of the MIN6 mouse pancreatic beta cell line that expresses destabilized GFP under the control of a 362 base pair fragment of the human insulin promoter and the mCherry red fluorescent protein under the control of the constitutively active rous sarcoma virus promoter. The ratio of the GFP to mCherry fluorescence of a cell indicates its insulin promoter activity. As G protein coupled receptors (GPCRs) have emerged as novel targets for diabetes therapies, we used this cell line to screen an siRNA library targeting all known mouse GPCRs. We identified several known GPCR regulators of insulin secretion as regulators of the insulin promoter. One of the top positive regulators was Gpr27, an orphan GPCR with no known role in beta cell function. We show that knockdown of Gpr27 reduces endogenous mouse insulin promoter activity and glucose stimulated insulin secretion. Furthermore, we show that Pdx1 is important for Gpr27's effect on the insulin promoter and insulin secretion. Finally, the over-expression of Gpr27 in 293T cells increases inositol phosphate levels, while knockdown of Gpr27 in MIN6 cells reduces inositol phosphate levels, suggesting this orphan GPCR might couple to Gq/11. In summary, we demonstrate a MIN6-based siRNA screening system that allows rapid identification of novel positive and negative regulators of the insulin promoter. Using this system, we identify Gpr27 as a positive regulator of insulin production

    Evaluation of Rehabilitation of Memory in Neurological Disabilities (ReMiND): a randomized controlled trial

    OBJECTIVE:The evidence for the effectiveness of memory rehabilitation is inconclusive. The aim was to compare the effectiveness of two group memory rehabilitation programmes with a self-help group control. DESIGN:Single-blind randomized controlled trial. PARTICIPANTS:Participants with memory problems following traumatic brain injury, stroke or multiple sclerosis were recruited from community settings. INTERVENTIONS:Participants were randomly allocated, in cohorts of four, to compensation or restitution group treatment programmes or a self-help group control. All programmes were manual-based and comprised two individual and ten weekly group sessions. MAIN MEASURES:Memory functions, mood, and activities of daily living were assessed at baseline and five and seven months after randomization. RESULTS:There were 72 participants (mean age 47.7, SD 10.2 years; 32 men). There was no significant effect of treatment on the Everyday Memory Questionnaire (P = 0.97). At seven months the mean scores were comparable (restitution 36.6, compensation 41.0, self-help 44.1). However, there was a significant difference between groups on the Internal Memory Aids Questionnaire (P = 0.002). The compensation and restitution groups each used significantly more internal memory aids than the self-help group (P 0.05). CONCLUSIONS:There results show few statistically significant effects of either compensation or restitution memory group treatment as compared with a self-help group control. Further randomized trials of memory rehabilitation are needed

    Reference Data for the Ruff Figural Fluency Test Stratified by Age and Educational Level

    The Ruff Figural Fluency Test (RFFT) was developed to avoid the difficulties that were encountered in earlier tests of figural fluency. Although the test characteristics of the RFFT seem to be good and it is a valuable addition to neuropsychological assessments, reference data are still scarce. To this aim, we required 2,404 community dwelling persons in Groningen, the Netherlands to perform the RFFT. All 1,651 persons with a complete RFFT and known educational level formed the reference sample. Their age ranged from 35 to 82 years and their educational level from primary school to university grade. Ninety-six percent of the persons were of Western European descent. All tests were analyzed by two independent examiners and subsequently three measures were calculated: number of unique designs, number of perseverative errors and error ratio. The main finding was that performance on the RFFT was dependent on age and educational level. This was not only observed in older persons but also in young and middle-aged persons. Reference data for the three RFFT measures are presented in groups of five years of age ranging from 35–39 years to 75 years or older
