    Hoy en día las empresas de entretenimiento que quieran mejorar su rentabilidad y crecer cada día más y ser mejores que sus competidores, tienen que mejorar sus productos y servicios, ofrecer mayor calidad, en definitiva, satisfacer las necesidades del cliente, de este modo captarlos y mantenerlos, ya que es una oportunidad para crecer rápidamente a través del “boca a boca”. Es dentro de este contexto, que el objetivo de esta investigación fue conocer el nivel de satisfacción del cliente, en la Discoteca Magno S.A.C a través del método o modelo SERVPERF. El enfoque fue cuantitativo y la investigación fue tipo de descriptiva. La población estuvo conformada, por 6600 personas que asistieron los fines de semana (viernes y sábados) a la referida Discoteca. Este es el número de clientes mensuales, al aplicar la fórmula se obtuvo un total de 363 personas encuestadas. En este trabajo se utilizó como instrumento de recolección de datos, el cuestionario de ítem cerrado, basándose en las preguntas planteadas por el método SERVPERF, para su posterior análisis, se utilizaron los programas microsoft word, microsoft excel 2016 y IBM SPSS Statistics 25. Se comprobó que el nivel de satisfacción en la calidad de servicio al cliente, en la Discoteca Mango fue satisfactorio.Tesi

    Se presentan los resultados obtenidos en los estudios de especiación de arsénico realizados sobre el alga comestible Hijiki. Los análisis se realizaron a través de métodos analíticos desarrollados en la UPM (España) y en la Universidad de Graz (Austria). La determinación de especies hidrosolubles se realizó mediante HPLC-(UV)-HG-AFS, previa extracción mediante MAE con agua desionizada. Por otro lado, la identificación y cuantificación de arsenolípidos se llevó a cabo mediante HPLC-ICPMS/ESMS, previa extracción con cloroformo:metanol y purificación mediante SPE

    Single-stranded DNA aptamers as affinity molecules for the rapid, reliable detection of intestinal bacteria are of particular interest to equip health systems with novel robust and cheap diagnostic tools for monitoring the success of supplementation strategies with selected probiotic gut bacteria in the fight against major widespread threats, such as obesity and neurodegenerative diseases. The human gut bacterium Parabacteroides distasonis (P. distasonis) is positively associated with diseases such as obesity, non-alcoholic fatty liver disease and multiple sclerosis with reduced cell counts in these diseases and is thus a promising potential probiotic bacterium for future microbial supplementation. In this paper we report on the evolution of a specific polyclonal aptamer library by the fluorescence based FluCell-SELEX directed against whole cells of P. distasonis that specifically and efficiently binds and labels P. distasonis. The aptamer library showed high binding affinity and was suited to quantitatively discriminate P. distasonis from other prominent gut bacteria also in mixtures. We believe that this library against a promising probiotic bacterium as a prototype may open new routes towards the development of novel biosensors for the easy and efficient quantitative monitoring of microbial abundance in human microbiomes in general

    Chromosomal characterization of Hyla bischoffi and Hyla guentheri (Anura, Hylidae)

    Hyla bischoffi and H. guentheri share some morphological and call characters with the H. pulchella and H. polytaenia groups. The inclusion of these two species in one of these two groups is still controversial. This study showed that both species have 2n = 24 chromosomes, consisting of five metacentric, five submetacentric, and two subtelocentric chromosome pairs. The nucleolus organizer region (NOR) was located on the long arm of chromosome 10, which also presented a block of heterochromatin in both species. The pericentromeric region of all the chromosomes was positively C-banded. There were no conspicuous differences in the karyotypes of these two species, except for an additional heterochromatic C-band on the short arms of chromosome 6 in H. bischoffi. The karyotypes of these two species were very similar to those of the H. pulchella group and indicate that neither species can be excluded from that group

    Differential activation of anti-erythrocyte and anti-DNA autoreactive B lymphocytes by the Yaa mutation

    An as-yet-unidentified mutation, Y-linked autoimmune acceleration (Yaa), is responsible for the accelerated development of lupus-like autoimmune syndrome in mice. In view of a possible role for Yaa as a positive regulator of BCR signaling, we have explored whether the expression of the Yaa mutation affects the development and activation of transgenic autoreactive B cells expressing either 4C8 IgM anti-RBC or Sp6 IgM anti-DNA. In this study, we show that the expression of the Yaa mutation induced a lethal form of autoimmune hemolytic anemia in 4C8 transgenic C57BL/6 mice, likely as a result of activation of 4C8 anti-RBC autoreactive B cells early in life. This was further supported, although indirectly, by increased T cell-independent IgM production in spleens of nontransgenic C57BL/6 mice bearing the Yaa mutation. In contrast, Yaa failed to induce activation of Sp6 anti-DNA autoreactive B cells, consistent with a lack of increased IgM anti-DNA production in nontransgenic C57BL/6 Yaa mice. Our results suggest that Yaa can activate autoreactive B cells in a BCR-dependent manner, related to differences in the form and nature of autoantigens

    Effects of climate change on grassland biodiversity and productivity: the need for a diversity of models

    There is increasing evidence that the impact of climate change on the productivity of grasslands will at least partly depend on their biodiversity. A high level of biodiversity may confer stability to grassland ecosystems against environmental change, but there are also direct effects of biodiversity on the quantity and quality of grassland productivity. To explain the manifold interactions, and to predict future climatic responses, models may be used. However, models designed for studying the interaction between biodiversity and productivity tend to be structurally different from models for studying the effects of climatic impacts. Here we review the literature on the impacts of climate change on biodiversity and productivity of grasslands. We first discuss the availability of data for model development. Then we analyse strengths and weaknesses of three types of model: ecological, process-based and integrated. We discuss the merits of this model diversity and the scope for merging different model types

    A Polyclonal Aptamer Library for the Specific Binding of the Gut Bacterium Roseburia intestinalis in Mixtures with Other Gut Microbiome Bacteria and Human Stool Samples

    Roseburia intestinalis has received attention as a potential probiotic bacterium. Recent studies have demonstrated that changes in its intestinal abundance can cause various diseases, such as obesity, enteritis and atherosclerosis. Probiotic administration or fecal transplantation alter the structure of the intestinal flora, offering possibilities for the prevention and treatment of these diseases. However, current monitoring methods, such as 16S rRNA sequencing, are complex and costly and require specialized personnel to perform the tests, making it difficult to continuously monitor patients during treatment. Hence, the rapid and cost-effective quantification of intestinal bacteria has become an urgent problem to be solved. Aptamers are of emerging interest because their stability, low immunogenicity and ease of modification are attractive properties for a variety of applications. We report a FluCell-SELEX polyclonal aptamer library specific for R. intestinalis isolated after seven evolution rounds, that can bind and label this organism for fluorescence microscopy and binding assays. Moreover, R. intestinalis can be distinguished from other major intestinal bacteria in complex defined mixtures and in human stool samples. We believe that this preliminary evidence opens new avenues towards aptamer-based electronic biosensors as new powerful and inexpensive diagnostic tools for the relative quantitative monitoring of R. intestinalis in gut microbiomes
