Localisation of the human hSuv3p helicase in the mitochondrial matrix and its preferential unwinding of dsDNA
We characterised the human hSuv3p protein belonging to the family of NTPases/helicases. In yeast mitochondria the hSUV3 orthologue is a component of the degradosome complex and participates in mtRNA turnover and processing, while in Caenorhabditis elegans the hSUV3 orthologue is necessary for viability of early embryos. Using immunofluorescence analysis, an in vitro mitochondrial uptake assay and sub‐fractionation of human mitochondria we show hSuv3p to be a soluble protein localised in the mitochondrial matrix. We expressed and purified recombinant hSuv3p protein from a bacterial expression system. The purified enzyme was capable of hydrolysing ATP with a Km of 41.9 µM and the activity was only modestly stimulated by polynucleotides. hSuv3p unwound partly hybridised dsRNA and dsDNA structures with a very strong preference for the latter. The presented analysis of the hSuv3p NTPase/helicase suggests that new functions of the protein have been acquired in the course of evolution
Perceptions of trends in Seychelles artisanal trap fisheries: comparing catch monitoring, underwater visual census and fishers' knowledge
Fisheries scientists and managers are increasingly engaging with fishers' knowledge (FK) to provide novel information and improve the legitimacy of fisheries governance. Disputes between the perceptions of fishers and scientists can generate conflicts for governance, but can also be a source of new perspectives or understandings. This paper compares artisanal trap fishers' reported current catch rates with landings data and underwater visual census (UVC). Fishers' reports of contemporary 'normal' catch per day tended to be higher than recent median landings records. However, fishers' reports of 'normal' catch per trap were not significantly different from the median CPUE calculated from landings data, and reports of 'good' and 'poor' catch rates were indicative of variability observed in landings data. FK, landings and UVC data all gave different perspectives of trends over a ten-year period. Fishers' perceptions indicated greater declines than statistical models fitted to landings data, while UVC evidence for trends varied between sites and according to the fish assemblage considered. Divergence in trend perceptions may have resulted from differences in the spatial, temporal or taxonomic focus of each dataset. Fishers may have experienced and understood behavioural changes and increased fishing power, which may have obscured declines from landings data. Various psychological factors affect memory and recall, and may have affected these memory-based estimates of trends, while different assumptions underlying the analysis of both interview data and conventional scientific data could also have led to qualitatively different trend perceptions. Differing perspectives from these three data sources illustrate both the potential for 'cognitive conflicts' between stakeholders who do not rely on the same data sources, as well as the importance of multiple information sources to understand dynamics of fisheries. Collaborative investigation of such divergence may facilitate learning and improve fisheries governance
Gene Combination Transfer to Block Autoimmune Damage in Transplanted Islets of Langerhans
Islet transplantation therapy would be applicable to a
wider range of diabetic patients if donor islet acceptance
and protection were possible without systemic immunosuppression
of the recipient. To this aim, gene transfer
to isolated donor islets ex vivo is one method that has
shown promise. This study examines the combined effect of
selected immunomodulatory and anti-inflammatory genes
known to extend the functional viability of pancreatic islet
grafts in an autoimmune system. These genes, indoleamine
2,3-dioxygenase (IDO), manganese superoxide dismutase
(MnSOD), and interleukin (IL)-1 receptor antagonist protein
(IRAP), were transferred to isolated NOD donor islets
ex vivo then transplanted to NODscid recipients and evaluated
in vivo after diabetogenic T-cell challenge. The length
of time the recipient remained euglycemic was used to measure
the ability of the transgenes to protect the graft from
autoimmune destruction. Although the results of these cotransfections
gave little evidence of a synergistic relationship,
they were useful to show that gene combinations can be used to more efficiently protect islet grafts from diabetogenic
T cells
Density functional theory calculations of the carbon ELNES of small diameter armchair and zigzag nanotubes: core-hole, curvature and momentum transfer orientation effects
We perform density functional theory calculations on a series of armchair and
zigzag nanotubes of diameters less than 1nm using the all-electron
Full-Potential(-Linearised)-Augmented-Plane-Wave (FPLAPW) method. Emphasis is
laid on the effects of curvature, the electron beam orientation and the
inclusion of the core-hole on the carbon electron energy loss K-edge. The
electron energy loss near-edge spectra of all the studied tubes show strong
curvature effects compared to that of flat graphene. The curvature induced
hybridisation is shown to have a more drastic effect on the
electronic properties of zigzag tubes than on those of armchair tubes. We show
that the core-hole effect must be accounted for in order to correctly reproduce
electron energy loss measurements. We also find that, the energy loss near edge
spectra of these carbon systems are dominantly dipole selected and that they
can be expressed simply as a proportionality with the local momentum projected
density of states, thus portraying the weak energy dependence of the transition
matrix elements. Compared to graphite, the ELNES of carbon nanotubes show a

Healthcare disparities for women hospitalised with myocardial infarction and angina
Ischaemic heart disease persists as the leading global cause of death. Myocardial infarction (MI) accounts for a large proportion of death due to cardiovascular disease. Between 2007 and 2016, age-sex standardised mortality for MI in Scotland has fallen by 42.5% from 129 to 74 per 100,000 population – a trend also apparent in other countries. Despite improvements in survival, considerable disparities exist according to sexin terms of delivery of guideline-recommended treatments and outcomes following MI suggesting women may be disadvantaged. Use of high-sensitivity troponin assays with sex-specific thresholds increases the detection of MI in women. However, women are less likely to undergo percutaneous coronary revascularisation (PCI) and are more often subject to underutilisation of evidence-based secondary preventative pharmacotherapy. Differences in adoption of invasive management may, in part, be explained by a perception held by clinicians and patients that outcomes are worse for women receiving PCI, as well as differences in symptoms and baseline risk profile which may impact clinical decision-making. Adverse events post-MI, including cardiogenic shock, heart failure and death, remain more common in women than in men, most notably in those with ST-elevation myocardial infarction (STEMI). Whether sex remains an independent predictor of adverse events despite adjustments for the higher risk-profile of women, notably age, is less clear. We hypothesised that sex-related differences in demographics and comorbidity underpin disparities in management and outcomes of women and men hospitalised with MI or angina. We investigated this hypothesis by analysis of a contemporary secondary care electronic registry (e-Registry) using electronic patient records (EPRs) for patients admitted to a complex regional healthcare network.PostprintPeer reviewe

Incorporating the effect of weather in construction scheduling and management with sine wave curves: application in the United Kingdom
The impact of (adverse) weather is a common cause of delays, legal claims and economic losses in construction projects. Research has recently been carried out aimed at incorporating the effect of weather in project planning; but these studies have focussed on either a narrow set of weather variables, or a very limited range of construction activities or projects. A method for processing a country’s historical weather data into a set of weather delay maps for some representative standard construction activities is proposed. Namely, sine curves are used to associate daily combinations of weather variables to delay and provide coefficients for expected productivity losses. A complete case study comprising the construction of these maps and the associated sine waves for the UK is presented along with an example of their use in building construction planning. Findings of this study indicate that UK weather extends project durations by an average
of 21%. However, using climatological data derived from weather observations when planning could lead to average reductions in project durations of 16%, with proportional reductions in indirect and overhead costs
Analysis of the capacity of google trends to measure interest in conservation topics and the role of online news
With the continuous growth of internet usage, Google Trends has emerged as a source of information to investigate how social trends evolve over time. Knowing how the level of interest in conservation topics--approximated using Google search volume--varies over time can help support targeted conservation science communication. However, the evolution of search volume over time and the mechanisms that drive peaks in searches are poorly understood. We conducted time series analyses on Google search data from 2004 to 2013 to investigate: (i) whether interests in selected conservation topics have declined and (ii) the effect of news reporting and academic publishing on search volume. Although trends were sensitive to the term used as benchmark, we did not find that public interest towards conservation topics such as climate change, ecosystem services, deforestation, orangutan, invasive species and habitat loss was declining. We found, however, a robust downward trend for endangered species and an upward trend for ecosystem services. The quantity of news articles was related to patterns in Google search volume, whereas the number of research articles was not a good predictor but lagged behind Google search volume, indicating the role of news in the transfer of conservation science to the public
