    Mapping and valuation of ecosystems and economic activities along the coast of Cameroon: implications of future sea level rise.

    The vulnerability of the coastal zone of Cameroon to flooding from sea level rise (SLR) was quantified using Geographic Information System (GIS) flooding analysis. The main economic activities and ecosystems along this area were iden tified using secondary data. Valuations of non- market values of ecosystems were based on the ecosystem service product method. The low-lying coastal areas were found to be physically and socio-economically susceptible to impacts of SLR due to their high ecological and economic value. A digitised land use/land cover (LULC) classification was produced from low resolution topographic maps and Google Earth images of the area. The digital elevation model (DEM) used was acquired by the shuttle radar topography mission. Evaluation of potential land loss due to inundation was based on empirical approaches using minimum and maximum scenarios of 2 and 10 m flooding. These were estimated considering the best available SLR data for the area, mean high water levels and wave heights during storms. The estimated SLR range from 2.3 m to 9.2 m for the low and high scenarios, respectively, by 2050 and from 2.6 m to 9.7 m for the low and high scenarios, respectively, by the year 2100. Results indicate that 112 km 2 (1.2 %) and 1,216 km 2 (12.6 %) of the coastal area will be lost from a 2 m (equivalent to a low scenario by 2050) and 10 m (equivalent to a high scenario by 2100) flooding, respectively. 0.3 % to 6.3 % of ecosystems worth US$ 12.13 billion/yr could be at risk of flooding by the years 2050 and 2100. The areas under a serious threat cont ain mangroves, sea and airport, residential and industrial areas of Douala. Main plantation crops of banana and palms will be slightly affected. The identification of the soci o-economic impacts of projected SLR on vulnerable coastlines and populations is important for timely actions to be taken in mitigating the effects of natural disasters in the coastal zone

    Stakeholder Perception of Global Warming, Rainfall Variability and Sea Level Rise Hazard Perils in Three Coastal Districts of Douala-Cameroon

    Present day climate change scientist fully embrace the concept of global warming and have repeatedly pointed out that a solution to global warming problem requires studies on climate change education, awareness and proper understanding of causes and impacts. The present study was designed to assess the level of knowledge and awareness of inhabitants of three sub divisions of Douala on the impacts of SLR hazards. 1200 questionnaires were administered to inhabitants of Douala I, IV and V by stratified purposive sampling technique. The Chi square (X2) statistical tests were performed to compare responses to survey questions across sub divisions as well as across demographic groups. The Cramer’s V test (nonparametric correlation) was performed to detect strength of associations of causes and impacts of global warming, rainfall variability, sea level rise perception and social determinants. Correlates of perception of global warming, rainfall variability, sea level rise and water quality parameters and environmental hazards were also assessed. Results show that awareness and perception to global warming, yearly variation of rainfall and knowledge of sea level rise were highly significant (p˂0.000) across districts. The Douala V inhabitants (37.8 %) were more aware of global warming and bothered about its impact than Douala IV and I respondents. Douala V respondents perceived that human beings are linked to the causes of global warming, rainfall variability and sea level rise than Douala I and IV. The Douala IV respondents were more knowledgeable in terms of SLR (37 %) than Douala I and V respondents. Impacts of exaggerated heat wave and increase in floods had significant differences across the district (p˂0.000). Sources of water during the rainy season (rivers/streams, wells, rainfall) were highly significant while differences in the use of water resources for domestic purposes across the three districts were also significant. Flooding and tidal effects were perceived to be the most devastating hazards in Douala I, IV and V as they were highly significant (p˂0.000). Highly significant association (P˂0.01) between the perceived human causes of sea level rise and knowledge of it for Douala I and IV were observed. Rising water tables, inundation, changed colour of water and odour of water as impacts of sea level rise were associated with the taste of water sources. The most devastating hazards were tidal effects and flooding (39 % each) for Douala IV and wave action (44 %) for Douala I. Awareness and knowledge on causes and impacts of climate change were low. A majority of the respondents perceived the will of God as being responsible to climate change causes and impacts on water resources quality in the study area. There is an urgent need for education of inhabitants on causes and impacts of climate change variables that are implicated on water source quality.     Quality and Safety of Beef Produced in Central African Sub-Region

    Aim of this research is to provide a general situation of cattle slaughtered in Cameroon, as a representative example for the Central African Sub-region. The quality and safety of beef from the abattoir of Yaoundé, the largest in Cameroon, were considered. From January 2009 to March 2012, the pre-slaughter conditions and characteristics of 1953 cattle carcasses were recorded, as well as the pH of m. longissimus thoracis 24 h after slaughter. From these carcasses, 60 were selected to represent the bulls slaughtered. The quality parameters and composition of m. longissimus thoracis were carried out. The origin of most of the cattle was the Guinea High Savannah (74.6%), and transhumance was the common production system (75.5%). Gudali (45.6%), White Fulani (33.3%) and Red Mbororo (20.3%) breeds were predominant. Carcass weight was affected by rearing system and cattle category, and it markedly varied during year. Considering meat quality, the fat content was low (1.2%) and similar between breeds, moreover Gudali showed the toughest meat. Of the cows slaughtered, 27% were pregnant and the most common abnormal conditions encountered were ectoparasites, fatigue, lameness, fungal-like skin lesions, enlarged lymph nodes, respiratory distress, nodular lesions. More than 20% of the carcasses had some organs condemned, mainly for liver flukes (5.17%), and 1.0% of them were completely condemned due to tuberculosis, that also affected 3.28% of lungs. These data could aid authorities draw up programmes with the aim to strengthen cattle production, improve beef supply, control and prevent the observed diseases, and promote the regional trade

    Retinal nerve fiber layer thickness predicts CSF amyloid/tau before cognitive decline

    Background: Alzheimer's disease (AD) pathology precedes symptoms and its detection can identify at-risk individuals who may benefit from early treatment. Since the retinal nerve fiber layer (RNFL) is depleted in established AD, we tested whether its thickness can predict whether cognitively healthy (CH) individuals have a normal or pathological cerebrospinal fluid (CSF) A f42 (A) and tau (T) ratio. Methods: As part of an ongoing longitudinal study, we enrolled CH individuals, excluding those with cognitive impairment and significant ocular pathology. We classified the CH group into two sub-groups, normal (CH-NAT, n = 16) or pathological (CH-PAT, n = 27), using a logistic regression model from the CSF AT ratio that identified >85% of patients with a clinically probable AD diagnosis. Spectral-domain optical coherence tomography (OCT) was acquired for RNFL, ganglion cell-inner plexiform layer (GC-IPL), and macular thickness. Group differences were tested using mixed model repeated measures and a classification model derived using multiple logistic regression. Results: Mean age (\ub1 standard deviation) in the CH-PAT group (n = 27; 75.2 \ub1 8.4 years) was similar (p = 0.50) to the CH-NAT group (n = 16; 74.1 \ub1 7.9 years). Mean RNFL (standard error) was thinner in the CH-PAT group by 9.8 (2.7) \u3bcm; p < 0.001. RNFL thickness classified CH-NAT vs. CH-PAT with 87% sensitivity and 56.3% specificity. Conclusions: Our retinal data predict which individuals have CSF biomarkers of AD pathology before cognitive deficits are detectable with 87% sensitivity. Such results from easy-to-acquire, objective and non-invasive measurements of the RNFL merit further study of OCT technology to monitor or screen for early AD pathology

    Simultaneous analysis of free amino acids and biogenic amines in honey and wine samples using in loop orthophthalaldeyde derivatization procedure

    This work presents a RP-HPLC method for the simultaneous quantification of free amino acids and biogenic amines in liquid food matrices and the results of the application to honey and wine samples obtained from different production processes and geographic origins. The developed methodology is based on a pre-column derivatization with o-phthaldialdehyde carried out in the sample injection loop. The compounds were separated in a Nova-Pack RP-C18 column (150 mm × 3.9 mm, 4 μm) at 35 °C. The mobile phase used was a mixture of phase A: 10 mM sodium phosphate buffer (pH 7.3), methanol and tetrahydrofuran (91:8:1); and phase B: methanol and phosphate buffer (80:20), with a flow rate of 1.0 ml/min. Fluorescence detection was used at an excitation wavelength of 335 nm and an emission wavelength of 440 nm. The separation and quantification of 19 amino acids and 6 amines was carried out in a single run as their OPA/MCE derivatives elute within 80 min, ensuring a reproducible quantification. The method showed to be adequate for the purpose, with an average RSD of 2% for the different amino acids; detection limits varying between 0.71 mg/l (Asn) and 8.26 mg/l (Lys) and recovery rates between 63.0% (Cad) and 98.0% (Asp). The amino acids present at the highest concentration in honey and wine samples were phenylalanine and arginine, respectively. Only residual levels of biogenic amines were detected in the analysed samples

    A gold-containing drug against parasitic polyamine metabolism: the X-ray structure of trypanothione reductase from Leishmania infantum in complex with auranofin reveals a dual mechanism of enzyme inhibition

    Auranofin is a gold(I)-containing drug in clinical use as an antiarthritic agent. Recent studies showed that auranofin manifests interesting antiparasitic actions very likely arising from inhibition of parasitic enzymes involved in the control of the redox metabolism. Trypanothione reductase is a key enzyme of Leishmania infantum polyamine-dependent redox metabolism, and a validated target for antileishmanial drugs. As trypanothione reductase contains a dithiol motif at its active site and gold(I) compounds are known to be highly thiophilic, we explored whether auranofin might behave as an effective enzyme inhibitor and as a potential antileishmanial agent. Notably, enzymatic assays revealed that auranofin causes indeed a pronounced enzyme inhibition. To gain a deeper insight into the molecular basis of enzyme inhibition, crystals of the auranofin-bound enzyme, in the presence of NADPH, were prepared, and the X-ray crystal structure of the auranofin–trypanothione reductase–NADPH complex was solved at 3.5 Å resolution. In spite of the rather low resolution, these data were of sufficient quality as to identify the presence of the gold center and of the thiosugar of auranofin, and to locate them within the overall protein structure. Gold binds to the two active site cysteine residues of TR, i.e. Cys52 and Cys57, while the thiosugar moiety of auranofin binds to the trypanothione binding site; thus auranofin appears to inhibit TR through a dual mechanism. Auranofin kills the promastigote stage of L. infantum at micromolar concentration; these findings will contribute to the design of new drugs against leishmaniasis
