    Extreme droughts and human responses to them: the Czech Lands in the pre-instrumental period

    The Czech Lands are particularly rich in documentary sources that help elucidate droughts in the pre-instrumental period (12th–18th centuries), together with descriptions of human responses to them. Although droughts appear less frequently before 1501, the documentary evidence has enabled the creation of a series of seasonal and summer half-year drought indices (Standardized Precipitation Index, SPI; Standardized Precipitation Evapotranspiration Index, SPEI; Z index) for the Czech Lands for the 1501–2017 period. Based on the calculation of return period for series of drought indices, extreme droughts were selected for inclusion herein if all three indices indicated a return period of ≥20 years. For further analysis, only those from the pre-instrumental period (before 1804) were used. The extreme droughts selected are characterized by significantly lower values of drought indices, higher temperatures and lower precipitation totals compared to other years. The sea-level pressure patterns typically associated with extreme droughts include significantly higher pressure over Europe and significantly lower pressure over parts of the Atlantic Ocean. Extreme droughts with a return period ≥ 50 years are described in detail on the basis of Czech documentary evidence. A number of selected extreme droughts are reflected in other central European reconstructions derived from documentary data or tree rings. Impacts on social life and responses to extreme droughts are summarized; analysis of fluctuations in grain prices with respect to drought receives particular attention. Finally, extreme droughts from the pre-instrumental and instrumental periods are discussed.</p

    "Soft and rigid" dithiols and Au nanoparticles grafting on plasma-treated polyethyleneterephthalate

    Surface of polyethyleneterephthalate (PET) was modified by plasma discharge and subsequently grafted with dithiols (1, 2-ethanedithiol (ED) or 4, 4'-biphenyldithiol) to create the thiol (-SH) groups on polymer surface. This "short" dithiols are expected to be fixed via one of -SH groups to radicals created by the plasma treatment on the PET surface. "Free" -SH groups are allowed to interact with Au nanoparticles. X-ray photoelectron spectroscopy (XPS), Fourier transform infrared spectroscopy (FTIR) and electrokinetic analysis (EA, zeta potential) were used for the characterization of surface chemistry of the modified PET. Surface morphology and roughness of the modified PET were studied by atomic force microscopy (AFM). The results from XPS, FTIR, EA and AFM show that the Au nanoparticles are grafted on the modified surface only in the case of biphenyldithiol pretreatment. The possible explanation is that the "flexible" molecule of ethanedithiol is bounded to the activated PET surface with both -SH groups. On the contrary, the "rigid" molecule of biphenyldithiol is bounded via only one -SH group to the modified PET surface and the second one remains "free" for the consecutive chemical reaction with Au nanoparticle.     Short-term streambed drying events alter amphipod population structure in a central European stream

    Temporary streams are becoming increasingly common, but ecological responses to streambed drying are poorly characterized in the temperate continental region of central Europe. In addition, global research has focused on community responses to drying, whereas effects on individual populations remain unknown. We explored the population structure of Gammarus fossarum (Crustacea: Amphipoda) in a central European temporary stream. Benthic gammarids were sampled on five dates during a flow recession, one date between two drying events (of 10 and 20 days, respectively), and five dates after flow resumed. Additional benthic samples were taken from isolated pools and dry sediments during drying events, and freeze cores were collected to compare the vertical distribution of amphipods during wet and dry phases. Gammarids were measured, adults distinguished from juveniles, adults sexed, and female reproductive state determined. Densities increased during flow recession, potentially reflecting both a decline in submerged habitat availability and seasonal increases in juvenile abundance. Persistence within dry benthic sediments was minimal, whereas pools and saturated subsurface sediments supported high population densities. Juveniles comprised 80 % of the subsurface population, suggesting that their ability to inhabit small interstices promotes persistence within the dry reach. Juveniles also comprised 92 % of pool inhabitants, despite their potential exposure to predation. Adults dominated after flow resumed, and population structure was altered post-drying by the loss of spring-recruited juveniles and reproductive females. Our results suggest that streambed drying may have longer-term effects than typically characterized by community-level studies. We recommend management actions that support populations of ecologically important species as they adapt to changing flow regimes

    Cyclin-dependent kinase 5 mediates pleiotrophin-induced endothelial cell migration

    Pleiotrophin (PTN) stimulates endothelial cell migration through binding to receptor protein tyrosine phosphatase beta/zeta (RPTPβ/ζ) and ανβ3 integrin. Screening for proteins that interact with RPTPβ/ζ and potentially regulate PTN signaling, through mass spectrometry analysis, identified cyclin-dependent kinase 5 (CDK5) activator p35 among the proteins displaying high sequence coverage. Interaction of p35 with the serine/threonine kinase CDK5 leads to CDK5 activation, known to be implicated in cell migration. Protein immunoprecipitation and proximity ligation assays verified p35-RPTPβ/ζ interaction and revealed the molecular association of CDK5 and RPTPβ/ζ. In endothelial cells, PTN activates CDK5 in an RPTPβ/ζ- and phosphoinositide 3-kinase (PI3K)-dependent manner. On the other hand, c-Src, ανβ3 and ERK1/2 do not mediate the PTN-induced CDK5 activation. Pharmacological and genetic inhibition of CDK5 abolished PTN-induced endothelial cell migration, suggesting that CDK5 mediates PTN stimulatory effect. A new pyrrolo[2,3-α]carbazole derivative previously identified as a CDK1 inhibitor, was found to suppress CDK5 activity and eliminate PTN stimulatory effect on cell migration, warranting its further evaluation as a new CDK5 inhibitor. Collectively, our data reveal that CDK5 is activated by PTN, in an RPTPβ/ζ-dependent manner, regulates PTN-induced cell migration and is an attractive target for the inhibition of PTN pro-angiogenic properties

    In the past decades, when the life on the Earth became interrelated and mutually influenced, the education of the citizens of the world has been pushed to the foreground of the priorities pursued by educational objectives. Should the teaching of geography pursue this educational demand? Do our students think in global dimensions? Do they realise that the life on the Earth is interrelated? Is this knowledge translated into their attitudes and values? The asked questions outline the topic that has become a focus of international research (Béneker et al. 2013). It is the main goal of this paper to highlight the level of Czech students’ world-mindedness as compared to students from selected European countries (the Netherlands, Finland, Germany, Hungary and Serbia)
