    Analysis of the tools used to ensure the well-being of students of a selected elementary school and their significance from the point of view of the pupils themselves

    This diploma thesis is devoted to the topic of well-being of pupils of a selected primary school in the Central Bohemian Region of the Czech Republic. The aim of the research was to find out what tools (measures) the given school implements to ensure the well-being of students in grades 1-7 of the given school and how these students evaluate the importance of the tools used to ensure their own well-being at school. Last but not least, the research also focused on finding out the opinions of these students on other measures they would like the school to take to ensure their well-being. The research was conducted in the form of a case study using a mixed qualitative-quantitative design method. With the help of the mind-map analysis of students' and teachers'work related to the determination of the types of well-being tools used in a given school, as well as an online questionnaire survey, which was answered by the students of the given school themselves, the answers to the research questions were found: 1. What tools (measures) does the school use to ensure the well-being of pupils 1 - 7 grades? 2. What weight do the students of 1 - 7 grade of the given school to individual tools to ensure their well-being in the given school? 3. Cílem výzkumu bylo zjistit, jaké nástroje (opatření) daná škola realizuje pro zajištění wellbeingu žáků 1. - 7. ročníku dané školy a jak tito žáci hodnotí významnost používaných nástrojů pro zajištění jejich vlastního wellbeingu ve škole. V neposlední řadě se výzkum zaměřil také na zjištění názorů těchto žáků na další opatření, která by si přáli, aby škola pro zajištění jejich wellbeingu učinila. Výzkum byl proveden formou případové studie za použití metody smíšeného designu kvalitativně- kvantitativního. Pomocí rozboru mind-map žáků a učitelů vztahujících se k určení druhů nástrojů wellbeingu používaných v dané škole a také online dotazníkového šetření, jehož respondi byli samotní žáci dané školy, byly zjištěny odpovědi na výzkumné otázky: 1. Jakou váhu přisuzují žáci 1. - 7. ročníku dané školy jednotlivým nástrojům pro zajištění jejich wellbeingu v dané škole? 3.     Analysis of the concept of education of employees in one private elementary school and kindergarten in the Czech Republic and a proposal for its changes.

    The diploma thesis deals with the issue of employee education in one primary and nursery school in the Czech Republic. This is a one-case study, which, using qualitative-quantitative research, aims to analyze the current concept of employee education of the given organization and also proposes changes to the existing concept so that it is in line with the requirements for the strategic concept of employee education. The diploma thesis includes a theoretical part with an explanation of relevant concepts from the field of andragogy and management, as well as a practical part, where the internal documents of the school are analyzed and the characteristics of the current concept of employee education are described. Furthermore, a questionnaire survey was also carried out using 3 questionnaires, the respondents were school employees and the school director. These questionnaires were designed to verify the functionality of the concept. Answers were obtained to the first research question, which reads: What is the characteristic of the current concept of employee education?, and which should help answer the second research question: What changes can be made to improve the quality of the current concept? Jedná se o jednopřípadovou studii, která za využití kvalitativně-kvantitativního výzkumu má za cíl provést analýzu stávající koncepce vzdělávání zaměstnanců dané organizace a také navrhnout změny stávající koncepce tak, aby byla v souladu s požadavky na strategickou koncepci vzdělávání zaměstnanců. Diplomová práce zahrnuje část teoretickou s vysvětlením relevantních pojmů z oblasti andragogiky a managementu a také část praktickou, kde je provedena analýza vnitřních dokumentů školy a popsána charakteristika stávající koncepce vzdělávání zaměstnanců. Dále bylo také provedeno dotazníkové šetření pomocí 3 dotazníků, jejichž respondenti byli zaměstnanci školy a ředitelka školy. Tyto dotazníky byly navrženy za účelem ověření funkčnosti koncepce.     VARIANCES OF LENGTH AND SURFACE AREA ESTIMATES BY SPATIAL GRIDS: PRELIMINARY STUDY

    Periodic spatial grids can be used for unbiased estimation of length and surface area of objects by counting or measuring intersections of the objects with the grids. The estimators are theoretically based on discrete approximation of well established integral geometric formulas. The variance of the estimates depends on properties of both the grid and the measured objects. Main results of the theory of variance of the isotropic uniform random (IUR) volume estimation by spatial grids, especially a formula relating the variance of the volume estimator with the object surface area and the grid constant, are recapitulated. To identify main features of length and surface area IUR estimates the variance due to rotation and simple asymptotic formulas for the residual variance of estimates of selected model objects is calculated. Surface area estimates by multiple grids of parallel lines in 3D and of the variance of length estimates by periodic grids of planes or spheres in ddimensional space are studied

    UNBIASED ESTIMATION OF NORWAY SPRUCE (PICEA ABIES L. KARST.) CHLOROPLAST STRUCTURE: HETEROGENEITY WITHIN NEEDLE MESOPHYLL UNDER DIFFERENT IRRADIANCE AND [CO2]

    The main objective of this study was to find out whether the selected chloroplast characteristics measured in the mesophyll layer nearest to the needle surface (i.e., the first mesophyll layer) could be representative for the whole needle cross section. Two chloroplast sampling approaches were applied on Norway spruce needles during the investigation of the effects of different levels of air CO2 concentration and irradiance: (i) sampling only from the first mesophyll layer, and (ii) systematic uniform random (SUR) sampling. The selected characteristics were: (i) chloroplast area, (ii) starch grain area, and (iii) starch areal density on median chloroplast cross sections, and (iv) chloroplast number per unit of needle volume. It was shown that the first mesophyll layer was not representative for estimating all evaluated characteristics except the chloroplast area. Sampling only there caused obtaining slightly biased results, while SUR sampling gave unbiased estimations at the cost of longer measuring time. The major effect of studied factors was in starch areal density and starch grain area, which were larger in sun needles in elevated CO2 concentration in comparison with sun needles in ambient CO2 concentration. In conclusion, it was demonstrated that the first layer of mesophyll is not always representative for the needle cross section. If technically feasible, SUR is recommended for analysis of chloroplast ultrastructure. The simplified sampling design can be applied, e.g., for comparisons of many different treatments. However, it should be combined with other approaches to characterize the chloroplast function and the results carefully considered and interpreted

    Testate Amoebae Examined by Confocal and Two-Photon Microscopy: Implications for Taxonomy and Ecophysiology

    Testate amoebae (TA) are a group of free-living protozoa, important in ecology and paleoecology. Testate amoebae taxonomy is mainly based on the morphological features of the shell, as examined by means of light microscopy or (environmental) scanning electron microscopy (SEM/ESEM). We explored the potential applications of confocal laser scanning microscopy (CLSM), two photon excitation microscopy (TPEM), phase contrast, differential interference contrast (DIC Nomarski), and polarization microscopy to visualize TA shells and inner structures of living cells, which is not possible by SEM or environmental SEM. Images captured by CLSM and TPEM were utilized to create three-dimensional (3D) visualizations and to evaluate biovolume inside the shell by stereological methods, to assess the function of TA in ecosystems. This approach broadens the understanding of TA cell and shell morphology, and inner structures including organelles and endosymbionts, with potential implications in taxonomy and ecophysiolog

    Analysis and three-dimensional visualization of collagen in artificial scaffolds using nonlinear microscopy techniques

    Extracellularly distributed collagen and chondro- cytes seeded in gelatine and poly-e-caprolactone scaffolds are visualized by two-photon excitation microscopy (TPEM) and second-harmonic generation (SHG) imaging in both for- ward and backward nondescanned modes. Joint application of TPEM and SHG imaging in combination with stereolog- ical measurements of collagen enables us not only to take high-resolution 3-D images, but also to quantitatively an- alyze the collagen volume and a spatial arrangement of cell-collagen-scaffold systems, which was previously im- possible.     This bachelor thesis deals with deficiencies that elementary school principals must solve in the field of ICT. In particular, there is a lack of the resources available, especially hard sources including ICT equipment, a lack of space for ICT equipment and inadequate connectivity on the Internet, but also soft resources in the field of human resources are missing. This thesis has two goals. The first goal is to find out what kind of deficiencies it is and how big they are and the second goal is to make recommendations for elementary school principals how to deal with these deficiencies. The research that is part of this thesis was focused on principals of elementary schools in the South Moravian Region. The research is conceived as a quantitative survey using an online Internet questionnaire. From the 66 expected respondents to 16 research questions, an analysis is found that leads to the conclusion that my four hypotheses were right. The questions are mainly focused on the finding the type of the shortage and its size from the point of view of elementary school principals, while there is 1 sorting question ( concerning school size) and the last question is focused on the type of education / training of elementary school principals in ICT.
