    In vitro activity of neem (Azadirachta indica) oil on growth and ochratoxin A production by Aspergillus carbonarius isolates

    Aspergillus carbonarius is a saprobic filamentous fungus, food spoiling fungus and a producer of ochratoxin A (OTA) mycotoxin. In this study, the in vitro antifungal activity of neem oil (0.12% p/p of azadirachtin) was evaluated against the growth of six strains of A. carbonarius and the production of OTA. Four different concentrations of neem oil were tested in addition to three incubation times. Only the concentration of 0.3% of neem oil inhibited more than 95% of the strain’s growth (97.6% ± 0.5%), while the use of 0.5% and 1.0% of neem oil showed lower antifungal activity, 40.2% ± 3.1 and 64.7% ± 1.1, respectively. There was a complete inhibition of OTA production with 0.1% and 0.3% neem oil in the four strains isolated in the laboratory from grapes. The present study shows that neem essential oil can be further evaluated as an auxiliary method for the reduction of mycelial growth and OTA production.Fil: Rodrigues, Mariana Paiva.     Design, Synthesis and Biological Evaluation of Novel Triazole N-acylhydrazone Hybrids for Alzheimer's Disease

    Alzheimer's disease (AD) is a multifactorial neurodegenerative disorder that involves different pathogenic mechanisms. In this regard, the goal of this study was the design and synthesis of new compounds with multifunctional pharmacological activity by molecular hybridization of structural fragments of curcumin and resveratrol connected by an N-acyl-hydrazone function linked to a 1,4-disubstituted triazole system. Among these hybrid compounds, derivative 3e showed the ability to inhibit acetylcholinesterase activity, the intracellular formation of reactive oxygen species as well as the neurotoxicity elicited by Aβ42 oligomers in neuronal SH-SY5Y cells. In parallel, compound 3e showed a good profile of safety and ADME parameters. Taken together, these results suggest that 3e could be considered a lead compound for the further development of AD therapeutics

    Genome wide scan for quantitative trait loci affecting tick resistance in cattle (Bos taurus × Bos indicus)

    <p><b>Abstract</b></p> <p><b>Background</b></p> <p>In tropical countries, losses caused by bovine tick <it>Rhipicephalus (Boophilus) microplus</it> infestation have a tremendous economic impact on cattle production systems. Genetic variation between <it>Bos taurus</it> and <it>Bos indicus</it> to tick resistance and molecular biology tools might allow for the identification of molecular markers linked to resistance traits that could be used as an auxiliary tool in selection programs. The objective of this work was to identify QTL associated with tick resistance/susceptibility in a bovine F2 population derived from the Gyr (<it>Bos indicus</it>) × Holstein (<it>Bos taurus</it>) cross.</p> <p>Results</p> <p>Through a whole genome scan with microsatellite markers, we were able to map six genomic regions associated with bovine tick resistance. For most QTL, we have found that depending on the tick evaluation season (dry and rainy) different sets of genes could be involved in the resistance mechanism. We identified dry season specific QTL on BTA 2 and 10, rainy season specific QTL on BTA 5, 11 and 27. We also found a highly significant genome wide QTL for both dry and rainy seasons in the central region of BTA 23.</p> <p>Conclusions</p> <p>The experimental F2 population derived from Gyr × Holstein cross successfully allowed the identification of six highly significant QTL associated with tick resistance in cattle. QTL located on BTA 23 might be related with the bovine histocompatibility complex. Further investigation of these QTL will help to isolate candidate genes involved with tick resistance in cattle.</p

    Influência do Tratamento por Realidade Virtual no Equilíbrio de um Paciente com Paralisia Cerebral

    The improvement of maintaining postural balances its essential i for those who have some motor impairment, such as Cerebral Palsy (CP). Virtual Reality (VR) is a feature that is standing out in neurological rehabilitation, but there are few studies regarding adult patients with CP. This study aimed to investigate the infl uence of the VR through the Wii on balance in standing position from a patient with Spastic Hemiplegic CP. It was used the equipment Nintendo® Wii, with the game Wii Fit Plus, where the training chosen stimulated the latero-lateral and anterior-posterior patient’s balance. The instruments used were: Berg Balance Scale, Functional Independence Measure, and 3 Neurocom Balance Master Systems tests: Limits of Stability, Tandem Walk and Step/ Quick Turn. The treatment protocol consisted of 10 sessions, with 30 minutes each, performed twice a week for 38 days. At the end of treatment, a signifi cant increase in the scores of various test items performed was observed, which resulted in an improvement of patient’s balance. Results suggest a benefi cial infl uence of the video game Wii regarding to postural balance and functional capacity of patients with PC, which become more confi dent to carry out their activities of daily living.A melhora da capacidade de manutenção do equilíbrio é essencial para quem apresenta algum comprometimento motor, como a Paralisia Cerebral (PC). A realidade virtual (RV) já se destaca na reabilitação neurológica, porém com poucos estudos que abordem pacientes adultos com PC. Este estudo teve como objetivo investigar a infl uência da RV por meio de jogos de vídeo Wii no equilíbrio em posição ortostática de um paciente com PC Espástica Hemiparética. Foi utilizado o equipamento Nintendo® Wii, com o jogo Wii Fit Plus, em treinamentos que estimularam o equilíbrio látero-lateral e ântero-posterior do paciente. Os instrumentos aplicados foram: Escala de Equilíbrio de Berg, Medida de Independência Funcional, além de 3 testes do Neurocom Balance Master: Limits of Stability, Tandem Walk e Step/Quick Turn. O protocolo de tratamento consistiu de 10 sessões, 30 minutos cada, duas vezes por semana por 38 dias. Ao fi nal foi possível observar um aumento expressivo na pontuação de vários itens dos testes realizados, implicando na melhora da capacidade de equilíbrio do paciente. Esses resultados sugerem a infl uência benéfi ca do Wii no que concerne o equilíbrio e capacidade funcional do portador de PC, tornando-o mais confi ante para realizar suas atividades diárias

    Design, synthesis, and biological evaluation of new thalidomide–donepezil hybrids as neuroprotective agents targeting cholinesterases and neuroinflammation

    A new series of eight multifunctional thalidomide–donepezil hybrids were synthesized based on the multi target-directed ligand strategy and evaluated as potential neuroprotective, cholinesterase inhibitors and anti neuroinflammatory agents against neurodegenerative diseases. A molecular hybridization approach was used for structural design by combining the N-benzylpiperidine pharmacophore of donepezil and the isoindoline 1,3-dione fragment from the thalidomide structure. The most promising compound, PQM-189 (3g), showed good AChE inhibitory activity with an IC50 value of 3.15 μM, which was predicted by docking studies as interacting with the enzyme in the same orientation observed in the AChE–donepezil complex and a similar profile of interaction. Additionally, compound 3g significantly decreased iNOS and IL-1β levels by 43% and 39%, respectively, after 24 h of incubation with lipopolysaccharide. In vivo data confirmed the ability of 3g to prevent locomotor impairment and changes in feeding behavior elicited by lipopolysaccharide. Moreover, the PAMPA assay evidenced adequate blood–brain barrier and gastrointestinal tract permeabilities with an Fa value of 69.8%. Altogether, these biological data suggest that compound 3g can treat the inflammatory process and oxidative stress resulting from the overexpression of iNOS and therefore the increase in reactive nitrogen species, and regulate the release of pro-inflammatory cytokines such as IL-1β.
