    Utilization of biological posts to reconstruct weakened roots

    Adequate reconstruction of endodontically treated teeth with severe loss of dentine tissue is one of the challenges of Restorative Dentistry. None of the commercially available pre-fabricated intraradicular posts meets all ideal biological and mechanical properties. Recently, a technique for the reconstruction of pulpless teeth with weakened roots has been indicated – the use of dental posts obtained from natural teeth.     High-energy diet does not overcome the negative impact of conjugated linoleic acid on young broiler performance

    The aim of this study was to evaluate the effect of conjugated linoleic acid (CLA) supplementation in diets with different energy levels in broiler performance. Birds were offered a starter (1-21 d), grower (22-35 d) and finisher (36-42 d) diets; wherein soybean oil was replaced by CLA. The study consisted of a 3 × 2 factorial arrangement with two CLA levels (0 and 1%) and three energy levels (3050, 3100 and 3150 ME kg-1 diet). During the grower and finisher periods, birds were fed diets with same energy level and CLA supplementation was maintained the same. Growth performance was assessed weekly, and carcass and cuts yield were assessed at 42d. Interaction effect of CLA by energy level was observed in broiler performance and carcass yield throughout the study (p > 0.05). During the overall period (1-42 d) broiler performance was not affected by CLA (p > 0.05).However, CLA supplementation (1%) decreased weight gain (p 0.05). The increase in dietary energy was not able to compensate the negative effect on growth performance of broilers supplemented with 1% CLA at the starter period

    Gene expression differences in relation to age and social environment in queen and worker bumble bees

    Eusocial insects provide special insights into the genetic pathways influencing aging because of their long-lived queens and flexible aging schedules. Using qRT-PCR in the primitively eusocial bumble bee Bombus terrestris (Linnaeus), we investigated expression levels of four candidate genes associated with taxonomically widespread age-related pathways (coenzyme Q biosynthesis protein 7, COQ7; DNA methyltransferase 3, Dnmt3; foraging, for; and vitellogenin, vg). In Experiment 1, we tested how expression changes with queen relative age and productivity. We found a significant age-related increase in COQ7 expression in queen ovary. In brain, all four genes showed higher expression with increasing female (queen plus worker) production, with this relationship strengthening as queen age increased, suggesting a link with the positive association of fecundity and longevity found in eusocial insect queens. In Experiment 2, we tested effects of relative age and social environment (worker removal) in foundress queens and effects of age and reproductive status in workers. In this experiment, workerless queens showed significantly higher for expression in brain, as predicted if downregulation of for is associated with the cessation of foraging by foundress queens following worker emergence. Workers showed a significant age-related increase in Dnmt3 expression in fat body, suggesting a novel association between aging and methylation in B. terrestris. Ovary activation was associated with significantly higher vg expression in fat body and, in younger workers, in brain, consistent with vitellogenin's ancestral role in regulating egg production. Overall, our findings reveal a mixture of novel and conserved features in age-related genetic pathways under primitive eusociality

    Handcycling: training effects of a specific dose of upper body endurance training in females

    Purpose: This study aims to evaluate a handcycling training protocol based on ACSM guidelines in a well-controlled laboratory setting. Training responses of a specific dose of handcycling training were quantified in a homogeneous female subject population to obtain a more in depth understanding of physiological mechanisms underlying adaptations in upper body training. Methods: 22 female able-bodied participants were randomly divided in a training (T) and control group (C). T received 7-weeks of handcycling training, 3 × 30 min/week at 65 % heart rate reserve (HRR). An incremental handcycling test was used to determine local, exercise-specific adaptations. An incremental cycling test was performed to determine non-exercise-specific central/cardiovascular adaptations. Peak oxygen uptake (peakVO2), heart rate (peakHR) and power output (peakPO) were compared between T and C before and after training. Results: T completed the training sessions at 65 ± 3 % HRR, at increasing power output (59.4 ± 8.2 to 69.5 ± 8.9 W) over the training program. T improved on handcycling peakVO2 (+18.1 %), peakPO (+31.9 %), and peakHR (+4.0 %). No improvements were found in cycling parameters. Conclusion: Handcycling training led to local, exercise-specific improvements in upper body parameters. Results could provide input for the design of effective evidence-based training programs specifically aimed at upper body endurance exercise in females

    Multicentric study of cervical cancer screening with human papillomavirus testing and assessment of triage methods in Latin America : the ESTAMPA screening study protocol

    Q1Q1Introduction Human papillomavirus (HPV) testing is replacing cytology in primary screening. Its limited specificity demands using a second (triage) test to better identify women at high-risk of cervical disease. Cytology represents the immediate triage but its low sensitivity might hamper HPV testing sensitivity, particularly in low-income and middle-income countries (LMICs), where cytology performance has been suboptimal. The ESTAMPA (EStudio multicéntrico de TAMizaje y triaje de cáncer de cuello uterino con pruebas del virus del PApiloma humano; Spanish acronym) study will: (1) evaluate the performance of different triage techniques to detect cervical precancer and (2) inform on how to implement HPV-based screening programmes in LMIC. Methods and analysis Women aged 30–64 years are screened with HPV testing and Pap across 12 study centres in Latin America. Screened positives have colposcopy with biopsy and treatment of lesions. Women with no evident disease are recalled 18 months later for another HPV test; those HPV-positive undergo colposcopy with biopsy and treatment as needed. Biological specimens are collected in different visits for triage testing, which is not used for clinical management. The study outcome is histological high-grade squamous intraepithelial or worse lesions (HSIL+) under the lower anogenital squamous terminology. About 50 000 women will be screened and 500 HSIL+ cases detected (at initial and 18 months screening). Performance measures (sensitivity, specificity and predictive values) of triage techniques to detect HSIL+ will be estimated and compared with adjustment by age and study centre. Ethics and dissemination The study protocol has been approved by the Ethics Committee of the International Agency for Research on Cancer (IARC), of the Pan American Health Organisation (PAHO) and by those in each participating centre. A Data and Safety Monitoring Board (DSMB) has been established to monitor progress of the study, assure participant safety, advice on scientific conduct and analysis and suggest protocol improvements. Study findings will be published in peer-reviewed journals and presented at scientific meetings. Trial registration number NCT01881659Revista Internacional - Indexad

    O presente estudo teve por objetivo a avaliação das consequências da expansão rápida da maxila em indivíduos com fissura labiopalatina unilateral nos seguintes desfechos: rizogênese, reabsorção radicular, espessura da tábua óssea e deiscência. Este estudo de coorte prospectivo foi composto por 30 participantes, sendo 20 homens e 10 mulheres, entre 8 e 15 anos. Os participantes foram alocados em 3 grupos, conforme o tipo de atresia maxilar, e tratados com diferentes tipos de aparelho expansor: G1, Hyrax; G2, Expansor em Leque; G3, Mini-Hyrax Invertido. Foram realizadas Tomografias Computadorizadas de Feixes Cônicos imediatamente antes do tratamento e 90 dias após contenção do aparelho. Medições lineares foram obtidas pelo mesmo examinador cegado. A estatística inferencial dos dados, após testes de normalidade e homogeneidade, foi realizada pela análise de regressão multinível. As raízes que apresentavam o ápice aberto ao início do tratamento demonstraram um aumento estatisticamente significante após o tratamento. Em contrapartida, não houve alterações significativas no comprimento radicular das raízes que apresentavam o ápice fechado no início do tratamento. Uma diminuição significativa da espessura da tábua óssea vestibular, bem como um aumento significativo da deiscência pode ser observado. Não houve diferença significante entre o lado com e sem a fissura para todas as variáveis apresentadas, assim como não houve diferença entre os grupos. Os achados neste estudo permitem concluir que as forças ortopédicas da expansão rápida da maxila não são capazes de interromper o processo de desenvolvimento radicular, tampouco causar reabsorção radicular apical externa significativa em indivíduos com fissura labiopalatina unilateral. Contudo, uma diminuição significativa de volume ósseo vestibular posterior é esperada, mas não deve ser considerada importante clinicamente.The present study aimed to evaluate the consequences of rapid maxillary expansion in subjects with unilateral cleft lip and palate in the following outcomes: root formation, root resorption, buccal bone thickness and dehiscence. Participants were allocated in 3 groups, according to the type of maxillary constriction, and were treated with different types of expanders: G1, hyrax; G2, fan-type; G3, inverted mini-hyrax. Cone Beam Computed Tomography scans were performed immediately before treatment and after 90 days of retention. Linear measurements were obtained by the same blinded examiner. The inferential statistics of the data, after normality and equality tests, was performed with a multilevel regression analysis. The roots presenting open apexes at the beginning of treatment showed a statistically significant increase in length after treatment. On the other hand, there were no significant changes in the root length of roots that had a closed apexes at the beginning of the treatment. A significant decrease in buccal bone thickness as well as a significant increase in dehiscence were observed. There was no significant difference between the cleft and non-cleft side for all variables, as there was no significant difference between groups. The findings in this study allow to conclude that the orthopedic forces of rapid maxillary expansion are not able to interrupt the root development process nor to cause significant external apical root resorption in subjects with unilateral cleft lip and palate. However, a significant decrease in posterior buccal bone volume is expected, but it should not be considered important in a clinical perspective
