Root to Kellerer
We revisit Kellerer's Theorem, that is, we show that for a family of real
probability distributions which increases in convex
order there exists a Markov martingale s.t.\ .
To establish the result, we observe that the set of martingale measures with
given marginals carries a natural compact Polish topology. Based on a
particular property of the martingale coupling associated to Root's embedding
this allows for a relatively concise proof of Kellerer's theorem.
We emphasize that many of our arguments are borrowed from Kellerer
\cite{Ke72}, Lowther \cite{Lo07}, and Hirsch-Roynette-Profeta-Yor

The development and cognitive testing of the positive outcomes HIV PROM: a brief novel patient-reported outcome measure for adults living with HIV
Background
People living with HIV experience burdensome multidimensional symptoms and concerns requiring person-centred care. Routine use of patient reported outcome measures can improve outcomes. There is no brief patient reported outcome measure (PROM) that currently reflects the breadth of concerns for people living with HIV. This study aimed to develop and cognitively test a brief novel patient reported outcome measure for use within routine adult HIV care– the “Positive Outcomes” HIV PROM.
Methods
Development followed the COSMIN taxonomy and guidance for relevance and comprehensiveness, and Rothrock guidance on development of valid patient reported outcome measures. The Positive Outcomes HIV PROM was developed by a steering group (people living with HIV, HIV professionals and health services researchers) using findings from a previously reported qualitative study of priority outcomes for people living with HIV. The prototype measure was cognitively tested with a purposive sample of people living with HIV.
Results
The Positive Outcomes HIV PROM consists of 23 questions (22 structured, and one open question) informed by the priorities of key stakeholders (n = 28 people living with HIV, n = 21 HIV professionals and n = 8 HIV commissioners) to ensure face and content validity, and refined through cognitive testing (n = 6 people living with HIV). Cognitive testing demonstrated high levels of acceptability and accessibility.
Conclusions
The Positive Outcomes HIV PROM is the first brief patient reported outcome measure reflecting the diverse needs of people living with HIV designed specifically for use in the clinical setting to support patient assessment and care, and drive service quality improvement. It is derived from primary data on the priority outcomes for people living with HIV and is comprehensive and acceptable. Further psychometric testing is required to ensure reliability and responsiveness
A review of the scientific knowledge of the seascape off Dronning Maud Land, Antarctica
Despite the exclusion of the Southern Ocean from assessments of progress towards achieving the Convention on Biological Diversity (CBD) Strategic Plan, the Commission for the Conservation of Antarctic Marine Living Resources (CCAMLR) has taken on the mantle of progressing efforts to achieve it. Within the CBD, Aichi Target 11 represents an agreed commitment to protect 10% of the global coastal and marine environment. Adopting an ethos of presenting the best available scientific evidence to support policy makers, CCAMLR has progressed this by designating two Marine Protected Areas in the Southern Ocean, with three others under consideration. The region of Antarctica known as Dronning Maud Land (DML; 20°W to 40°E) and the Atlantic sector of the Southern Ocean that abuts it conveniently spans one region under consideration for spatial protection. To facilitate both an open and transparent process to provide the vest available scientific evidence for policy makers to formulate management options, we review the body of physical, geochemical and biological knowledge of the marine environment of this region. The level of scientific knowledge throughout the seascape abutting DML is polarized, with a clear lack of data in its eastern part which is presumably related to differing levels of research effort dedicated by national Antarctic programmes in the region. The lack of basic data on fundamental aspects of the physical, geological and biological nature of eastern DML make predictions of future trends difficult to impossible, with implications for the provision of management advice including spatial management. Finally, by highlighting key knowledge gaps across the scientific disciplines our review also serves to provide guidance to future research across this important region.publishedVersio
Recent translational research: Oncogene discovery by insertional mutagenesis gets a new boost
Knowledge of the genes and genetic pathways involved in onco-genesis is essential if we are to identify novel targets for cancer therapy. Insertional mutagenesis in mouse models is among the most efficient tools to detect novel cancer genes. Retrovirus-mediated insertional mutagenesis received a tremendous boost by the availability of the mouse genome sequence and new PCR methods. Application of such advances were limited to lympho-magenesis but are now also being applied to mammary tumourigenesis. Novel transposons that allow insertional muta-genesis studies to be conducted in tumors of any mouse tissue may give cancer gene discovery a further boost
Contrasting phylogeographic pattern among Eudyptes penguins around the Southern Ocean
Since at least the middle-Miocene, the Antarctic Polar Front (APF) and the Subtropical Front (STF) appear to have been the main drivers of diversification of marine biota in the Southern Ocean. However, highly migratory marine birds and mammals challenge this paradigm and the importance of oceanographic barriers. Eudyptes penguins range from the Antarctic Peninsula to subantarctic islands and some of the southernmost subtropical islands. Because of recent diversification, the number of species remains uncertain. Here we analyze two mtDNA (HVRI, COI) and two nuclear (ODC, AK1) markers from 13 locations of five putative Eudyptes species: rockhopper (E. filholi, E. chrysocome, and E. moseleyi), macaroni (E. chrysolophus) and royal penguins (E. schlegeli). Our results show a strong phylogeographic structure among rockhopper penguins from South America, subantarctic and subtropical islands supporting the recognition of three separated species of rockhopper penguins. Although genetic divergence was neither observed among macaroni penguins from the Antarctic Peninsula and sub-Antarctic islands nor between macaroni and royal penguins, population genetic analyses revealed population genetic structure in both cases. We suggest that the APF and STF can act as barriers for these species. While the geographic distance between colonies might play a role, their impact/incidence on gene flow may vary between species and colonies
Immunostimulatory Motifs Enhance Antiviral siRNAs Targeting Highly Pathogenic Avian Influenza H5N1
Highly pathogenic avian influenza (HPAI) H5N1 virus is endemic in many regions around the world and remains a significant pandemic threat. To date H5N1 has claimed almost 300 human lives worldwide, with a mortality rate of 60% and has caused the death or culling of hundreds of millions of poultry since its initial outbreak in 1997. We have designed multi-functional RNA interference (RNAi)-based therapeutics targeting H5N1 that degrade viral mRNA via the RNAi pathway while at the same time augmenting the host antiviral response by inducing host type I interferon (IFN) production. Moreover, we have identified two factors critical for maximising the immunostimulatory properties of short interfering (si)RNAs in chicken cells (i) mode of synthesis and (ii) nucleoside sequence to augment the response to virus. The 5-bp nucleoside sequence 5′-UGUGU-3′ is a key determinant in inducing high levels of expression of IFN -α, -β, -λ and interleukin 1- β in chicken cells. Positioning of this 5′-UGUGU-3′ motif at the 5′- end of the sense strand of siRNAs, but not the 3′- end, resulted in a rapid and enhanced induction of type I IFN. An anti-H5N1 avian influenza siRNA directed against the PB1 gene (PB1-2257) tagged with 5′-UGUGU-3′ induced type I IFN earlier and to a greater extent compared to a non-tagged PB1-2257. Tested against H5N1 in vitro, the tagged PB1-2257 was more effective than non-tagged PB1-2257. These data demonstrate the ability of an immunostimulatory motif to improve the performance of an RNAi-based antiviral, a finding that may influence the design of future RNAi-based anti-influenza therapeutics
