    Role of the enterocyte in fructose-induced hypertriglyceridaemia

    Abstract: Dietary fructose has been linked to an increased post-prandial triglyceride (TG) level, which is an established independent risk factor for cardiovascular disease. Although much research has focused on the effects of fructose consumption on liver-derived very-low density lipoprotein (VLDL), emerging evidence also suggests that fructose may raise post-prandial TG levels by affecting the metabolism of enterocytes of the small intestine. Enterocytes have become well recognised for their ability to transiently store lipids following a meal and to thus control post-prandial TG levels according to the rate of chylomicron (CM) lipoprotein synthesis and secretion. The influence of fructose consumption on several aspects of enterocyte lipid metabolism are discussed, including de novo lipogenesis, apolipoprotein B48 and CM-TG production, based on the findings of animal and human isotopic tracer studies. Methodological issues affecting the interpretation of fructose studies conducted to date are highlighted, including the accurate separation of CM and VLDL. Although the available evidence to date is limited, disruption of enterocyte lipid metabolism may make a meaningful contribution to the hypertriglyceridaemia often associated with fructose consumption

    Nutrient Patterns are Associated with Discordant Apolipoprotein B and Low-Density Lipoproteins: A Population-Based Analysis

    Individuals with discordantly high ApoB to LDL-C levels carry a higher risk of atherosclerotic cardiovascular disease compared to those with average or discordantly low ApoB to LDL-C. We aimed to determine associations between ApoB and LDL-C discordance in relation to nutrient patterns (NPs) using National Health and Nutrition Examination Survey data. Participants were grouped by established LDL-C and ApoB cut-offs (Group 1: Low ApoB/Low LDL-C, Group 2: Low ApoB/High LDL-C, Group 3: High ApoB/Low LDL-C, Group 4: High ApoB/High LDL-C). Principle component analysis was used to define NPs. Machine learning (ML) and structural equation models were applied to assess associations of nutrient intake with ApoB/LDL-C discordance using the combined effects of ApoB and LDL-C. Three NPs explained 63.2% of variance in nutrient consumption. These consisted of NP1 rich in saturated fatty acids, carbohydrate and vitamins, NP2 high in fibre, minerals, vitamins and PUFA and NP3 rich in dietary cholesterol, protein, and sodium. The discordantly high ApoB to LDL-C group had the highest consumption of the NP1 and the lowest consumption of the NP2. ML showed nutrients which had the greatest unfavourable dietary contribution to individuals with discordantly high ApoB to LDL-C were total fat, saturated fatty acids and thiamine and the greatest favourable contributions were MUFA, folate, fibre and selenium. Individuals with discordantly high ApoB in relation to LDL-C had greater adherence to NP1, whereas those with lower levels of ApoB, irrespective of LDL-C, are more likely to consume NP3

    Personalised nutrition advice reduces intake of discretionary foods and beverages: findings from the Food4Me randomised controlled trial

    Background: The effect of personalised nutrition advice on discretionary foods intake is unknown. To date, two national classifications for discretionary foods have been derived. This study examined changes in intake of discretionary foods and beverages following a personalised nutrition intervention using these two classifications. Methods: Participants were recruited into a 6-month RCT across seven European countries (Food4Me) and were randomised to receive generalised dietary advice (control) or one of three levels of personalised nutrition advice (based on diet [L1], phenotype [L2] and genotype [L3]). Dietary intake was derived from an FFQ. An analysis of covariance was used to determine intervention effects at month 6 between personalised nutrition (overall and by 10 levels) and control on i) percentage energy from discretionary items and ii) percentage contribution of total fat, SFA, total sugars and salt to discretionary intake, defined by Food Standards Scotland (FSS) and Australian Dietary Guidelines (ADG) classifications. Results: Of the 1607 adults at baseline, n=1270 (57% female) completed the intervention. Percentage sugars from FSS discretionary items was lower in personalised nutrition vs control (19.0 \ub1 0.37 vs 21.1 \ub1 0.65; P=0.005). Percentage energy (31.2 \ub1 0.59 vs 32.7 \ub1 0.59; P=0.031), percentage total fat (31.5 \ub1 0.37 vs 33.3 \ub1 0.65; P=0.021), SFA (36.0 \ub1 0.43 vs 37.8 \ub1 0.75; P=0.034) and sugars (31.7 \ub1 0.44 vs 34.7 \ub1 0.78; P<0.001) from ADG discretionary items were lower in personalised nutrition vs control. There were greater reductions in ADG percentage energy and percentage total fat, SFA and salt for those randomised to L3 vs L2. 21Conclusions: Compared with generalised dietary advice, personalised nutrition advice achieved greater reductions in discretionary foods intake when the classification included all foods high in fat, added sugars and salt. Future personalised nutrition approaches may be 24 used to target intakes of discretionary foods

    Higher vegetable protein consumption, assessed by an isoenergetic macronutrient exchange model, is associated with a lower presence of overweight and obesity in the web-based Food4me European study

    The objective was to evaluate differences in macronutrient intake and to investigate the possible association between consumption of vegetable protein and the risk of overweight/obesity, within the Food4Me randomised, online intervention. Differences in macronutrient consumption among the participating countries grouped by EU Regions (Western Europe, British Isles, Eastern Europe and Southern Europe) were assessed. Relation of protein intake, within isoenergetic exchange patterns, from vegetable or animal sources with risk of overweight/obesity was assessed through the multivariate nutrient density model and a multivariate-adjusted logistic regression.A total of 2413 subjects who completed the Food4Me screening were included, with self-reported data on age, weight, height, physical activity and dietary intake. As success rates on reducing overweight/obesity are very low, form a public health perspective, the elaboration of policies for increasing intakes of vegetable protein and reducing animal protein and sugars, may be a method of combating overweight/obesity at a population level

    Controlled creation of a singular spinor vortex by circumventing the Dirac belt trick

    Persistent topological defects and textures are particularly dramatic consequences of superfluidity. Among the most fascinating examples are the singular vortices arising from the rotational symmetry group SO(3), with surprising topological properties illustrated by Dirac’s famous belt trick. Despite considerable interest, controlled preparation and detailed study of vortex lines with complex internal structure in fully three-dimensional spinor systems remains an outstanding experimental challenge. Here, we propose and implement a reproducible and controllable method for creating and detecting a singular SO(3) line vortex from the decay of a non-singular spin texture in a ferromagnetic spin-1 Bose–Einstein condensate. Our experiment explicitly demonstrates the SO(3) character and the unique spinor properties of the defect. Although the vortex is singular, its core fills with atoms in the topologically distinct polar magnetic phase. The resulting stable, coherent topological interface has analogues in systems ranging from condensed matter to cosmology and string theory

    Summer and winter differences in zooplankton biomass, distribution and size composition in the KwaZulu-Natal Bight, South Africa

    Zooplankton biomass and distribution in the KwaZulu-Natal Bight were investigated in relation to environmental parameters during summer (January–February 2010) and winter (July–August 2010). Mean zooplankton biomass was significantly higher in winter (17.1 mg dry weight [DW] m–3) than in summer (9.5 mg DW m−3). In summer, total biomass was evenly distributed within the central bight, low off the Thukela River mouth and peaked near Durban. In winter, highest biomass was found offshore between Richards Bay and Cape St Lucia. Zooplankton biomass in each size class was significantly, negatively related to sea surface temperature and integrated nitrate, but positively related to surface chlorophyll a and dissolved oxygen. Zooplankton biomass was significantly related to bottom depth, with greatest total biomass located inshore (<50 m). Distribution across the shelf varied with zooplankton size. Seasonal differences in copepod size composition suggest that a smaller, younger community occupied the cool, chlorophyll-rich waters offshore from the St Lucia upwelling cell in winter, and a larger, older community occurred within the relatively warm and chlorophyll-poor central bight in summer. Nutrient enrichment from quasi-permanent upwelling off Durban and Richards Bay appears to have a greater influence on zooplankton biomass and distribution in the bight than the strongly seasonal nutrient input from the Thukela River.DHE

    Diabetes Health, Residence & Metabolism in Asians: the DHRMA study, research into foods from the Indian subcontinent - a blinded, randomised, placebo controlled trial

    <p>Abstract</p> <p>Background</p> <p>Coronary heart disease (CHD) is highly prevalent amongst the South Asian communities in Britain. The reasons for this excess CHD risk are multifactorial, but in part relate to a susceptibility to diabetes mellitus - where the aberrant metabolism of non-esterified fatty acids (NEFA) and glucose are likely to underpin vascular disease in this population. Dietary intervention is an important and first line approach to manage increased CHD risk. However, there is limited information on the impact of the South Asian diet on CHD risk.</p> <p>Methods/Design</p> <p>The Diabetes Health, Residence & Metabolism in Asians (DHRMA) study is a blinded, randomised, placebo controlled trial that analyses the efficacy of reduced glycaemic index (GI) staples of the South Asian diet, in relation to cardio-metabolic risk factors that are commonly perturbed amongst South Asian populations - primarily glucose, fatty acid and lipoprotein metabolism and central adiposity. Using a 10-week dietary intervention study, 50 healthy South Asians will be randomised to receive either a DHRMA (reduced GI) supply of chapatti (bread), stone ground, high protein wheat flour and white basmati rice (high bran, unpolished) or commercially available (leading brand) versions chapatti wheat flour and basmati rice. Volunteers will be asked to complete a 75g oral glucose tolerance test at baseline and at 10-weeks follow-up, where blood metabolites and hormones, blood pressure and anthropometry will also be assessed in a standardised manner.</p> <p>Discussion</p> <p>It is anticipated that the information collected from this study help develop healthy diet options specific (but not exclusive) for South Asian ethnic communities.</p> <p>Trial registration</p> <p>Current Controlled Trials <a href="http://www.trialregister.nl/trialreg/admin/rctview.asp?TC=ISRCTN02839188">ISRCTN02839188</a></p

    P. falciparum Modulates Erythroblast Cell Gene Expression in Signaling and Erythrocyte Production Pathways

    Global, genomic responses of erythrocytes to infectious agents have been difficult to measure because these cells are e-nucleated. We have previously demonstrated that in vitro matured, nucleated erythroblast cells at the orthochromatic stage can be efficiently infected by the human malaria parasite Plasmodium falciparum. We now show that infection of orthochromatic cells induces change in 609 host genes. 592 of these transcripts are up-regulated and associated with metabolic and chaperone pathways unique to P. falciparum infection, as well as a wide range of signaling pathways that are also induced in related apicomplexan infections of mouse hepatocytes or human fibroblast cells. Our data additionally show that polychromatophilic cells, which precede the orthochromatic stage and are not infected when co-cultured with P. falciparum, up-regulate a small set of genes, at least two of which are associated with pathways of hematopoiesis and/or erythroid cell development. These data support the idea that P. falciparum affects erythropoiesis at multiple stages during erythroblast differentiation. Further P. falciparum may modulate gene expression in bystander erythroblasts and thus influence pathways of erythrocyte development. This study provides a benchmark of the host erythroblast cell response to infection by P. falciparum
