Vitamin D deficiency among children and adolescents living in sunny South Texas
Background: Exposure to sunlight is essential to produce Vitamin D (ViD). Recent studies suggest obesity is associated with low ViD concentration. Living in South Texas with 220 sunny days a year should be enough to maintain adequate ViD levels. We aimed to analyze ViD levels and obesity in children and adolescents.
Methods: We included 1239 pediatric (1.5 to 18.8 years old) participants (primary care clinic from Laredo) with registered CDC percentiles of BMI (pBMI) and serum concentrations of ViD (Atellica™). Data are described as median (p25, p75), Loess correlation between pBMI and ViD, ANCOVA to adjust by age, sex, and pBMI. We used the program Stata v16.1. The size of effects is expressed as Cohen-d and eta squared (eta2).
Results: The median age was 12.5 (9.5, 15.1) years, pBMI was 94 (80, 98), 49% females (n=611). The pBMI showed small differences by sex (M 82.1±24 vs M 84.5±23, Cohen-d 0.14, p,0.001). The Loess showed an inverse relationship between pBMI with a rapid drop of ViD from p90. The ANCOVA coefficients were negative for sex (b=- 0.32 for females p=0.007, eta2=0.03) and pBMI (b=-0.001, p=0.025, eta2=0.15) on ViD concentration.
Conclusion: We conclude obesity and female are related to low concentration VitD in sunny Laredo. Perhaps participants with more pBMI have less outdoor physical activity and increased sequester of ViD from adipose tissue. Future research should analyze the effect of these findings on adulthood morbidity

4. RESUMEN
La OMS clasifica la obesidad como una pandemia. La prevalencia de sobrepeso y obesidad es mayor en mujeres, representando un 63% de mujeres por grupo de edad, así como el 50% de las mujeres embarazadas presentan obesidad o sobrepeso. En mujeres gestantes el sobrepeso y la obesidad se han asociado a un mayor número de complicaciones durante el embarazo, como son la diabetes gestacional, la enfermedad hipertensiva del embarazo, la preeclampsia, las infecciones maternas (urinarias o endometritis) la enfermedad tromboembólica, el asma y la apnea del sueño.
OBJETIVO: Conocer la asociación existente en pacientes con obesidad y sobrepeso con las complicaciones maternas en el embarazo y puerperio en el Hospital General de Cuautitlán “General José Vicente Villada”
Se realizó un estudio: Descriptivo, Observacional, Retrospectivo, Transversal.
Se analizaron expedientes, de las pacientes con obesidad y sobrepeso durante el embarazo, las cuales fueron captadas a su ingreso a la unidad toco quirúrgica de esta unidad, para analizar la evolución así como la resolución del embarazo en un periodo de 01 Julio 2016 al 31 de Diciembre 2016
Genetic and Phenotypic Features to Screen for Putative Adherent-Invasive Escherichia coli
To date no molecular tools are available to identify the adherent-invasive Escherichia coli (AIEC) pathotype, which has been associated with Crohn’s disease and colonizes the intestine of different hosts. Current techniques based on phenotypic screening of isolates are extremely time-consuming. The aim of this work was to search for signature traits to assist in rapid AIEC identification. The occurrence of at least 54 virulence genes (VGs), the resistance to 30 antibiotics and the distribution of FimH and ChiA amino acid substitutions was studied in a collection of 48 AIEC and 56 non-AIEC isolated from the intestine of humans and animals. χ2 test was used to find frequency differences according to origin of isolation, AIEC phenotype and phylogroup. Mann–Whitney test was applied to test association with adhesion and invasion indices. Binary logistic regression was performed to search for variables of predictive value. Animal strains (N = 45) were enriched in 12 VGs while 7 VGs were more predominant in human strains (N = 59). The prevalence of 15 VGs was higher in AIEC (N = 49) than in non-AIEC (N = 56) strains, but only pic gene was still differentially distributed when analyzing human and animal strains separately. Among human strains, three additional VGs presented higher frequency in AIEC strains (papGII/III, iss and vat; N = 22) than in non-AIEC strains (N = 37). No differences between AIEC/non-AIEC were found in FimH variants. In contrast, the ChiA sequence of LF82 was shared with the 35.5% of AIEC studied (N = 31) and only with the 7.4% of non-AIEC strains (N = 27; p = 0.027). Binary logistic regression analysis, using as input variables all the VGs and antibiotic resistances tested, revealed that typifying E. coli isolates using pic gene and ampicillin resistance was useful to correctly classify strains according to the phenotype with a 75.5% of accuracy. Although there is not a molecular signature fully specific and sensitive to identify the AIEC pathotype, we propose two features easy to be tested that could assist in AIEC screening. Future work using additional strain collections would be required to assess the applicability of this method

Analyses of chondrogenic induction of adipose mesenchymal stem cells by combined co-stimulation mediated by adenoviral gene transfer
INTRODUCTION: Adipose-derived stem cells (ASCs) have the potential to differentiate into cartilage under stimulation with some reported growth and transcriptional factors, which may constitute an alternative for cartilage replacement approaches. In this study, we analyzed the in vitro chondrogenesis of ASCs transduced with adenoviral vectors encoding insulin-like growth factor-1 (IGF-1), transforming growth factor beta-1 (TGF-β1), fibroblast growth factor-2 (FGF-2), and sex-determining region Y-box 9 (SOX9) either alone or in combinations. METHODS: Aggregate cultures of characterized ovine ASCs were transduced with 100 multiplicity of infections of Ad.IGF-1, Ad.TGF-β1, Ad.FGF-2, and Ad.SOX9 alone or in combination. These were harvested at various time points for detection of cartilage-specific genes expression by quantitative real-time PCR or after 14 and 28 days for histologic and biochemical analyses detecting proteoglycans, collagens (II, I and X), and total sulfated glycosaminoglycan and collagen content, respectively. RESULTS: Expression analyses showed that co-expression of IGF-1 and FGF-2 resulted in higher significant expression levels of aggrecan, biglycan, cartilage matrix, proteoglycan, and collagen II (all P ≤0.001 at 28 days). Aggregates co-transduced with Ad.IGF-1/Ad.FGF-2 showed a selective expression of proteoglycans and collagen II, with limited expression of collagens I and × demonstrated by histological analyses, and had significantly greater glycosaminoglycan and collagen production than the positive control (P ≤0.001). Western blot analyses for this combination also demonstrated increased expression of collagen II, while expression of collagens I and × was undetectable and limited, respectively. CONCLUSION: Combined overexpression of IGF-1/FGF-2 within ASCs enhances their chondrogenic differentiation inducing the expression of chondrogenic markers, suggesting that this combination is more beneficial than the other factors tested for the development of cell-based therapies for cartilage repair
