    NASA's current human space flight research is directed towards enabling human space exploration beyond Low Earth Orbit (LEO). The Space Flight Payload Projects; Rodent Research, Cell Science, and Microbial Labs, flown on the International Space Station (ISS), benefit both the global life sciences and commercial space communities. Verified data sets, science results, peer-reviewed publications, and returned biospecimens, collected and analyzed for flight and ground investigations, are all part of the knowledge base within NASAs Human Exploration and Operations Mission Directorates Space Life and Physical Sciences Research and Applications (SLPSRA) Division, specifically the Human Research and Space Biology Programs. These data and biospecimens are made available through the public LSDA website. The Ames Institutional Scientific Collection (ISC), or ARC Biobank, stores flight and ground biospecimens from Space Shuttle and ISS programs. These specimens are curated and managed by the Ames Life Sciences Data Archive (ALSDA), an internal node of NASA's Life Sciences Data Archive (LSDA). The ARC Biolbank stores over 15,000 specimens from experiments dating from 1984 to present. Currently available specimens include tissues from the circulatory, digestive, endocrine, excretory, integumentary, muscular, neurosensory, reproductive, respiratory and skeletal systems. The most recent contributions include RNA, DNA and protein extracts from Rodent Research 1 and tissues from Rodent Research 4. NASA's biospecimen collection represents a unique and limited resource. The use of these biospecimens maximizes utilization and scientific return from these unique spaceflight payload and ground control research subjects. These biospecimens are harvested following complex, costly NASA research activities to meet primary scientific objectives. Once the primary scientific objectives have been met, the remaining specimens are made available to provide secondary opportunities for complementary studies or new investigations to broaden research without large expenditures of time or resources. Innovative ways of sharing this information ultimately advances the frontiers of human space exploration as well as scientific understanding of the effects of gravity on life on earth

    Use of Portable Infrared Cameras to Facilitate Detection and Capture Success of Montezuma Quail

    Survey and trapping methods for Montezuma quail (Cyrtonyx montezumae) require means not traditionally used for other quail species (e.g., northern bobwhite, Colinus virginianus). Trapping Montezuma quail is most effective using pointing dogs at night when coveys can be located and captured by net during roosting. However, reduced visibility at night, cryptic coloration of plumage, and behavioral adaptive stillness reduce detection rates and increase accidental flushing of birds while searching for roost locations. Forward-looking infrared (FLIR) cameras have been used to aid in detection of cryptic wildlife, including avifauna roosting sites. We conducted 25 survey and night-trapping sessions for Montezuma quail in southeast Arizona using a combination of trained pointing dogs and a portable FLIR camera. Detection of coveys on a roost was less successful when ambient climate conditions were freezing (below 3.88 8C), when residual heat signatures from surrounding soils and rocks were greater than 18.33 8C, or when density of grass cover exceeded 40% and the distance to covey was . 2.5 m. The small thermal signatures of quail were often obstructed by vegetative cover or confused with residual thermal signatures reflected by inanimate objects (e.g., rocks, bare ground). Successful detection of coveys combining the use of dogs and FLIR before trapping was 6.06%. Trapping success and detection of coveys with FLIR was improved when used with radiotelemetry and coveys which included radio-marked individuals. Proper tuning of FLIR camera sensitivity to a limited thermal bandwidth, or isotherm range, may effectively narrow covey locations approximated by a pointing dog. The FLIR camera was of limited benefit when actively trapping coveys with dogs and a team of 2–3 people, but may be beneficial for non-invasive monitoring and estimating covey size of marked birds on roosts in landscapes with reduced vegetative cover

    La transformación digital de la Administración Aduanera del Perú presentó un impulso inesperado con la aparición de la pandemia por Covid-19, donde la integración y uso de nuevas tecnologías de información y comunicación a disposición de los usuarios se aplique aceleradamente. Esta investigación tiene como objetivo, analizar e identificar el impacto de la transformación digital de la Aduana en la gestión de las agencias de aduanas del Callao, en el periodo 2018 al 2022; para ello, primeramente se estudió los cambios suscitados respecto a la integración y modernización de tecnologías en la entidad aduanera para que continúe con su rol facilitador del comercio exterior, también se investigó a los agentes de aduanas como actores principales en la operatividad de los servicios de agenciamiento. Por tanto, se establecieron como objetivos específicos: analizar e identificar de qué manera la transformación digital de la Aduana facilita los servicios, influye en el despacho aduanero, influye en los recursos, impacta en la logística de transporte e influye en el almacenamiento de las agencias de aduanas del Callao en el periodo 2018 al 2022. No obstante, el estudio presenta un enfoque cuantitativo y un tipo de investigación explicativa-descriptiva, con una población de estudio de 315 agencias de aduanas con Jurisdicción Marítima del Callao y una muestra de 87 encuestados, aplicando como instrumento un cuestionario constituido por 34 interrogantes.     Scleractinian Coral Recruitment to Reefs Physically Damaged by Ship Groundings

    The southeast Florida reef system faces a number of stress factors, among which ship groundings are one of the most physically damaging. Portions of the Florida reef tract located near Port Everglades, Broward County, Florida, USA have been damaged by ship groundings. In 2004, physical damage of more than 30,000 m2 was caused by the groundings of two large cargo ships, the MV Eastwind and MV Federal Pescadores. The present study was designed to measure differences of scleractinian coral recruitment patterns (recruit diversity and richness) and rates to these injured sites in comparison to undamaged reef sites. Coral recruitment rates were measured on unglazed ceramic tiles deployed for a period of one year from February 2007 to February 2008 at five different locations: three control sites (including a high coral cover site), and the two ship grounding sites. Morphology and genetic markers including CO1 and cytb were used to identify the coral recruits. A whole genome amplification kit (REPLI-g, Qiagen) was used to obtain sufficient amounts of DNA. Results revealed low recruitment rates (0.5-2.7 recruitsm-2 yr-1) to the studied sites, suggesting a low potential for recovery of the damaged areas

    Characterisation and expression of microRNAs in developing wings of the neotropical butterfly Heliconius melpomene.

    BACKGROUND: Heliconius butterflies are an excellent system for studies of adaptive convergent and divergent phenotypic traits. Wing colour patterns are used as signals to both predators and potential mates and are inherited in a Mendelian manner. The underlying genetic mechanisms of pattern formation have been studied for many years and shed light on broad issues, such as the repeatability of evolution. In Heliconius melpomene, the yellow hindwing bar is controlled by the HmYb locus. MicroRNAs (miRNAs) are important post-transcriptional regulators of gene expression that have key roles in many biological processes, including development. miRNAs could act as regulators of genes involved in wing development, patterning and pigmentation. For this reason we characterised miRNAs in developing butterfly wings and examined differences in their expression between colour pattern races. RESULTS: We sequenced small RNA libraries from two colour pattern races and detected 142 Heliconius miRNAs with homology to others found in miRBase. Several highly abundant miRNAs were differentially represented in the libraries between colour pattern races. These candidates were tested further using Northern blots, showing that differences in expression were primarily due to developmental stage rather than colour pattern. Assembly of sequenced reads to the HmYb region identified hme-miR-193 and hme-miR-2788; located 2380 bp apart in an intergenic region. These two miRNAs are expressed in wings and show an upregulation between 24 and 72 hours post-pupation, indicating a potential role in butterfly wing development. A search for miRNAs in all available H. melpomene BAC sequences (~2.5 Mb) did not reveal any other miRNAs and no novel miRNAs were predicted. CONCLUSIONS: Here we describe the first butterfly miRNAs and characterise their expression in developing wings. Some show differences in expression across developing pupal stages and may have important functions in butterfly wing development. Two miRNAs were located in the HmYb region and were expressed in developing pupal wings.     Dosing regimen of meropenem for adults with severe burns : a population pharmacokinetic study with Monte Carlo simulations

    Objectives To develop a population model to describe the pharmacokinetics (PK) of intravenous meropenem in adult patients with severe burns and investigate potential relationships between dosage regimens and antimicrobial efficacy.Patients and methods A dose of 1 g every 8 h was administered to adult patients with total body surface area burns of ≥15%. Doses for subsequent courses were determined using results from the initial course and the patient's clinical condition. Five plasma meropenem concentrations were typically measured over the dosage interval on one to four occasions. An open, two-compartment PK model was fitted to the meropenem concentrations using NONMEM and the effect of covariates on meropenem PK was investigated. Monte Carlo simulations investigated dosage regimens to achieve a target T>MIC for ≥40%, ≥60% or ≥80% of the dose interval.Results Data comprised 113 meropenem concentration measurements from 20 dosage intervals in 12 patients. The parameters were CL (L/h) = 0.196 L/h/kg × [1 − 0.023 × (age − 46)] × [1 − 0.049 × (albumin − 15)], V1 = 0.273 L/kg × [1 − 0.049 × (albumin − 15)], Q = 0.199 L/h/kg and V2 = 0.309 L/kg × [1 – 0.049 × (albumin − 15)]. For a target of ≥80% T>MIC, the breakpoint was 8 mg/L for doses of 1 g every 4 h and 2 g every 8 h given over 3 h, but only 4 mg/L if given over 5 min.Conclusions Although 1 g 8 hourly should be effective against Escherichia coli and CoNS, higher doses, ideally with a longer infusion time, would be more appropriate for empirical therapy, mixed infections and bacteria with MIC values ≥4 mg/L

    Dasatinib inhibits CXCR4 signaling in chronic lymphocytic leukaemia cells and impairs migration towards CXCL12

    Chemokines and their ligands play a critical role in enabling chronic lymphocytic leukaemia (CLL) cells access to protective microenvironmental niches within tissues, ultimately resulting in chemoresistance and relapse: disruption of these signaling pathways has become a novel therapeutic approach in CLL. The tyrosine kinase inhibitor dasatinib inhibits migration of several cell lines from solid-organ tumours, but effects on CLL cells have not been reported. We studied the effect of clinically achievable concentrations of dasatinib on signaling induced by the chemokine CXCL12 through its' receptor CXCR4, which is highly expressed on CLL cells. Dasatinib pre-treatment inhibited Akt and ERK phosphorylation in CLL cells upon stimulation with CXCL12. Dasatinib also significantly diminished the rapid increase in actin polymerisation observed in CLL cells following CXCL12 stimulation. Moreover, the drug significantly inhibited chemotaxis in a transwell assay, and reduced the percentage of cells able to migrate beneath a CXCL12-expressing murine stromal cell line. Dasatinib also abrogated the anti-apoptotic effect of prolonged CXCL12 stimulation on cultured CLL cells. These data suggest that dasatinib, akin to other small molecule kinase inhibitors targeting the B-cell receptor signaling pathway, may redistribute CLL cells from protective tissue niches to the peripheral blood, and support the investigation of dasatinib in combination strategies
