    Mangrove bacterial diversity and the impact of oil contamination revealed by pyrosequencing:Bacterial proxies for oil pollution

    BACKGROUND: Mangroves are transitional coastal ecosystems in tropical and sub-tropical regions and represent biologically important and productive ecosystems. Despite their great ecological and economic importance, mangroves are often situated in areas of high anthropogenic influence, being exposed to pollutants, such as those released by oil spills. METHODOLOGY/PRINCIPAL FINDINGS: A microcosm experiment was conducted, which simulated an oil spill in previously pristine mangrove sediment. The effect of the oil spill on the extant microbial community was studied using direct pyrosequencing. Extensive bacterial diversity was observed in the pristine mangrove sediment, even after oil contamination. The number of different OTUs only detected in contaminated samples was significantly higher than the number of OTUs only detected in non-contaminated samples. The phylum Proteobacteria, in particular the classes Gammaproteobacteria and Deltaproteobacteria, were prevalent before and after the simulated oil spill. On the other hand, the order Chromatiales and the genus Haliea decreased upon exposure to 2 and 5% oil, these are proposed as sensitive indicators of oil contamination. Three other genera, Marinobacterium, Marinobacter and Cycloclasticus increased their prevalence when confronted with oil. These groups are possible targets for the biomonitoring of the impact of oil in mangrove settings. CONCLUSIONS/SIGNIFICANCE: We suggest the use of sequences of the selected genera as proxies for oil pollution, using qPCR assessments. The quantification of these genera in distinct mangrove systems in relation to the local oil levels would permit the evaluation of the level of perturbance of mangroves, being useful in field monitoring. Considering the importance of mangroves to many other environments and the susceptibility of such areas to oil spills this manuscript will be of broad interest

    ORGANIZAÇÃO E CONEXÕES: cultura, gestão e liderança

    Resumo: Este estudo se propõe a analisar as evidências científicas que integram os conceitos de cultura organizacional, gestão e liderança. A busca pelas pesquisas com estas temáticas ocorreu de forma on-line, observando os critérios de inclusão: língua portuguesa e temporização das publicações, de 01 de janeiro de 2008 até 31 de dezembro de 2018. As bases de dados consideradas acessadas foram: Base de Periódicos da Fundação Getúlio Vargas, SciELO e Coordenação de Aperfeiçoamento de Pessoal de Nível Superior, com a utilização dos descritores: cultura organizacional, gestão e liderança conforme critérios metodológicos. Após a análise, três artigos foram inclusos na revisão, os quais abordam a cultura, gestão e liderança por uma perspectiva da influência no comprometimento, autenticidade e criatividade, tópicos relacionados às competências humanas e ciências sociais aplicadas. Conclui-se que esta influência é comprovada e estudada nas organizações, através dos achados de pesquisa, e que estes estudos são de importante valia para compreender os movimentos comportamentais dos grupos dentro das organizações. Palavras-chave: Cultura Organizacional.     Composição química de oito cultivares de feijão-caupi

    O feijão é um dos alimentos mais produzidos em todo o território nacional, sendo intensa a busca por cultivares produtivas, com boas características culinárias e diferentes propriedades nutricionais. Com o presente estudo, visou-se avaliar a influencia do cozimento na composição centesimal de oito cultivares de feijão-caupi: Costela de Vaca, BRS Marataoã, BRS Itaim, BR 17-Gurguéia, BRS Novaera, Paulistinha, Setentão e Patativa. Os grãos foram submetidos a dois procedimentos diferentes para realização das análises: in natura (farinha) e cozido (com maceração) sob pressão. Foram realizadas as determinações de umidade, cinzas, lipídios, proteínas, carboidratos, valor energético, pH, acidez e açúcares. Todas as análises foram realizadas em quadruplicata nas cultivares in natura e cozidas, sendo os resultados expressos como média ± desvio-padrão. Os resultados mostraram que o conteúdo de umidade variou de 7-11% nas cultivares cruas, com aumento após o cozimento (60-70%). Ao passo que, o conteúdo de proteínas, cinzas, carboidratos, valor energético e açúcares, diminuíram em todas as cultivares cozidas. Pode-se concluir que, dentre as cultivares analisadas, a Setentão foi a que apresentou o maior teor de proteína, antes e após o cozimento. Mesmo com redução nos demais parâmetros, as cultivares de feijão-caupi mantiveram características nutritivas e funcionais relevantes, apresentando um tempo de cocção entre 8 a 10 minutos, sendo consideradas de rápido cozimento e, portanto adequadas para atender a demanda atual dos consumidores

    IntroductionMannose-binding lectin (MBL) promotes opsonization, favoring phagocytosis and activation of the complement system in response to different microorganisms, and may influence the synthesis of inflammatory cytokines. This study investigated the association of MBL2 gene polymorphisms with the plasma levels of MBL and inflammatory cytokines in COVID-19.MethodsBlood samples from 385 individuals (208 with acute COVID-19 and 117 post-COVID-19) were subjected to real-time PCR genotyping. Plasma measurements of MBL and cytokines were performed by enzyme-linked immunosorbent assay and flow cytometry, respectively.ResultsThe frequencies of the polymorphic MBL2 genotype (OO) and allele (O) were higher in patients with severe COVID-19 (p< 0.05). The polymorphic genotypes (AO and OO) were associated with lower MBL levels (p< 0.05). IL-6 and TNF-α were higher in patients with low MBL and severe COVID-19 (p< 0.05). No association of polymorphisms, MBL levels, or cytokine levels with long COVID was observed.DiscussionThe results suggest that, besides MBL2 polymorphisms promoting a reduction in MBL levels and therefore in its function, they may also contribute to the development of a more intense inflammatory process responsible for the severity of COVID-19

    Pervasive gaps in Amazonian ecological research

    Biodiversity loss is one of the main challenges of our time,1,2 and attempts to address it require a clear un derstanding of how ecological communities respond to environmental change across time and space.3,4 While the increasing availability of global databases on ecological communities has advanced our knowledge of biodiversity sensitivity to environmental changes,5–7 vast areas of the tropics remain understudied.8–11 In the American tropics, Amazonia stands out as the world’s most diverse rainforest and the primary source of Neotropical biodiversity,12 but it remains among the least known forests in America and is often underrepre sented in biodiversity databases.13–15 To worsen this situation, human-induced modifications16,17 may elim inate pieces of the Amazon’s biodiversity puzzle before we can use them to understand how ecological com munities are responding. To increase generalization and applicability of biodiversity knowledge,18,19 it is thus crucial to reduce biases in ecological research, particularly in regions projected to face the most pronounced environmental changes. We integrate ecological community metadata of 7,694 sampling sites for multiple or ganism groups in a machine learning model framework to map the research probability across the Brazilian Amazonia, while identifying the region’s vulnerability to environmental change. 15%–18% of the most ne glected areas in ecological research are expected to experience severe climate or land use changes by 2050.
