Bio Mind and Techno Nature in the Performance of Memory: Arts-Based-Research and Human Enhancement
This paper intends to examine two recent projects Enhancing the Mind’s I and Emerging Self that address notions of self-identity, the desire for freedom of form and the greater cognitive capacities promised by neuro-enhancement technologies. It is a bid on critical evaluation of the production of the artworks; departing from an understanding that the observer is not independent of reality and that observation and experience are constructed. Consciousness is considered as resulting from the organism capacities to understand its emotions and interacting context, thus the research explores the possibilities that memory and knowledge do not reflect a real exterior world, but a concrete interior world attempting to play with possibilities to generate affect and empathy in the audiences. The text reflects on the ethical side of Human Enhancement (i.e. the potentiation of biological characteristics of Human Body) and the technologies, such as Brain Computer Interaction (BCI) or digital tattoos (tattoos that have technological interactive properties), promising a refinement of nature by technology. It raises questions surrounding memory and identity through art installation. It explores whether is it possible to translate one’s emotions directly into matter, as a memory. The result is the opening of a critical gap between the way sciences produce knowledge about the subject and the affect produced by the experience of the viewer on the installation artworks
Role of the endocytic recycling pathway in lysosome exocytosis
Until recently, lysosomes were considered as the end-point of the endocytic pathway, to where cargo is delivered to be degraded. However, several studies showed that conventional lysosomes can undergo regulated exocytosis in response to an increase in intracellular calcium concentration, an important process for plasma membrane repair and secretion of lysosomal contents. Nevertheless, the molecular machinery involved in lysosome transport and fusion with the plasma membrane is not fully understood. Our group found recently that the small GTPases Rab11a and Rab11b are essential for calcium-triggered lysosome exocytosis in HeLa cells.
Therefore, we aim to find the Rab11a/b effectors that mediate lysosome exocytosis process. To this end, we silenced several known effectors such as Rab11 family of interacting proteins (FIPs), Myosin Va/b or subunits of the exocyst tethering complex (Sec8, Sec15 and Exo70). After stimulation with the calcium ionophore ionomycin, we investigated the cell surface expression levels of the late endosome/lysosome marker LAMP1, as well as the release of the lysosomal enzyme β-hexosaminidase. We found that the silencing of Sec15 impairs lysosome exocytosis, while in the absence of FIP1-C or FIP2 there is an increase in LAMP1 cell surface levels and β-hexosaminidase release. Moreover, we confirmed the interaction and co-localization of Rab11a/b with the effector proteins studied using co-immunoprecipitation assays and confocal immunofluorescence microscopy, respectively. Finally, using live cell imaging, we found that Rab11-positive vesicles interact transiently with late endosomes/lysosomes near the plasma membrane, upon ionomycin stimulation.
Thus, our results provide new insights into the role of Rab11 and its effectors in the regulation of conventional lysosome exocytosis
Elucidating the role of exosomes in Candida – macrophage interaction
Dissertação de mestrado em Genética MolecularOs fungos têm sido identificados como causadores de graves doenças humanas, sendo as espécies do
género Candida e Aspergillus as principais causas de infeções fúngicas oportunistas em todo o mundo. Ao
longo dos anos, houve um aumento na incidência destas infeções e no número de espécies resistentes aos
medicamentos, o que contribuiu para maiores percentagens de morbilidade e mortalidade, principalmente
entre indivíduos imunocomprometidos. Uma vez que as infeções por Candida têm um efeito significativo na
saúde pública, a necessidade de melhor compreender a sua patogénese de forma a guiar o desenvolvimento
de medicamentos antifúngicos otimizados é premente. No decorrer da última década, tem sido dado mais
destaque aos tratamentos cujo alvo é específico à interação hospedeiro-patógeno, o que em teoria pode
possibilitar um impacto mínimo dos medicamentos na sobrevivência dos organismos comensais, tornando-os menos suscetíveis a conduzir uma resistência universal no microbioma. Assim sendo, o melhor
conhecimento sobre as interações hospedeiro-espécies de Candida pode levar a novos alvos para os
medicamentos antifúngicos e consequentemente auxiliar no controlo destas infeções. Recentemente, as
vesículas extracelulares ganharam muito interesse na investigação como um mecanismo omnipresente na
transferência de informação entre células e organismos em todos os três domínios da vida. Para além das
funções na fisiologia normal, estas partículas têm sido expostas como mediadores de um mecanismo
anteriormente não reconhecido para empacotar e proteger carga para absorção e integração noutras células,
nomeadamente produtos secretados por patógenos, emergindo assim como um tipo de interação
hospedeiro-patógeno funcionalmente diverso. Considerando as potenciais funções relatadas para estas
vesículas nas interações hospedeiro-Candida, há motivação significativa para aumentar a nossa
compreensão sobre a biologia e funções das mesmas, o que pode resultar num melhor entendimento da
patogénese de Candida e ajudar a elucidar possíveis alvos terapêuticos para o desenvolvimento de vacinas
ou imunoterapias contra este tipo de infeções.
O objetivo desta dissertação foi estudar a biologia dos exossomas secretados por C. albicans SC5314 e
C. albicans 124A e as suas funções num ambiente de infeção, através do seu isolamento para posterior
caraterização, avaliação de citotoxicidade, análise de fagocitose e produção de citocinas quando em cultura
com uma linha celular de macrófagos de murinos. Adicionalmente, este trabalho teve como objetivo
estabelecer um modelo de embrião de peixe-zebra para estudar as interações hospedeiro-Candida,
permitindo estudos adicionais sobre o papel de exossomas na patogénese de Candida na escala de um
organismo inteiro.Since the 1980s, fungi have been revealed as major causes of human disease, where Candida species
represent the most common cause of opportunistic fungal infections worldwide. Through the years, there has
been an increase in the occurrence of these infections, and in the number of drug-resistant species which
contributed to greater percentages of morbidity and mortality, particularly amongst the immunocompromised.
Accordingly, Candida infections have become a significant influence in public health which leadsto a necessity
of better understanding their pathogenesis to improve therapeutic options and facilitate the development of
enhanced antifungal drugs. Over the past decade, more prominence has been placed on treatments aiming
to target specifically the host-pathogen interaction which, in theory, could enable a minimal impact from the
drugs on the survival of commensal organisms, being less susceptible to eliciting resistance in the
microbiome. Therefore, improved knowledge about host-Candida species interactions may identify new
targets for the antifungal drugs and consequently assist in controlling these infections. More recently,
extracellular vesicles (EVs) have gained much interest in research as a ubiquitous mechanism for transferring
information between cells and organisms across all three domains of life. In addition to their influence on
normal physiology, these particles have also been pinpointed as mediators of a previously unrecognized
mechanism to package and protect cargo for uptake and integration into other cells, namely pathogen
secretion products, emerging as a functionally diverse type of host-pathogen interaction. Considering the
potential roles reported for EVs in host-Candida interactions, improving existing comprehension on the biology
and functions of Candida EVs could open new views for understanding Candida pathogenesis and help
elucidate possible therapeutic targets and develop potential vaccines or immunotherapy against this infection.
The aim of this thesis was to study the biology of C. albicans SC5314 and C. albicans 124A exosomes,
and their roles in an infection environment, by isolating them for further characterization, cytotoxicity
evaluation, phagocytosis assessment and cytokine production analysis when in culture with a murine
macrophage-like cell line. Furthermore, this work aimed to establish an effective zebrafish embryo model to
study host-Candida interactions, to allow further studies on the role of exosomes in Candida pathogenesis in
the scale of a whole organis

associated, and little is known about this relationship during COVID-19 pandemic. This
study examines the mediating role of quality of life and emotional well-being in the
relationship between sexual functioning and sexual satisfaction in women during COVID19 outbreak in Portugal. A sample of 301 women was recruited in May 2020, and women
answered to a sociodemographic questionnaire, the Female Sexual Functioning Index, the
Sexual Satisfaction Scale for Women, the brief WHOQOL, and the Positive Affect –
Negative Affect Scale (emotional well-being). Results indicated that quality of life was the
only significant mediator for the relationship between sexual functioning and sexual
satisfaction. Overall, the results emphasize the role of perceived quality of life in women’s
sexuality, by extending previous knowledge to a crisis context (COVID-19 pandemic),
enhancing quality of life promotion during adverse moments to improve women's sexual
satisfaction

O regresso ao trabalho directo com crianças faz nascer a necessidade de reflectir
sobre a própria prática de forma a ter presente os valores e fragilidades dessa
acção educativa. Surge, assim, um projecto de investigação-acção centrado
na própria prática.
Sendo a prática educativa um campo demasiado vasto e complexo para se
constituir como objecto de investigação, situou-se o enfoque da investigação
na organização do ambiente educativo como suporte da acção do educador.
É apresentado o projecto profissional que assenta num quadro teórico com
diferentes vertentes, crivo por onde passaram as reflexões que se iam fazendo,
analisando o que já se tinha conseguido e procurando as estratégias necessárias
para melhorar a acção.
Deste processo de auto-formação nasce um conhecimento identificado como
um conhecimento profissional não standard, um tipo de conhecimento
particular e complexo que, não sendo transmissível, é ensinável.
Como cooperante, o que se aprendeu ao longo deste estudo adquire todo o
sentido no processo de supervisão, em que se procura incentivar um saber
reflexivo, que tem de se tornar pessoal e que deverá ser desenvolvido ao longo
da vida profissional
Interactive Voice Response systems for informing citizens about the COVID-19 pandemic: A study on Brazil's Disque Saúde
In order to slow down the spread of the coronavirus SARS-CoV-2, it is vital to adopt measures to inform citizens about preventive actions. Such an operation requires a wide-ranged system that comprises a variety of interfaces as channels between citizens and healthcare authority’s information services. Amongst such interfaces, the Interactive Voice Response (IVR) systems can present benefits for informing citizens about the pandemic. Although the literature shows that IVR systems have been used for healthcare, the extent of the COVID-19 pandemic demands new examinations on the role of IVR systems on a multiplatform system for delivering information. This paper aimed to identify gaps and opportunities for the use of IVR systems to inform citizens about the COVID-19 pandemic. A case study was conducted by mapping the Brazilian Ministry of Healthcare’s channels of information about the coronavirus and analyzing the Disque Saúde IVR system – a phone-based ombudsman channel - based on literature recommendations. The results showed that while IVR systems have great potential for accessibility, it is essential that all types of information are available and continuously updated for citizens. Furthermore, the vast and mutable availability of information in a pandemic scenario may be a challenge for the usability of such systems.

Benefícios do envolvimento familiar no acompanhamento dos TPC dos alunos do 1º ciclo do ensino básico
O presente Relatório Final contempla uma breve descrição das Práticas de Ensino Supervisionadas (PES), desenvolvidas no âmbito de mestrado em Educação Pré-Escolar e Ensino do 1º Ciclo do Ensino Básico. O mesmo resulta de uma observação e reflexão sustentada, produto da experiência das PES, no ano letivo de 2021/2022. A partir da observação direta participante de uma turma de 3º ano do ensino básico, constituída por 23 alunos, surgiu a pergunta de partida: Quais os benefícios do envolvimento familiar no acompanhamento dos TPC dos alunos do 1º ciclo do Ensino Básico? Desta forma, foi levado a cabo um processo investigativo, assente numa abordagem mista de caráter qualitativo / interpretativo, com recurso a instrumentos de caráter quantitativo, com o fito de analisar os benefícios desse envolvimento. Esta investigação apoiou-se num referencial teórico relacionado com a problemática e foi sustentada pela recolha de informação através de inquérito por questionário e por entrevista semiestruturada. Com o presente estudo exploratório pretendeu-se perceber como é que o envolvimento dos pais / encarregados de educação é realizado, quais as vantagens e desvantagens segundo o olhar e perceção das famílias que acompanham os alunos, a docente e os próprios alunos. Esta análise passou, também, pela perceção dos intervenientes supra indicados sobre o envolvimento familiar nas tarefas escolares, durante o período de confinamento por Covid-19. Os participantes no estudo em questão são os encarregados de educação / família dos alunos de uma turma de 3º ano do 1º ciclo do ensino básico, os próprios alunos e a professora titular dessa mesma turma. Os resultados do tratamento dos dados recolhidos apontam que, tanto os alunos, como os encarregados de educação e docente atribuem grande importância ao envolvimento familiar no processo de aprendizagem das crianças, nomeadamente na realização dos TPC. Os resultados da informação recolhida, permite-nos compreender que o envolvimento das famílias espelha algumas objeções, todavia, são apontadas bastantes mais vantagens. Como principais vantagens estes apontam o facto dos TPC serem uma ferramenta importante de aprendizagem e solidificação de conhecimentos, uma maior proximidade entre as crianças e famílias, um maior rendimento e sucesso escolar, e maior motivação na realização desta tarefa
