    Plasma technology increases the efficacy of prothioconazole against fusarium graminearum and fusarium proliferatum contamination of maize (Zea mays) seedlings

    The contamination of maize by Fusarium species able to produce mycotoxins raises great concern worldwide since they can accumulate these toxic metabolites in field crop products. Further-more, little information exists today on the ability of Fusarium proliferatum and Fusarium graminearum, two well know mycotoxigenic species, to translocate from the seeds to the plants up to the kernels. Marketing seeds coated with fungicide molecules is a common practice; however, since there is a growing need for reducing chemicals in agriculture, new eco-friendly strategies are increasingly tested. Technologies based on ionized gases, known as plasmas, have been used for decades, with newer material surfaces, products, and approaches developed continuously. In this research, we tested a plasma-generated bilayer coating for encapsulating prothioconazole at the surface of maize seeds, to protect them from F. graminearum and F. proliferatum infection. A minimum amount of chemical was used, in direct contact with the seeds, with no dispersion in the soil. The ability of F. graminearum and F. proliferatum species to translocate from seeds to seedlings of maize has been clearly proven in our in vitro experiments. As for the use of plasma technology, the combined use of the plasma-generated coating with embedded prothioconazole was the most efficient approach, with a higher reduction of the infection of the maize seminal root system and stems. The debated capability of the two Fusarium species to translocate from seeds to seedlings has been demonstrated. The plasma-generated coating with embedded prothioconazole resulted in a promising sustainable approach for the protection of maize seedlings

    Vertically-resolved particle size distribution within and above the mixing layer over the Milan metropolitan area

    Vertical aerosol profiles were directly measured over the city of Milan during three years (2005–2008) of field campaigns. An optical particle counter, a portable meteorological station and a miniaturized cascade impactor were deployed on a tethered balloon. More than 300 vertical profiles were measured, both in winter and summer, mainly in conditions of clear, dry skies. <br><br> The mixing height was determined from the observed vertical aerosol concentration gradient, and from potential temperature and relative humidity profiles. Results show that inter-consistent mixing heights can be retrieved highlighting good correlations between particle dispersion in the atmosphere and meteorological parameters. Mixing height growth speed was calculated for both winter and summer showing the low potential atmospheric dispersion in winter. <br><br> Aerosol number size distribution and chemical composition profiles allowed us to investigate particle behaviour along height. Aerosol measurements showed changes in size distribution according to mixing height. Coarse particle profiles (<i>d</i><sub><i>p</i></sub>>1.6 μm) were distributed differently than the fine ones (<i>d</i><sub><i>p</i></sub><1.6 μm) were, at different heights of the mixing layer. The sedimentation process influenced the coarse particle profiles, and led to a reduction in mean particle diameter for those particles observed by comparing data above the mixing height with ground data (−14.9±0.6% in winter and −10.7±1.0% in summer). Conversely, the mean particle diameter of fine particles increased above the mixing height under stable atmospheric conditions; the average increase, observed by comparing data above the mixing height with ground data, was +2.1±0.1% in winter and +3.9±0.3% in summer. A hierarchical statistical model was created to describe the changes in the size distribution of fine particles along height. The proposed model can be used to estimate the typical vertical profile characterising launches within pre-specified groups starting from: aerosol size and meteorological conditions measured at ground-level, and a mixing height estimation. The average increase of fine particle diameter, estimated on the basis of the model, was +1.9±0.5% in winter and +6.1±1.2% in summer, in keeping with experimental findings

    Comparison of initial oral microbiomes of young adults with and without cavitated dentin caries lesions using an in situ biofilm model

    Dental caries is caused by acids released from bacterial biofilms. However, the in vivo formation of initial biofilms in relation to caries remains largely unexplored. The aim of this study was to compare the oral microbiome during the initial phase of bacterial colonization for individuals with (CC) and without (NC) cavitated dentin caries lesions. Bovine enamel slabs on acrylic splints were worn by the volunteers (CC: 14, NC: 13) for in situ biofilm formation (2 h, 4 h, 8 h, 1 ml saliva as reference). Sequencing of the V1/V2 regions of the 16S rRNA gene was performed (MiSeq). The relative abundances of individual operational taxonomic units (OTUs) were compared between samples from the CC group and the NC group. Random forests models were furthermore trained to separate the groups. While the overall heterogeneity did not differ substantially between CC and NC individuals, several individual OTUs were found to have significantly different relative abundances. For the 8 h samples, most of the significant OTUs showed higher relative abundances in the CC group, while the majority of significant OTUs in the saliva samples were more abundant in the NC group. Furthermore, using OTU signatures enabled a separation between both groups, with area-under-the-curve (AUC) values of ~0.8. In summary, the results suggest that initial oral biofilms provide the potential to differentiate between CC and NC individuals

    Full-length soluble urokinase plasminogen activator receptor down-modulates nephrin expression in podocytes

    Increased plasma level of soluble urokinase-type plasminogen activator receptor (suPAR) was associated recently with focal segmental glomerulosclerosis (FSGS). In addition, different clinical studies observed increased concentration of suPAR in various glomerular diseases and in other human pathologies with nephrotic syndromes such as HIV and Hantavirus infection, diabetes and cardiovascular disorders. Here, we show that suPAR induces nephrin down-modulation in human podocytes. This phenomenon is mediated only by full-length suPAR, is time-and dose-dependent and is associated with the suppression of Wilms' tumor 1 (WT-1) transcription factor expression. Moreover, an antagonist of alpha v beta 3 integrin RGDfv blocked suPAR-induced suppression of nephrin. These in vitro data were confirmed in an in vivo uPAR knock out Plaur(-/-) mice model by demonstrating that the infusion of suPAR inhibits expression of nephrin and WT-1 in podocytes and induces proteinuria. This study unveiled that interaction of full-length suPAR with alpha v beta 3 integrin expressed on podocytes results in down-modulation of nephrin that may affect kidney functionality in different human pathologies characterized by increased concentration of suPAR

    Mass and charge identification of charged products detected with Silicon-CsI(Tl) telescopes of the Chimera apparatus is presented.     124Sn+64Ni and 112Sn+58Ni reactions at 35 AMeV incident energy were studied with the forward part of CHIMERA multi-detector. The most central collisions were selected by means of a multidimensional analysis. The characteristics of the source formed in the central collisions, as size, temperature and volume, were inspected.     Adherence issues related to sublingual immunotherapy as perceived by allergists

    Objectives: Sublingual immunotherapy (SLIT) is a viable alternative to subcutaneous immunotherapy to treat allergic rhinitis and asthma, and is widely used in clinical practice in many European countries. The clinical efficacy of SLIT has been established in a number of clinical trials and meta-analyses. However, because SLIT is self-administered by patients without medical supervision, the degree of patient adherence with treatment is still a concern. The objective of this study was to evaluate the perception by allergists of issues related to SLIT adherence. Methods: We performed a questionnaire-based survey of 296 Italian allergists, based on the adherence issues known from previous studies. The perception of importance of each item was assessed by a VAS scale ranging from 0 to 10. Results: Patient perception of clinical efficacy was considered the most important factor (ranked 1 by 54% of allergists), followed by the possibility of reimbursement (ranked 1 by 34%), and by the absence of side effects (ranked 1 by 21%). Patient education, regular follow-up, and ease of use of SLIT were ranked first by less than 20% of allergists. Conclusion: These findings indicate that clinical efficacy, cost, and side effects are perceived as the major issues influencing patient adherence to SLIT, and that further improvement of adherence is likely to be achieved by improving the patient information provided by prescribers. © 2010 Scurati et al, publisher and licensee Dove Medical Press Ltd
